Ecdysteroid Stimulates Virus Transmission in Larvae Infected with Bombyx mori Nucleopolyhedrovirus

  • Most baculoviruses have an ecdysteroid UDP-glucosyltransferase (egt) gene, whose product inactivates ecdysteroid within the infected host. Bomhyx mori larvae infected with BmEGTZ, a mutant B. mori nucleopolyhedrovirus (BmNPV) in which the egt gene has been inactivated, die more rapidly compared to larvae infected with wild-type BmNPV. In this study, the profile of hemolymph proteins, and progression of virus infection in BmEGTZ- and BmNPV-infected B. mori larvae, was analyzed by SDS-PAGE and histochemically. These analyses showed that virus-encoded and virus-induced proteins were expressed quicker in BmEGTZ-infected larvae than in BmNPV-infected larvae. This suggests that the decrease in time to death, following BmEGTZ infection, results from the stimulation of virus-specific protein expression. In order to examine the effect of ecdysteroid on virus transmission, the profile of hemolymph proteins, and progression of virus infection, were analyzed following an ecdysteroid injection of BmEGTZ- or BmNPV-infected larvae. In the BmNPV-infected larvae, ecdysteroid treatment had no apparent effect on hemolymph protein expression. This suggests that the injected ecdysteroid was inactivated by the BmNPV-expressed ecdysteroid UDP-glucosyltransferase. An Ecdysteroid injection into BmEGTZ-infected larvae increased the speed of virus-specific protein expression and virus transmission. These results suggest that ecdysteroid stimulates protein expression, which in tum results in the stimulation of virus transmission.

동중국해 북부해역에서의 살오징어(Todarodes pacificus) 유생의 분포 (Distribution of Larvae of the Common Squid Todarodes pacificus in the Northern East China Sea)

  • The common squid Todarodes pacificus is an ecologically and commercially important species in Korea and Japan. However, the distribution pattern of its eggs and larvae in Korean waters has not yet been clarified. To determine the horizontal and temporal distribution of common squid larvae in the northern East China Sea, samples collected using paired, 60 cm diameter Bongo nets from the three surveys conducted in August 2003, May 2004 and November 2005 were examined. In addition, the vertical distribution was examined from the samples collected using a $1\;m^2$ MOCNESS in April 1999 (20 m interval down to 100 m). A total of 218 larvae ranging in mantle length (ML) from 1.2 to 17.0 mm were counted at 27 stations. Larval abundance was highest in May 2004. The larvae mainly occurred in the southeastern area of Jeju Island, where the water temperature and salinity at 50 m deep ranged from $15-23^{\circ}C$ and 34-34.6 psu, respectively. Most larvae were collected in the frontal zone, where the Tsushima Warm Current and inshore waters meet. The results from the MOCNESS samples showed that the larvae occurred mostly in 20-80 m depth ranges (about 90%), although collections were only conducted above 100 m. No significant differences in larval mantle lengths (ANOVA, P>0.05) were found among each depth interval. Given the occurrence sites of the larvae <2.0 mm ML, the spawning ground of this species appears to be within the northern East China Sea, mainly to the southeast and northeast of Jeju Island, in early spring.

볼락류 Sebastes spp.에 있어서 선충류 아니사키스 유충의 감염 (Infection of Parasitic Anisakis Type Larvae (Nematoda) from Some Rockfishes, Sebastes spp.)

  • 2002년 2월부터 3월까지 오레건 주에 위치한 오레건 해안 수조와 뉴포트 어시장으로부터 16마리의 볼락류 Sebastes spp.를 채집하여 검사한 결과, 3종의 볼락류로부터 총 157개체의 아니사키스 유충이 검출되었으며 50.0%의 감염률을 나타내었다. 3종의 볼락류인 Sebastes flavidus, S. crameri, S. pinniger로부터 Anisakis simplex유충 110 개체, Contracaecum app. 유충 30개체 그리고 Pseudoteryanova decipiens 유충 17개체가 각각 검출되었다. 아니사키스 유충의 감염부위는 장관, 장간막과 위벽이었다. A. simplex 유충은 S. crameri에서 60%, S. flavidus와 S. pinniger에서 각각 33.3%의 감염률을 나타내었다. Contracaecum spp. 유충과 P. decipiens 유충은 S. pinniger에서 33.3%의 감염률을 나타내었다. A. simplex 유충의 감염강도는 5. flavidus, S. crameri 그리고 S. pinniger에서 1에서 87이었고 Contracaecum spp.유충과 P. decipiens 유충은 S. pinniger에서 0에서 30까지 나타내었다.

Genetical identification and morphological description of the larvae and juveniles of Porocottus leptosomus (Pisces: Cottidae) from Korea

  • The larvae and juveniles of Porocottus leptosomus belonging to the family Cottidae were collected (n = 95, 3.9-16.5 mm in body length, BL) from Busan, Korea, in March 2015. The larvae and juvenile were identified using DNA barcoding as P. leptosomus, and their morphological description was presented in detail. The yolk-sac larvae (3.9-5.6 mm BL) body was slightly compressed, the head was large, the eye was round and large, and the anus was before the middle of the body. The preflexion larvae (5.2-10.0 mm BL) body length drastically increased; caudal fin rays began to occur. The flexion larvae (9.4-11.8 mm BL) notochord flexion started; dorsal, pectoral, and anal fin rays began to occur; pelvic fin buds are seen; they possessed a pair of parietal spine; and a pair of supraocular cirri was first to develop. At 12 mm BL, the notochord was completely flexed. The larva stage (3.9-12.6 mm SL) had the stellate melanophores in the head, isthmus, gut, and tail (along to the ventral midline). During the juvenile stage (11.4-16.5 mm BL), melanophores covered the head and began to form five black bands on the side of the body. The larvae of P. leptosomus spent pelagic life, but moved to the bottom during the juvenile stage. The larvae and juveniles of P. leptosomus differ from other cottid larval fishes by body shape, melanophore head pattern, and spine development. P. leptosomus can be distinguished from Porocottus allisi by morphological development and the occurrence of larval fish: preopercular spine development, melanophore pattern, and caudal fin development.

The effect of sawdust fermentation period and storage period after sawdust fermentation on the development of Protaetia brevitarsis larvae

  • To investigate the effect of sawdust fermentation period and storage period after fermentation on development as food for P. brevitarsis larvae, individual and group breeding were conducted on berry sawdust and oak sawdust. In individual breeding, the growth period of P. brevitarsis larvae was reduced by 12 days from the 60-day fermentation of berry sawdust to the 40-day fermentation of berry sawdust, 30 days from the 90-day fermentation of oak sawdust, and the weight of the larvae was the heaviest. In group breeding, the time it takes for P. brevitarsis larvae to change from 1st to 3rd instar is about 30 days after hatching from 60-day fermentation of berry sawdust, while 90-day fermentation of oak sawdust took more than 60 days, so the growth speed was fast and the survival rate was good. The results of the farmhouse demonstration test were the same trend, and it was judged that it would be possible to produce P. brevitarsis larvae with berries fermented sawdust, and it would be advantageous in terms of economy. In addition, for both individual and group breeding, the growth period of P. brevitarsis larvae was longer as the storage period was longer, the weight of the larvae decreased, and the survival rate was no different. The development period of P. brevitarsis larvae was the longest in the storage period of 18 months for berries fermented sawdust, and the storage period of oak fermented sawdust was longer in the storage period of 12 months and 18 months. Therefore, considering the results of individual breeding and group breeding, the fermentation period was appropriate for 60 days for berries sawdust, and the storage period for berries sawdust was stable from 0 to 12 months, and 0 to 6 months for oak sawdust.

In Vivo Effects of Antibiotics on Silkworm Bombyx Mori L Infected with Bacillus Coagulans

  • The present study was aimed to screen different antibiotics in vivo for their effects against the bacterium Bacillus coagulans in silkworm Bombyx mori in three experimental conditions viz., a) healthy larvae treated with the antibiotics b) larvae which were first inoculated with Bacillus coagulans immediately after the $2^{nd}$ moult and treated with the antibiotics from 24 hour after inoculation up to the end of the $5^{th}$ instar(pre-inoculated larvae) and c) larvae which were treated with the antibiotics right from hatching up to the end of the $5^{th}$ instar and inoculated with Bacillus coagulans immediately after the $2^{nd}$ moult(post-inoculated larvae). All the antibiotics used in this study stimulated better performance in improving the rearing performance and economic characters of the cocoons in healthy, pre-inoculated and post-inoculated larvae, besides reducing the mortality of infected larvae. Cephelexin and Tetracycline were more effective than the others in their overall performance under all the three experimental conditions. Chloramphenical caused greater increase in the length of the reelable silk filament, but was not as good in elevating the other parameters compared to the remaining antibiotics. Of the four concentrations of antibiotics tested, 2.0% performed better under all the three experimental conditions.

감성돔, Acanthopagrus schlegeli 수정란과 부화자어의 발달에 미치는 3,5,3'-triiodo-L-thyronine (T3)의 침지처리 효과 (Effects of 3,5,3'-triiodo-L-thyronine (T3) Immersion on Development of Fertilized Egg and Newly Hatching Larva of Black Seabream, Acanthopagrus schlegeli)

  • The hatching of fertilized egg and early development of larval black seabream, Acanthopngrus schlegeli by immersion of 3,5,3'-triiodo-L-thyronine ($T_3$) with doses of 0.01~0.5 ppm in tank were examined for early development periods and compared with control fish immersed by 0.1 N NaOH and 99% alcohol. Larvae were fed with rotifers for first 5 days during experimental period. Yolk absorption of larvae from $T_3$ immersion groups were significantly faster than that of control. Although hatching times among each groups were not significant difference. the hatching rates of larvae in higher doses (0.1 ppm and 0.5 ppm $T_3$ groups) were significantly lower than control. The results of $T_3$ immersion experiments to newly hatching larvae indicated that exogenous $T_3$induced significant faster the absorption of yolk and oil globule, faster the development of myotome and digestive duct and higher the growth rate of larvae than larvae of control. These results indicate that $T_3$ immersion supplements appear to confer a distinct advantage to eggs and larvae of black seabream, which is in early fragile developmental stage.

Longevity of Toxocara cati Larvae and Pathology in Tissues of Experimentally Infected Chickens

  • This study was conducted to determine the distribution patterns and duration of stay of Toxocara cati larvae in organs of chickens and to investigate chronic phase and potential zoonotic risk of toxocariasis in chickens. Chickens were orally infected with 1,000 embryonated T. cati eggs and necropsied 240 days post-infection. Organs of the chickens were examined at gross and microscopic levels; tissues were digested to recover larvae. Peribronchiolitis with infiltration of lymphocytes, and hyperplasia of bronchiolar associated lymphatic tissues (BALT) and goblet cells, were evident in the lungs of infected chickens. There were mild hemorrhages and infiltration of lymphocytes and a few eosinophils in the meninges. Larvae were recovered from 30% of the exposed chickens. Larvae recovery indicated that T. cati larvae stay alive for at least 240 days in the chicken brain. Therefore, chickens may potentially act as a paratenic host in nature and transfer T. cati larvae to other hosts.

Proactive Approach for Biofouling Control: Consequence of Chlorine on the Veliger Larvae of Mytilus edulis under Laboratory Condition

  • Macro fouling due to blue mussels (Mytilus edulis) has affected negatively on the operation efficiency and eventual system failure of offshore structures and coastal power stations. A certain range of chlorine (0.05, 0.1, 0.3, 0.5, 0.7 and 1.0 mg/L) was applied on the mussel larvae to identify the survival rate with respect to various exposure times under laboratory condition. The ciliary movement of the larvae was used to check their survival. The 1.0 mg/L of chlorine shows to 97% of larvae mortality whereas 0.7 mg/L of chlorine shows only 16% of larvae mortality. Minimum exposure times for 100% larvae mortality ranged from 300 to 20 min for increasing concentrations of chlorine (0.05~1.0 mg/L). It was found that 1 mg/L of chlorine was 4 times more efficient than 0.7 mg/L of that, and 15 times more than 0.05 mg/L of chlorine dose. Data collected and analyzed here will help plant operators to optimize chlorine dosage and its scheduling.

Feeding the Larvae of the Sea Urchin Strongylocentrotus intermedius on a Red-Tide Dinoflagellate Cochlodinium polykrikoides

  • This study is the first attempt to understand the feeding physiology of a sea-urchin larva on a red-tide dinoflagellate. Fifteen day old larvae of S. intermedius capture C. polykrikoides cells by localized reversal of ciliary beats. No failure to transporte the algal cells from theciliated band to mouth and no rejection at the mouth suggest that C. polykrikoides has no feeding deterrence to S. intermedius larvae. The trend obtained for the clearance rate of S. intermedius larvae is similar to that of other sea urchin larvae. Thus, the clearance rate decreased as the algal concentration increased. Maximum clearance rate of S. intermedius on C. polykrikoides was 17.7 $\mu l$/larva/hr. Ingestion rate rapidly increased at lower algal concentrations and saturated at higher concentrations. There was no inhibition in ingestion rate at the highest prey concentration of ca. 3000 cells/ml. Maximum ingestion rate of S. intermedius on C. polykrikoides was 131 ngC/larva/d, which is higher than that reported for the larvae of the mussel Mytilus gal-lotrovincialis, but lower than that of the ciliate Strombidinopsis sp. The grazing rate, calculated by combining the field data on algal abundances with experimental data on ingestion rate, suggests that due to its low abundance, sea urchin Iarva has no significant grazing impact on C. polykrikoides population.
