Biochemical Changes in the Hemolymph of the Larvae of Thecodiplosis japonensis Uchi. et Inouye (솔잎혹파리 유충 체액의 생화학적 변화)

  • The concentration of amino acids, total nitrogen, trehalose, lipids and the activities of respiratory, acid$\cdot$alkaline phosphatase, glutamic oxalozcetic transaminase and glutamic pyruvic transaminase during larval stage in Pine leaf gall midge, Thecodiplosis janensis Uchi. et Inouye were measured using Paper chromatographic method, micro-Kjeldahl method, Thin layer chromatographic method, Warburg's manometric method, Bessey-Lowry method and Reitman-Frankel method, respectively. Healthy specimens )yore chosen as samples of each larval stages; alrva in gall and larva in soil. Amino acids present in the alcoholic extracts were alanine, glutamic acid, glycine, histidine, methionine, proline, threonine, tryptophan and valine. The total nitrogen concentration reached to 31.348mg/g during the larva in gall and the larval stage in soil of the value was decreased to 29.027mg/g. The hemolymph sugar, trehalose value for larva in soil was about two times of the value for larva in gall. Total lipid, phospholipid,monoacylglycerol, triacylglycerol, sterol, free fatty acid and ester cholesterol were identified at larval stages in gall and soil. Triacylglycerol concentration reached high level in contrast with other lipid contents during larvae in gall and larva in soil. Free fatty acid, sterol except decreased lipids during larval stage in soil. Endogenous respiration, succinate of respiratory activities decreased at larval stage in soil compare with larva in gall. The activities of acid phosphatase decreased larval stage in soil but the activities of alkaline phosphatase increased remarkably. The activities of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase reached high level of the larva in gall.

  • Imago's flight activities of Protaetia brevitarsis were from mid June to early September, and those of Allomyrina dichotoma were from mid June to late August. These activities were mainly influenced by amount and duration of rain. They were more active when there was small amount and short period of rain in a year. Distribution depth of the 3$^{rd}$ larvae of P. brevitarsis were not affected to the temperature. But A. dichotoma were sensitively react to the temperature, almost larvae were near the surface when high temperature. The distribution of P. brevitarsis was not affected by humidity, but most of A. dichotoma were near the surface when high humidity. Larvae of P. brevitarsis usually ate their dead individuals. When we supplied as food the pork, chicken, fresh mackerel, bread, apple, and pear. They took all of these food but larvae of A. dichotoma took only bread. Two species were lived in similar environment but those feeding habit was very different.

LIN-23, an E3 Ubiquitin Ligase Component, Is Required for the Repression of CDC-25.2 Activity during Intestinal Development in Caenorhabditis elegans

  • Caenorhabditis elegans (C. elegans) utilizes two different cell-cycle modes, binucleations during the L1 larval stage and endoreduplications at four larval moltings, for its postembryonic intestinal development. Previous genetic studies indicated that CDC-25.2 is specifically required for binucleations at the L1 larval stage and is repressed before endoreduplications. Furthermore, LIN-23, the C. elegans ${\beta}$-TrCP ortholog, appears to function as a repressor of CDC-25.2 to prevent excess intestinal divisions. We previously reported that intestinal hyperplasia in lin-23(e1883) mutants was effectively suppressed by the RNAi depletion of cdc-25.2. Nevertheless, LIN-23 targeting CDC-25.2 for ubiquitination as a component of E3 ubiquitin ligase has not yet been tested. In this study, LIN-23 is shown to be the major E3 ubiquitin ligase component, recognizing CDC-25.2 to repress their activities for proper transition of cell-cycle modes during the C. elegans postembryonic intestinal development. In addition, for the first time that LIN-23 physically interacts with both CDC-25.1 and CDC-25.2 and facilitates ubiquitination for timely regulation of their activities during the intestinal development.

  • Due to consumer reluctance to take synthetic drugs for nematode infections and the appearance of resistance to anthelminthic drugs, new drugs from natural products must be developed. Caenorhabditis elegans is one of the freely living nematodes and serves as a good model system for screening anthelminthic drugs. In this study, thirteen flavonoid derivatives were tested for anthelminthic activity and the relationships between their activities and structures were investigated. The structural information combined with the data for the larval growth inhibition of C. elegans provided meaningful structural insights in the search for new anthelminthic drugs.

Effect of Anti-juvenile Hormone Analogue (AJH) on the Larval Ecdysis of the Silkworm, Bombyx mori L. (항유약호르몬 활성물질이 누에탈피에 미치는 영향)

  • To clarify the effect of anti-juvenile hormone analogue (AJH) on the larval ecdysis by feeding at early stage of the 4th instar, the total amount of protein and activity of chitinolytic enzymes in the integument of Bombyx mori were analyzed, PAGE pattern of the protein was observed and the morphological changes of integument during molting period were also observed and the morphological changes of integument during molting period were also observed by means of TEM. The total amount of protein was greatly increased in premolting, then reached maximum level just before ecdysis, and rapidly decreased after the larval ecdysis in the control, while in the AJH treatment, increased 12 hr later than the control and its maximum was only 82.6% of the control. Two specific proteins, which were presumed as the protein originated from endocuticle, also appeared 12 hr later than the control and were maintained to 132 hr after AJH treatment from the aspects of the Native- and SDS-PAGE patterns, although those of the control disappeared instantly after ecdysis. Chitinase and $\beta$-N-acetylglucosaminidase activities were also suppressed and delayed by AJH treatment. Furthermore, it was observed that the apolysis took place 12 hr later than the control but new epicuticle was not formed at least until 132 hr after AJH treatment. From these results, it is suggested that the larval molting process of silkworm develops 12 hr later than the control but new epicuticle was not formed at least until 132 hr after AJH treatment. From these results, it is suggested that the larval molting process of silkworm develops 12 hr later than the control by AJH treatment but no further processing takes place just after apolysis.

  • Crude fermentation broths of two bacterial isolates, S-11 and S-25, from about 200 soil samples, showed inhibitory activities against the crude chitinase prepared from alimentary canals of the silkworm, Bombyx mori, as well as against reagent chitinase from Streptomyces griseus, in vitro. The chitinase inhibitors also exhibited insecticidal activities by preventing larval-pupal ecdysis when these broths were injected into the silkworm larvae during spinning.

Pheophytin Content and Fibrinolytic Activity of Silkworm Feces in the Different Larval Stages of Silkworms

  • In order to find potential anticancer agents, we extracted pheophytin in the silkworm feces from various larval stages by water, chloroform and methanol extraction. The cytotoxicity of the pheophytin extracts of various silkworm feces was measured in the CT-26 cells originated from murine metastatic colon cancer, by dye uptake assay. The cytotoxicity of those pheophytins in 2nd, 3rd and 4th instars was better than remaining larval stages. The in vitro anticoagulant and fibyinolytic activities of ethanol extract from varietal mulberry leaves, mulberry branches and silkworm feces and pheophytin extracts from silkworm feces obtained at various larval stages were evaluated in order to find effective therapeutic drugs for the treatment of myocardial and cerebral thrombosis. The fibrinolytic activity was tested using the activated partial thromboplastin time (APTT) and thrombin time (TT) was measured for blood clotting activity. With regards to the fibrinolytic system, ethanol extracts of silkworm feces were better than varietal mulberry leaves and mulberry branches. The pheophytin extracts from 7th days of 5th instar contained the highest percentage of pheophytin and good fibrinolytic activity.

Comparison of Feed Efficiency Between Rotifers Enriched Lipid-contents to Enrichment and Enhanced Digestive Enzymes Activity to Starch (영양강화 Rotifer와 효소활성 향상 Rotifer의 먹이효율 비교)

  • In this study, we carried out an experiment for estimation the larval digestibility in aspects which digestive enzymatic activities and nutrition of the rotifers, Brachionus rotundiformis. Thus we enhanced the digestive enzymatic activity through the addition of starch for the increase of digestibility of rotifer (starch-rotifer), and compared with the feed efficiency through rearing of the olive flounder, Paralichthys olivaceus used rotifer lipid-enriched with Algamac $2000^{(R)}$ (CE-rotifer). The digestive enzyme activities (except for TG-lipase), total protein contents, total essential amino acid, essential amino acids (methionin and phenylalanine) of starch-rotifer (the rotifer used a starch as additive, and enriched not) was assayed significantly higher than CE-rotifer (P<0.05). And total lipid, lipid classes (except for sterol) and fatty acids as DHA and EPA showed higher in CE-rotifer than starch-rotifer (P<0.05). But, sterol contents and ST/TG ratio were shown significantly higher in starch-rotifer (P<0.05). The flounder larvae supplied the two rotifers showed standard length and body weight that not significantly differed with ranges $3.72{\sim}3.79\;mm$ and $32.9{\sim}37.8\;mg$/larva on 6 days after hatching (DAH), respectively (P>0.05). However, these of 12 DAH showed the values of significantly higher to $5.94{\pm}0.249\;mm$, $144.0{\pm}23.86\;mg$/larva and $26.2{\pm}12.13%$ in standard length, body weight and survival in CE-flounder than that of starch-flounder (P<0.05). The hydrolytic enzymatic activities of flounder larvae severally supplied the two rotifers showed the significantly higher activities in acidic -amylase, neutral -amylase, TG-lipase, lysozyme and acidic phosphatase in starch-flounder on 5 DAH (P<0.05). But neutral $\alpha$-amylase, three proteases and two phosphatases of CE-flounder on 11 DAH showed the significantly higher activities than that of starch-flounder (P<0.05). Therefore, for the flounder, Paralichthys olivaceus larvae just depleted yolk was more beneficial to supply the feed, rotifer, enhanced the digestibility than to supply the feed lipid-enriched for aspect of larval digestibility up to 6 DAH, thereafter nutrition of absorption due to the development of digestive organs suggested that enrichment effect appeared with larval somatic growth. Consequently, investigation more detailed about the larval digestive physiological and nutritional requirement variations after 6 DAH will be necessary, thereafter.

Accumulation of Dietary Conjugated Linoleic Acid (CLA) in Silkworm, Bombyx mori

  • Conjugated linoleic acid (CLA) exhibits potent anti-carcinogenic and other biological activities in several animal models. We report here that dietary CLA, chemically synthesized from corn oil, accumulates I silkworm, Bombyx mori, which is used as therapeutic agent for diabetes in Korea and Japan. Mulberry leaves treated with 0.1 or 10% CLA in ethanol were supplied to silkworms from the end of 3rd instar to the 3rd day of the 5th instar. Fresh mulberry leaves or leaves treated with 10% corn oil in ethanol were fed as a check treatment. the amount of total lipids in the larval body ranged from 17.4 to 19.1mg/g of body tissue, which was not significantly affected by the source of the diets. No CLA was found in the control silkworms. But the level of CLA significantly increased to 83.5 mg/g of fat, when fed with mulberry leaves treated with 10% CLA. Only trace amounts of CLA were detected in the larvae reared with check leaves and 0.1% CLA-treated leaves. Mulberry leaves treatd with corn oil or CLA were not palatable to the larvae, resulting in a reduction of larval weight. These results suggest that silkworms containing CLA in body lipids could be producted by dietary CLA.

담배나방의 발생과 휴면에 따른 혈림프 생체물질의 변화

  • Changes of biological components in haemolymph of Helicoverpa assulta Guenee were investigated during the development and pupae of cold-stress periods. Protein content was the highest in the larval stage, whereas it showed somewhat lower level at the cold-stressed pupae. The level of free amino acids was high in the larval and adult stages. Althougth decresed in pupal stage, the level of free amino acid remarkably increased at the cold-stressed pupae. Total lipids were a little increased at the pupal stage, after small quantity decreased in I-day old adults, and then remarkably decreased at the cold-stressed pupae. Glycogen was increased at the pupal stage, and somewhat decreased in the stage of adult, but remarkably decreased at the cold-stressed pupae. From thin layer chromatographic analysis, glucose was detected in small quantity at 1-day old pupae, one-day old adults, and cold-stressed pupae, respectively. Each .activity of acid and alkaline phosphatases was the highest in the 1 and 7-day old pupae, respectively, and both enzyme activities were increased about two-fold compared to the activity in the cold-stressed pupae. A total of 2 phosphatase isozymes were observed in throughout the developmental stage, and a new isozyme specific to cold-stressed pupae was identified.
