Rhizospheric fungi of Panax notoginseng: diversity and antagonism to host phytopathogens

  • Background: Rhizospheric fungi play an essential role in the plantesoil ecosystem, affecting plant growth and health. In this study, we evaluated the fungal diversity in the rhizosphere soil of 2-yr-old healthy Panax notoginseng cultivated in Wenshan, China. Methods: Culture-independent Illumina MiSeq and culture-dependent techniques, combining molecular and morphological characteristics, were used to analyze the rhizospheric fungal diversity. A diffusion test was used to challenge the phytopathogens of P. notoginseng. Results: A total of 16,130 paired-end reads of the nuclear ribosomal internal transcribed spacer 2 were generated and clustered into 860 operational taxonomic units at 97% sequence similarity. All the operational taxonomic units were assigned to five phyla and 79 genera. Zygomycota (46.2%) and Ascomycota (37.8%) were the dominant taxa; Mortierella and unclassified Mortierellales accounted for a large proportion (44.9%) at genus level. The relative abundance of Fusarium and Phoma sequenceswas high, accounting for 12.9% and 5.5%, respectively. In total,113 fungal isolates were isolated from rhizosphere soil. They were assigned to five classes, eight orders (except for an Incertae sedis), 26 genera, and 43 species based on morphological characteristics and phylogenetic analysis of the internal transcribed spacer. Fusarium was the most isolated genus with six species (24 isolates, 21.2%). The abundance of Phoma was also relatively high (8.0%). Thirteen isolates displayed antimicrobial activity against at least one test fungus. Conclusion: Our results suggest that diverse fungi including potential pathogenic ones exist in the rhizosphere soil of 2-yr-old P. notoginseng and that antagonistic isolates may be useful for biological control of pathogens.

Changes in bound water and microstructure during consolidation creep of Guilin red clay

  • Creep of soils has a significant impact on mechanical properties. The one-dimensional consolidation creep test, thermal analysis test, scanning electron microscope (SEM) test, and mercury compression test were performed on Guilin red clay to study the changes in bound water and microstructure during the creep process of Guilin red clay. According to the results of the tests, only free and weakly bound water is discharged during the creep of Guilin red clay. When the consolidation pressure p is in the 12.5-400.0 kPa range, it is primarily the discharge of free water; when the consolidation pressure p is in the 800.0-1600.0 kPa range, the weakly bound water is converted to free water and discharged. After consolidation creep, the microstructure of soil changes from granular overhead contact structure to flat sheet-like stacking structure, with a decrease in the number of large and medium pores, an increase in the number of small and micro pores, and a decrease in the fractal dimension of pores. The creep process of red clay is the discharge of weakly bound water as well as the compression of large pores into small pores and the transition of soil particles from loose to dense.

연약지반상 고속철도 노반 축조시 지오셀 시스템의 효과 (Reinforcement of Soft Soil Subgrade for High-Speed Railroad Using Geocell)

  • This paper presents the results of plate load test and dynamic load test performed to evaluate the performance of geocell where it is used to reinforce soft subgrade for high-speed railroad. Efficacy of geocell was observed in increase in bearing capacity of subgrade and reduction of thickness of reinforced sub-ballast. Plate load tests were carried out at four different places with varying foundation soil strength as a function of number of geocell layer, type of filler material, thickness of cover soil, and the presence of non-woven geotextile. Dynamic load tests were performed in a laboratory. The test soil chamber consists of, from the bottom, 50 cm thick clayey soil, one layer of geocell filled with crushed stone, 10 cm thick crushed stone cover, reinforced sub-ballast of varying thickness, 35 cm thick ballast. This configuration was determined based on the results of numerical analysis and plate load tests. For each set of the dynamic load tests, loads were applied more than 80,000 times. One layer of geocell underlying a 10 cm thick cover soil led to an increase in bearing capacity three to four times compared to a crushed stone layer of the same thickness substituted for the geocell and cover soil layer. Given the test conditions, the thickness of reinforced sub-ballast can be reduced by approximately 35 cm with the presence of geocell.

  • When a nail pulled out in dense, granular soil, the soil in the vicinity of the nail tends to dilate, but its dilatancy results in a normal stress concentration at the soil/nail interface, thereby increasing the pull-out resistance of the inclusion. It is thought to be occurring within the resistance zone where the soil mass is at stationary state and the reinforcement are held in position by the soil, due to the friction or bond. In this paper, A series of direct shear and interface tests were conducted by using so called‘Constant Normal Stiffness Test Apparatus’which was modified and improved from the conventional direct shear box test rig. Unlikely the normal shear box test, this enables to simulate the different constraint effects of surrounding soil during shear under the conditions of constant stress and volume, constant normal stiffness. The aim of the research programme is to get better understanding of pull-out bond mechanism, thus to explore the possibility of evaluating the pull-out bond capacity of soil/reinforcement at the preliminary design stage from the laboratory test.

Mechanical behaviour of biocemented sand under triaxial consolidated undrained or constant shear drained conditions

  • Biocementation based on the microbially induced calcite precipitation (MICP) process is a novel soil improvement method. Biocement can improve significantly the properties of soils by binding soil particles to increase the shear strength or filling in the pores to reduce the permeability of soil. In this paper, results of triaxial consolidated undrained (CU) tests and constant shear drained (CSD) tests on biocemented Ottawa sand are presented. In the CU tests, the biocemented sand had more dilative behaviour by showing a higher stress-strain curves and faster pore pressure reducing trends as compared with their untreated counterparts. In the CSD tests, the stress ratio q/p' at which biocemented sand became unstable was higher than that for untreated sands, implying that the biocementation will improve the stability of sand to water infiltration or liquefaction.

모형실험에 의한 토공구조물의 침투거동특성 (The Characteristics for Seepage Behaviour of Soil Structure by Modeling Tests)

  • In parallel flow condition, to estimate the stability of the extended embankment constructed on a permeable foundation ground, a laboratory model test was performed due to extended materials and water level increasing velocity of a flood period. A laboratory model test was peformed for different permeability coefficients ($K_1=2.0{\times}10^{-5}cm/sec,\;K_2=1.5{\times}10^{-4}cm/sec,\;K_3=2.3{\times}10^{-3}cm/sec$) using seepage. The fluctuation of water level occurring to an extended embankment was analyzed by laboratory model tests as vary the increasing velocity of water level with 0.6cm/min, 1.2cm/min, 2.4cm/min respectively. In analysis results, the increase of water level into embankment occurs rapidly because seepage water moving along with a permeable soil flow into embankment. The larger the permeability coefficient of an extended part is the longer initial seepage distance, and the exit point of downstream slope is gradually increased and then shows unstable seepage behavior as occurring partial collapse. As the increasing velocity of water level increase, the initial seepage line is formed low, and the discharge increases. Therefore, the embankment extended by a lower permeable soil than existing embankment shows stable seepage behavior because an existing embankment plays a role as filter for an extended part.

A simplified directly determination of soil-water retention curve from pore size distribution

  • Numbers fitting-curve equations have been proposed to predict soil-water retention curve (SWRC) whose parameters have no definitude physical meaning. And these methods with precondition of measuring SWRC data is time-consuming. A simplified directly method to estimate SWRC without parameters obtained by fitting-curve is proposed. Firstly, the total SWRC can be discretized into linear segments respectively. Every segment can be represented by linear formulation and every turning point can be determined by the pore-size distribution (PSD) of Mercury Intrusion Porosimetry (MIP) tests. The pore diameters governing the air-entry condition (AEC) and residual condition (RC) can be determined by the PSDs of MIP test. The PSD changes significantly during drying in SWR test, so the determination of AEC and RC should use the PSD under corresponding suction conditions. Every parameter in proposed equations can be determined directly by PSD without curve-fitting procedure and has definitude physical meaning. The proposed equations give a good estimation of both unimodal and bimodal SWRCs.

단순회귀분석에 의한 토층지반의 투수계수 산정모델 (Estimation model of coefficient of permeability of soil layer using linear regression analysis)

  • To derive easily the coefficient of permeability from several other soil properties, the estimation model of coefficient of permeability was proposed using linear regression analysis. The coefficient of permeability is one of the major factors to evaluate the soil characteristics. The study area is located in Kangwon-do Pyeongchang-gun Jinbu-Myeon. Soil samples of 45 spots were taken from the study area and various soil tests were carried out in laboratory. After selecting the soil factor influenced by the coefficient of permeability through the correlation analysis, the estimation model of coefficient of permeability was developed using the linear regression analysis between the selected soil factor and the coefficient of permeability from permeability test. Also, the estimation model of coefficient of permeability was compared with the results from permeability test and empirical equation, and the suitability of proposed model was proved. As the result of correlation analysis between various soil factors and the coefficient of permeability using SPSS(statistical package for the social sciences), the largest influence factor of coefficient of permeability were the effective grain size, porosity and dry unit weight. The coefficient of permeability calculated from the proposed model was similar to that resulted from permeability test. Therefore, the proposed model can be used in case of estimating the coefficient of permeability at the same soil condition like study area.

불포화 준설매립 지반의 흙-수분 특성곡선 및 불포화 투수계수 예측 (Prediction of Soil-water Characteristic Curve and Unsaturated Permeability Coefficient of Reclaimed Ground)

  • 불포화지반에서 발생되는 부 간극수압은 함수비 변화에 따라 비선형적인 거동을 보이며, 이 관계를 흙-수분 특성곡선(soil-water characteristic curve)이라고 정의한다. 현재까지 우리나라의 흙에 대한 흙-수분 특성의 연구결과는 거의 전무한 실정이기 때문에 불포화 지반의 응력해석 및 침투해석 시, 마땅히 입력할 계수나 특성값이 없다. 본 연구에서는 준설매립지반의 주요 토사인 준설토와 모래, 그리고 복토재로 사용되는 화강풍화토에 대한 흙수분 특성시험을 실시하여 흙-수분 특성을 규명하였으며, 해석적 검증을 통하여 불포화 지반의 응력해석 및 침투해석 시 필요한 계수를 분석하였다. 또한, 입도분포곡선으로부터 흙수분 특성곡선 예측모델을 적용하고 흙수분 특성시험 결과와 비교 검토하였다. 흙-수분 특성시험결과, 세 가지 시료 모두 뚜렷한 히스테리시스 현상을 나타내었으며, 기존에 제안된 경험방정식에 실험결과를 적용한 결과 Fredlund and Xing의 경험방정식은 세 가지 시료 모두에 대하여 높은 정확도를 나타내었다. 한편, 입도분포를 이용한 예측모델은 Fredlund and Wilson의 방법이 가장 높은 정확도를 나타내었다. 흙-수분 특성시험결과와 다양한 예측모델을 통하여 불포화 투수계수를 예측하여 상호 비교한 결과, 모래와 화강풍화토는 Fredlund and Wilson의 방법, 준설토는 Arya and Paris의 예측모델이 높은 정확도를 나타내었다.

실내모형실험을 통한 히빙 이론에 관한 연구 (A Study on Heaving Phenomenon by Model Test)

  • In this paper, heaving phenomenon is analyzed by laboratory tests. A laboratory test is consist of building soft clay foundation in plane-strain soil tank, construction of retaining wall, and excavation work. And range of shear strain, and destruction shape about soft clay foundation is compared, and analyzed with results of proposal formula. Using this study, safety factor is suggested for heaving phenomenon in the construction of wall on the soft clay. Actual theory is suggested by this suggested safety factor. There are various proposal formula for heaving phenomenon. For example, Terzaghi & Peck, Tschebotarioff, Bjerrum & Eide(Experience formula) and so on. Terzaghi & Peck's proposal formula is chosen, compared with laboratory test's result and analyzed in this study. A soft clay used in study is assumed homogeneous. A Depth of foundation is enough to observe shear strain by heaving phenomenon. Retaining wall is enough hard not to have vertical displacement.
