Anti-platelet Effects of Isoscopoletin through Regulation of PI3K/Akt and MAPK on Collagen-induced Human Platelets (콜라겐-유도의 사람 혈소판에서 PI3K/Akt 및 MAPK 조절을 통한 Isoscopoletin의 항혈소판 효과)

  • When blood vessels are damaged, a rapid hemostatic reaction occurs to minimize blood loss and maintain normal circulation. Platelet activation and aggregation is essential in this process. However, excessive platelet aggregation or abnormal platelet aggregation may be the cause of cardiovascular disease, such as thrombosis, stroke and atherosclerosis. Therefore, it is important to prevent and treat cardiovascular disease by finding substances that can regulate platelet activation and suppress aggregation reactions. Isoscopoletin, which is mainly found in the roots of plants Artemisia or Scopolia, has been reported to have potential pharmacological effects on anticancer and Alzheimer's disease, but its role and mechanisms for platelet aggregation and thrombus formation are unknown. This study confirmed the effect of isoscopoletin on major regulation of collageninduced human platelet aggregation, TXA2 production and intracellular granular secretion (ATP and serotonin release). In addition, the effects of isoscopoletin on phosphorylation of phosphorylated proteins PI3K/Akt and MAPK involved in signal transduction in platelet aggregation was studied. As a result, isoscopoletin significantly inhibited the phosphorylation of PI3K/Akt and MAPK, significantly inhibiting platelet aggregation through TXA2 production and intracellular granular secretion (ATP and serotonin release). Therefore, we suggest that isoscopoletin is an anti-platelet substance that regulates phosphorylation of phosphorus proteins such as PI3K/Akt and MAPK and is valuable as a preventive and therapeutic agent for platelet-derived cardiovascular disease.

Computed Tomographic Features of Blunt Abdominal Trauma in a Dog (개에서 컴퓨터단층촬영을 이용한 복부 장기 열상 진단 1례)

  • A 10 months old, male Poongsan dog was referred with a history of right forelimb lameness due to hit by a car a few hours before presentation. On the physical examination, the dog showed pale mucous membrane, respiration distress, and skin abrasions. The main laboratory finding was leukocytosis, while hematocrit was normal. Abdominal radiography revealed the loss of abdominal serosal detail, which was diagnosed as peritoneal hemorrhage on ultrasonography (US) and aspiration. On computed tomography (CT), hepatic and splenic injuries were seen characterized by the discontinuity of the capsule with irregular margin and lower density lesions than parenchyma. No contrast enhancing area were shown in the laceration lesion. The accessory spleen was incidentally found by US and CT. In this case, acute intra-abdominal hemorrhage from splenic and hepatic laceration was diagnosed using CT.

A Case of Struvite Urolithiasis in a One-month-old Korean Sapsal Dog (1개월령 삽살개에서 발생한 Struvite요도결석 1례)

  • A one-month-old, 0.9 kg BW, male Korean Sapsal dog with anorexia, anuria, ataxia and depression was referred. On physical examination, the dog was showed tachycardia, hypothermia and pale mucous membrane. Hematologic values showed leukocytosis and mild anemia. Serum chemistry profile results revealed increased BUN (57.3 mg/dl) and $NH_3$ ($584\;{\mu}g/dl$), decreased albumin, sodium and potassium. Urinalysis showed hematuria, proteinuria, glucosuria, bacteriuria (Staphylococcus spp.) and magnesium ammonium phosphate crystalluria. On radiographic finding, urinary bladder was enlarged and 2 mm diameter radiopaque urolith was showed between os penis and prescrotal region. Urolithiasis was surgically corrected by urethrostomy.

Morphological and molecular finding of Longicollum pagrosomi (Acanthocephala: Pomphorhynchidae) in cultured red sea bream from Korea

  • Acanthocephalan worms were harvested from the posterior intestines of the cultured marine fishes from January to July 2016 in a fish market located in Daejeon metropolitan city, Republic of Korea. Totally 450 cultured fishes (rock fish 100, olive flounder 250, red sea bream 100) were surveyed. Of the 100 red sea bream fish, 37 fishes (37%) were diagnosed as positive for Longicollum pagrosomi by light, electron microscopic and molecular examination. The number of worm was 25~78 (mean $51{\pm}13$). However, we can't found any worm from the cultured rock fish and olive flounder. After sequencing, none of Pomphorhynchidae family are not identical based on 18S rRNA gene, and this data were identified with the first report for 18S rRNA gene sequence of L. pagrosomi. Furthermore, we confirmed that L. pagrosomi of the cultured red sea bream in Republic of Korea is very common parasite.

Effects of Running Training on Superoxide Production in Spontaneously Hypertensive Rats (트레드밀달리기 훈련이 자연발생고혈압쥐의 Superoxide 생성에 미치는 효과)

  • The purpose of this study was to examine effects of treadmill running training on superoxide product of spontaneously hypertensive rats. 20 males aged five week old rats were used for the experiments. They were randomly selected into experimental group (10 rats) and control group (10 rats) after being housed in laboratory for two weeks. The rats of experimental group performed treadmill exercise from 15 minutes for the first time to 35 minutes for the last one in order to familiar with experimental protocol ($VO_{2}max\;65{\sim}70%$, 0.9km/h, 5 times/week, 35 minutes/session). They began to perform treadmill running at the age of eight weeks for eight weeks. Superoxide production and NADPH oxidase activities were measured on the aortal from end of experiment. All statistical analyses and description methods were computed by SPSS Version 10.0. Differences in the values between experimental group and control group were calculated using t-tests. The results of this study were: there wewe no significant differences in superoxide production, NADPH oxidase-drivened superoxide activity, NADPH oxidase activity between two groups (p>.05). Spontaneously hypertensive rats of experimental group were characterized by higher superoxide production and lower NADPH oxidase-drivened superoxide activity and NADPH oxidase activity in comparison with the control group. One possible explanation of this finding was that NADPH oxidase activity of experimental group was lower than the control group. It can be concluded that treadmill running for eight weeks was associated positively with superoxide product of spontaneously hypertensive rats.

  • The aim of this article is to propose a framework for standardization of knowledge based on a case study of clinical operation to guide initial hospital knowledge management projects. Both quantitative and qualitative data for the project from 2009 through 2012 were collected for analysis. In a study of knowledge management project of a lab in S Hospital's Eye Center, this research examines the standardization of operational knowledge for competence enhancement of the lab. The lab defined and standardized the knowledge concern of the lab operation and the in-group from 2008. Organizations are often unaware of the potential value of tacit knowledge in both clinical and administrative area. The main finding from the case study is that analysis of such tacit knowledge of a lab in a hospital can be transformed into explicit knowledge for organizational capabilities such as improved quality, responsiveness and competitive advantage. Concepts from Nonaka's theory are used to investigate processes of knowledge standardization, examining the strategic value of tacit knowledge and the need to rely primarily on internal documentation processes. Since there has been limited research on hospital knowledge management. it may also be useful to researchers in generating propositions for further study.

  • The coadsorption of carbon monoxide and oxygen on polycrystalline nickel surface has been studied using XPS at the room temperaure. The adsorption of CO on the nickel surface precovered partially with oxygen is found to take place by the following steps: The CO molecules react with the preadsorbed oxygen atoms to liberate $CO_2$ gas at the initial stage of low CO exposures, and they are coadsorbed gradually with the increasing CO exposures. The extent of coadsorption at the higher CO exposures is found to decrease with the increasing degree of oxygen preadsorption. This finding is explained in terms of the reduced adsorption site for CO as a consequence of oxygen preadsorption. The CO molecules preadsorbed on the nickel surface inhibited the adsorption of $O_2$ molecules. The increase of oxygen exposure led to the dissociation of preadsorbed CO, and the NiO layers were formed concurrently. The dissociation was rendered to arise from an oxygen-to-CO energy transfer.

RFLP Analysis of cry1 and cry2 Genes of Bacillus thuringiensis Isolates from India

  • The PCR-RFLP method has been useful for detection of known genes and identification of novel genes. In the present study, degenerate primers were designed from five groups of cry1 genes for PCR-RFLP analysis. Bacillus thuringiensis (Bt) isolates from different regions were evaluated for PCR amplification of various cry1 genes using newly designed primers and cry2 genes using reported primers. PCR analysis showed an abundance of cry1A genes and especially cry1Ac genes in isolates from all regions. RFLP analysis revealed the presence of multiple cry1A genes in isolates from central and southern regions. Unique digestion patterns of cry1A genes were observed in isolates from each region. Few of the isolates represented a digestion pattern of cry1A genes that did match to any of the known cry1A genes. RFLP analysis suggested an abundance of cry2Ab along with a novel cry2 gene in Bt isolates from different regions of India. Sequence analysis of the novel cry2 gene revealed 95% sequence identity to cry2Ab and cry2Ah genes. Phylogenetic analysis revealed that the novel cry2 gene could have diverged earlier than the other cry2 genes. Our results encourage finding of more diverse cry2 genes in Bt isolates. Rarefaction analysis was used to compare cry1A gene diversity in isolates from different soil types. It showed a higher degree of cry1A gene diversity in isolates from central region. In the present study, we propose the use of novel degenerate primers for cry1 genes and the PCR-RFLP method using a single enzyme to distinguish multiple cry1A and cry2 genes as well as identify novel genes.

The Panax ginseng Flowering Locus T Shows Age Specific Expression Pattern in Ginseng and Increases Root Length in Transgenic Arabidopsis

  • Panax ginseng Meyer is a perennial medicinal plant, the roots of which has been used in the traditional formulations in Oriental countries. To understand its floral transition, we isolated Flowering Locus T (FT) from ginseng, the bioinformatics analysis of PgFT has revealed a considerable homology to the higher plants, with the essential amino acids for FT function are conserved. The phylogenetic analysis has shown that the PgFT is belonged to the shrub classes of plants and closest kin to Jatropha curcas FT. The expression profiling from juvenile (2-year-old) were abundant in leaves as well as in root and was concentrated in the secondary leaflet and stem bottom in adult (4-year-old) ginseng plant tissues, moreover PgFT transcript displayed photoperiod dependent oscillation. The ectopic expression of PgFT in Arabidopsis thaliana, exhibit precocious flowering and several floral pathway integrators were up-regulated, interestingly their root length was increased in the transgenic seedlings. Therefore, we could conclude that PgFT encodes a florigen that acts as a key regulator in the flowering pathway in ginseng and hypothesize that, it might involve in the underground organ development as well. We believe our finding could provoke future studies on the physiology and development in P. ginseng.

Verifications of the Impact-echo Technique for Integrity Evaluations of the Drilled Shaft Using Full Scale Tests (현장시험에 의한 충격반향기법의 말뚝 건전도 검사 적용성 평가)

  • The reflected signals from the defects of a pile and the boundaries between the pile and soils are analyzed to evaluate the integrity of drilled shafts in the impact-echo test. Signals varied according to both of the stiffness ratio of the pile to defects and that of the pile to surrounding soils. Model tests using the small size pile in the laboratory and numerical analyses have limitations in finding the characteristics of the signals due to different stress wave characteristics and unreliable modelling for the interaction between the pile and soils respectively. Full scale testing piles which have artificial defects are installed by the actual construction method and they were used to investigate the characteristics of reflected signals according to defects and the stiffness ratios of the pile to soils around.
