Physicochemical and microbial characteristics of domestic commercial semi solid type yogurt

  • Yogurt is a food produced by bacterial fermentation of milk and the bacteria used to make it are known as "yogurt cultures". Most of them belong to probiotics such as Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus thermophilus bacteria. Domestic fermented milk market is increasing and about 30 companies are producing yogurt. The purpose of this study was to analyze the quality characteristics of domestic commercial semisolid type yogurt. We collected 20 types of commercial yogurt at local markets. Physicochemical properties including pH, sugar content, acidity, viscosity and microbial characteristics of lactic acid bacteria counts were measured. The yogurt showed pH 4.5, 7.4~18.1% of sugar contents, 0.6~1.3% of total acids and 282~748 cP of viscosities. In the microorganism populations, lactic acid bacteria count were 6.5~11.5 Log CFU/mL and anaerobic lactic acid bacteria count were 7.2 ~ 11.1 Log CFU/mL. The quality characteristics were different depending on the constituents of the sample and the microorganisms used. These results are related to the quality characteristics of yogurts which are useful information about identifying new trends in domestic fermented milk industry.

블루베리 유산균 발효물을 첨가한 양갱의 제조 및 품질특성 (Quality Characteristics of Yanggaeng Prepared with Fermented Blueberry by Lactic Acid Bacteria)

  • Blueberry juice extraction (JE), hot water extract (HE), and 50% ethanol extract (EE) were fermented with Lactobacillus plantarum CGKW3. We investigated the quality characteristics and antioxidative activities of yanggaeng prepared with different amounts of fermented blueberry extract (JE, HE, EE). The viable cells in fermented HE was higher (7.49 log CFU/mL) than JE (7.28 log CFU/mL) and EE (6.99 log CFU/mL), during the fermentation period. The viable cells and acidity in yanggaeng increased significantly with increasing levels of fermented blueberry extract (p<0.05). In terms of color, the lightness and yellowness decreased significantly, but redness increased with increasing levels of fermented blueberry extract. In the texture profile analysis, control showed the highest result in hardness. Cohesiveness did not show significant differences, according to amount of fermented blueberry extract. The springiness decreased with the increasing levels of fermented blueberry extract. Antioxidant activity, which was measured by DPPH and reducing power, was significantly higher than those of control; and it increased proportionally according to the amount of fermented blueberry extract. Anthocyanin contents were increased proportionally with the increasing levels of fermented blueberry extract. Sensory evaluation showed that the color, taste, flavor, texture, and overall acceptability of yanggaeng containing the JE, HE, and EE were higher than those of the control.

Fermentation Characteristics and Lactic Acid Bacteria Succession of Total Mixed Ration Silages Formulated with Peach Pomace

  • The objective of this study was to assess the use of peach pomace in total mixed ration (TMR) silages and clarify the differences in aerobic stability between TMR and TMR silages caused by lactic acid bacteria (LAB). The TMR were prepared using peach pomace, alfalfa hay or Leymus chinensis hay, maize meal, soybean meal, cotton meal, limestone, a vitamin-mineral supplement, and salt in a ratio of 6.0:34.0:44.4:7.0:5.0:2.5:1.0:0.1 on a dry matter (DM) basis. Fermentation quality, microbial composition, and the predominant LAB were examined during ensiling and aerobic deterioration. The results indicated that the TMR silages with peach pomace were well fermented, with low pH and high lactic acid concentrations. The aerobic stability of TMR silages were significantly higher than that of TMR. Compared with TMR silages with alfalfa hay, TMR silage with Leymus chinensis hay was much more prone to deterioration. Although the dominant LAB were not identical in TMR, the same dominant species, Lactobacillus buchneri and Pediococcus acidilactici, were found in both types of TMR silages after 56 d of ensiling, and they may play an important role in the aerobic stability of TMR silages.

Bacterial Inoculant Effects on Corn Silage Fermentation and Nutrient Composition

  • The survival and effect of three new probiotic inoculants (Lactobacillus plantarum CCM 4000, L. fermentum LF2, and Enterococcus faecium CCM 4231) on the nutritive value and fermentation parameters of corn silage was studied under laboratory conditions. Whole corn plants (288.3 g/kg DM) were cut and ensiled at $21^{\circ}C$ for 105 days. The inoculants were applied at a concentration of $1.0{\times}10^{9}$ cfu/ml. Uninoculated silage was used as the control. The chopped corn was ensiled in 40 plastic jars (1 L) divided into four groups (4${\times}$10 per treatment). All corn silages had a low pH (below 3.55) and 83-85% of total silage acids comprised lactic acid after 105 days of ensiling. The probiotic inoculants in the corn silages affected corn silage characteristics in terms of significantly (p<0.05-0.001) higher pH, numerically lower crude protein content and ratio of lactic to acetic acid compared to control silage. However, the inoculants did not affect the concentration of total silage acids (acetic, propionic, lactic acids) as well as dry matter digestibility (IVDMD) of corn silages in vitro. In the corn silages with three probiotic inoculants, significantly (CCM 4231, CCM 4000) lower n-6/n-3 ratio of fatty acids was detected than in control silage. Significant decrease in the concentration of $C_{18:1}$, and significant increase in the concentration of $C_{18:2}$ and $C_{18:3}$ was mainly found in the corn silages inoculated with the strains E. faecium CCM 4231 and L. plantarum CCM 4000. At the end of ensiling, the inoculants were found at counts of less than 1.0 log10 cfu/g in corn silages.

산양유를 섭취한 성인 여성의 분변에서 분리한 Bifidobacteria와 Lactobacilli의 탄수화물 발효특성 (Carbohydrate Fermentation Character of Bifidobacteria and Lactobacilli isolated from Feces of the Adult Women supplied with Goat Milk)

  • 산양유을 섭취한 성인의 분변 내 bifidobacteria의 증식 효과 및 분변에서 분리된 bifidobacteria와 lactobacilli의 당 발효 능력을 조사하였다. 1. 산양유를 섭취한 처리군이 대조군보다 bifidobacteria 세균수가 상대적으로 높았으며, 8주에서 95% 신뢰한계에서 유의성이 있었다. 2. 분변에서 분리한 균주들을 13 균주의 16S rRNA 염기서 열로 동정한 결과, 산양유를 섭취한 처리구는 B. adolescentis, B. longum, B. pseudocatenulatum, B. dentium, L. sakei 순으로 분리되었다. 대조군의 12 균주는 B. adolescentis, B. longum, L. ruminis, L. sakei, B. pseudocatenulatum 순으로 분리되었다. 3. 산양유 올리고당과 lactulose는 모든 bifidobacteria 및 lactobacilli 균주가 발효하였다. Fructooligosaccaride는 B. adolescentis 7 균주 모두 발효하였다. 조사한 B. pseudocatenulatum 4 균주, L. sakei 3 균주, B. longum 7 균주 중에서 각각 3, 2, 1 균주가 fructooligosaccharide를 발효하였다.

김치 젖산균의 In Vitro에서 콜레스테롤 저하 효과 (In Vitro Evaluation of Cholesterol Reduction by Lactic Acid Bacteria Extracted from Kimchi)

  • 본 연구는 김치에 분포하는 것으로 알려져 있는 대표적인 젖산균에 관한 콜레스테롤 저하 효과에 관하여 연구하였다. Lactobacillus 1종, Leuconostoc 2종과 Weissella 2종을 대상으로 콜레스테롤 저하 효과를 측정하였다. 김치 초기 발효균인 Leu. citreum은 55.46% 콜레스테롤 감소율을 보였고, 대표적인 산패균인 Lac. plantarum은 55.91%, Leu. mesenteroides은 56.37%였으며 W. kimchii,, W. confusa 등에서도 55% 이상의 콜레스테롤 저하 효과가 나타났다. 김치의 발효 숙성 과정에 나타나는 젖산균들이 시험관 내에서 콜레스테롤 저하 효과가 확인되어, 향후 in vivo 조건하에서 콜레스테롤 저하에 어느 정도 영향을 미치는지 검토해볼 필요가 있다고 사료된다.

  • Bitid"bactcria를 이용한 요구르트 제조를 위하여 B. bitidwη의 우유내에서 최시 배양조간과 B. bitidum. L. acidophilus 및 Str. thermophihilus의 혼합배양시의 최적 조건을 조사하고 요구르트 제조에 적합한 조건을 조사하여 다음과 같은 결과을 얻었다. B. bitidum의 성장을 위한 탈지유아 농도는,15% 농도가 12%농도보다 산진전은 잘 되었으나 균의 생존력에서 12% 농도가 적합한 것으보 나타났다. B. bifidum 의 성장에 대한 L-cysteine.HCl, yeast extract, glucose와 BIOS 2000의 성장촉진물 침가효과는 BIOS 2000을 첨가한 것이 산 진전면과 균수 유지면에서 B. bhifidum익 성장에 큰 효과가 있었다. B. bifidum, L.acidophilus와 Str. thermophilus를 혼합하여 배양한 것이 산 진전과 균수면에서 B. bifidum 단독 배양한 것보다 좋게 나타났다. B. bifidum. L. acidophilus 와 SIr. thermophilus의 세 균주 혼합배양액을 이용한 요구르트 제조시 접송량은 5%. 배양온도는 $37^{\circ}C$와 배양시간은 8시간이 석당하였다 . 이 경우 세 균주의 비율은 B. bifidu: L. acidophilus. Str. thermophilus가 2:1:1의 비율이 균수유지와 산 진전면에서 가상 좋은 결과를 보였다.

김치의 보존성 증진을 위한 자초.감초의 혼합 첨가와 Chitosan 침지 효과 (Effect of Lithospermum erythrorhizon, Glycyrrhiza uralensis and Dipping of Chitosan on Shelf-life of Kimchi)

  • 한약재 혼합 효과와 chitosan 병용효과가 김치의 보존성에 미치는 영향을 조사하기 위하여 자초·감초 추출액 혼합 첨가(LG)와 절임 배추를 1% chitosan 용액에 침지 후 혼합 한약재를 첨가한 김치(LGDC)를 $10^{\circ}C$에서 25일간 발효시키면서 김치의 특성을 검토하였다. 김치 숙성 중 pH, 산도 총균수, 유산균수, 유산균 분포의 변화, 색택, 기호성을 대조구와 비교한 결과, LG군과 LGDC군은 대조구와 비교시, pH 감소속도는 완만하였고, 산도증가도 더 낮게 나타났으며, 총균수, 유산균수, Leuconostoc sp., L. plantarum 균수의 증식은 억제되었으며, LGDC군은 그 효과가 더욱 크게 나타났다. 특히 Leuconostoc sp., L. plantarum과 김치 유산균에 대해 초기부터 높은 억제능을 나타내었으며 이로써 chitosan에 대한 유산균 특이성에 대한 연구가 더욱 진행되어야 할 것으로 사료되었다. 김치의 관능검사 결과 신맛은 대조구에 비해 낮게 나타났으며 향, 종합적 기호도 등은 대조구와 뚜렷한 차이를 보이지 않아 자초, 감초 및 chitosan은 김치 숙성을 지연시키기 위한 첨가제로서 적당한 것으로 나타났다.

Fermentation Characteristics of Exopolysaccharide-Producing Lactic Acid Bacteria from Sourdough and Assessment of the Isolates for Industrial Potential

  • Lactic acid bacteria (LAB) with antimicrobial activity and high exopolysaccharide (EPS) production ability isolated from sourdough were studied for their fermentation characteristics as potential new starter cultures. The values of pH, titratable acidity, and viable cell counts were $4.06{\pm}0.009-4.50{\pm}0.015,\;0.787{\pm}0.020%-1.172{\pm}0.018%,\;and\;8.78{\pm}0.08-8.98{\pm}0.06$ log CFU/ml, respectively. In order to select probiotics with a high survival rate in the gut, isolates were tested to assess resistance against the artificial gastric acid and bile juice. Viable LAB counts were significantly (p<0.05) affected by the acidity. At pH 2.0, the total declines in the initial bacterial counts were 4.52$\pm$0.07 log for S. thermophilus St-Body-1, >7.98$\pm$0.03 log for E. flavescens DU-10, >7.95$\pm$0.05 log for E. faecium DU-12, and 3.15$\pm$0.06 log for L. amylovorus DU-21. Among the strains, L. amylovorus DU-21 was the only strain that had bile tolerance under simulated gastrointestinal conditions. In order to improve EPS production by L. amylovorus DU-21, the influence of carbon source was studied. When glucose was used as a carbon source, EPS production dramatically increased to 17.19$\pm$0.28 g/l (p<0.05). The maximum cell growth (10.012$\pm$>0.012 log CFU/ml) and EPS production (18.71$\pm$0.19 g/l) were achieved when 15 g/l of glucose was employed as the carbon source.

Hepatoprotective Effects of Potato Peptide against D-Galactosamine-induced Liver Injury in Rats

  • The effect of some peptides on hepatoprotection and cecal fermentation against D-galactosamine (GalN)-treated rats was studied. In acute hepatic injury tests, serum alanine aminotransferase (ALT), aspartate aminotranferase (AST), and lactic dehydrogenase (LDH) activities were remarkably increased after injection of GalN. However, potato and soybean peptides significantly decreased GalN-induced alterations of serum ALT and AST activities. Hepatic thiobarbituric acid-reactive substance (TBARS) concentration in GalN-treated groups fed potato and soybean peptides was significantly lower than that in GalN-treated control group. Hepatic glutathione level in the GalN-treated group fed potato peptide was significantly higher than that in GalN-treated control group. Furthermore, cecal Lactobacillus level in GalN-treated groups fed potato and soybean peptides was significantly higher than that in GalN-treated control group, and cecal short-chain fatty acid concentrations in GalN-treated group fed potato peptide were significantly higher than in GalN-treated control group. These results indicate that potato peptide may improve the cecal fermentation and prevent the GalN-induced liver damage in rats.
