Antimutagenic Activities of Cell Wall and Cytosol Fractions of Lactic Acid Bacteria Isolated from Kimchi

  • Cell wall (lactic acid bacteria-sonicated precipitate ; LAB-SP) and cytosoll(lactic acid bacteria-sonicated supernatant ; LAB-SS) fractions were prepared from kimchi fermenting lactic acid bacteria such as Leuconostoc mesenteroides, Lactobacillus brevis, Lactobacillus fermentum , Lactobacillus plantarum and Pediococcus acidilactici, with Lactobacillus acidophillus isolated from yogurt. Using the Ames mutagenicity test and SOS chormotest system, the antimutagenic acitivity of those cell fractions was studied . One hundered eighty $\mu$l of LAB-SP from lactic acid bacteria isolated from kimchi, excepting Pediococcus acidilactici, supressed the mutagenicity of 4-nitroquinoline-1-oxide(4-NQO) in Ames mutagenicity test and SOS chromotes system , by above 90% and 60% , respectively. LAB-SP from lactic acid bacteria also inhibited the mutagenicity mediated by 3-amino-1-methyl-5H-pyrido [4,3-b]indole (Trp-P-2). Lactobacillus fermentum, Lactobacillus plantarum, and Lactobacillus acidphillus had higher antimutagenicity against Trp-P-2). Lactobacillus fermentum , Lactobacillus plantarum , and Lactobacillus acidphillus had higher antimutagenicity against Trp-P-2 than the other lactic acid bacteria. However, LAB-SS of lactic acid bacteria did not show any mutagenic activity against 4-NQO in Ames mutagenicity test and SOS chromotest systems. On the mutagenicity of MEIQ and Trp-P-2 , LAB-SS of lactic acid bacteria from kimchi or dairy products exhibited a weaker inhibitory effect than LAB-SP of those bacteria. These results represent that, whether the lactic acid bacteria from kimchi are viable or nonviable, antimutagenic acitivity was still effective. We suggest that the strong, antimutaganic activity of lactic acid bacteria might be found in the cell wall fraction , rather than in the cytosol fraction.

Identification of Bacteriocin-producing Lactic Acid Bacteria from Kimchi and Partial Characterization of their Bacteriocin

  • Nineteen strains of bacteriocin-producing lactic acid bacteria were isolated from 432 Kimchi samples, and identified by the comprehensive biochemical and morphological tests verifying their cellular fatty acid composition. Using partially purified bacteriocins from these isolates, their inhibitory activities against other lactic acid bacteria and some pathogens, and sensitivity to enzyme and heat treatments were tested. The isolates were identified as Lactobacillus plantarum (2 strains), L curvatus (2 starins), L brevis (2 strains), Enterococcus faecium (6 strains), Leuconostoc mesenteroides subsp. mesenteroides (1 strain) and Lactobacillus sp. (6 strains). The bacteriocins produced by E. faecium strains provided the broadest spectrum of inhibition, affecting against other Gram-positive bacteria including lactic acid bacteria and health-threatening bacteria such as Clostridium perfringens and Listeria monocytogenes. The bacteriocins of Lactobacillus sp., L plantarum and L brevis strains were capable of inhibiting many strains of the lactic acid bacteria, whereas those of L curvatus and L mesenteroides subsp. mesenteroides strains were only inhibitory to a few strains. Generally, the inhibitory activities of both E. faecium and Lactobacillus sp. strains were greater than those of other producer strains. The bacteriocins from the isolates were sensitive to several proteolytic enzymes, and those of L curvatus and L mesenteroides subsp. mesenteroides were also sensitive to lipase and $\alpha$-amylase as well as to proteolytic enzymes. The bacteriocins from the strains of Lactobacillus sp. and a strain of L. brevis were resistant to autoclaving.

병원성 세균과 젖산균에 대한 마늘의 항균작용 (The Antibacterial Activity of Garlic Juice Against Pathogenic Bacteria and Lactic Acid Bacteria.)

  • This study was carried out to determine the inhibitory effect of garlic juice against Escherichia coli, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhimurium, Shigella flexneri. Staphylococcus aureus, Streptococcus mutans, Virio. parahaemolyticus which are food pathogenic bacteria and Lactobacillus acidophilus, Lactobacillus brevis, Lactobacillus casei, Lactobacillus plantarum, Lactococcus. lactis, Leuconostoc mesenteroides which are lactic acid bacteria. An aqueous extract of garlic was bacteriocidal against Gram-positive and Gram-negative bacteria in all concentrations (0.1∼2.5(w/v)%) tested in this experiment. Especially 0.5(w/v)% garlic juice inactivated completely E. coli, S. typhimurium, S. flexineri, V. parahaemolyticus and 1.0(w/v)% garlic juice perfectly reduced P. aeruginosa, S. mutans. Generally, the experiment result indicate that garlic juice restrains the growth of the pathogenic bacteria better than the lactic acid bacteria. Therefore, garlic has potential for the preservation of processed foods.

혐기적 조건하에서 젖산균의 cadmium 축적 (Accumulation of Cadmium in Lactic Acid Bacteria under the Anaerobic Conition)

  • In this study, authors investigated the cadmium tolerance, the accumulation of cadmium, and the cellular distribution of accumlated cadmium in lactic acid bacteria under the anaerobic condition. Lactic acid bacteria grew fairly well in modified EG medium containing 10 ppm of cadmium but could hardly grow at 50 ppm of cadmium. Tolerance to cadmium of genus Lactobacillus was greater than that of genus Streptococcus, and Lactobacillus acidophilus showed the higest cadmium tolerance amomg the bacteria tested. The capacity of cadmium accumlation (9.304-12.428 mg/g wet cell) of lactic acid bacteria was higher than that (6.775 mg/g wet cell) of Escherichia coli. Lactobacillus casei of them took up the largest amount of cadmium. The cadmium elimination amount (28.46-29.25%) of lactic acid bacteria from modified EG medium containing cadmium were also higher than that (14.43%) of Escherichia coli. Accumulated cadmium in Lactobacillus acidophilus was distributed by 42.41% at cell wall, 28.97% at cytoplasm, and 28.62% at plasma membrane, respectively.

Naturally Occurring Lactic Acid Bacteria Isolated from Tomato Pomace Silage

  • Silage making has become a significant method of forage conservation worldwide. To determine how tomato pomace (TP) may be used effectively as animal feed, it was ensilaged for 90 days and microbiology counts, fermentation characteristics and chemical composition of tomato pomace silage (TPS) were evaluated at the 30th, 60th, and 90th days, respectively. In addition, 103 lactic acid bacteria were isolated from TPS. Based on the phenotypic and chemotaxonomic characteristics, 16S rDNA sequence and carbohydrate fermentation tests, the isolates were identified as 17 species namely: Lactobacillus coryniformis subsp. torquens (0.97%), Lactobacillus pontis (0.97%), Lactobacillus hilgardii (0.97%), Lactobacillus pantheris (0.97%), Lactobacillus amylovorus (1.9%), Lactobacillus panis (1.9%), Lactobacillus vaginalis (1.9%), Lactobacillus rapi (1.9%), Lactobacillus buchneri (2.9%), Lactobacillus parafarraginis (2.9%), Lactobacillus helveticus (3.9%), Lactobacillus camelliae (3.9%), Lactobacillus fermentum (5.8%), Lactobacillus manihotivorans (6.8%), Lactobacillus plantarum (10.7%), Lactobacillus harbinensis (16.5%) and Lactobacillus paracasei subsp. paracasei (35.0%). This study has shown that TP can be well preserved for 90 days by ensilaging and that TPS is not only rich in essential nutrients, but that physiological and biochemical properties of the isolates could provide a platform for future design of lactic acid bacteria (LAB) inoculants aimed at improving the fermentation quality of silage.

  • 멸치젓갈로부터 ν-aminobutyric acid(GABA)을 생성하는 유산균주를 분리 동정하였다. 멸치젓갈로부터 분리한 9종의 유산균 중 GABA 생산능이 우수한 균주를 순수 분리하여 형태학적 특성과 gram positive identification(GPI) card 와 API 50kit로 동정한 결과 Lactobacillus brevis BH-21, Lactobacillus rhamnosus BH-28 및 Lactobacillus plantarum BH-38로 동정하였다. 이들 Lactobacillus 속 균주중 Lactobacillus brevis BH-21이 GABA생성능력이 제일 우수하였고, GABA 생성함량은 배지(0.1% glucose, 0.1% yeast extracts, 0.05% polypeptone, 0.002 % MgSO$_4$$.$4$H_2O$, 0.001% FeSO$_4$$.$7$H_2O$, 0.01% NaCl, 0.1% monosodium glutamate, pH 6.0)에서 Lactobacillus brevis BH-21을 배양하였을 때, 배양 24시간에 43.2mg/mL의 GABA을 생성하였다.

  • Bacteriocins from lactic acid bacteria have attracted much attention in recent years because of their useful worth in increasing safety and extending shelf life of foods. These substances show an inhibitory effect against some food spoilage bacteria and food-borne pathogens. The inhibitory effect fo the bacteriocin produces by lactic acid bacteria against Listeria monocytogenes(L. monocytogenes) was examined in this study. The culture supernatants of 5 kinds of bacteria among the 10 kinds of testes lactic acid bacteria had the inhibitory activity against Listeria sp., various Gram positive and Gram negative bacteria. Bacteriocin produced by Lactobacillus plantarum(Lact. plantarum) LMG 7945 was the most active toward L. monocytogenes. Bacteriocin production of the Lact. plantarum LMG 7945 cultured on MRS broth was increased late logarithmic phase over early stationary phase. This bacteriocin was stable at heat treatment and acidic pH relatively; The activity was retained after heating at 121$^{\circ}C$ for 15min and was active in the pH range of 2~4 but was lost above pH 5.

시판 유제품 중에서 분리된 젖산균의 감마선 조사에 의한 생존 효과 (The Effect of Gamma Irradiation on the Survival of Lactic Acid Isolated from Commercial Daily Product)

  • This study was conducted with effect of lactic acid bacteria by gamma irradiation. Lactic acid bacteria were exposed to irradiation with a single absorbed dose of 1, 2, 3, 4, 5, 6, 8, and 10 kGy. Possible lactic acid bacteria, including Lactobacillus paracasei KCTC 13169, Lactobacillus casei KCTC 3109, Lactobacillus acidophilus KCTC 3140, Lactobacillus plantarum subsp plantarum KCTC 3103, Lactobacillus debruekii subsp bulgaricus KCTC 3635, Streptococcus thermophilus KCTC 3658 were selected. The radiation sesitivities of lactic acid bacteria were expressed as $D_{10}$ values. The $D_{10}$ values of Lactobacillus paracasei, Lactobacillus casei, Streptococcus thermophilus, Lactobacillus acidophilus, Lactobacillus plantarum, and Lactobacillus debruekii were calculated as 0.42, 0.51, 0.47, 0.90, 0.44, and 0.61 kGy, respectively. Results suggest that L. acidiphilus has the highly resistant to gamma irradiation.

혐기적 산성조건하에서 젖산균의 생존과 그 특성 (Survivals of Lactic Acid Bacteria and its Characteristics under the Acidic and Anaerobic Condition)

  • We investigated the survival, $\beta$-galactosidase activity and cellular permeability of lactic acid bacteria such as Lactobacillus acidophilus ATCC 4356, Lactobacillus casei subsp. casei IFO 3533, Streptococcus thermophilus KCTC 2185, Lactobacillus delbrueckii subsp. lactis ATCC 4797, and Lactobacillus delbrueckii subsp. bulgaricus ATCC 11842 in anaerobic condition of pH 1.5-3.5 range. Numbers of all tested viable cells did not decrease at pH 3.5, but decreased rapidly at pH 1.5 and pH 2.5 during 2 hour incubation at modified EG medium. Immediately after 2 hour incubation, the decrease in population at pH 1.5 and pH 2.5 was about 6-8 and 5-7 log cycles/ml, respectively. Lactobacillus acidophilus ATCC 4356 showed the higest survival of all tested bacteria. The $\beta$-galactosidase activity from Lactobacillus delbrueckii subsp. bulgaricus ATCC 11842 and Streptococcus thermophilus KCTC 2185 decreased rapidly at pH 1.5 and 2.5, but there was a little decrease at pH 3.5. The cellular permeability that was measured by the leakage of intracellular materials increased with decrease of pH. These results suggest that the ingested lactic acid bacteria may be destroyed in contact with low pH of gastric acid.

Lactobacillus farciminis로부터 미지의 작은 플라스미드의 분리와 염기서열 분석 (Isolation and sequence analysis of a small cryptic plasmid from Lactobacillus farciminis KCTC3681)

  • From the extensive screening for small cryptic plasmid among about 23 lactic acid bacteria (LAB), 2.4 kb of cryptic plasmid was isolated from Lactobacillus farciminis strain KCTC 3681 and named as pLF24. The plasmid pLF24 was a circular molecule of 2,396 base-pairs in length with a G+C content of 38%. Two protein-coding sequences could be predicted. ORF1 and ORF2 showed homologies to plasmids of gram-positive bacteria. The replication protein coded by ORF2 and the plus origin, were similar to replication regions of other gram-positive bacteria as shown in plasmids such as pLH2, pLS141-1 and pLC2. The nucleotide sequence of pLF24 was deposited into Genbank data base with an accession number of EU429343. The newly isolated plasmid can be used for construction of shuttle vector in Lactobacillus bacteria.
