Porous Ti-systems with unidirectionally aligned channels were synthesized by freeze-drying and a heat treatment process. $TiH_2$ powder and camphene were used as the source materials of Ti and sublimable vehicles, respectively. Camphene slurries with $TiH_2$ content of 10 and 15 vol% were prepared by milling at $50^{\circ}C$ with a small amount of oligomeric polyester dispersant. Freezing of the slurry was done in a Teflon cylinder attached to a copper bottom plate cooled at $-25^{\circ}C$ while unidirectionally controlling the growth direction of the camphene. Pores were generated subsequently by sublimation of the camphene during drying in air for 48 h. The green body was heat-treated at $1100^{\circ}C$ for 1 h in a nitrogen and air atmosphere. XRD analysis revealed that the samples composed of TiN and $TiO_2$ phase were dependent on the heat-treatment atmosphere. The sintered samples showed large pores of about 120 mm which were aligned parallel to the camphene growth direction. The internal wall of the large pores had relatively small pores with a dendritic structure due to the growth of camphene dendrite depending on the degree of nucleation and powder rearrangement in the slurry. These results suggest that a porous body with an appropriate microstructure can be successfully fabricated by freeze-drying and a controlled sintering process of a camphene/$TiH_2$ slurry.
Al-Li 8090 합금을 재료로 인장과 파괴 시험 중의 음향방출 거동을 조사하여, 미세조직과 방향성에 따른 변형 기구를 규명해 보고자 하였다. 이것을 위해 시편은 미세조직(${\delta}'$상이 지배적인 조직과 $S'+{\delta}'$조직)과 방향성(L방향과 ST방향)에 따라 제조하였으며, 인장과 파괴 시험을 수행하면서 음향방출 거동을 관찰하였고, 이를 투과 및 주사 전자 현미경의 결과와 비교하여 전위의 미세기구에 관하여 고찰해 보았다. ${\delta}'$ 조직에서는 L방향과 ST방향의 시편 모두에서 낮은 RMS의 연속형 방출 신호가 나타났고, $S'+{\delta}'$조직아서는 돌발형 방출 신호들만이 관찰되었다. 여러 연구 결과를 통해 ${\delta}'$조직에서의 연속형 형태의 음향방출은 주로 정합인 ${\delta}'$상의 전단에 기인한 것인 반면, $S'+{\delta}'$조직에서의 돌발형 형태의 음향방출은 부정합 ${\delta}'$상의 전단 또는 microcracking때문인 것으로 추론할 수 있었다. 시편의 방향성에 관해서는, ST방향의 시편들의 경우 LT 방향의 시편들보다 많은 돌발형 신호들을 보였다. ST방향 시편들에서 나타나는 이러한 돌발형 신호들은 주로 균열 전파 방향에 평행하게 놓여져 있는 입계를 따라, 급격하게 진행되는 균열 전파에 의해서 나타나는 것으로 보인다.
Porous W with controlled pore characteristics was fabricated by a freeze-drying process. $WO_3$ powder and camphene were used as the source materials of W and sublimable vehicles, respectively. Camphene slurries with $WO_3$ contents of 10 and 15 vol% were prepared by milling at $50^{\circ}C$ with a small amount of oligomeric polyester dispersant. Freezing of a slurry was done in a Teflon cylinder attached to a copper bottom plate cooled at $-25^{\circ}C$ while the growth direction of the camphene was unidirectionally controlled. Pores were generated subsequently by sublimation of the camphene during drying in air for 48 h. The green body was hydrogen-reduced at $800^{\circ}C$ for 30 min and sintered in a furnace at $900^{\circ}C$ for 1 h under a hydrogen atmosphere. Microstructural observation revealed that all of the sintered samples were composed of only W phase and showed large pores which were aligned parallel to the camphene growth direction. The porosity and pore size increased with increasing camphene content. The difference in the pore characteristics depending on the slurry concentration may be explained by the degree of powder rearrangement in the slurry. The results strongly suggest that a porous metal with the required pore characteristics can be successfully fabricated by a freeze-drying process using metal oxide powders.
Generally vascular grafts with a relatively large inner diameter (> 5 mm) have been successfully employed for replacement in the human body. However, the use of small diameter grafts is limited, because these grafts rapidly occlude due to the thrombosis. The ideal blood-contacting surface of a prosthesis would be an endothelial cell (EC) lining, because the confluent monolayer of healthy ECs that culture natural blood vessels represents the ideal nonthrombogenic surface. For vascular graft application, the stable EC adhesion on surface under How conditions is very important. In this study, the adhesive strength of ECs attached on polymer surfaces coated with collagen type IV (Col IV), fibronectin (Fn), laminin (Ln), and treated with corona was investigated onto polyurethane (PU) films. The EC-attached PU surfaces were mounted on parallel-plate flow chambers in a How system prepared for cell adhesiveness test. Three different shear stresses (100, 150, and 200 dyne/㎠) were applied to the How chambers and each shear stress was maintained for 120 min to investigate the effect of shear stress and surface treatment condition on the EC adhesion strength. It was observed that the EC adhesion strength on the surface-modified PU films was in the order of Ln≡Fn > Col IV > corona 》 control. More than 70% of the adhered cells were remained on surface-modified PU surface after applying the shear stress,200 dyne/㎠ for 2 hrs, whereas the cells were completely detached on the control PU surface within 10 min after applying the same shear stress. It seems that the type of adsorbed proteins and hydrophilicitv onto the PU surfaces play very important roles for cell adhesion strength.
This paper proposes a novel way of fabricating aligned porous Sn by freeze-drying of camphene slurry with stannic oxide ($SnO_2$) coated Sn powders. The $SnO_2$ coated Sn powders were prepared by surface oxidation of the initial and ball-milled Sn powders, as well as heat treatment of tin chloride coated Cu powders. Camphene slurries with 10 vol% solid powders were prepared by mixing at $50^{\circ}C$ with a small amount of oligomeric polyester dispersant. Freezing the slurry was done in a Teflon cylinder attached to a copper bottom plate cooled at $-25^{\circ}C$. Improved dispersion stability of camphene slurry and the homogeneous frozen body was achieved using the oxidized Sn powder at $670^{\circ}C$ in air after ball milling. The porous Sn specimen, prepared by freeze-drying of the camphene slurry with oxidized Sn powder from the heat-treated Sn/tin chloride mixture and sintering at $1100^{\circ}C$ for 1 h in a hydrogen atmosphere, showed large pores of about $200{\mu}m$, which were aligned parallel to the camphene growth direction, and small pores in their internal walls. However, $100{\mu}m$ spherical particles were observed in the bottom part of the specimen due to the melting of the Sn powder during sintering of the green compact.
Rhinomonas reticulata var. atrorosea G. Novarino is a photosynthetic marine flagellate that is known to have typical characteristics of cryptomonads. We examined the flagellar apparatus of R. reticulata var. atrorosea by transmission electron microscopy. The major components of the flagellar apparatus of R. reticulata var. atrorosea consisted of four types of microtubular roots (1r, 2r, 3r, and mr), a non-keeled rhizostyle (Rhs), mitochondrion-associated lamella (ML), two connections between basal bodies, a striated fibrous root (SR) and a striated fiber-associated microtubular root (SRm). Four types of microtubular roots originated near the ventral basal body and extended toward the left side of the basal bodies. The non-keeled Rhs originated at the Rhs-associated striated fiber, which was located between two basal bodies and extended into the middle of the cell. The ML was a plate-like fibrous structure associated with mitochondria and originating from a Rhs-associated fiber. It split into two parts and extended toward the dorsal-posterior of the cell to a mitochondrion. The SR and SRm extended parallel to the anterior lobe of the cell. The overall configuration of the flagellar apparatus in R. reticulata var. atrorosea was similar to the previously reported descriptions of those of Cryptomonas paramecium, C. pyrenoidifera, C. ovata, Hanusia phi, Guillardia theta, and Proteomonas sulcata. However, the flagellar apparatus system of R. reticulata var. atrorosea was more complex than those of other cryptomonad species due to the presence of an additional microtubular root and other distinctive features, such as a rhizostyle-associated striated fiber and large ML.
In this study, the mechanical properties of the Al-Mg-X (X=Cr, Si) alloy, which clearly showed the influence of the specimen and grain size, were investigated by changing the specimen size extensively. In addition, the effect on the specimen size, grain size and aging condition on the mechanical properties of the grain refining alloy according to the addition of Cr was clarified, and the relationship between these factors was studied. As the specimen size decreased, the yield stress decreased and the fracture elongation increased. This change was evident in alloys with coarse grain sizes. Through FEM analysis, it was confirmed that the plastic deformation was localized in the parallel part of specimen S2. Therefore, when designing a tensile specimen of plate material, the W/L balance should be considered along with the radius of curvature of the shoulder. In the case of under-aged materials of alloys with coarse grain size, the fracture pattern changed from intergranular fracture to transgranular fracture as W/d decreased, and δ increased. This is due to the decrease in the binding force between grains due to the decrease in W. In the specimen with W/d > 40 or more, intergranular fracture occurred, and local elongation did not appear. Under-aged materials of alloys with fine grain size always had transgranular fracture over a wide range of W/d = 70~400. As W/d decreased, δ increased, but the change was not as large as that of alloys with coarse grain sizes. Compared to the under-aged material, the peak-aged material did not show significant dependence on the specimen size of σ0.2 and δ.
Lovastatin은 근사형성 균류인 Aspergillus terreus가 생합성하는 이차대사산물로 강력한 고지혈증 치료제로 널리 이용되는 물질이다. 본 연구에서는 lovastatin 고생산변이주를 이용하여 포자배지 최적화를 통한 miniature 배양 방법을 확립하고자 하였다. 우선 miniature 배양에 필수적인 효과적인 포자 형성 방법을 개발하고자 포자 형성 배지의 통계학적 배지 최적화를 수행하였다. Miniature 배양의 inoculum으로 이용되는 대량의 포자를 획득하기 위해 Plackett-Burman 실험법을 이용하여 포자 형성을 향상시키는 성분을 조사한 결과, glucose, sucrose, yeast extract 그리고 $KH_2PO_4$가 주목할 만한 효과를 보였다. 상기 성분의 최적 농도를 확인하기 위해 반응표면분석법 (RSM)을 이용한 결과, PDA 포자 형성 배지와 비교하여 볼 때, 최적 성분 농도에서 포자 형성이 약 190배 증가하였다. 최적화된 포자형성 배지를 이용하여 lovastatin 고생산성 변이주의 대량 선별을 위한 miniature 배양 방법을 확립하기 위해 기존의 실험 과정에 'PaB (adaptation)'라는 한 번의 계대배양을 더 추가한 결과 생산균주의 안정성과 재현성이 큰 폭으로 증가하는 주목할 만한 결과를 얻을 수 있었다. 단기간에 가능한 한 다량의 균주를 스크리닝하기 위해 성장배양과 생산배양 모두 조업부피가 7 ml인 tube를 이용해 miniature 배양을 반복 수행하여, lovastatin 생산성과 배양형태가 훌륭한 변이주를 선별할 수 있었는데, 이 균주는 7 ml tube배양과 250 ml flask배양 (조업부피 50 ml) 모두에서 생산성이 높은 것으로 보아 산소 의존도가 비교적 낮고 생산 안정성이 높은 균주인 것으로 판단되었다. 한편 miniature 배양을 이용해서 lovastatin 고생산성을 보이는 균주를 신속 선별하기 위해서는 균주의 적절한 배양형태 유도가 매우 중요한 것으로 관찰되었다. 즉 생산배양으로의 고활성 균주의 접종을 위해서, 또한 생산배양에서 pellet의 배양형태 유도를 위해서 성장배양 시에는 반드시 고농도의 균사모양을, 생산배양 시에는 직경 1 mm 이하의 pellet모양의 배양 형태를 유지해야만, 생산균주가 lovastatin을 안정적으로 고생산할 수 있는 것으로 관찰되었다. 초기에 선별된 균주를 이용하여 miniature 배양에 의해 고속 균주선별 실험을 반복함으로써 고생산성 균주들을 다량 선별할 수 있었는데, 이들의 lovastatin 생산성을 조사한 결과, 기존의 flask 배양대비 오차범위가 $\pm$20% 이내의 생산성을 보이는 균주가 초기 선별시의 32%에 비해 81%로 크게 증가함을 확인할 수 있었다. 이와 같은 결과는 lovastatin 고생산성, 고안정성 균주의 고속 스크리닝을 위해서 본 연구에서 개발한 tube를 이용한 miniature 배양이 기존의 flask 배양을 대체할 수 있는 훌륭한 배양방법임을 제시해 준다.
계룡산국립공원으로부터 기증된 야생오소리의 췌장에서 오소리구충 (Tetragomphius procyonis)을 분리하여 광학 및 주사전자현미경으로 그 형태를 관찰하였다. 충체의 길이는 수컷 15.0-18.8 mm, 암컷 21.5-25.5 mm에 달하였다. 암수 모두 구강 복면 변연에 한 쌍의 절판과 배면에 하나의 절판이 존재하였다. 컵모양의 구강 기저부에 한쌍의 분기된 아복치 및 한쌍의 아배치가 존재하였다. 충체 전체에 가로 줄무늬가 존재하였고 경부유두 선상의 복측 중앙에 배설공이 개구하였다. 가늘고 긴 교접자 (8.7-9.3 mm)는 그 끝이 서로 융합되어 있었다. 교접낭은 잘 발달되어 있었으며 잘 발달된 배늑은 평행으로 주행하였다. 음문은 충체 후방 4/5 부위에 개구하였고 꼬리에는 미돌기 및 미유두가 존재하지 않았다. 배구 및 부교접자가 존재하지 않았으며 음문은 체후방 4/5에 개구하였다. 자궁내의 충란은 16 세포기였고 직장내의 충란은 32 세포기였으며 충란의 크기는 77.48-83.45 ${\times}$ 50.75-63.38 ${\mu}m$였다.
전자선의 선량을 측정하기 위하여 수종의 측정기와 팬톰의 조합에 대해서 실험 및 결과를 분석하였다. 전자선의 선축상 선량분포가 조사면의 크기에 좌우되는지를 알기 위하여 실리콘 PN 접합형 다이오드를 사용하였다. 50 및 80, $90\%$ 선량점의 깊이가 작은 조사면에 대해서는 조사면의 크기에 따라 증가하지만, 큰 조사면에 대해서는 거의 일정했다. 그러나 그 깊이가 일정한 경우 최소 조사면의 크기가 뚜렷하지는 않으나 전자선의 에너지가 증가함에 따라 증가하였다. $6\~18MeV$의 전자선에 대해 조사면의 크기가 $10\times10cm^2$ 이상인 경우 그 깊이 가 어느 에너지에 대해서도 조사면의 크기에 무관함이 측정치에서 관찰되었다. 그래서 수종의 측정기와 팬톰의 결합에 따른 전자선의 선축상 선량분포의 차이점을 관찰하고자 하는 실험에서 조사면의 크기로 $10\times10cm^2$을 선택하였다. 원주형 전리함과 평판형 전리함, 필름은 폴리스티렌 팬톰과 함에, 실리콘다이오드는 물팬톤과 함께 선량측정에 이용되었다. 원주형 전리함은 표면선량이나 6MeV처럼 낮은 에너지의 선량증가 영역에서 선량을 측정할 수 없었다. 몇 가지를 제외하고는 측정된 변수들은 서로 다른 측정기 및 팬톰의 결합에 관계없이 거의 동일하였다. 어떤 에너지에서는 서로 다른 측정기에 의한 표면선량이 $4\%$ 정도 차이가 났으며, 에너지가 증가함에 따라 그 대소가 반전되기도 하였다. 18MeV의 경우 필름에 의한 80 및 $90\%$ 선량점의 깊이가 다른 측정기에 의한 것보다 꽤 얕았다. 평판형 전리함과 실리콘 다이오드는 전자선의 선량분포측정에 사용될 수 있겠으나 평판형 전리함은 큰 조사면에서만 사용하는 것이 바람직할 것이 다. 표면선량 측정이나 저 에너지 전자선의 선량증가 부분에서 선량측정에는 원주형 전리함을 사용하지 않는 것이 바람직할 것이다. 18 MeV와 같이 높은 에너지의 전자선의 선량분포 측정에 필름이 사용되어도 좋을지 의심스럽다.
