과메기에서 histamine 분해능을 나타내는 세균의 분리 동정 (Isolation and Identification of Histamine Degrading Bacteria from Kwamegi)

  • 과메기에서 histamine분해능을 가진 세균을 분리 동정하기 위하여 histamine만을 첨가한 제한배지에서 균을 분리했다. 기본적인 형태 및 생화학적 동정시험을 거쳐 10종의 시험균주를 선택하여 16S rDNA 염기서열 비교에 의한 계통발생학적 분석으로 동정하였다. 동정된 세균의 histamine분해능은 탁도측정법과 효소법에 의해 재 확인하였으며 그 실험 결과는 다음과 같다. 16S rDNA 염기서열 비교에 의한 계통 발생학적 분석을 이용하여 동정한 결과, E americana B791, Arthrobacter sp. R45S, H. marisflava, P. sp. 9B-7, Bacillus sp. LMG 21002, P. cibarius JG-220, B. megaterium KL-197 7종이 동정되었고, 각각 $99\%,\;95\%,\;98\%,\;99\%,\;99\%,\;99\%,\;98\%$의 상동성을 보였으며, 3종은 미동정되었다. 동정된 세균의 histamine분해능을 탁도측정법과 효소법에 의해 재 확인한 결과, Arthrobacter sp. R45S, H. marisflava, Bacillus sp. LMG 21002, B. megaterium KL-197이 분해능을 나타내었고, Psychrobacter sp. 9B-7은 미약한 분해능을 보였으며 미동정된 3종도 분해능이 있는 것으로 나타났다. 그리고 E. americana B791과 P. cibarius JG-220은 분해능이 불확실한 것으로 확인되었다 자반고등어에서 분리한 균주보다 시험균주 모두가 생육과 histamine의 분해속도가 비교적 빨랐다.

하수슬러지로부터 페놀분해세균의 분리 및 동정에 관한 연구 (Isolation and identification of a phenol-degrading bacterium from the sewage sludge)

  • 난지도 하수처리장내 슬러지로부터 페놀을 유일한 탄소 및 에너지원으로 이용하는 세균을 분리하여 그 특성을 조사하고 동정하였다. 본 세균은 그람 음성 구균으로 운동성을 가지며 페놀의 농도가 0.05%, 0.10%, 0.15%인 배지에서 성장을 보였으며, BBL Test를 실시한 결과 유사성을 가진 세균이 나타나지 않았다. 세균의 계통분류학적 분석을 위하여 세균의 염색체로부터 16s rDNA를 클로닝한 후 DNA 염기서열을 파악하고 이를 비교 및 분석한 결과 본 세균은 Proteobacteria의 Gamma (${\gamma}$) Subdivision에 속한 Xanthomonas Group의 Stenotrophomonas maltophilia strain과 99%의 유사성을 보이는 것으로 나타났다. 또한 Nitrogen-Fixing bacterium MAGDE3, Pseudomonas cissicola strain과는 98%의 상동성을, Stenotrophomonas sp. LMG 198, Xanthomonas cucurbitae과는 97%의 상동성을 보였다.

Caulobacter ginsengisoli sp. nov., a Novel Stalked Bacterium Isolated from Ginseng Cultivating Soil

  • A Gram negative, aerobic, nonspore-forming, straight or curved rod-shaped bacterium, designated Gsoil $317^T$, was isolated from soil of a ginseng field in Pocheon Province (South Korea) and was characterized using a polyphasic approach. Cells were dimorphic, with stalk (or prostheca) and nonmotile or nonstalked and motile, by means of a single polar flagellum. Comparative analysis of 16S rRNA gene sequences revealed that strain Gsoil $317^T$ was most closely related to Caulobacter mirabilis LMG $24261^T$ (97.2%), Caulobacter fusiformis ATCC $15257^T$ (97.1 %), Caulobacter segnis LMG $17158^T$ (97.0%), Caulobacter vibrioides DSM $9893^T$ (96.8%), and Caulobacter henricii ATCC $15253^T$ (96.7%). The sequence similarities to any other recognized species within Alphaproteobacteria were less than 96.0%. The detection of Q-10 as the major respiratory quinone and a fatty acid profile with summed feature 7 ($C_{18:1}\;{\omega}7c$ and/or $C_{18:1}\;{\omega}9t$ and/or $C_{18:1}\;{\omega}12t;$ 56.6%) and $C_{16:0}$ (15.9%) as the major fatty acids supported the affiliation of strain Gsoil $317^T$ to the genus Caulobacter. The G+C content of the genomic DNA was 65.5 mol%. DNA-DNA hybridization experiments showed that the DNA-DNA relatedness values between strain Gsoil $317^T$ and its closest phylogenetic neighbors were below 11%. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain Gsoil $317^T$ should be classified as representing a novel species in the genus Caulobacter, for which the name Caulobacter ginsengisoli sp. novo is proposed. The type strain is Gsoil $317^T$ (=KCTC $12788^T=DSM\;18695^T$).

흰쥐의 자궁에 대한 Tamoxifen과 성스테로이드 호르몬 상호작용에 관한 연구 (Studies on Interaction of Tamoxifen with Sex Steroid Hormones in Rat Uterus)

  • These studies were undertaken to examine the interaction of tamoxifen with sex steroid hormones in rat uterine activity. The uterine wet weights of the immature Tat uterus were examined after the administration of estradiol-l7$\beta$(1$\mu$g), tamoxifen(50$\mu$g), progesterone(lmg). The uterotropic activity in immature ovariectomized rats was observed under various treatment conditions following pretreatment with above drugs. The results obtained were as follows:1) Tamoxifen produced significant increase (p <0.01) in uterine wet weight compared with control group, although the increase was not as great as that seen with estradiol-17$\beta$. Administration of estradiol-17$\beta$ together with tamoxifen inhibited significantly the increase of uterine wet weight by estradiol-17$\beta$ (p < 0.01). Coadministration of progresterone with tamoxifen partly blocked the increase of tamoxifen-induced uterine wet weights by progesterone. 2) Estradiol-17$\beta$after the estradiol-17$\beta$ pretreatment discontinued the declining uterine wet weights due to the absence of estrogen support, but uteri continued to increase in weight if daily estradiol-17 $\beta$ was maintained. Administration of tamoxifen on the fourth day of estradiol-17$\beta$ treatment reduced uterine wet weights within 24 hours, and the weights continued to decline with additional tamoxifen. 3) The modest growth of the uterus induced by three daily injections of 5Opg tamoxifen remained stable for five days, with or without additional tamoxifen treatment. Coadministration of tamoxifen with estradiol17$\beta$ increased slightly the increase of uterine wet weight by tamoxifen. Coadministration of tamoxifen with progesterone inhibited the increase of uterine wet weight by tamoxifen. 4) The modest growth of the uterus induced by three daily injections of lmg progesterone reduced uterine wet weight to the control level for five days. Commencement of tamoxifen or estadiol-17 $\beta$ injections on the fourth day of progesterone treatment rapidly elevated uterine wet weight.

추출방법들에 의한 Malachite Green과 Leucomalachite Green 회수율 (Extraction Methods for Recovering Malachite Green and Leucomalachite Green)

  • Malachite green (MG), a triphenylmethane dye, is carcinogenic, mutagenic, teratogenic, a respiratory toxin, and causes chromosomal fractures. It is not permitted for use as an aquaculture veterinary drug in a number of countries. Sensitive extraction methods for MG and leucomalachite green (LMG), which have long residence times in fish tissues, were developed. For LMG, the average recovery of liquid extraction (LE) ranged from 41.71 (yellowtail) to 71.60% (snakehead); the recovery of liquid-liquid extraction (LLE) was between 67.68 (yellowtail) and 83.68% (snakehead); and the average recovery of solid-phase extraction (SPE) ranged from 84.16 (yellowtail) to 92.92% (shrimp). The recovery of MG was less than 30% with SPE. However, the dye is found primarily as the colorless reduced leuco form in fish tissues.

유럽낙엽송의 형성층조직 배양으로부터 줄기의 재분화 (Shoot Regeneration from Cambial Tissue Culture of European Larch (Larix decidua))

  • 3년생 유럽낙엽송의 실생묘로부터 형성층조직을 분리하여 부정아를 유기하였다. BLG배지에 4.5$\mu$M의 BA가 첨가된 배지에서 형성층 절편체 당 약 25개의 부정아가 형성되어 시험된 여러 가지 BA와 NAA농도 중 최고의 발생율을 보였다. 유기된 부정아는 1/2 GD배지와 LMG배지에서 10주 후 약 2.5cm의 신장을 보였다. GD배지에서는 이보다 낮은 약 1,8cm의 신장을 나타내었다. 발근은 NAA가 $0.54\;\mu\textrm{M}$ 함유된 1/2 GD배지에 2주간 배양하였을때 20% 정도 나타났다. 발근된 shoot들은 NAA가 함유되지 않은 1/2 GD 배지에서 뿌리를 신장시킨 후 온실로 옮겨져 순화되었다. 본 실험의 결과 그동안 낙엽송의 조직배양시료로 사용되지 않았던 형성층 조직의 뛰어난 형태형성능을 보여줌으로써 기내 급속대량증식 뿐 아니라 향후 낙엽송의 체세포 변이주선발이나 형질전환 및 그와 관련된 연구분야에 유용하게 사용될 수 있을 것으로 사료된다.

Isolation and Identification of Lactobacillus kimchicus sp.nov and Bioconversion of Ginseng Saponin

  • Ginseng contained many different kinds of saponin which was the most valuable for people, but its yield cannot satisfy the demand using traditional extract methods. Enzyme transformation is a conformable and highly performed method which was fit for today. A ${\beta}$-glucosidase producing bacterium ($DCY51^T$) was isolated from Korean fermented-vegetable food kimchi. The 16S rRNA gene sequence analysis revealed that the strain $DCY51^T$ belongs to the genus Lactobacillus. The highest sequence similarity was found with Lactobacillus paracollinoides LMG $22473^T$ and Lactobacillus collinoides LMG $9194^T$ with levels of 16S rDNA similarity of 97.4% and 97.3%, respectively. Based on the above results the strain $DCY51^T$ placed in the genus Lactobacillus and proposed a new species, Lactobacillus kimchicus sp. nov. $DCY51^T$ (= KCTC $12976^T$ = JCM $15530^T$). It was culture solution reacted with Red Ginseng extract and $Rb_1$, respectively. The medium of bacteria was the liquid of MRS, the temperatures of growing and reacting between bacteria liquid and saponin were samely $37^{\circ}C$, there spective reacting time were 12 hours and 48 hours. Thus we got different saponins, and TLC and HPLC analysis showed that: enzyme respectively reacted with $Rb_1$ and Red Ginseng extract got the transformed saponin, respectively. The polarity position in TLC was a little higher than Rd; and the polarity position was the same as that of Compound K's, the saponin obtained from HPLC and other experimental results was not Compound K. The constitution of its saponin was hoped to be further confirmed.

감귤과피를 함유하는 유과의 품질특성 (Quality Characteristics of Yukwa with Citrus Peel Powder)

  • 유과의 품질을 향상과 기능성 부여를 목적으로 감귤과피(0, 2, 4 및 6%)를 첨가하여 제조한 유과의 품질특성을 조사하였다. 팽화율은 대조구(1,740%)보다 첨가구(855~l,517%)에서 낮았다. 감귤과피의 첨가량이 증가할수록 경도는 증가한 반면 아삭아삭한 정도는 감소하였으나 전통유과의 텍스쳐 범위에 속하였다. 색상은 대조구(L*값 70, a*값 3.30, b*값 12.6)의 경우는 백색을 띄었으나 첨가구(L*값 63.3~49.9, a*값 10.6~17.5, b*값 12.6~56.4)는 연황~황색을 띄었다. 감귤과피를 첨가한 유과의 carotenoid, hesperidin 및 naringin의 함량은 각각 0.18~0.5lmg%, 36.55~101.60mg%, 24.65~70.8lmg%범위였다. 관능검사에 의하여 펑가된 색상에 대한 기호도 및 종합적 기호도는 감글 과피륵 4% 첨가한 유과에서 가장 양호하였으며 감귤의 향긋한 냄새가 있었다. 그러나 대조구와 첨가 구간의 단맛과 신맛은 유의적인 차이를 보이지 않았다.

이상유영 증상 국내산 양식 넙치에서의 슈도알테로모나스 분리·동정 및 항생제 감수성 연구 (Isolation and Identification from Korean Olive Flounders (Paralichthys olivaceus) Showing Abnormal Swimming Behavior, and Sstudy of Antibiotic Susceptibility)

  • 2020년 11월, 국내의 넙치 양식장에서 양식 중이던 넙치가 이상 유영 소견을 보이다가, 지속적으로 폐사하였다. 질병 진단 과정 중, 폐사어의 신장에서 세균(KNCFKW-PN1)이 분리되었다. gyrase B subunit 유전자의 시퀀스 분석 결과, KNCFKW-PN1 분리주는 기존에 보고된 LMG 2227T 균주의 해당 유전자 시퀀스와 99.59% 유사도를 보여 Pseudoalteromonas nigrifaciens 로 동정이 되었다. 항생제 감수성 실험 결과에 따르면, KNCFKW-PN1 분리주는 ciprofloxacin에 대하여 중등도의 내성을 나타내었고, ampicillin, cefepime, cefotaxime, ceftazidime, amikacin에 내성을 나타내었다. 본 사례는 다제 내성 Pseudoalteromonas nigrifaciens 세균이 넙치로부터 분리된 최고의 보고이다.

Comparative Genome analysis of the Genus Curvibacter and the Description of Curvibacter microcysteis sp. nov. and Curvibacter cyanobacteriorum sp. nov., Isolated from Fresh Water during the Cyanobacterial Bloom Period

  • The three Gram-negative, catalase- and oxidase-positive bacterial strains RS43T, HBC28, and HBC61T, were isolated from fresh water and subjected to a polyphasic study. Comparison of 16S rRNA gene sequence initially indicated that strains RS43T, HBC28, and HBC61T were closely related to species of genus Curvibacter and shared the highest sequence similarity of 98.14%, 98.21%, and 98.76%, respectively, with Curvibacter gracilis 7-1T. Phylogenetic analysis based on genome sequences placed all strains within the genus Curvibacter. The average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between the three strains and related type strains supported their recognition as two novel genospecies in the genus Curvibacter. Comparative genomic analysis revealed that the genus possessed an open pangenome. Based on KEGG BlastKOALA analyses, Curvibacter species have the potential to metabolize benzoate, phenylacetate, catechol, and salicylate, indicating their potential use in the elimination of these compounds from the water systems. The results of polyphasic characterization indicated that strain RS43T and HBC61T represent two novel species, for which the name Curvibacter microcysteis sp. nov. (type strain RS43T =KCTC 92793T=LMG 32714T) and Curvibacter cyanobacteriorum sp. nov. (type strain HBC61T =KCTC 92794T=LMG 32713T) are proposed.
