A Study on the Kinetic Parameters of Alkaline Phosphatase by Algae (조류에 의한 유기인산염분해효소의 Kinetic Parameters에 관한 연구)

  • Alkaline phosphatase ($AP_{ase}$) activity was studied for the pure cultures and natural communities of phytoplankton. The Michaelis-Menten constant ($K_m$) showed large variation with species. Some green algae showed large $K_m$ ($650\;{\mu}M$ for Selenastrum capriconutum). Chlorella sp. and Nitzschia palea showed smaller Km (respectively1.7, 2.0 HM) than those of other species examined. The extracellular free enzyme in the filtrate of Anabaena flos-aquae showed smaller Km ($52\;{\mu}M$) than that of cellbound form ($276\;{\mu}M$). The $K_m$ ($12.0\;{\mu}M$) of summer phytoplankton in Lake Soyang. when Anabaena sp. was dominant species, was larger than that (1.5 HM) of spring phytoplankton when Asterionella sp. was the dominant. Although maximum activity($V_{max}$) in Lake Soyang was affected by the concentration of DIP within the lake, but the $V_{max}$ always varied not with the DIP concentration of the lake. Induction of $AP_{ase}$ may be more affected by the phosphate content within the cell of phytoplankton than by the concentration of DIP within the lake, The extracellular free $AP_{ase}$ activity accounted for $36{\sim}97%$ of total activity from fall to spring turnover in Lake Soyang. The $K_m$ ($1.1{\mu}3.5\;M$) of extracelluar free enzyme were simillar to those ($0.7{\mu}3.5\;M$) of the total activity. This indicates that the extracelluar free enzyme was derived from phytoplankton.

  • To define the structure and diversity of bacterial communities in the aqutic ecosystem, Lake Soyang, the largest artificial reservoir in Korea, a new method, fluorescent in situ hybridization was applied. This technique relies on the specific hybridization of the nucleic acid probes to the naturally amplified intracellular rRNA. By this method, the bacterial community composition of Lake Soyang and bacterial numbers belong to eubacteria, proteobacteria and Cytophaga-Flavobacterium group were estimated. Total bacterial numbers ranged from $0.3{\times}10^6{\sim}2.0{\times}10^6cells{\cdot}ml^{-1}$, and vertical profile of total bacteria showed the peak at 2 and 5 m depths. The ratio of eubacteria to total bacteria were 22~100% and varied with depth and season. The percentage of Proteobacteria ${\alpha}$-group ranged 2.6~66.7%, ${\beta}$-group 4.5~53.5%, ${\gamma}$-group 4.6~76.7% and Cytophaga-Flavobacterium group 2.1~35.9%. Also, bacteria] community had spatial and temporal characteristics. The dominant groups were ${\beta}$-group in winter, ${\gamma}$-group in spring and early summer and ${\alpha}$-group in summer.

Bacterial Community Analysis of Lake Soyang in Winter by Using 16S and 23S rRNA-targeted Probes (16S와 23S rRNA에 결합하는 probe를 이용한 겨울철 소양호 세균 군집 구조의 분석)

  • To scrutinize the bacterial community composition of Lake Soyang in winter, bacterial numbers belonging to Eubacteria, Proteobacteria and Cytophaga-Flavobacterium group were estimated by using 16S and 23S rRNA targeted oligonucleotide probes. Total bacterial numbers ranged from $0.7{\times}10^6$ to $1.1{\times}10^6cells{\cdot}ml^{-1}$, and vertical profile of total bacteria showed a peak at 5 m depth. The ratio of eubacteria to total bacteria were 34~90% and at 5 m and 10 m depths those were low exhibiting, 39 and 34%, respectively. The percentage of proteobacteria ${\alpha}$-group ranged 10.8~28.7%, ${\beta}$-group 4.5~53.5%, ${\gamma}$-group 4.9~35.5% and Cytophaga-Flavobacterium group 6.1~21.1%. The dominant groups were ${\beta}$-group at 0, 2 and 5 m, ${\gamma}$-group at 10 m, ${\alpha}$-group at 30 m and Cytophaga-Flavobacterium group at 50 m depth. In winter season, Lake Soyang can be divided into three layer, 0~2 m, 5~10 m and 30~50 m, by the bacteria community composition. By this method, new informations about aquatic ecosystem were developed.

  • Structure and dynamics of the phytoplankton communities of Uiam lake, Korea, was investigated. In the Uiam lake four dominant species were found Oscillatoria limosa at Chuncheon City side, O. tenuis at Soyang river side, Melosira italica at south-east side and Asterionella gracillima at west side of the lake. By cluster analysis, based on the similarity index and dissimilarity index, the phytoplanktons in this lake were grouped into three communities; i.e. Oscillatoria, Melosira and Asterionella community. And also the same groups obtained by the cluster analysis were recognized by polar ordination technique along polluted degree gradient. It is clear that oscillatoria community occur in polluted site, Asterionella community in unpolluted site and Melosira community in less polluted site.

Phosphorus Cycle in a Deep Reservoir in Asian Monsoon Are3 (Lake Soyang, Korea) and the Modeling with a 2-D Hydrodynamic Water Quality Model [CE-QUAL-W2] (아시아 몬순지역의 대형댐(소양호)에서의 인순환과 2차원모델의 적용)

  • Phosphorus cycle was studied in a deep stratified reservoir in summer monsoon area (Lake Soyang, Korea) by surveying phosphorus input from the watershed and the movement of phosphorus within the reservoir. And the spatial and temporal distribution of phosphorus was modeled with a 2-dimensional water quality model (CE-QUAL-W2), Phosphorus loading was calculated by measuring TP in the main inflowing river (the Soyang River) accounting for 90% of watershed discharge. TP of the Soyang River showed a large daily variation with the flow rate. High phosphorus loading occurred during a few episodic storm runoff laden with suspended sediments and phosphorus. Because storm runoff water on rainy days have lower temperature, it plunges into a depth of same temperature (usually below 20m depth), forming an intermediate turbidity layer with a thickness of 20 ${\sim}$ 30 m. Because of stable thermal stratification in summer the intermediate layer water of high phosphorus content was discharged from the dam through a mid-depth outlet without diffusing into epilimnion. The movement of runoff water within the reservoir, and the subsequent distribution of phosphorus were well simulated by the water quality model showing a good accuracy. The major parameter for the calibration of phosphorus cycle was a settling velocity of detritus, which was calibrated to be 0.75 m ${\cdot}$ $day^{-1}$. It is concluded that the model can be a good simulator of limnological phenomena in reservoirs of summer monsoon area.

Long-Term Annual Trend Analysis of Epilimnetic Water Quality and Their Longitudinal Heterogeneities in Lake Soyang (소양호 표층수 수질의 연별 추이 및 상 ${\cdot}$ 하류 이질성 분석)

  • The spatial and temporal trends of water qualities in Lake Soyang was statistically analyzed in this study. The water qualities include nutrients, ionic contents and chlorophyll-a (Chl-a) measured during 1993${\sim}$2000. The rainfall intensity and runoff from the catchment appeared to play an important role in water quality trends in the lake. According to seasonal Mann-Kendall test, conductivity, TP, and Ctl-a did not show any trends of increase or decrease over the 8 year period, while TN declined slightly. It was found that the variation of TP was a function of interannual inflow and rainfall. In the analyses of spatial trend, conductivity, based on the mean by site, showed a downlake decline over the eight year period. Minimum conductivity was found in the headwaters during summer monsoon of July to August and near the dam during October. This result indicates a time-lag phenomenon that the headwater is diluted by rainwater immediately after summer monsoon rain and then the lake water near the dam is completely diluted in October. During summer period, TP and TN had an inverse relation with conductivity values. Concentrations of TP peaked during July to September in the headwaters and during September in the downlake. Also, TN increase during the summer and was more than 1.5 mg/L regardless of season and location, indicating a consistent eutrophic state. Values of Chl-a varied depending on location and season, but peaked in the midlake rather than in the headwaters during the monsoon. Regression analyses of log-transformed seasonal Chl-a against TP showed that value of $R^2$ was below 0.003 in the premonsoon and monsoon seasons but was 0.82 during the postmonsoon, indicating a greater algal response to the phosphorus during the postmonsoon. In contrast, TN had no any relations with Chl-a during all seasons.

Comparison of Settleable and Non-settleable Suspended Sediment in Streams and Lakes (강우시 하천과 호수에서 침강성 부유토사와 비침강성 부유토사의 비교)

  • Total suspended solids (TSS), settleable suspended solids (SSS) and nonsettleable suspended solids (NSSS) were compared in streams and lakes during rain events. Samples were collected in Lake Soyang and nine agricultural streams during storm events over two years. To measure NSSS, samples were settled in 1 L cylinders for one hour and then 50% of the total sample volume was taken from the center of the cylinder. The SSS/TSS ratio in streams (0.50) was higher than in Lake Soyang (0.20). The ratio of TSS to turbidity decreased from 1.21 in the inflowing river to 0.69 in the reservoir outlet. Turbidity and NSSS showed higher correlation ($r^2$=0.82) compared with the correlation between turbidity and TSS ($r^2$=0.66). Median particle size decreased from $11.4\;{\mu}m$ in the inflowing river to $4.2\;{\mu}m$ in the lake outlet. The results indicate that only fine particles of less than $5{\sim}10\;{\mu}m$ can be transported downstream of the lake. Because fast-flowing water in streams includes not only fine particles but also coarse particles, an overestimation of suspended sediment concentration results if only TSS is measured. Consequently, TSS and NSSS should be measured separately to evaluate the effect of suspend sediment loadings of turbid streams on aquatic habitat during the monsoon season.
