Identification of Lactobacillus spp. associated with nematodes in peach farm soil (복숭아 농장 토양에서 Nematodes와 연관된 Lactobacillus spp.의 분리 및 동정)

  • Strains D4 and D5 were isolated from peach-rotten soil during the peach harvest season. The isolates were identified based on morphological and biochemical characterization, and identification was determined by 16S rRNA gene sequencing. Results showed that D4 has high similarity to Lactobacillus plantarum ATCC $14917^T$ and Lactobacillus pentosus ATCC $8041^T$ at 99.05% and 98.98%, respectively. D5 was also similar to Lactobacillus pentosus ATCC $8041^T$ and Lactobacillus plantarum ATCC $14917^T$ at 98.71% and 98.64%, respectively. In contrast, isolates showed differences in carbohydrate utilization in comparison to Lactobacillus plantarum ATCC $14917^T$ and Lactobacillus pentosus ATCC $8041^T$. In view of this we performed VITEK MS matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) analysis, multiplex PCR fingerprinting, and random amplified polymorphic DNA (RAPD)-PCR to further confirm the identification of D4 and D5. The results of these analyses showed that both strains were most similar to Lactobacillus plantarum.

Characterization of Lactobacillus cellobiosus D37 Isolated from Soybean Paste as a Probiotic with Anti-Cancer and Antimicrobial Properties

  • The probiotic characteristics of a total of 137 lactic acid bacterial strains isolated from soybean paste were investigated. Among those tested, the D37 strain was selected as a probiotic bacteria due to its acid and bile tolerance, and its strong anti-cancer and antibacterial activities. The D37 strain showed highly stable viability at acidic pH for 2 hr, and was very stable in 10% bovine bile. The viability of human colon cancer HT-29 cells was inhibited more than 60% at a $200\;{\mu}/mL$ concentration of D37 cell-free culture supernatant, and the degree of inhibition was concentration-dependent. The D37 strain showed a wide range of antibacterial activities against food-borne pathogenic bacteria such as Escherichia coli O157, Listeria spp., Vibrio spp., Salmonella spp., and Staphylococcus aureus. According to phenotypic characteristics and the utilization of various sugars, the D37 strain was identified as Lactobacillus cellobiosus.

Antibacterial Activities of Fermented Sayuksan Ingredient Extracts for Multidrug-resistant Strains (한약재발효액의 항생제 다제내성균에 대한 항균활성 및 항산화활성)

  • Sayuksan has been widely applied to treat a variety of diseases such as acute hepatitis, gastritis, and colitis. Sayuksan consists of medicinal herbs such as Glycrrhizae uralensis Fischer, Paeonia lactiflora Pallas, Bupleurum falcatum Linne, and Poncirus trifoliata Rafinesqul. Methanol extracts (1 mg/mL) from the four kinds of medicinal herbs did not show antibiotic activities against general test strains and multi-drug resistant strains. The antibacterial activity of fermented medicinal herbs extracts with Lactobacillus spp. strain was confirmed as Gram-positive bacteria which are higher than Gram-negative bacteria. Extracts of Glycrrhizae uralensis Fischer fermented with Lb. casei KCTC 3109 displayed inhibitory diameters of 16 mm against Pseudomonas aeruginosa P01828. Superoxide dismutase (SOD)-like activity of the medicinal herb extracts was not determined, but the extract of Paeonia lactiflora Pallas fermented with six strains of Lactobacillus spp. had the highest antioxidant activity. SOD-like activity of Paeonia lactiflora Pallas extracts fermented by Lb. brevis KCTC 3498 was $41.4{\pm}0.8%$, which was the highest antioxidant activity among the fermented extracts with the other medicinal herbs.

Characterization and Identification of Lactic Acid Bacteria Isolated from Fermented Milks in Iran (이란 발효 유제품에서 분리한 유산균의 특성)

  • This study aimed to identify lactic acid bacteria isolated from eight fermented milk products in Iran. We enumerated Lactobacillus species using De Man-Rogosa-Sharpe (MRS)-maltose and MRS agar with pH adjusted to 5.2, as well as assessment at 37℃ for 48 hr, studied Streptococcus spp. using M17 agar at 43℃ for 24 hr, and assessed Bifidobacterium species using nalidixic acid, paromomycin sulfate, neomycin sulfate, and lithium chloride (BL-NPNL) agar at 37℃ for 48 hr. The total viable Streptococcus spp. cell in fermented milk varied at 4.73-8.83 log CFU/mL. However, Bifidobacterium spp. were not detected in any of the tested samples. Lactobacilli were not detected in four of the eight samples, and viable Lactobacilli cells in the remaining four samples ranged 2.48-3.85 log CFU/mL. The pH of the tested samples ranged 3.53-4.19, and soluble solids (Brix measurement) ranged 7.5%-17.9%. A total of 130 isolates of gram-positive catalase-positive bacteria were characterized at the species level using 16S rRNA sequencing. Sequence analysis identified six species: Streptococcus thermophilus, Lactobacillus delbrueckii subsp. sunkii, Lactobacillus delbrueckii subsp. indicus, Lactiplantibacillus plantarum, Lacticaseibacillus rhamnosus, and Levilactobacillus brevis.

  • Lactobacillus spp. PAP1 과 Lactobacillus spp. MG-G2를 혼합구조로 사용하여 밤 발효제품을 제조하기 위한 제반 조건을 조사하였다. 밤 발효액은 phytone peptone, yeast exract, glucose를 각각 0.4, 0.2. 0.5%이상으로 첨가할 때 산도와 생균수가 가장 컸다. 첨가물 중 phytone peptone은 대두박이나 순물로 대체해도 같은 효과가 관찰되었다. 버섯추출물의 첨가는 산도와 생육을 약간 촉진하였으나 안정제는 약간의 감소를 가져왔다. 기본 발효액은 관능성적이 나빴으나 산도를 0.54%로 낮추고, 당을 10%로 보강하고, so-dium alginate를 0.2% 첨가하였을 때, 산미, 감미, 음용감이 크게 개선되었다

Studies on the Bacteriological Properties of Loin Ham during Curing Period (염지경과에 따른 Loin Ham의 세균학적 특성에 관한 연구)

  • In order to examine related bacteria on the maturation of cured loin ham, bacteria isolated during curing periods from ham, which counted coliform group, psychrotrophic and halo-tolerant bacteria. The results are as follows; The isolated bacteria from ham during the curing period were Staphylococcus spp. 24 strains, Bacillus spp. 21 strains, Lactobacillus spp. 10 strains, Coryneform 2 strains. Microbacterium spp. 2 strains and Gram negative rods 8 strains. Micrococcus spp. were identified M. varians 12 strains and M. luteus 3 strains, and Streptococcus spp. identified S. faecium 14 strains, S. lactis 2 strains. Lactobacillus spp. were isolated L. Plantarum 4 strains, L. brevis and L. casei 1 strains. In the case of cured ham, the number of coliform group and psychrotrophic bacteria were decreased but halo-tolerant bacteria were increased for 10 days of curing period. On the brine solution. the number of coliform group, psychrotrophic and halo-tolerant bacteria were increased for 10 days. 4 days and 20 days, respectively.

Analysis of Microbial Diversity in Nuruk Using PCR-DGGE (PCR-DGGE를 이용한 누룩에서의 미생물 다양성 분석)

  • Nuruk plays a significant role in the flavor and quality of Takju and Yakju, which are produced through saccharification and alcohol fermentation by various microorganisms. In this study, we identified microbial strains isolated from a plate count and PCR-denaturing gradient gel electrophoresis (DGGE) analysis targeting the 16S and 28S rRNA genes, in order to characterize bacterial and fungal diversity in Sansung Nuruk. The numbers of bacteria and fungi in Nuruk were $1.5{\times}10^9$ CFU/g and $2.2{\tims}10^8$ CFU/g, respectively. The 16S rRNA gene sequence indicated that the predominant bacteria in the isolates and PCR-DGGE profile of Nuruk were Kocuria spp., Pantoea spp., Lactobacillus spp., Pediococcus spp., Weissella spp., Staphylococcus spp., endophytic bacterium, uncultured Gamma-proteobacteria, uncultured Cyanobacteria, and Actinobacteria. Dominant bacteria from the PCR-DGGE profile were Pediococcous pentosaceus and uncultured Cyanobacteria. The 28S rRNA gene sequence indicated the predominant fungi in the isolates and PCR-DGGE profile to be Trichomonascus spp. Pichia spp., Torulaspora spp., Wickerhamomyces spp., Sacharomycopsis spp., Lichtheimia spp., Mucor spp., Rhizopus spp. Aspergillus spp., and Cladosporium spp. Dominant fungi from the PCR-DGGE profile were Pichia kudriavzevii and Aspergillus oryzae. The PCR-DGGE technique was used for the first time in this study to assess a microbial community in Nuruk and proved to be an effective protocol for profiling microbial diversity.

Probiotic Properties of Lactobacillus spp. Isolated from Gajami Sikhae (가자미식해에서 분리한 유산균의 프로바이오틱스 특성)

  • Forty species of lactic acid bacteria isolated from gajami sikhae were identified as Lactobacillus plantarum, Leuconostoc mesenteroides, Lactobacillus brevis, and Weissella paramesenteroides. 10 of the 40 strains were selected and used for the test. In this study, experiments such as those using acid and artificial gastric juice resistance, bile acid resistance, autoaggregation, coaggregation, and cell surface hydrophobicity were conducted to utilize lactic acid bacteria separated from gajami sikhae as probiotics. The separated lactic acid strains showed high survival rates through displaying resistance to acidic and artificial gastric juices; L. plantarum GS11 showed the best resistance. Also, as a result of a measurement of bile acid resistance, all lactic acid bacteria stocks showed survival of more than 100% with a probiotic number of 108 to 109 log CFU/ml. After evaluating cohesion to indirectly measure cell surface adhesion, autoaggregation ability was shown to be more than 46%. Measurement of xylene adhesion for cell surface hydrophobicity evaluation revealed better cell adhesion than B. subtilis, which has 32.2% hydrophobicity in isolated lactic acid strains. Antibacterial force measurement found antibacterial activity in lactic acid bacteria, excluding L. plantarum GS12 and L. plantarum GS13. Therefore, it was judged that lactic acid bacteria separated from gajami sikhae could be used as probiotics with various probiotic properties.

Comparison of Fermentation Properties of Winter Kimchi Stored for 6 Months in a Kimchi Refrigerator Under Ripening Mode or Storage Mode (김치냉장고의 숙성 후 저장 및 저온 저장 모드에서 6개월간 저장한 김장 김치의 발효특성 비교)

  • The purpose of this study is to determine long-term storage conditions for winter kimchi. Kimchi was stored in a kimchi refrigerator for 6 months with or without fermentation. Four different temperature systems used were as follows: 5 days at $10^{\circ}C$ followed by storage at $-2.5^{\circ}C$ (F1), 1 day at $15^{\circ}C$ followed by storage at $-2.5^{\circ}C$ (F2), storage at $-1^{\circ}C$ (S1), or at $-2.5^{\circ}C$ (S2). Time periods required for F1, F2, S1, or S2 kimchi to reach pH 4.4 and acidity 0.6% were 2, 8, 12, and 22 weeks, respectively. Lactobacillus spp. growth on F1 and F2 kimchi was faster and greater than that on S1 and S2 kimchi, revealing a maximum concentration of 8~9 verses 6.8 log CFU/mL, respectively. However, Leuconostoc spp. were fully grown (8~9 log CFU/mL) on all four kimchi samples regardless of temperature, even at $-2.5^{\circ}C$, although the times required to reach maximum growth were different. Growth of Lactobacillus and Leuconostoc spp. both decreased after reaching maximum levels, except for F1 kimchi. Sensory evaluation results for 3 month storage showed that F1 kimchi was the best among kimchi samples in terms of appearance, acidic taste, carbonated taste, crispiness, and moldy smell. For 6 months of storage, F1 and S1 kimchi were the most highly evaluated among the kimchi samples. Sensory evaluation result for S1 kimchi stored at $-1^{\circ}C$ was comparable to that of F1 kimchi due to fully grown Leuconostoc spp. Acidities of F1 and S1 kimchi after 6 months of storage were 0.8 and 0.7%, respectively. Taken together, fermentation of kimchi at $10^{\circ}C$ for 5 days followed by storage at $-2.5^{\circ}C$ for 6 months was optimal for high quality kimchi. Sensory properties of winter kimchi were significantly influenced by the degree of fermentation.

Functionality Analysis of Rhus javanica Fermented by Lactobacillus spp. (Lactobacillus spp. 이용 발효 붉나무의 기능성물질 검색에 대한 연구)

  • To determine the potential function of Rhus javanica in Korean medicine, it was fermented with each strain of Lactobacillus spp. Each strain of Lactobacillus spp. was inoculated in lactobacilli MRS broth, and 5 mg/ml of methanol extract of Rhus javanica was added. In mouse hippocampal HT22 cells, ethyl acetate extract of R. javanica fermented with L. brevis KCTC 3498 induced heme oxygenase-1 expression and showed a significant cytoprotective effect on glutamate-induced oxidative damage. The cytoprotective effect was related to the transcription of the nuclear factor E2-related factor2 (Nrf2), which is responsible for the induction of heme oxygenase-1 within the nucleus. The antimicrobial, antioxidant, and heme oxygenase-1 expression activities of fermented R. javanica were measured after extraction with ethyl acetate. R. javanica fermented with L. plantarum subsp. plantarum KCTC 3108, L. fermentum KCTC 3112, and L. brevis KCTC 3498 had higher antioxidant activity than nonfermented R. javanica. The fermented R. javanica with L. plantarum subsp. plantarum KCTC 3108, L. casei KCTC 3109 after ethyl acetate extraction showed antibacterial activity against Bacillus subtilis PCI 219, Escherichia coli KCTC 1682, Shigella flexneri KCTC 2517, Vibrio parahaemolyticus KCTC 7471, and Pseudomonas aeruginosa KCTC 2004. An ethyl acetate extract of the fermented R. javanica with Lactobacillus brevis KCTC 3498 exhibited stronger antibacterial activity than a nonfermented one against strains of B. subtilis PCI 219, E. coli KCTC 1682, S. flexneri KCTC 2517, and V. parahaemolyticus KCTC 7471.
