Effects of Predator Addition to the Algicidal Bacterium in Controlling Diatom Sephanodiscus hantzschii Dominating the Eutrophic Pal′tang Reservoir, Korea (살조세균과 초식성 섭식자의 혼합배양에 의한 녹조제어효과)

  • An algicidal effect of endemic algicidal bacterium (Pseudomonas putida) and rotifer zooplankton (Brachionus calyciflorus) on diatom Stephanodiscus hantzschii were examined in the filtered water, and were compared with those of bacterium plus ciliate. Bacteria removed as 80% of the diatom within 3.5 days, while ciliate and zooplankton suppressed 57% and 40% of diatom during the same period, respectively. Mixed treatment of bacteria plus ciliate removed as 54% of diatoms, while that of bacteria plus zooplankton decreased as 85%. Although single bacteria and mixed treatment of bacteria plus zooplankton quickly decreased the diatom in the initial of experiment, bacteria plus ciliate perfectly removed the diatom in culture flask within 5.5 days of the study. On the other hand, other single and mixed treatments did not clear the diatom during the same period, and over 10% of them still remain in culture flask. Predator biomass in the presence of algicidal bacteria showed the growth patterns; zooplankton gradually decreased, and ciliate sustained over 0.5 cells/ml. These results indicated that the addition of ciliate to the algicidal bacterium in controlling the diatom Stephanodiscus hantzschii is more effective than that of zooplankton. Therefore, this synergistic interaction relationship between the bacterium and ciliate play an important role in the bio-manipulation using bio-agents to control the diatomal bloom in freshwater lakes and streams.

Development of Multiple Regression Equation for Estimation of Suspended Solids in Unmeasurable Watershed (미계측 유역의 부유물질 산정을 위한 다중회귀식 개발)

  • The purpose of this study is to present quantitatively the influence of variables that had the largest effect on the changes in suspended solids(SS), which would cause turbid water phenomenon, among water quality factors of the non-point pollution source, and then to develop a multiple regression equation of SS and predict the water quality of ungaged watersheds so as to provide basic data to establish efficient management plans for SS which flow in rivers and lakes. To identify the correlation of SS with the amount of rainfall and the state of land use, a simple correlation analysis and a simple regression analysis were conducted respectively. Finally, a multiple regression analysis was conducted to provide that SS were set as dependent variables while the amount of rainfall, paddy fields and dry fields were set as independent variables. As a result, the amount of rainfall had the most significant influence on changes in SS, followed by dry fields and paddy fields. In addition, the multiple regression equation was developed to predict SS in unmeasurable watersheds.

Change of Groundwater Quality derived from Contaminant Sources (오염원에 의한 지하수 수질의 변화)

  • In order to provide for the guidance on groundwater quality monitoring network design and also, to suggest the index to the solution of the contaminated groundwater remediation problems in the lake watershed, it is necessary to analyze the contaminant transport in the groundwater. The solute transport was analyzed in the lake watershed to investigate the behavior of the injected contaminant sources depend on the relationships between the point of contaminant sources and position of the lake. Three hypothetical groundwater flow systems, which is composed of a flow-through lake and two solute sources, were considered. The lakes located in the upper, middle, and lower portions of a watershed respectively. The transported contaminant was numerically simulated for five years by using MT3D contaminant transport model under the three-dimentional steady state conditions. From the above simulations, it can be concluded that the contaminant concentration was high as the contaminant source located at the upper position of a watershed, and the influence of the contaminant injection was large as the solute source located at the lower position. When the injection of contaminant was continued for one year without regard to the position of contaminant source and the lake, the influence of contaminant source was reached to bedrock.

  • The different methods such as HPLC, indirect- and direct-ELISA were employed for the analysis of microtoxins and the results of each method were compared in terms of the detection limit and accurary. Three toxins, microcystin-RR, -LR and -YR were clearly separated by HPLC using 0.05 M methanol and phosphate buffer used as a solvent system. The calibration curves for the toxins were linear in the range of 5 ng to 50 ng. The standard curves for the immunoassay of microcystin obtained by direct and indirect ELISA are compared. The linear responses of inhibitions of binding by microcystin in the direct and indirect competitive ELISA were in the range of 10 ng to 1000 ng and 50 pg to 160 pg, respectively. Distribution of microtoxins at 11 sites in the Nakdong river and several lakes in Korea was also studied. The most dominant microcystin variant in the test sites was found to be microcystin-RR.

The avifauna at Chunsoo bay (Seosan A and B area)

  • Seosan A, B area is located on 36°42' of north parallel and 126°27' of east longitude. It consisted of broad artificial lakes and reclaimed agricultural lands; there is Ganwalho lake in A area and Bunamho lake in B area. Total birds surveyed four times in 2001 at the A area are 105,580 of 11 orders, 28 families, 89 species. Among them, resident species were 21 species including Pica pica. Twenty species including Cuculus canorus were summer migrants, and 30 species including Platalea leucorodia were winter migrants. Seventeen species including Limosa limosa were occasionally species. And Rostratula benghalensis was an unconfirmed species. Species diversity index (H') was 0.72; species equally common index (e/sup H'/) was 2.06. In January, maximum observed 102,121 individuals. However in July 52 species were observed most variously. There were total 81,152 birds observed with 11 orders, 22 families, and 71 species at the B area. Fifteen species including Falco tinnumculus were resident species. Summer migrants were 18 species including Ixobychus sinensis. Winter migrants were 25 species including Ciconia boyciana. Thirteen species including Tringa glareola were occasionally species. Species diversity index (H') was 0.281; species equally common index (e/sup H'/) was 1.325. In January, maximum observed 78,433 individuals. However in Silly 42 species were observed most variously.

Evaluation of Water Quality in the Keum River using Statistics Analysis (통계분석 기법을 이용한 錦江水系의 水質評價)

  • This study was conducted to evaluate water quality in the Keum River using multivariate analysis. The analysis data in Keum river made use of surveyed data by the ministry of environment from January 1994 to December 2001. Thirteen water quality parameter were determined on each sample. The results was summarized as follow; Water quality in the Keum River could be explained up to 71.39% by four factors which were included in loading of organic matter and nutrients by the tributaries (32.88%), seasonal variation (16.09%), loading of pathogenic bacteria by domestic sewage of Gapcheon (13.39%) and internal metabolism in estuary as lakes(9.03%). For spatial variation of factor score, four group was classified by each factor characterization. Station 1 and 2 was influenced by Daechung dam, station 3 was affected by domestic sewage of Gapcheon, station 10~12 was affected by estuary dyke and the rest station. The result of cluster analysis by station was classified into four group that has different water quality characteristics. In monthly cluster analysis, three group was classified according to seasonal characteristic. Also, in yearly cluster analysis, three group was classified. It is necessary to control the pollutant loadings by Gapcheon inflow domestic sewage in Daejeon city for the sake of water quality management of Keum river.

A Basic Study of Thermal-Fluid Flow Analysis Using Grid Computing (그리드 컴퓨팅을 이용한 열유동 해석 기법에 관한 기초 연구)

  • Simulation of three-dimensional turbulent flow with LES and DNS lakes much time and expense with currently available computing resources and requires big computing resources especially for high Reynolds number. The emerging alternative to provide the required computing power and working environment is the Grid computing technology. We developed the CFD code which carries out the parallel computing under the Grid environment. We constructed the Grid environment by connecting different PC-cluster systems located at two different institutes of Pusan National University in Busan and KISTI in Daejeon. The specification of PC-cluster located at two different institutes is not uniform. We run our parallelized computer code under the Grid environment and compared its performance with that obtained using the homogeneous computing environment. When we run our code under the Grid environment, the communication time between different computer nodes takes much larger time than the real computation time. Thus the Grid computing requires the highly fast network speed.

Characterization of Choristoneura fumiferana Genes of the Sixth Subunit of the Origin Recognition Complex: CfORC6

  • A new protein was cloned and identified as the sixth subunit of Choristoneura fumiferana origin recognition complex (CfORC6). The newly identified 43 kDa protein CfORC6 is much bigger than DmORC6 (25.7 kDa) and HsORC6 (28.1 kDa), though it's 23.85% identical to DmORC6 and 23.81% identical to HsORC6. Although the molecular weight of CfORC6 is close to ScORc6 (50 kDa), CfORC6 is only 14.03% identical to ScORC6. By alignment, it was found that the N-terminal of CfORC6 has about 30% identities with other ORC6s, but about 100aa of C-terminal of CfORC6 has no identity with other ORC6s. Like ScORC6, CfORC6 has many potential phosphorylation sites, (S/T)PXK. Like DmORC6, CfORC6 has leucine-rich region in the relevant site. Northern Blot showed that CfORC6 mRNA is about 2,000nt. Southern Blot confirmed that there is one copy of CfORC6 gene in spruce budworm genome. Western blot showed that infection of Cf124T cells with CfMNPV didn't affect the expression levels of CfORC6, at least up to 26 hr post infection.

Archaeal Diversity in Tidal Flat Sediment as Revealed by 16S rDNA Analysis

  • During the past ten years, Archaea have been recognized as a widespread and significant component of marine picoplankton assemblages. More recently, the presence of novel archaeal phylogenetic lineages has been discovered in coastal marine environments, freshwater lakes, polar seas, and deep-sea hydrothermal vents. Therefore, we conducted an investigation into the archaeal community existing in tidal flat sediment collected from Ganghwa Island, Korea. Phylogenetic analysis of archaeal 16S rDNA amplified directly from tidal flat sediment DNA revealed the presence of two major lineages, belonging to the Crenarchaeota ($53.9\%$) and Euryarchaeota ($46.1\%$) phyla. A total of 102 clones were then sequenced and analyzed by comprehensive phylogenetic analysis. The sequences determined in our samples were found to be closely related to the sequences of clones which had been previously obtained from a variety of marine environments. Archaeal clones exhibited higher similarities ($83.25 - 100\%$) to sequences..from other environments in the public database than did those ($75.22 - 98.46\%$) of previously reported bacterial clones obtained from tidal flat sediment. The results of our study suggest that the archaeal community in tidal flat sediment is remarkably diverse.
