Stability Test and Quantitative and Qualitative Analyses of the Amino Acids in Pharmacopuncture Extracted from Scolopendra subspinipes mutilans

  • Objectives: Scolopendra subspinipes mutilans (S. subspinipes mutilans) is known as a traditional medicine and includes various amino acids, peptides and proteins. The amino acids in the pharmacopuncture extracted from S. subspinipes mutilans by using derivatization methods were analyzed quantitatively and qualitatively by using high performance liquid chromatography (HPLC) over a 12 month period to confirm its stability. Methods: Amino acids of pharmacopuncture extracted from S. subspinipes mutilans were derived by using O-phthaldialdehyde (OPA) & 9-fluorenyl methoxy carbonyl chloride (FMOC) reagent and were analyzed using HPLC. The amino acids were detected by using a diode array detector (DAD) and a fluorescence detector (FLD) to compare a mixed amino acid standard (STD) to the pharmacopuncture from centipedes. The stability tests on the pharmacopuncture from centipedes were done using HPLC for three conditions: a room temperature test chamber, an acceleration test chamber, and a cold test chamber. Results: The pharmacopuncture from centipedes was prepared by using the method of the Korean Pharmacopuncture Institute (KPI) and through quantitative analyses was shown to contain 9 amino acids of the 16 amino acids in the mixed amino acid STD. The amounts of the amino acids in the pharmacopuncture from centipedes were 34.37 ppm of aspartate, 123.72 ppm of arginine, 170.63 ppm of alanine, 59.55 ppm of leucine and 57 ppm of lysine. The relative standard deviation (RSD %) results for the pharmacopuncture from centipedes had a maximum value of 14.95% and minimum value of 1.795% on the room temperature test chamber, the acceleration test chamber and the cold test chamber stability tests. Conclusion: Stability tests on and quantitative and qualitative analyses of the amino acids in the pharmacopuncture extracted from centipedes by using derivatization methods were performed by using HPLC. Through research, we hope to determine the relationship between time and the concentrations of the amino acids in the pharmacopuncture extracted from centipedes.

Load Transfer Characteristics of Pile Foundation for Lightweight Pavement in Sand Soil using Laboratory Chamber Test (모형챔버시험을 이용한 사질토 지반의 경량포장체용 기초의 하중전달 특성)

  • In this study, small scaled (1/30) laboratory chamber tests of the pile foundation for a lightweight concrete pavement system were carried out to evaluate the safety of a pile foundation on sandy soil. The testing ground was simulated in the field and a standard pile-loading test was conducted. The test piles were divided into 3 types, Cases A, B and C, which is the location from the center of the slab by applying a vertical load. The interval between the piles was set to 8 cm. As a result of the pile foundation model test, the pavement settled when the vertical load was increased to 12kg from 1.5kg in sandy soil ground, particularly the maximum settlement of 0.04mm. Judging from the model chamber test, Case A showed compressive deformation, whereas Case B represented the compression and tensile forces with increasing vertical load. Case C showed an increase in tensile strain.

  • In this paper, a series of laboratory chamber tests was carried out to evaluate promoting consolidation of a porous concrete pile fabricated with recycled aggregates (RAPP) method for soft ground improvement. Performing the laboratory chamber tests for the RAPP, characteristics of the surface settlement with time and the consolidation time were compared with those of SCP and GCP provided by You (2009) under the same experimental condition. In addition, the experimental results were compared with the numerical analysis in this study. As a test result, the effects for settlement reduction in both the primary and the secondary consolidation and promoting consolidation by RAPP were prominent comparatively.

Characteristics of flow field of nose-only exposure chamber for inhalation toxicity evaluation (흡입독성평가를 위한 비부노출 챔버의 유동흐름 특성)

  • In this work, we evaluated the characteristics of flow field and uniformity of the nose-only exposure chambers for the inhalation toxicity test. Computational fluid dynamics (CFD) modeling was carried out to demonstrate uniformity of the nose-only exposure chambers. Because it is very important in the inhalation toxicity experiments that test materials are distributed uniformly to each holder of the chamber. The test was done with these 3 types of chamber with different form to develop inhalation toxicity evaluation system, easy-to-operate system among exposure chamber used for evaluating inhalation toxicity of environmental chemical mixtures. Through CFD interpretation, nose-only exposure chamber was made with the selection of the optimal conditions. For its evaluation, one type of fragrance was selected and measured particle size distribution of each port. The gene becoming luminous to green fluorescence was combined with GPT-SPE, a type of tGFP vector, to be inhaled to the mouse. Based on this, luminous intensity was checked. As a result, total particle number concentration of each port had average value of $3.17{\times}10^6{\sharp}/cm^3$ and range of the highest and lowest concentration value was approximately ${\pm}4.8%$. Autopsy of lung tissues of mouse showed that it had clearly better delivery of gene compared to the control group.

  • Antenna Passive Intermodulation (PIM) level measurement results are rarely credited to be due to external signal receiving characteristics of the antennas or serious effects from external PIM sources, such as the anechoic chamber absorber and antenna tower. This paper presents an antenna PIM model for a reflected PIM measurement method. Based on the findings of null point generation and the behavior of the third order PIM values obtained by theoretical predictions and experimental results, we concluded that the results of the antenna PIM level test were influenced by the external PIM sources generated by the anechoic chamber absorber and the path differences of PIM signals coming into the antenna.

Degradation of Coatings under Atmospheric Tropical Conditions

  • The weather resistance of five coatings systems based on alkyd, chlorinated rubber, epoxy, polyurethane and fluoropolymer were studied by natural exposure test and accelerated test. The coatings were exposed at Hanoi station with urban industry atmosphere and at Baichay station with marine atmosphere. The degradation of coatings was evaluated by gloss measurement and surface analysis by scanning electronic microscopy. The results obtained show that among coatings tested the gloss of polyurethane and fluoropolymer coatings remained highly and those of alkyd, chlorinated rubber and epoxy coatings were very low after two years of atmospheric exposure. Under accelerating conditions the gloss of fluoropolymer coatings remained highly after 80 cycles of testing. By comparison with accelerating test in UV-condensation chamber the conditions at atmospheric stations are more aggressive.

  • The purpose of this research is to assess the application of recycled-aggregate that is gained from construction wastes as the material of compaction pile method. At the same time, the development of the new technique rectifies defects of the existing compaction pile method for soft ground improvement. In this research, laboratory chamber tests were carried out analyzing the effect of the soft ground improvement by porous concrete pile using recycled aggregate. Through the results of the laboratory chamber tests, the variations of settlement, excess pore pressure, and increment of the vertical stress with time and the behavior of the composite ground were elucidated.

Effects of Hyperbaric Oxygen Intervention on Alcohol-Induced Oxidative Stress and Hangover in Hyperbaric Oxygen Chamber (고압산소챔버 중재가 알코올에 의한 산화적 스트레스 및 숙취 해소에 미치는 영향)

  • The purpose of this study is to evaluate the reliability and effectiveness of hyperbaric oxygen chamber on alcohol-induced oxidative stress and hangover. In order to evaluate them, 20 healthy adults were tested for blood and hangover stress tests. When an exponential change was examined after 3 interventions (hangover drink, hyperbaric oxygen chamber, and normal chamber as placebo effect) of 1 hour, the heart rate of subjects experiencing a hyperbaric oxygen chamber showed a statistically significant decrease (p < 0.001). The tests of blood alcohol concentration (BAC) as for exquisite hematology analysis, glucose, creatinine, and AST (aspartate aminotransferase) as well as ALT (alanine aminotransferase) used as liver-damage indicator show that the hyperbaric chamber has the effective effect. In the test of lactic acid, CRP (c-reactive protein), cortisol, and creatinine, the hyperbaric chamber shows much more excellent effect than the hangover drink and normal chamber as control groups. Further studies on hyperbaric oxygen chamber intervention including this study could be very helpful for improving lives of both the patients and healthy people.

Helium Leak Test for the PLS Storage Ring Chamber (포항가속기 저장링챔버의 헬륨누설검사)

  • The storage ring vacuum system for the Pohang Light Source (PLS) has been designed to maintain the vacuum pressure of $10^{-10}Torr$ which requires UHV welding to have helium leak rate less than $1{\times}10^{-10}Torr{\cdot}{\ell}/sec$. In order to develop new technique (PLS) welding technique), a prototype vacuum chamber has been welded by using Tungsten Inert Gas welding method and all the welded joints have been tested with a non-destructive method, so called helium leak detection, to investigate the vacuum tightness of the weld joints. The test was performed with a detection limit of $1{\times}10^{-10}Torr{\cdot}{\ell}/sec$ for helium and no detectable leaks were found for all the welded joints. Thus the performance of welding technique is proven to meet the criteria of helium leak rate required in the PLS Storage Ring. Both the principle and the procedure for the helium leak detection are also discussed.

An Analysis of Pile Foundation Load Transfer for Lightweight Pavement System in Clay Soil using Lab Chamber Test (모형챔버시험을 이용한 점성토 지반에 설치된 경량포장체용 기초의 하중전달 특성)

  • The main purpose of this study is to analyze and evaluate the feasibility of ligthweight pavement system with pile foundation on soft soil by laboratory small chamber test. In order to verify the stability of lightweight pavement system, the 1/30 scaled downed model system was tested at lab. The soft soil condition was simulated and group piles for skin friction resistance were used. Within the limited lab test, the settlements of pavement system were 0.86 mm for Case A, 0.70 mm for Case B, and 0.50 mm for Case C. The converted maximum settlement differential settlement were 25.8 mm and 10.8 mm. These values meet the inside of specification of Bridge Design Guide in Korea. The use of lightweight pavement systems on soft soils could be an alternative construction method on soft soils to reduce the challenges of conventional design and constructions.
