Chemical Composition and Size of Floating and Sunken Eggs of Olive Flounder Paralichthys olivaceus

  • Olive flounder (Paralichthys olivaceus) is one of the most commercially important species of farmed marine fish in Korea. Although techniques for rearing the larva of this species are improving, production costs are also increasing due to microbial influences and genetic degradation caused by successive culturing. Since the demand for healthy seed, which requires eggs of good quality of, is high, we examined the effects of nutrient composition on the size of P. olivaceus eggs. We analyzed floating (live) and sunken (dead) eggs of P. olivaceus from five different hatcheries for their size and amino and fatty acid composition. The sizes of eggs and oil globules from floating vs. sunken eggs were significantly different at p<0.05. No significant relationships were observed, however, among larval length, hatching percentage, and egg and oil globule size. The dry weight and amino acid levels of floating eggs were greater than those of sunken eggs (p<0.05), but no difference in fatty acid content was observed.

  • his study has been carried out to investigate amylase activity of digestive juice in the 5th day of the 5th instar influenced by the dietary composition (quantitative ratio between carbohydrate and protein) and the rearing temperature during the 4th moulting period. The larvae grew on three kinds of semi-synthetic diets. The A-diet has more carbohydrate than the others, the B-diet has carbohydrate in 1 : 2 with protein, and the C-diet has more protein than the others. All the diets were kept at 16$^{\circ}C$, 25$^{\circ}C$ and 32$^{\circ}C$ during the 4th moulting period. Amylase activity of digestive juice at the 5th day of the 5th instar was analyzed by Fuwa's method. The results were as follows. 1. Both A and C-diets were worse than B-diet in the larval growth and development. 2. The dietary composition influencing amylase activity of digestive juice was not related to the rearing temperature during the 4th moulting period. Amylase activity was stronger in C-diet, B-diet and A-diet order. 3. It was found that amylase activity at 32$^{\circ}C$ was stronger than that at 16$^{\circ}C$ in all kinds of diets. 4. There was an inter-action in amylase activity of male larval digestive juice between the dietary composition and the rearing temperature during 4th the moulting period.

Growth Performance and Nutrient Composition in the White-spotted Flower Chafer, Protaetia brevitarsis (Coleoptera: Scarabaeidae) Fed Agricultural By-product, Soybean Curd Cake (비지박 첨가 먹이원 급여에 따른 흰점박이꽃무지 유충의 생육과 영양성분 변화)

  • Insects are gaining recognition as an alternative source of protein. As a result, more and more domestic farms have begun mass rearing of edible insects. In order to produce high quality insects, studies on the development of safe and nutritious feed sources are needed. Given the cost of rearing insects, agricultural and industrial by-products are good sources for feed. The efficient utilization of these by-products can help in reducing the cost of production and preventing environmental pollution. In the current study, Citrus unshiu peel (CP), soybean curd cake (SCC), soybean oil meal (SOM), and brewers dried grain (BDG) were investigated for their effects on larval growth and development of Protaetia brevitarsis. Interestingly, the feed with 10% SCC increased larval weight by 3.5 times. For the larval period, the group of 10% SCC was significantly shorter than the control. Furthermore, minerals such as Zn, Ca, K, Mg, Na, and P were recorded to be high in 10% SCC. A total of 17 amino acids were present in 10% SCC, of which tyrosine and arginine were predominant. The heavy metal contents were very small amounts or not detected in any of the investigated groups. These findings provided a scientific basis for the utilization of soybean curd cake as a nutritional feed source to promote larval growth and produce quality insects.

Distribution of Larval Fishes off the East Sea, Korea (동해 근해에 출현하는 어류 자치어 분포 특성)

  • In order to understand the species composition and distribution characteristics of larval assemblages in the East Sea, Korea, larvae were collected at 13 stations every other month from February to December 2018. Fish larvae were identified through DNA barcoding along with morphological methods, and as the result, a total of 104 taxonomic groups appeared during the survey. Among these, Engraulis japonicus, which accounted for 76.2% of the total population, was the most dominant species, followed by Maurolicus japonicus, which accounted for 15.0%. In addition, Ammodytes personatus, Champsodon snyderi, Scomber japonicus and Echelus uropterus appeared more frequently than other taxa. The above six species accounted for 93.2% of the total catch. The number of taxa and the amount of larvae collected in the survey area were higher during the high water temperature periods (June to October) than during the low water temperature periods (February, April and December). Analysis of similarity (ANOSIM) showed a statistically significant difference in monthly larval assemblages. The results of the canonical correspondence analysis (CCA) show that the distributions of larval community were mainly affected by sea surface temperature during low water temperature periods, and various environmental factors such as salinity, dry weight of zooplankton, and the concentrations of nutrients during high water temperature periods.

  • An electron microscopical study on hemocytes of last instar larvae of Orthoptera (Acrida cinerea Thunberg, Paratenodera sinensis De Saussure, Atractomorpha lata Hotschulsk was performed to observe cell types, structures and their origins. Prohemocytes are the smallest round cells with large, nucleus, poorly developed cytoplasmic organelles and lower composition ratio, accouting for less than 1% of all cell types. Little remarked differences were revealed among different species. Plasmatocytes are relatively large cells with higher composition ratio, accounting for 5 to 20%. The majority of the plasmatocytes are oval forms with numerous external cytophismic projections., relatively well developed cytoplasmic organelles. Granular cells are characterized by various granules in cytoplasm. Some considerable remarked differences were revealed among different species. Furthermore the granules themselves differ in fine structures. Adipohemocytes were found only in Acrida cinerea Thunberg.

Potential Usage of Food Waste as a Natural Fertilizer after Digestion by Hermetia illucens (Diptera: Stratiomyidae)

  • The black soldier fly larvae are able to decompose various organic wastes such as livestock manures and food wastes. We tested whether the quality of the insect derived compost, i.e. larval feces, was comparable to that of a commercial fertilizer. The results show that the chemical composition and the growth rate of cabbages grown on the insect derived compost are virtually identical to those on the commercial fertilizer. Therefore the insect derived compost will be an ideal substitute to commercial fertilizers.

Community Structures of Macrobenthos in Chonsu Bay, Korea

  • Based on 53 quantitative samples collected in April (23 stations) and August (30 stations) of 1993, the species composition, distribution and abundance of macrobenthic invertebrates and bottom characteristics in Chonsu Bay were conducted. Bottom sediments consisted of mud at the close to the dyke, sandymud in the central bay, and coarse sand in the mouth of the bay. A total of 273 species (177 species in April and 200 species in August) were identified. Mean density per sampling station was 480 ind./m$^2$ in April and 1126 ind./m$^2$ in August, respectively. Number of species and densities decreased in the northern part of the bay. The macrobenthos showed a temporal difference in species composition, range of distribution, and community delimitation caused by larval settling. Although during the settling period in summer, large numbers of juveniles added in most of regions, unrelated to environmental conditions, but, successful recruitment may has been a rare event locally Theora fragilis (bivalve), Lumbrineris longifolia(polychaete), Neptys oligobranchia (polychaete) were numerically dominant, but their densities varied seasonally. Community structures of macrobenthos in Chonsu Bay were affected by several bottom environmental conditions, related to tidal current speed, regionally.

Distribution and Species Composition of Larval Fish during Winter Season in Jinhae Bay, Korea (동계 진해만에서의 자어 분포 및 종조성)

  • We investigated the spacial distribution and species composition of fish larvae during winter season (January 2013~March 2013) in Jinhae Bay, Korea. During the study period, a total of 12 larvae species belonging to 8 families were collected, and the dominant species were Pholis crassispina, Clupea pallasii, Liparis tanakae, Hexagrammos otakii. These four species accounted for 93% of the total number of individual collected. High density and species diversity occurred around Jam-do and eastern area of Chilcheon-do, suggesting that these areas are the main spawning ground of fish larvae in Jinhae Bay.

Purification and Biochemical Properties of Storage Protein SP-2 in Tobacco Budworm (Helicoverpa assulta Guenee) (담배나방 저장단백질 SP-2의 정제 및 생화학적 특성)

  • A storage protein(SP-2) was confirmed in haemolymph during larval-pupal-adult development of tobacco burdworm(Helicoverpa assulta Guenee), and its biochemical characteristics were investigated. The titer of SP-2 showed a peak at mature larva, decreased gradually through the late pupal stage, and became undetectable at adult period. As the results from electrophoretic mnysis, SP-2 was confirmed to be glycolipoprotein(M.W. 332kDa) relatively stable to heat( $\leq$ 68$^{\circ}C$ ). This storage protein was determined to be a tetramer composed of a single subunit with MW of 83kDa, and the isoelectric point was 5.7. The amino acid composition of the SP-2 was characterized. It has relatively high content of methionine and histidine, whereas the contents of tyrosine and phenylalanine were relatively low.

  • The "crescent-egg" a new spontaneous mutant was detected in a white egg strain k37. Studies were carried out the linkage analysis, to investigate phenotypic characteristics and biochemical analysis of haemolymph and ovarian protein. The mutant, ore was independent from 20 linkage groups P(2), Ze(3),L(4), oc(5), sn8), Ia(9), w-1(10), K(11), ch(13), U(14), bl(15), cts(16), bts(17), mln(18), nb(19), oh(20),Lan(21), or(22), tub(23) and Xan(27). The fertilization, hatchability and larval growth were not different from the those of normal eggs. The content and composition of yolk protein were similared to normal eggs. Scanning electron microscopy revealed the areal specific structure in dorsal region of egg-shell of cre mutant. Analysis of chorion protein by isoelectrofocusing(IEF), was resolved no difference in the composite of the chorion protein. We conclude that the egg mutant ere is expressed only in the egg-shape formation and region specific determination.
