A Study was conducted to investigate the effect of feeding Lactobacillus casei YS on the growing performance and gastrointestinal flora of the suckling piglets, which were delivered from 2 heads of three-way crossbred(Landrace$\times$Large White$\times$Duroc) pigs, for 4 weeks. The results from the present study was summarized as follows. Average body weight gains of feeding group was slightly better than that of control group and diarrhea was prevented by successive 7 days feeding. Population levels of lactic acid bacteria were maintained about 107 colony forming unit(cfu) per gram of the contents in both feeding and control group at upper parts of small intestine. In this part, coliform count was greatly reduced in (ceding group but not in control group. pH values of the intestinal contents were gradually decreased especially at the upper part of alimentary track of feeding group. Among lactic acid bacteria, L. salivarius, L. cases and L. fermentum were found most predominant strains in feeding group, Wheareas L. salivarius, L. acidophilus and L. cunts in control group. In the other hand, Escherichia coli recovered from scouring pigs were resistant to the drug such as streptomycin, ampicillin and sensitive to gentamycin and neomycin in vitro test.
The recombinant DNA pLR5cat_PSAB, in which pediocin PA-1 structural and immunity genes (pedAB) fused with the promoter and deduced signal sequence of an ${\alpha}$-amylase gene from a bifidobacterial strain were inserted in Escherichia coli-lactobacilli shuttle vector pLR5cat, was transferred to Lactobacillus reuteri KCTC 3679 and the transformant presented bacteriocin activity. The recombinant L. reuteri KCTC 3679 transformed with the shortened pLR5cat(S)_PSAB, where a nonessential region for the lactobacilli replicon was removed, also showed bacteriocin activity. The molecular mass of the secreted pediocin PA-1 from the recombinant bacteria was the same as that of native pediocin PA-1 (~4.6 kDa) from Pediococcus acidilactici K10 on a sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE) gel. In cocultures with Listeria monocytogenes, the recombinant L. reuteri KCTC 3679 effectively reduced the viable cell count of the pathogenic bacterium by a 3 log scale compared with a control where L. monocytogenes was incubated alone.
In order to determine a proper amount of sodium pyrophosphate (SPP) which protect Lactobacilli from phage attack, Lactobacillus casei YIT-9018 was grown in $Ca^{++}$ free Murata medium having various concentrations of SPP after infections with phage Jl at different multiplicities of infection (MOI). it was found that the level of SPP to inhibit phage activity depended upon the level of MOIs. When the bacteria were infected by less than 10$^{-1}$ of MOI, 0.15% of SPP was sufficient, while with 10$^{0}$ of MOI, 0.175% of SPP was needed to inhibit the lytic activity completely.
The effects of Haematococcus pluvialis extracellular products on microbial growth and bacteriocin production were investigated to improve bacteriocin synthesis during the growth cycle of Lactobacilli. Lactobacillus pentosus KJ-108, L. plantarum KJ-10311, and L. sakei KJ-2008 were cultured in MRS and enriched medium (ERM) with or without supplement of the extracellular products obtained from a late exponential phase culture of Haematococcus pluvialis in modified Bold's basal medium (MBBM). In both MRS and ERM, the extracellular products strongly enhanced the growth as well as the bacteriocin production of all the lactic acid bacteria tested. The enhancing effect was observed in ERM with pH adjusted at 5 and 6. In addition, some difference in growth effects with the extracellular products of H. pluvialis was observed between pH 5 and 6 in ERM, but no effect was observed in the minimal medium. The final biomass and the final concentration of bacteriocin activity were associated with the cell growth that was promoted by the extracellular products of H. pluvialis, and the enhanced cell growth of the three lactic acid bacterial strains induced the increase of the specific bacteriocin production. Therefore, bacteriocin production and activity were influenced by the addition of the extracellular products of H. pluvialis in the culture medium.
This study was done to evaluate the effect of fixed orthodontic patients on the level of oral streptococci, Streptococcus mutans, lactobacilli, yeasts in saliva. 35 patients wearing bands were compared with age-matched 35 non-banded control group by conlony counting method on the specially designed culture medium. The following results were obtained ; 1. The colony forming unit(CFU) of total streptocci per militer of saliva in subjects with or without orthodontic treatment showed no significant statistical difference between them(p>0.05). 2. The colony forming unit(CFU) of total Streptococcus mutans per mililiter of saliva in subjects with orthodontic treatment showed significantly higher than those without orthodontic treatment(p<0.05). 3. The colony forming unit(CFU) of total lactobacilli per mililiter of saliva in sujects with or without orthodontic treatment showed no significant statistical difference between them but higher tendency in those with orthodontic treatment(p=0.052). 4. The colony forming unit(CFU) of total yeasts per mililiter of saliva in subjects with or without orthodontic treatment showed no significant statistical difference between them(p>0.05).
The antagonistic activities of 30 strains of lactobacilli against Helicobacter pylori were determined and Lactobacillus helveticus CU631 has been selected as the strain which possesses the strongest inhibitory effect in the disc diffusion assay showing inhibition zone diameter of $10{\pm}1.5mm$, whereas those of L. plantarum and L. fermentum have been shown to be $4.0{\pm}0.6mm$. H. pylori G88016 revealed the highest vacuolating toxin producing activity among the 8 strains, the inhibitory activity of L. helveticus CU631 in vacuolating toxin producing activity of H. pylori manifested in the co-culture of two strains and in the 5:5 mixture of supernatant of the two strains. Both L. helveticus CU631 and cell free culture supernatant had a strong inhibitory activities in urease and cytotoxin producing activities of H. pylori NCTC11637 and CJH12. An accelerated proteolytic activity of water soluble peptides by L. helveticus CU631 during the refrigeration storage has been manifested in the cream cheese. DNA seqences of 16S-23S ribosomal RNA spacer region showed typical pattern among the various strains of L. helveticus, which could be used in the identification of L. helveticus CU 631.
Reliable PCR based identification of lactobacilli has been described utilizing the sequence of 16S-23S rRNA intergenic spacer region. Those sequence comparisons showed a high degree of difference in homology among the strains of L. rhamnosus, L. casei, L. acidophilus and L. helveticus whose 16S-23S rRNA intergenic small SR's sizes were 222 bp, 222 bp, 206 bp and 216 bp respectively. The sequence of 16S-23S rRNA intergenic spacer region of L. rhamnosus ATCC 53103 revealed the close relatedness to those of L. casei strains by the homology ranges from 95.4% to 97.2%. 16S-23S rRNA intergenic spacer region nucleotide sequence of L. acidophilus showed some distant relatedness with L. rhamnosus ATCC 53103 with the homology ranges from 40.3% to 41.8% and that with L. helveticus was shown to be 30% of homology, which exists at the most distant phylogenetic relatedness. The identification of species and strain of lactobacilli was possible on the basis of these results. The common sequences among the 17 strains were CTAAGGAA located in the initiating position of the DNA and some discrepancies were found between the same strains based on these results.
In vivo antagonistic effect of Lactobacillus helveticus CU 631 and Lactobacillus spp. against typical enteritis causing pathogen Salmonella enteritidis KU 101 have been determined, which showed an increase in survival rate and the decline in viable cell numbers of pathogen in liver and spleen at sacrifice. A signifcant difference in the antagonistic effect against KU 101 were observed, which was species and/or strain dependent of Lactobacillus (p<0.01), the survival rate of the mice in the Salmonella infection by feeding L. helveticus CU 631 has been shown to be 157%, whereas those of L. rhamnosus GG ATCC 53103, L. acidophilus ATCC 4356, L. johnsonii C-4 were 137%, 132%, 119% respectively on the basis of lactobacilli non-associated control KU101 fed mice to be 100%. Viable cells of S. enteritidis KU101 in the liver and in the spleen at sacrifice were decreased in Lactobacillus spp. fed group with no significant difference. The higher level of total secretory IgA concentration in the intestinal fluid of lactobacilli fed mice than control mice have been observed. In vitro antagonistic activity of Lactobacillus spp. against KU101 have been determined, a prominent antagonistic activity of CU 631 against KU 101 were demonstrated.
Lactobacillus acidophilus is known to have an inhibitory activity on growth of Helicobacter pylori and this activity has been attributed to lactic acid and antibacterial agents produced by Lactobacillus. Since every Lactobacilli produces lactic acid, an another factor must exist for L. acidophilus to inhibit H. pylori growth. In this work, the inhibitory activity of L. acidophilus on H. pylori adherence was studied. An immunoabsorbent assay using TLC plate was developed and used for screening the inhibitory activity of various Lactobacilli on H. pylori adherence. Glycolipid, the attachment site for H. pylori, was isolated from blood type O red blood cells and spotted on a TLC plate. The H. pylori adherence increased linearly with increasing amounts of glycolipid spotted on the TLC plate. Various L. acidophilus strains, but not L. casei, appeared to inhibit H. pylori adherence to glycolipid, and the adherence decreased linearly as the concentration of the Lactobacillus increased. The results show that the inhibitory activity of L. acidophilus on H. pylori adherence is an another factor for L. acidophilus to inhibit H. pylori growth.

The antioxidative properties of lactobacilli originating from humans (Lactobacillus acidophilus KCTC 3111, Lactobacillus jonsonii KCTC 3141, Lactobacillus acidophilus KCTC 3151, and Lactobacillus brevis KCTC 3498) were investigated using in vitro methods, including inhibition of lipid peroxidation, resistance to hydrogen peroxide and hydroxyl radical, hydroxyl radical scavenging activity, and glutathione peroxidase (GPX) activity. L. acidophilus KCTC 3111 showed the highest inhibition of lipid peroxidation in both intact cells (49.7%) and cell lysate (65.2%). This strain exhibited resistance to hydrogen peroxide and hydroxyl radical, which was viable for 7 h in the concentration of 1.0 mM hydrogen peroxide. In addition, this strain showed high hydroxyl radical scavenging activity. In the GPX activity assay, the highest activity was measured in L. brevis 3498. GPX activity of L acidophilus 3111 was lower than that of L. brevis 3498.
