A feeding trial was conducted to study the effect of tannin rich Pakar (Ficus infectoria) leaves on microbial profile, rumen fermentation and nutrient utilization in goats. Eight goats divided in two groups were fed pakar leaves (experimental group) and green oats (control group) as sole roughage source along with a fixed quantity of concentrate mixture for a period of 3 months. Two metabolic trials of six days duration were conducted after 30 and 90 days of experimental feeding. The dry matter intake was significantly higher (p<0.05) and digestibility's of DM, OM, CP, EE, NDF and ADF were reduced in experimental as compared with the control group. The TDN intake was similar (236.52 vs. 240.39 g/d) in both the groups. All the animals were in positive nitrogen balance. The concentration of ammonia nitrogen, TVFA, lactic acid and activities of xylanase and protease were reduced in pakar leaves fed goats. The rumen microbial profile as obtained by MPN technique showed no change in total bacterial population but total fungi and cellulolytic bacteria were reduced (p<0.05), whereas, tannin degrading/tolerant bacteria increased with the feeding of pakar leaves. Real time PCR data revealed a decrease in Ruminococcus flavefaciens, an increase in methanogens and no change in the Fibrobacter succinogenes population by feeding of pakar leaves.

Objective: Although the efficacy of Rubus coreanus (RC) byproducts as a feed additive has been recognized, its effects on intestinal microorganisms and the immune system are still unknown. Methods: Six-week-old male rats were treated with 0.5% RC (T1), 1.0% RC (T2), and 1.5% RC (T3) for 4 weeks. Results: We found that treatment with RC byproducts significantly increased the daily gain of body weight and feed intake. Treg-cell differentiation was enhanced in the mesenteric lymph nodes and spleen from the rats fed with RC byproducts. Illumina sequencing showed that bacteria in the phylum Firmicutes decreased and while those in the phylum Bacteroidetes increased in RC-treated groups. Particularly, the pathogenic microorganisms in the family Peptococcaceae decreased, and the non-pathogenic families Lachnospiraceae and S24-7 increased. Quantitative polymerase chain reaction analysis showed that the RC byproducts increased the lactic acid bacteria Bifidobacterium spp., Oscillospira spp., Leuconostoc citreum, and Weissella cibaria in a concentration-dependent manner. Conclusion: RC byproducts may be effective in immunomodulation by affecting intestinal microorganisms.
Myeolchi jeotgals (MJs) were prepared with purified salt (PS), solar salt aged for 1 year (SS), and bamboo salt (BS) melted 3 times at 10% and 20% (w/w) concentrations, and fermented for 28 weeks at $15^{\circ}C$. BS MJ showed higher pH and lower titratable acidities than the other samples because of the alkalinity of bamboo salt. Lactic acid bacteria counts increased until 4-6 weeks and then decreased gradually, and were not detected after 20 weeks from MJs with 10% salt. Yeast counts of PS MJs were higher than those of BS and SS MJs. Bacilli were detected in relatively higher numbers throughout the 28 weeks, like marine bacteria, but archae were detected in lower numbers during the first 10 weeks. When 16S rRNA genes were amplified from total DNA from PS MJ (10% salt) at 12 weeks, Tetragenococcus halophilus was the major species. However, Staphylococcus epidermidis was the dominant species for BS MJ at the same time point. In SS MJ, T. halophilus was the dominant species and S. epidermidis was the next dominant species. BS and SS MJs showed higher amino-type nitrogen, ammonia-type nitrogen, and volatile basic nitrogen contents than PS MJs. SS and BS were better than PS for the production of high-quality MJs.
Objective: This study investigated the association of enzyme-producing microbes and their enzymes with starch and hemicellulose degradation during fermentation of total mixed ration (TMR) silage. Methods: The TMRs were prepared with soybean curd residue, alfalfa hay (ATMR) or Leymus chinensis hay (LTMR), corn meal, soybean meal, vitamin-mineral supplements, and salt at a ratio of 25:40:30:4:0.5:0.5 on a dry matter basis. Laboratory-scale bag silos were randomly opened after 1, 3, 7, 14, 28, and 56 days of ensiling and subjected to analyses of fermentation quality, carbohydrates loss, microbial amylase and hemicellulase activities, succession of dominant amylolytic or hemicellulolytic microbes, and their microbial and enzymatic properties. Results: Both ATMR and LTMR silages were well preserved, with low pH and high lactic acid concentrations. In addition to the substantial loss of water soluble carbohydrates, loss of starch and hemicellulose was also observed in both TMR silages with prolonged ensiling. The microbial amylase activity remained detectable throughout the ensiling in both TMR silages, whereas the microbial hemicellulase activity progressively decreased until it was inactive at day 14 post-ensiling in both TMR silages. During the early stage of fermentation, the main amylase-producing microbes were Bacillus amyloliquefaciens (B. amyloliquefaciens), B. cereus, B. licheniformis, and B. subtilis in ATMR silage and B. flexus, B. licheniformis, and Paenibacillus xylanexedens (P. xylanexedens) in LTMR silage, whereas Enterococcus faecium was closely associated with starch hydrolysis at the later stage of fermentation in both TMR silages. B. amyloliquefaciens, B. licheniformis, and B. subtilis and B. licheniformis, B. pumilus, and P. xylanexedens were the main source of microbial hemicellulase during the early stage of fermentation in ATMR and LTMR silages, respectively. Conclusion: The microbial amylase contributes to starch hydrolysis during the ensiling process in both TMR silages, whereas the microbial hemicellulase participates in the hemicellulose degradation only at the early stage of ensiling.
높은 유산생성 능력을 가진 lactobacillus bulgaricus NLS-4주를 대창 배양하기 위하여 배양조건을 개선하였다. 이제까지 보고된 많은 배지중에서 7개를 선정하고 흔히 사용되고 있는 11% 탈지유 배지를 대조군으로 하여 사용 균주의 증식과 유산 생성능을 비교한 결과 TIP배지를 최종 선정하였다. 이 배지를 이용하여 38$^{\circ}C$에서 통기없이 200rpm의 속도로 교반해 주면서 IN KOH용액을 사용하여 PH를 6.5로 자동 조절해준 결과 18시간 동안의 회분배양에서 얻은 균체수는 생균수로 $m\ell$ 당 1.0$\times$$10^{9}$ 이었으며 이를 다시 연속 배양한 결과 희석 배율 D=0.17$hr^{-1}$에서 $m\ell$당 총 2.3$\times$$10^{9}$ 의 균체를 얻을 수 있었다.
숙성 정도가 오래된 김장 배추 김치에서 분리되어 16S rDNA 염기 서열 분석을 통해 부분 동정된 KNUC25 분리 균주를 단백질 전기 영동 패턴과 생리적 특징 그리고 염기 서열의 유사성을 비교하여 Lactobacillus paraplantarum로 동정하였다. L. paraplantarum KNUC25의 농축 상등액은 그람 양성균과 음성균에 넓은 범위의 항균 활성을 나타냈다. 전자 현미경을 통해 KNUC25가 만들어내는 항균 물질은 세균의 표면에 작용하여 생육을 억제하는 것으로 확인되었다. 항균 활성 물질을 생산하는 최적 온도는 섭씨 30도이고, 높은 온도에서도 항균 활성을 보였으며 단백질 분해 효소에 안정하며 비단백질성 항균 물질일 가능성을 보여주었다.

An experiment of 10 weeks duration was carried out to study the influence of supplemental effective microorganism (EM) culture, yeast culture and enzymes on nutrient digestibility and gut microflora in rabbit gastrointestinal (GI) tract. Twenty four eight to nine weeks old, New Zealand White rabbits were allotted to four dietary treatments; a basal (control) feed, basal feed supplemented with either EM (1%), yeast culture or enzymes (400 ppm). Nutrient flow in digesta and their digestibility at ileum, caecum, colon and in the total tract as well as gut microflora distribution were studied. Feed dry matter was diluted from 92% to about 14% up to the ileum and about 95% of this water was reabsorbed by the colonic rectal segment followed by caecum (25%). EM and yeast improved protein digestibility at a lower rate than enzymes. Ileal, caecal, colonic and total tract digestibility of crude protein with enzymes were higher by 10.8, 9.4, 11.3 and 10.7%, respectively, as compared to the control. Yeast and enzymes increased crude fiber digestibility at ileum, caecum, colon and in the total tract by 8.5, 9.6, 9.0 and 8.3%, respectively, while EM improved them at a lower rate. Irrespective of treatments, total tract digestibility of crude protein (0.698-0.773) and fiber (0.169-0.183) were greater (p<0.05) than the ileal digestibility. Even though a post-caecal protein digestibility was observed, fiber digestion seemed to be completed in the caecum especially with yeast and enzymes. High precaecal digestibility of crude fiber (97%) and protein (95%) were observed even without additives probably due to caecotrophy. EM and yeast culture promoted the growth of lactic acid bacteria especially in the caecum but they did not influence gut yeast and mould. Present findings reveal that even though rabbits digest nutrients efficiently through hind gut fermentation, they can be further enhanced by EM, yeast and enzymes. Of the three additives tested, enzymes found to be the best.

Influences of forage DM and addition of cane molasses on silage characteristics of berseem (Trifolium alexandrium) and lucerne (Medicago sativa) and their ruminal digestion kinetics in Nili buffaloes were studied. Berseem and lucerne fodders (at one tenth bloom) were ensiled with wheat straw in laboratory silos to achieve 20, 30, 40% forage DM and without wheat straw (control); each forage DM level was supplemented with 2, 4 and 6% of cane molasses at ensiling. The pH and lactic acid contents of berseem and lucerne silages were affected by both forage DM and addition of molasses. Dry matter, CP and true protein (TP) of berseem and lucerne silages were affected by forage DM at ensiling but were not affected by the addition of cane molasses. Higher DM, CP and TP losses were observed when berseem and lucerne fodders were ensiled either without wheat straw or with wheat straw to achieve 20% and 40% forage DM at ensiling compared with 30% DM at ensiling. Fiber fractions (NDF, ADF, hemicellulose and cellulose) of berseem silage and lucerne silage were significantly increased with increasing forage DM at ensiling. Addition of cane molasses did not affect the DM, CP, TP and fiber fractions of both berseem and lucerne silages. Berseem and lucerne ensiled at 30% DM with 2% cane molasses were screened for comparative ruminal digestion kinetics with their respective fodders. Addition of wheat straw to berseem or lucerne fodder at ensiling depressed DM and NDF ruminal degradability. However, ruminal lag time, rate of degradation and extent of digestion of silages were similar to their respective fodders. In conclusion, berseem and lucerne could be ensiled with wheat straw to increase their DM to 30% along with 2% molasses for buffaloes.

As a functional additive for intestinal microflora, panose ($6^2-{\alpha}$-D-glucopyranosylmaltose) was synthesized during kimchi fermentation using the glucose transferring reaction of glucansucrase from Leuconostoc mesenteroides. For the glucose transferring reaction, sucrose and maltose were added ($2\%$ each, w/v) to dongchimi-kimchi as the glucosyl donor and acceptor molecule, respectively. After five days of incubation at $10^{\circ}C$, referring to the initial phase for the production of lactic acid in kimchi, over $60\%$ (w/v) of the total sugars were converted into panose and other branched oligosaccharides. Thereafter, the kimchi was stored at $4^{\circ}C$ and the amount of panose remained at a constant level for three weeks, thereby indicating the stability of panose to microbial degradation during the period of kimchi consumption. The use of maltose as the acceptor molecule in the kimchi also facilitated a lower viscosity in the kimchi-juice by preventing the synthesis of a dextran-like polymer which caused an unfavorable taste. Accordingly, the application of this new method of simultaneous biocatalytic synthesis of oligosaccharides during lactate fermentation should facilitate the extensive development of new function-added lactate foods.
