  • All vertebrates other than lampreys exhibit multiple loci encoding lactate dehydrogenase (EC 1.1.1.27,LDH). From the result shown by cellulose acetate and starch gel electrophoresis, the lampreys were-reported to have only one isozyme. However in our results the LDH of skeletal muscle, heart and kidney in Lampetra japonica were separated into three isozymes and that of liver was separated into two isozymes by polyacrylamide gel electrophoresis. The LDH of skeletal muscle and heart were separated into four isozymes and that of liver was separated into two isozymes by polyacrylamide gel isoelectric focusing (PAGlEF). The LDH of skeletal muscle were separated into four isozymes through the chromatofocusing. The molecular weight of LDH isozymes in skeletal muscle was approximately estimated to be 140,000 by Sephadex G-200 gel filtration. The LDH isozymes of skeletal muscle, heart and liver were inhibited by pyruvate to the nearly similar degree. And the degree of inhibition by pyruvate showed the value between LDH A$_4$and LDH B$_4$isozyme. And the LDH isozymes in heart, liver and skeletal muscle were thermostable. The results mentioned above indicate that the LDH isozyme in lamprey (Lampetra japonica) has not one isozyme but isozymes. And it is also found out that the two structures of their subunits are similar each others.

  • The lactate dehydrogenase (EC 1.1.1.27, LDH) in liver of Lempetra japonica was purified in buffer of affinity chromatography. The liver-specific $C_4$ isozyme of Gadus macrocephalus was purified by heat treatment, affinity chromatography, and DEAE-Sephacel chromatography. The liver-specific $C_4$ isozyme was eluted in a buffer containing NAD+ and was coeluted with $B_4$isozyme in plain buffer of affinity chromagraphy. Liver-specific $C_4$ isozyme in G. macrocephalus was the most thermostable, and$B_4$isozyme was more stable than $A_4$. The LDH in the fraction of pH 7.45 purified from the liver of L. iaponica by chromatofocusing was more inhibited by pyruvate than purified LDH. The optimum pH of the LDH isozyme in the liver of L. japonica was 7.5 and that of liver-specific$C_4$ isozyme was 8.5. The LDH in liver of L. japonica made complexes more with antibody against Coreoperca herzi$A_4$ and liver-specific $C_4$ than with that against eye-specific $C_4$. Therefore, the structure of the LDH in liver of L. japonica might be similarly evolved to that of subunit A and liver-specific $C_4$ isozyme in liver tissue of G. macrocephalus. The evolution rate of subunit C is faster than that of subunit A. LDH in liver of L. japonica has not one isozyme but isozymes and it was also found out to have not only subunit A and B but also subunit C.

  • Lj-RGD3, an RGD (Arg-Gly-Asp) toxin protein from the salivary gland of Lampetra japonica, exhibits antifungal activity against Candida albicans. Lj-RGD3 has three RGD motifs and shows homology to histidine-rich glycoprotein. We synthesised two mutant derivatives of Lj-RGD3: Lj-26, which lacks all three RGD motifs and contains no His residues; and Lj-112, which lacks only the three RGD motifs. We investigated the effects of the wild-type and mutated toxins on a gram-positive bacterium (Escherichia coli), a gram-negative bacterium (Staphylococcus aureus), and a fungus (C. albicans). rLj-RGD3 and its mutants exhibited antifungal but not antibacterial activity, as measured by a radial diffusion assay. The C. albicans inhibition zone induced by rLj-112 was larger than that induced by the other proteins, and its inhibitory effect on C. albicans was dose-dependent. In viable-count assays, the rLj-112 MIC was $7.7{\mu}M$, whereas the MIC of the positive control (ketoconazole) was $15{\mu}M$. Time-kill kinetics demonstrated that rLj-112 effectively killed C. albicans at $1{\times}$ and $2{\times}$ MIC within 12 and 6 h, respectively. Electron microscopy analysis showed that rLj-RGD3 and rLj-112 induced C. albicans lysis. Our results demonstrate a novel anticandidal activity for rLj-RGD3 and its mutant derivatives.

  • Chromosome numbers of 3 species of Korea lampreys(Genus Lampetra) were investigated. Approximately 160 small and mainly acrocentric chromsomes were present in the three species of Korean lampreys. And the diploid chromosome numbers of three species were not significantly different between the species.

  • The ichthyofauna and fish community were studied in Miho Stream from May 2004 to August 2005. During the surveyed period 41 species belonging to 10 families were collected. Twelve of the species (29.3%) are endemic to Korea. Exotic fishes were Carassius cuvieri, Lepomis macrochirus, and Micropterus salmoides. Dominant species was Zacco platypus (54.7%). Comparing the present result with that of Son (1982), the indices of diversity, evenness and richness were lower and the dominance index was higher; Lampetra reissneri, Anguilla japonica, Acheilognathus macropterus, Coreoleuciscus splendidus, Pseudopungtungia nigra, Gobiobotia naktongensis, Lefua costata, Pseudobagrus koreanus, Monopterus albus and Iksookimia choii did not appear; Carassius auratus, Rhodeus uyekii, Rhodeus notatus, Acheilognathus lanceolatus, Pseudogobio esocinus, Hemibarbus longirostris, Microphysogobio yaluensis, Cobitis lutheri, Iksookimia koreensis, Pseudobagrus fulvidraco, and Rhinogobius brunneus, remarkably decreased, and Z. platypus, Pseudorasbora parva, and Squalidus gracilis majimae increased.

  • Pineal organs of poikilotherm vertebrates transform the environmental light information into a humoral message and a neuronal activity. The former is melatonin, and the latter is modulation of the impulse in ganglion cells. The ganglion cells are physiologically classified into luminosity (achromatic) type and chromatic one, as the neural activity is modulated in two ways. We attempted to classify the pineal ganglion cells with morphological characteristics by means of the three- dimensional reconstruction method. In the pineal ganglion cells of river lamprey, there are two different features, oval and spherical. For comparison of their projection region in the brain, the tracing investigation was also carried out. The application of the neural tracer near mesencephalic tegmentum showed that only oval-shaped ganglion cells were labeled in the pineal organ. These results suggest that the oval-shaped ganglion cell is functionally different from the spherical one.
