Production of Essential Oils by Tissue Culture of Schizonepeta tenuifolia (형개의 조직배양에 의한 정유생산)

  • The callus was induced from the seedlings of Schizonepeta tenuifolia Brig. (Labiatae) and the effects of culturing conditions on growth rate and essential oil formation of the callus were experimented. It was found in the experiments, that the proper culturing temperature is $23^{\circ}C$ and the addition of biosynthetic precursors(leucine, mevalonic acid lactone) inhibits the growth of the callus. The growth rate of the callus and the amount of essential oils of the callus in the medium containing NAA were higher than the medium containing 2,4-D. The essential oils from the callus and the leaves of the cultivated Schizonepeta tenuifolia showed different GC pattern, but pulegone was found in both oils.

  • The volatile components of the barks of Eucommia ulmoides Oliver were extracts simutaneous steam distillation-extraction apparatus, and analyzed by combined GC and GC-MS. Forty nine componets, including 4 acids, 11 alcohols, 9 aldehydes, 4 esters, 3 ketones, 16 hydrocarbons, 1 lactone were confirmed in Eucommiae cortexs. Among total volatiles the most component was caproic acid comprising about 18.1%.

  • The phytochemical study of the aerial parts of Artemisia sylvatica (Asteraceae) led to the isolation of nine sesquiterpene lactones, l$\alpha$-hydroperoxy-rupicolin A acetate (1), rupicolin B acetate (2), rupicolin A acetate (3), l$\alpha$-hydroxy-4$\alpha$-hydroperoxy-bishopsolicepolide (4), 1$\alpha$-hydroperoxy-4$\beta$-hydroxy-8$\alpha$-acetoxy-guaia-2,9,11 (13)-triene-6$\alpha$,12-oxide (5), 1$\alpha$-hydroperoxy-4$\alpha$-hydroxybishopsolicepolide (6), l$\alpha$,4$\beta$-dihydroxy-8$\alpha$-acetoxy-guaia-2,9,11(13)-triene-6$\alpha$,12-olive (7), rupicolin A (8) and l$\alpha$,4$\alpha$-dihydroxy-bishopsolicepolide (9). Their structures were established by chemical and spectroscopic methods.

  • 4-(p-Chlorophenyl)tetralone (6) and 7-chloro-5-(p-chlorophenyl)tetralone (9) are key intermediates for the development of benzazepinone derivative haftens. These compounds could be synthesized from 4-phenyltetralone derivatives by triflic acid catalyzed Friedel-Crafts reaction. The reaction mechanism of Friedel-Crafts alkylation/acylation with lactones in triflic acid is presented. According to our tentative research, ring opening of protonated lactone (2) occurs in alkyl cleavage and the rate of the reaction is not dependent on concentration of triflic acid. So, alkylation of lactone in Friedel-Crafts reaction is presumed to be $A_{AL}$ 1. In second step, intramolecular acylation of the intermediates 4 to 6, 4 can be transformed to a triflic acid-carboxylic anhydride and then the cyclization is undergone after leaving of the triflate anion.

  • The inhibitory effect of Inula sesquiterpene lactones on the growth of Phaseolus vulgaris was tested and the abnormality of the stem organization caused by the lactones was also examined. The longitudinal growth of the young stem and the expansion of the young leaf were stopped by the application of the lactones. However, this inhibitory effect was appeared and strictly restricted within the treated area. So the young shoot was observed for possible bending as a result of the unilateral application of the lactones. When the application of the lactones into the medium, the growth of the plant was entirely repressed. However, the growth of shoot and re-initiation of root were started after the plant was transfered to the lactone free medium. And partial reversal of inhibition of the stem growth was achieved by the additions of gibberelline and the lactones.

  • The characteristic aroma of Gardenia Absolute Oil made from Gardenia Jasminoides Ellis was investigated by GC and GC-MSD and by sensory evaluation The method of GC Analysis was used Double Column System by Carbowax 20M /SPB-1 of Pola/nonPola columns. And the result data of analysis was checked automatic Relent ion Indexs Match System by GC Workstation. A total of 51 compounds were identified in Gardenia Absolute Oil, including t entatively erstimated 9 compounds. The major components were Linalool, Farnesene of various form, Jasmin Lactone, Gamma undecalactone and Cis-3-hexone derivatives.

  • Soy curd was prepared using imported soybean(U.S.grade A) and changes in quality were evaluated using different types and mixtures of coagulants(CaCl$_2$, CaSO$_4$, MgCl$_2$, MgSO$_4$, and glucono-$\delta$-lactone). Quality of soy curd was determined by rheological properties, yield, and color. Based upon the characteristics of each coagulant, mixtures of two coagulants were prepared in various ratios. The mixture of CaSO$_4$or MgCl$_2$with glucono-$\delta$-lactone(GDL), and that of CaSO$_4$or MgCl$_2$with MgSO$_4$resulted in soy curd of good quality in terms of rheological properties and yield.

  • (3S,1'S)-3-(1'-Carboxy-2'-phenyl)ethylamino-2-oxetanone (1a) and (3R,1'S)-3-(1'-carboxy-2'-phenyl)ethylamino-2-oxetanone (1b) were designed, synthesized, and evaluated as inhibitors for carboxypeptidase A, a prototypical zinc protease that removes the C-terminal amino acid having an aromatic side chain from oligopeptide substrate. It was concluded from the analysis of inhibition kinetics that while 1a inactivates CPA irreversibly, its diastereoisomer, 1b is a weak competitive inhibitor for CPA. A possible explanation for the observed difference in inhibition mode that is dependent on the inhibitor stereochemistry is offered.
