Overexpression of Arabidopsis Homogentisate Phytyltransferase or Tocopherol Cyclase Elevates Vitamin E Content by Increasing γ-tocopherol Level in Lettuce (Lactuca sativa L.)

  • Tocopherols, essential components of the human diet, are synthesized exclusively by photosynthetic organisms. To increase tocopherol content by increasing total flux to the tocopherol biosynthetic pathway, genes encoding Arabidopsis homogentisate phytyltransferase (HPT/V-TE2) and tocopherol cyclase (TC/VTE1) were constitutively overexpressed in lettuce (Lactuca sativa L.). Total tocopherol content of the transgenic plants overexpressing either of the genes was increased by more than 2-fold mainly due to an increase in ${\gamma}$-tocopherol. However, chlorophyll content in the HPT/VTE2 and TC/VTE1 transgenic lines decreased by up to 20% and increased by up to 35%, respectively (P < 0.01). These results demonstrate that manipulation of the tocopherol biosynthetic pathway can increase or decrease chlorophyll content depending on the gene introduced.

Plant Growth-Promoting Rhizobacteria Improved Salinity Tolerance of Lactuca sativa and Raphanus sativus

  • Salinity stress is an important environmental problem that adversely affects crop production by reducing plant growth. The impacts of rhizobacterial strains to alleviate salinity stress on the germination of Lactuca sativa and Raphanus sativus seeds were assessed using different concentrations of NaCl. Plant growth-promoting rhizobacteria (PGPR) strains were also examined to improve the early germination of Chinese cabbage seeds under normal conditions. Lactobacillus sp. and P. putida inoculation showed higher radicle lengths compared with non-inoculated radish (Raphanus sativus) seeds. LAP mix inoculation increased the radicle length of lettuce (Lactuca sativa) seedlings by 2.0 and 0.5 cm at salinity stress of 50 and 100 mM NaCl concentration, respectively. Inoculation by Azotobacter chroococcum significantly increased the plumule and radicle lengths of germinated seeds compared with non-inoculated control. A. chroococcum increased the radicle length relative to the uninoculated seeds by 4.0, 1.0, and 1.5 cm at 50, 100, and 150 mM NaCl concentration, respectively. LAP mix inoculation significantly improved the radicle length in germinated radish seeds by 7.5, 1.3, 1.2, and 0.6 cm under salinity stress of 50, 100, 150, and 200 mM NaCl concentration, respectively. These results of this study showed that PGPR could be helpful to mitigate the salinity stress of different plants at the time of germination.

드론 초분광 스펙트럼과 분광각매퍼를 적용한 생태계교란식물 탐지 (Detection of Ecosystem Distribution Plants using Drone Hyperspectral Spectrum and Spectral Angle Mapper)

  • Ecological disturbance plants distributed throughout the country are causing a lot of damage to us directly or indirectly in terms of ecology, economy and health. These plants are not easy to manage and remove because they have a strong fertility, and it is very difficult to express them quantitatively. In this study, drone hyperspectral sensor data and Field spectroradiometer were acquired around the experimental area. In order to secure the quality accuracy of the drone hyperspectral image, GPS survey was performed, and a location accuracy of about 17cm was secured. Spectroscopic libraries were constructed for 7 kinds of plants in the experimental area using a Field spectroradiometer, and drone hyperspectral sensors were acquired in August and October, respectively. Spectral data for each plant were calculated from the acquired hyperspectral data, and spectral angles of 0.08 to 0.36 were derived. In most cases, good values of less than 0.5 were obtained, and Ambrosia trifida and Lactuca scariola, which are common in the experimental area, were extracted. As a result, it was found that about 29.6% of Ambrosia trifida and 31.5% of Lactuca scariola spread in October than in August. In the future, it is expected that better results can be obtained for the detection of ecosystem distribution plants if standardized indicators are calculated by constructing a precise spectral angle standard library based on more data.

음식물찌꺼기 퇴비의 시용에 따른 토양의 화학성 변화 밑 작물체내 염류의 흡수 (Changes of Soil Chemical Properties and Uptake of Salts by the Plants according to the Application of the Food Waste Compost)

  • 이 연구는 대규모의 pilot plant에서 생산된 음식물찌꺼기 퇴비의 시용에 따른 작물(열무, 상추)에 의한 염류의 흡수와 토양의 화학적 특성 변화를 조사하기 위하여 수행되었다. 일반적으로 음식물찌꺼기 퇴비(FWC)는 전기전도도(EC)가 높고 Na, K, Ca 그리고 Mg와 같은 많은 염류를 함유하고 있다. 이러한 FWC가 토양에 시용될 경우 토양의 염류집적과 작물의 생육저해 가능성을 검증하지 않으면 안된다. FWC를 건물중으로 1/5,000a pot에 0, 20, 40, 80 그리고 160Mg/ha의 비율로 시용한 후 60일 동안 작물을 재배하여 토양의 pH와 EC, 작물에 의한 염류의 흡수를 조사하여 얻어진 결과를 요약하면 다음과 같다. 1. 열무와 상추를 재배한 후 토양의 pH는 증가하였으며, EC는 감소하였다. 2. Na와 K의 흡수량은 FWC의 시용에 의해 약간 증가하였다. Ca의 흡수량에 있어서, 열무와 상추는 각각 감소, 증가하였다. 열무에서 Mg의 흡수량은 40Mg/ha 시용했을 때 가장 높았으며, 상추에서는 FWC의 시용에 따라 지속적으로 증가하였다.

왕고들빼기 어린잎과 성체의 수확 후 특성과 온도별 저장성 비교 (Comparison of Post-Harvest Character and Storability at Several Temperature for Lactuca indica L. Baby and Adult Leaves)

  • 왕고들빼기(선향 품종)을 4주 간격으로 파종하여, 4주 동안 재배하여 어린잎(초장 10cm 이하)과 8주 동안 재배하여 성체(초장이 $20{\pm}5cm$) 상태에서 수확하였다. 수확직후 조사한 호흡률과 에틸렌 발생률은 모두 어린잎이 성체보다 높았으며, DPPH 라디컬 소거능도 성체에 비해 어린잎에서 높았다. 잎의 $L^*$, $a^*$, $b^*$값은 어린잎과 성체가 유의적인 차이를 보이지 않았고 엽록소 함량은 성체에서 높았다. 왕고들빼기 성체와 어린잎을 미세천공 필름으로 포장하여 $2^{\circ}C$, $8^{\circ}C$, 및 $20^{\circ}C$에서 저장하였을 때, 저장온도가 높을수록 생체중 감소율이 높았으며, $2^{\circ}C$$8^{\circ}C$에서는 성체의 생체중 감소율이 낮았다. 저장 중 패널테스트를 통해 진행한 외관상 품질도 저장온도가 높을수록 빠르게 나빠졌다. 또한 외관상 품질이 3점을 유지되었던 기간으로 계산한 저장수명은 성체가 $2^{\circ}C$에서는 1.6일, $8^{\circ}C$에서는 1.4일, 그리고 $20^{\circ}C$에서는 1.5일 어린잎보다 더 길었다. 저장 종료일 조사한 포장 내 산소 및 이산화탄소 농도는 대기 중 농도와 유사한 수준이었다. 저장 종료일의 엽록소 함량은 저장온도가 낮을수록 높게 유지되었고, 패널테스트로 조사한 이취는 어린잎에서 더 높은 수준이었다.

The Liver Protective Activities of Some Iranian Medicinal Plants Against Liver Damage in Mice Induced by $CCL_4$ Intoxication

  • The aim and objective of this study are to carry out the liver protective activities against the $CCl_4$ intoxication in mice with some Iranian medicinal plants traditionally used for liver injuries. The methanol extracts of Cichorium intybus, Lactuca scarJoia, Eucalyptus camadulensis were evaluated. With various doses of these plants, liver protective activities were performed after $CCl_4$ administration to mice. The serum aminotransferases activites, liver sizes, and histopatological examinations of liver were studied. At a dose of 50 mg/ kg, all three plants were able to protect liver damages induced by $CCl_4$.

식물성 생약의 도파민 베타 수산화효소에 대한 저해활성 (Dopamine ${\beta}-Hydroxylase$ Inhibitory Activity of Medicinal Plants)

  • Dopamine ${\beta}-hydroxylase$ (DBH) catalyses the enzymatic reaction of dopamine to norepinephrine. For the purpose of isolating DBH inhibitors from natural resources, thirty one kinds of medicinal plants were screened by tracing the inhibitory activities against bovine adrenal DBH, utilizing tyramine as a substrate. Among the crude drugs tested, leaves of Lactuca sativa L., Gardeniae Fructus, Magnoliae Flos and Scutellariae Radix showed potent enzyme inhibitory activities against DBH.

First Report of Sclerotinia Rot Caused by Sclerotinia sclerotiorum on Some Vegetable Crops in Korea

  • Sclerotinia rot occurred severely on some vegetable crops grown in Namyangju, Yangpyung, and Yangiu areas in Korea in 2001-2002. The crops infected with Scterotinia sp. were Adenophora remotiflora, Armoracia lapathfolia, Angelica acutiloba, Angelica archangelica, Anthriscus sylvestris, Aster tataricus, Beta vulgaris var. cicla, Brassica campestris var. marinosa, Brassica juncea var. laciniata, Chicholium intybus, Lactuca indica var. dracoglossa, Lactuca sativa var. oak-leaf, Petroselinum crispum, and Phyteuma japonicum. The fungus associated with the disease was identified as Sclerotinia sclerotiorum, based on the morphological characteristics of the pathogen. The symptoms were water-soaked spots that enlarged later and became a watery soft rot. Infected parts became yellow and then turned brown, followed by death of the whole plant. White mycelia developed on the upper petioles and leaves and on the soil where these plant parts lay. Then black sclerotia in variable size and shape formed from the mycelial mass. Pathogenicity of the fungus was proven by artificially inoculating each crop. This is the first report of Sclerotinia rot on the listed vegetable crops in Korea.

Antioxidant Potential and Chlorogenic Acid Level of Lettuce (Lactuca sativa L.) Cultivars

  • Lettuce (Lactuca sativa L.) is known to contain water-soluble substances that improve antioxidant status due to the richness in antioxidants. Greenhouse experiment was carried out under different shading conditions during spring lettuce growing season. Shade significantly reduced shoot weight, number of leaves and chlorophyll content, while it increased shoot length of lettuce plants. The antioxidant potential of the individual fraction was in order of n-butanol > ethyl acetate > water > n-hexane fraction, although was less than that of commonly used antioxidants, BHT and ascorbic acid. Fractions from lettuce plants dose-dependently increased DPPH free radical scavenging activity, in vitro test. By means of HPLC analysis, BuOH fraction of cultivar 'Hwahyang' (57.93 mg $100g^{-1}$) had the highest amount of antioxidant chi orogenic acid. Shading treatment increased average amount of chlorogenic acid of all cultivars in BuoH, EtOAc, hexane and water fractions by 33, 120, 144, and $58\%$, respectively. These results suggest that lettuce plants had potent antioxidant activity, and their activities were differently exhibited depending on cultivar and fraction.
