Development & Assessment of Alkyl Chain Modified Aptamers as Potential PET Radiotracers for Lymphoma Diagnosis

  • The Td05 and Sgc8c, DNA-based aptamers, are well-known to target internalized surface markers (IGHM and PTK7) of Burkitt's lymphoma and acute lymphoblastic leukemia (ALL). Thus, Td05 and Sgc8c labeled with metallic radioisotope 64Cu can be evaluated as potential diagnostic PET imaging agents. In this study, we modified the carbon chain length of the last adenosine of aptamer (n = 3, 6, 12) to increase tumor cell uptake and select the best candidate among six types of aptamer analogues and one adenosine of aptamer. After labeling of 64Cu, [64Cu]Cu-DOTA-aptamer analogues were evaluated in vitro studies (serum stability, Log P values, cell uptake, biodistribution). Then, we evaluate in vivo PET imaging study for two candidates (64Cu-DOTA-C12-Sgc8c, 64Cu-DOTA-C6-Td05). PET images clearly visualize tumors at 24 h post-injection rather than at an early time point and the tumor-to-background ratio also increases at the delay time point. 64Cu-DOTA-C12-Sgc8c and 64Cu-DOTA-C6-Td05 could be used as potential radiotracers for lymphoma.

Comparison of Kinematics and Myoelectrical Activity during Deadlift, with and without Variable Banded Resistance, in Healthy, Trained Athletes

  • Background: The conventional deadlift is a popular exercise for enhancing trunk, core, and lower extremity strength. However, its use in sports medicine is constrained by concerns of lumbar injuries, despite evidence supporting its safety and rehabilitative benefits. To optimize muscle activation using resistive bands in variable resistance therapy, we explored their feasibility in the deadlift. Design: Comparative experimental design Methods: Surface electromyography recorded muscle activity in the trunk and lower extremities during lifting, with normalization to the isometric Floor Lift using Maximal Voluntary Contraction. Kinematics were measured using inclinometer sensors to track hip and trunk sagittal plane angles. To prevent fatigue, each subject only used one of the three pairs of bands employed in the study. Results: Our study involved 45 healthy subjects (mean age: 30.4 ± 6.3 years) with similar baseline characteristics, except for years of lifting and strength-to-years-of-lifting ratio. Various resistance band groups exhibited significantly higher muscle activity than conventional deadlifts during different phases. The minimal resistance band group had notably higher muscle activity in the trunk, core, and lower extremity muscles, particularly in the end phase. The moderate resistance band group showed increased muscle activity in the mid-and end-phases. The maximum resistance band group demonstrated greater muscle activity in specific muscles during the early phase and overall higher activity in all trunk and lower extremity muscles in the mid and end phases of the deadlift (p<0.05). Conclusion: Our findings provide valuable insights into muscle activation with various resistance bands during deadlift exercise in clinical and gym settings. There appears to be a dose-response relationship between increased resistance bandwidth, external load, myoelectric activation, and range.

연구실 위험성평가방법 개선에 관한 연구 (A Study on the Improvement of Risk Assessment Method in Laboratory)

  • 연구목적: 연구실험실에는 사전유해인자위험분석을 실시하도록 하고 있으나 실행하지 않는 연구실험실이 많다. 연구실험실의 안전을 확보하기 위한 목적으로 좀 더 쉽게 위험성평가를 적용 할 수 있도록 개선점을 찾고자 한다. 연구방법: 선행논문 및 과학기술정보통신부 연구실 안전관리 실태조사 보고서 자료를 활용하였다. 사전유해인자위험분석과 산업안전보건법에 의한 위험성평가를 비교하여 연구하였다. 연구결과: 연구실험실에 대한 위험성을 평가할 수 있는 기법을 제시하였다. 연구실험실 위험성평가시 가능성(빈도)과 중대성(강도)의 크기를 추정할 수 있도록 방법을 제시하였다. 결론: 연구실험실에서 쉽게 적용할 수 있도록 체크리스트 형태의 위험성평가 기법을 제시하였고, 실제 평가해 보았다. 연구실험실에서 위험성평가를 통한 개선을 실시하여 안전사고 예방에 도움이 될 것으로 기대한다.

Research Progress on Strategies for Improving the Enzyme Properties of Bacteriophage Endolysins

  • Bacterial resistance to commonly used antibiotics is one of the major challenges to be solved today. Bacteriophage endolysins (Lysins) have become a hot research topic as a new class of antibacterial agents. They have promising applications in bacterial infection prevention and control in multiple fields, such as livestock and poultry farming, food safety, clinical medicine and pathogen detection. However, many phage endolysins display low bactericidal activities, short half-life and narrow lytic spectrums. Therefore, some methods have been used to improve the enzyme properties (bactericidal activity, lysis spectrum, stability and targeting the substrate, etc) of bacteriophage endolysins, including deletion or addition of domains, DNA mutagenesis, chimerization of domains, fusion to the membrane-penetrating peptides, fusion with domains targeting outer membrane transport systems, encapsulation, the usage of outer membrane permeabilizers. In this review, research progress on the strategies for improving their enzyme properties are systematically presented, with a view to provide references for the development of lysins with excellent performances.

  • The Japanese Good Laboratory Practice (GLP) Standard on Drugs was finalized as a guideline and implemented in April, 1983. This standard is intended to ensure the quality and integrity of the data from nonclinical toxicity studies submitted to the Ministry of Health and Welfare in support of applications for approval to manufacture or import new drugs or to be used in the reevaluation of previously approved drugs. The standard includes a guideline for organizational matters, personnel, facility, equipment, testing operation, documentation and conduct of studies. Principles and influences of implementation of Japanese GLP will be discussed briefly in comparison with foreign GLPs.

Measurement and Modeling of Personal Exposure to the Electric and Magnetic Fields in the Vicinity of High Voltage Power Lines

  • Background: This work presents an experimental and modeling study of the electromagnetic environment in the vicinity of a high voltage substation located in eastern Algeria (Annaba city) specified with a very high population density. The effects of electromagnetic fields emanating from the coupled multi-lines high voltage power systems (MLHV) on the health of the workers and people living in proximity of substations has been analyzed. Methods: Experimental Measurements for the Multi-lines power system proposed have been conducted in the free space under the high voltage lines. Field's intensities were measured using a referenced and calibrated electromagnetic field meter PMM8053B for the levels 0 m, 1 m, 1.5 m and 1.8 m witch present the sensitive's parts as organs and major functions (head, heart, pelvis and feet) of the human body. Results: The measurement results were validated by numerical simulation using the finite element method and these results are compared with the limit values of the international standards. Conclusion: We project to set own national standards for exposure to electromagnetic fields, in order to achieve a regional database that will be at the disposal of partners concerned to ensure safety of people and mainly workers inside high voltage electrical substations.

Subchronic Oral Toxicity Study of Acanthopanax divaricatus var. albeofructus in Rats

  • Acanthopanax divaricatus (Siebold & Zucc.) Seem. var. albeofructus (ADA), a traditional medical herb, has been used to treat arthritis and muscular injury, to strengthen muscle and bone, and to get vital energy. However, information regarding its toxicity is limited. ADA was administered by oral gavage to groups of rats at doses of 0 (control), 1,000, 1,500, 2,000, 2,500, and 3,000 mg/kg five times per week for 13 weeks. Mortality, clinical signs, body weights, food consumption, hematology, serum chemistry, urinalysis, organ weights, necropsy, histopathological finding, vaginal cytology, and sperm morphology were compared between control and ADA-treated groups. Salivation was intermittently observed in both sexes receiving 2,500 and 3,000 mg/kg directly after dosing. Absolute liver weights increased in females receiving 2,000, 2,500, and 3,000 mg/kg ADA (P < 0.05, P < 0.01, and P < 0.01, respectively) and so did the relative liver weights (P < 0.001). Salivation and increased liver weight were ADA-related changes but not considered to be adverse effects. Salivation was intermittent and transient, and the liver weight increase was minor and not accompanied by other changes such as hepatic morphological or functional alterations. The no-observed-adverse-effect-level was determined to be at least 3,000 mg/kg in both sexes of rats.

Worker Safety in Modular Construction: Investigating Accident Trends, Safety Risk Factors, and Potential Role of Smart Technologies

  • Modular building is a fast-growing construction method, mainly due to its ability to drastically reduce the amount of time it takes to construct a building and produce higher-quality buildings at a more consistent rate. However, while modular construction is relatively safer than traditional construction methods, workers are still exposed to hazards that lead to injuries and fatalities, and these hazards could be controlled using emerging smart technologies. Currently, limited information is available at the intersection of modular construction, safety risk, and smart safety technologies. This paper aims to investigate what aspects of modular construction are most dangerous for its workers, highlight specific risks in its processes, and propose ways to utilize smart technologies to mitigate these safety risks. Findings from the archival analysis of accident reports in Occupational Safety and Health Administration (OSHA) Fatality and Catastrophe Investigation Summaries indicate that 114 significant injuries were reported between 2002 and 2021, of which 67 were fatalities. About 72% of fatalities occurred during the installation phase, while 57% were caused by crushing and 85% of crash-related incidents were caused by jack failure/slippage. IoT-enabled wearable sensing devices, computer vision, smart safety harness, and Augment and Virtual Reality were identified as potential solutions for mitigating identified safety risks. The present study contributes to knowledge by identifying important safety trends, critical safety risk factors and proposing practical emerging methods for controlling these risks.

탄소나노튜브 성장 실험실에서 CVD 밀폐 여부에 따른 공기 중 나노입자 농도 비교 (Comparison of Airborne Nanoparticle Concentrations between Carbon Nanotubes Growth Laboratories based on Containment of CVD)

  • Although the usage of nanomaterials including carbon nanotubes (CNTs) has increased in various fields, scientific researches on workers' exposures and controls of these materials are very limited. The purpose of this study was to compare the airborne nanoparticles concentrations from two university laboratories conducting experiments of CNTs growth based on containment of thermal chemical vapor deposition (CVD). Airborne nanoparticle concentrations in three metrics (surface area concentration, particle number concentration, and mass concentrations) were measured by task using three direct reading instruments. In a laboratory where CVD was not contained, the surface area concentration, number concentration and mass(PM$_1$) concentration of airborne nanoparticles were 1.5 to 3.5 times higher than those in the other laboratory where CVD was confined. The ratio of PM$_1$ concentration to total suspended particles(TSP) in the laboratory where CVD was not confined was about 4 times higher than that in the other laboratory. This indicates that CVD is a major source of airbone nanoparticles in the CNTs growth laboratories. In conclusion, researchers performing CNTs growth experiments in these laboratories were exposed to airborne nanoparticles levels higher than background levels, and their exposures in a laboratory with the unconfined CVD were higher than those in the other laboratory with the confined CVD. It is recommended that in the CNTs growth laboratories adequate controls including containment of CVD be implemented for minimizing researchers' exposures to airborne nanoparticles.
