Face Image Synthesis using Nonlinear Manifold Learning (비선형 매니폴드 학습을 이용한 얼굴 이미지 합성)

  • This paper proposes to synthesize facial images from a few parameters for the pose and the expression of their constituent components. This parameterization makes the representation, storage, and transmission of face images effective. But it is difficult to parameterize facial images because variations of face images show a complicated nonlinear manifold in high-dimensional data space. To tackle this problem, we use an LLE (Locally Linear Embedding) technique for a good representation of face images, where the relationship among face images is preserving well and the projected manifold into the reduced feature space becomes smoother and more continuous. Next, we apply a snake model to estimate face feature values in the reduced feature space that corresponds to a specific pose and/or expression parameter. Finally, a synthetic face image is obtained from an interpolation of several neighboring face images in the vicinity of the estimated feature value. Experimental results show that the proposed method shows a negligible overlapping effect and creates an accurate and consistent synthetic face images with respect to changes of pose and/or expression parameters.

A Simple and Efficient Method to Determine Rivaroxaban in Rat Plasma Using Liquid-Liquid Extraction and LC-MRM

  • Rivaroxaban (RRN) is the first available active direct factor Xa inhibitor (anticoagulant) with oral administration. Due to its success in market, there have been efforts to develop various RRN formulations, and the development of good analytical methods for its in vivo evaluation is an essential prerequisite. Thus, here, a simple and efficient method to determine RRN in rat plasma using liquid-liquid extraction (LLE) and liquid chromatography and multiple reaction monitoring (LC-MRM) was presented. The use of ethyl acetate as the LLE solvent results appropriate extraction and purification of RRN and it also helps the significant reduction of rat plasma volume required for RRN quantitation. The developed method showed good analytical performance including specificity, linearity ($r^2{\geq}0.999$ within 0.5 - 500 ng/mL), sensitivity (the lower limit of quantitation at 0.5 ng/mL), accuracy (89.3 - 107.0%), precision (${\geq}12.7%$), and recovery (89.2 - 105.7%). Additionally, RRN in sample extracts showed good stability. Finally, the applicability of the validated method to the PK evaluation of RRN was confirmed after its oral administration to normal rats. The present method is the first analytical method employing LLE for the simple and efficient extraction and purification of RRN in rat plasma. Therefore, the present method can contribute to the development of new RRN formulations as well as to the monitoring of RRN in special clinical situations through its efficient determination in various samples with or without minor modification.

A Sensitive, Efficient, and Cost-Effective Method to Determine Rotigotine in Rat Plasma Using Liquid-Liquid Extraction (LLE) and LC-MRM

  • Rotigotine (RTG) is a non-ergot dopamine agonist used to manage the early stage of Parkinson's disease (PD) as transdermal patch. However, the poor medication compliance of PD patients and skin issues related with repeated applications of RTG patches lead to the search for alternative formulations and it also requires appropriate analytical methods for their in vivo evaluation. Thus, here, a sensitive, efficient, and cost-effective method to determine RTG in rat plasma using liquid-liquid extraction (LLE) and multiple reaction monitoring was developed. The use of 20 µL of rat plasma for sample treatment, 8-OH-DPAT as the internal standard, and methyl tert-butyl ether as the LLE solvent in the present method gives it advantages over previous methods for the analysis of RTG in biological samples. The good analytical performance of the developed method was confirmed in specificity, linearity (the coefficient of determination ≥0.999 within 0.1-100 ng/mL), sensitivity (the lower limit of quantitation at 0.1 ng/mL), accuracy (81.00-115.05%), precision (≤10.75%), and recovery (81.00-104.48%) by following the FDA guidelines. Finally, the applicability test of the validated method to the in vivo evaluation of a RTG formulation showed that the present method is the only method which can be accurately applied to that longer than 24 hours, critical for the development of formulations with reduced dosing frequencies. Therefore, the present method could contribute to the development of new RTG formulations helpful to people suffering from PD.

Rapid and Simple Analytical Method for Removing Patulin from Apple Juice Using Solid Phase Extraction (고체상 추출법(SPE: Solid Phase Extraction)을 이용한 국내 사과주스 중 Patulin 간편.신속 분석방법)

  • Patulin, a secondary metabolite of mold, is commonly found in rotten apples. Many countries regulate patulin at levels ranging from 30 to $50\;{\mu}g/L$. Most analytical methods for removing patulin from apple juice include liquid-liquid extraction (LLE), which is time and labor intensive. To replace the LLE method, a solid-phase extraction (SPE) method has been developed for apple juice and unfiltered apple juice. A portion of the test sample was applied to a macroporous copolymer cartridge and washed with 5 mL of 1% sodium bicarbonate, followed by 5 mL of 1% acetic acid. Patulin was eluted with 5 mL of 2% acetonitrile in anhydrous ethyl ether. The mobile phase was tetrahydrofuran in water (0.8:99.2) and was detected with a UV detector at 276 nm. Recoveries ranged from 95 to 101% in test samples, and the minimum detectable level was 30 ppb. Because this SPE method is fast, easy, reliable, and inexpensive, it could be applicable for companies or analytical agencies to analyze patulin concentrations in apple juice.

An extraction method and residues of benzimidazole fungicides in soybean sprouts (콩나물중 benzimidazole계 농약의 추출방법과 잔류성)

  • An analytical method was approached to determine the residues of benzimidazole fungicides in soybean sprouts and was compared with traditional solvent-based method of extraction, solid phase extraction(SPE) and matrix solid phase dispersion(MSPD). The recoveries of carbendazim for liquid-liquid extraction (LLE), SPE and MSPD in soybean sprouts were $69.25{\sim}79.16$, $79.99{\sim}90.42$ and $92.40{\sim}98.48%$, respectively. The recoveries of thiabendazole for LLE, SPE and MSPD in soybean sprouts were $64.86{\sim}83.26$, $79.12{\sim}94.04$ and $65.44{\sim}73.92%$, respectively. The residues of carbendazim by elapsed time under our cultivated soybean sprouts program found $0.10{\sim}0.25$ mg/kg after 5 days and trace concentration of carbendazim was detected after 7 days. Of the 448 samples analyzed, less than 1.5% had residues of carbendazim ranged from 0.05 to 0.37 mg/kg.

Establishment of Analytical Method for Pymetrozine Residues in Crops Using Liquid-Liquid Extraction(LLE) (액-액 분배법을 활용한 작물 중 pymetrozine의 잔류분석법 확립)

  • Polar pesticides like pymetrozine (log $P_{ow}$: -0.18) are known to be difficult to analyze. The analytical method of pymetrozine using hydromatrix included in the official method of KFDA was uncommon and provided ambiguous evidence to confirm both the identity and the quantity. Therefore, precise single residue analytical method was developed in representative crops for using liquid-liquid extraction (LLE). The pymetrozine residue was extracted with methanol from 11 representative crops which comprised apple, blueberry, broccoli, cabbage, cherry, crown daisy, hulled rice, Korean cabbage, potato, rice and watermelon. The extract was purified serially by liquid-liquid extraction (LLE) and silica solid phase extraction (SPE). For rice and hulled rice samples, n-hexane partition was additionally adopted to remove nonpolar interferences, mainly lipids. The residue levels were analyzed by HPLC with DAD, using $C_8$ column. LOQ (limit of quantitation) of pymetroizinie was 1 ng (S/N > 10) and MQL (method quantitation limit) was 0.01 mg/kg. Mean recoveries from 11 crop samples fortified at three levels (MQL, 10 ${\times}$ MQL and 50 ${\times}$ MQL) in triplicate were in the range of 83.1~98.5% with coefficients of variation (CV) of less than 10%, regardless of sample type, which satisfies the criteria of KFDA. The method established in this study could be applied to most of crops as an official and general method for analysis of pymetrozine residue.

  • In order to find an useful condition under which the mutants defective in sexual development could be isolated, the effects of several cultural conditions on the developments of Aspergillus nidulans were examined. Among several conditions found to restrict the asexual sporulation but enhance the sexual process, the interference of aeration by sealing the plates with sealing film was the most useful one for the purpose of mutant isolation. Sealing at any time before 20 hours from inoculation prevented both sexual and asexual process. When the seal was removed after 24 hours, however, the mycelia developed only to sexual organs. Using this properity, the early morphogenic process of sexual development could be easily observed and a number of mutants that showed some defects in the process could be isolated. The mutants were divided into 3 groups, NSD (never in sexual development), BSD (block in sexual development) and ASD (abnormal in sexual development). NSD mutants never produced either the $H{\ddot{u}}lle$ cells or cleistothecia and some produced the asexual organs even when the aeration was restricted. BSD mutants were blocked in the process of $H{\ddot{u}}lle$ cell, cleistothecia, crozier, asci or ascospore formation. ASD mutants had defects in the amount of cleistothecia, time of cleistothecial maturation or color of ascospores.

Determination of Corticosteroids in Moisturizers by LC-MS/MS

  • Simultaneous determination of three corticosteroids (clobetasol propionate, betamethasone dipropionate, fluticasone propionate) in moisturizers was performed by using liquid chromatography (LC) coupled with tandem mass spectrometry (MS/MS). Sample preparation was conducted by the liquid-liquid extraction (LLE). Moisturizers include emulsifying agent and it forms micelles. In order to improve the extraction efficiency of corticosteroids trapped in micelle, newly developed-optimized extraction conditions which can remove the matrix effect from moisturizers was applied with various pH conditions in LLE extraction stage of sample preparation. Thus, the addition of 10 μL of 1 M HCl into moisturizers sample before extraction could improve the extraction efficiency. For the quantitative analysis, SRM table that contained specific transition of all of target corticosteroids was created. The developed method was validated for linearity, accuracy, precision, limit of detection (LOD), limit of quantization (LOQ) and recovery. Over the 0.99 r2 value was obtained in calibration standard range. Effective accuracy and precision were also obtained. LODs were below 31 ng/mL and LOQs were estimated below 94 ng/mL for all corticosteroids tested.

A Novel Approach of Feature Extraction for Analog Circuit Fault Diagnosis Based on WPD-LLE-CSA

  • The rapid development of large-scale integrated circuits has brought great challenges to the circuit testing and diagnosis, and due to the lack of exact fault models, inaccurate analog components tolerance, and some nonlinear factors, the analog circuit fault diagnosis is still regarded as an extremely difficult problem. To cope with the problem that it's difficult to extract fault features effectively from masses of original data of the nonlinear continuous analog circuit output signal, a novel approach of feature extraction and dimension reduction for analog circuit fault diagnosis based on wavelet packet decomposition, local linear embedding algorithm, and clone selection algorithm (WPD-LLE-CSA) is proposed. The proposed method can identify faulty components in complicated analog circuits with a high accuracy above 99%. Compared with the existing feature extraction methods, the proposed method can significantly reduce the quantity of features with less time spent under the premise of maintaining a high level of diagnosing rate, and also the ratio of dimensionality reduction was discussed. Several groups of experiments are conducted to demonstrate the efficiency of the proposed method.

Comparative Study of Dimension Reduction Methods for Highly Imbalanced Overlapping Churn Data

  • Retention of possible churning customer is one of the most important issues in customer relationship management, so companies try to predict churn customers using their large-scale high-dimensional data. This study focuses on dealing with large data sets by reducing the dimensionality. By using six different dimension reduction methods-Principal Component Analysis (PCA), factor analysis (FA), locally linear embedding (LLE), local tangent space alignment (LTSA), locally preserving projections (LPP), and deep auto-encoder-our experiments apply each dimension reduction method to the training data, build a classification model using the mapped data and then measure the performance using hit rate to compare the dimension reduction methods. In the result, PCA shows good performance despite its simplicity, and the deep auto-encoder gives the best overall performance. These results can be explained by the characteristics of the churn prediction data that is highly correlated and overlapped over the classes. We also proposed a simple out-of-sample extension method for the nonlinear dimension reduction methods, LLE and LTSA, utilizing the characteristic of the data.
