The Effects of Different Wavelengths of Light-Emitting Diodes on the Expression of Reproduction-Related Genes in Goldfish Carassius auratus

  • We investigated the differences in the expression of the neurohormones kisspeptin (Kiss) and gonadotropin-inhibitory hormone (GnIH) and cytochrome P450 aromatase (P450arom), gonadotropin hormones (GTHs), and sex steroids in the goldfish Carassius auratus exposed to light-emitting diodes (LEDs). The expression levels of Kiss1, Kiss2, G-protein-coupled receptor 54 (GPR54), GTHs, GnIH, and P450arom were compared between the control (white light) and LED-treated goldfish. Furthermore, we measured the plasma levels of follicle-stimulating hormone (FSH) and luteinizing hormone (LH). The levels of Kiss1 mRNA and protein; Kiss2, GPR54, and $GTH{\alpha}$ protein; GTH mRNA; and plasma FSH and LH in the hypothalamus and cultured hypothalamus cells were significantly higher in the green and purple LED treatment groups than in the other groups. These results suggested that red LEDs inhibit the sex maturation hormones, Kiss, GPR54, GTHs, and P450arom, and that GnIH plays a role in the negative regulation of reproductive function in goldfish.

  • Purpose : This study is to examine what are the effects of the Guisinhwan(GSH) on the ovulation and ovary in rats. Methods : 4weeks Female Sprague-Dawley 12 rats of weighting 160-l80g, were divided into three groups including the GSH oral administration(4ml/kg) groups(4heads) and GSH oral administration(8ml/kg) groups(4heads). Then we observed changes in the serum concentrations of FSH, LH, and estradiol($E_2$) and the histological changes of ovary and the immunohistochemical staining for progesterone receptor in ovary of rats. Results : 1. GSH didn't make a difference as compared with control group in serum FSH level. 2. GSH didn't make a difference as compared with control group in serum LH level. 3. GSH significantly increased serum $E_2$ level. 4. GSH significantly increased ovulation in histological observations of ovary. 5. GSH tended to decrease immunohistochemical staining score (ISS) of atretic follicles in immunohistochemical staining for progesterone receptor in ovary. Conclusion : GSH influences ovary to increase the ovulation of rats.

Effect of DDT on Testosterone Production by Modulator Aromatase (CYP 19) in R2C

  • Various pesticides known or suspected to interfere with steroid hormone function were screened toy effects in leydig cells on catalytic activity and mRNA expression of aromatase. Dichlorodiphenyltrichloroethane (DDT) is a widespread environmental pollutant. In this study, we investigated the effect of DDT on testosterone production through aromatase activity and its molecular mechanism in testicular leydig cell, R2C by using radioimmunoassay (RIA). As the results, the potent leydig: cell activator LH increased testosterone production compared to the control. DDT exposure significantly decreased testosterone production in R2C cell. In addition, DDT was found to increase aromatase gene expression and activity in R2C cell in a dose dependent manner. In order to assess whether the suppressive effects of DDT on LH-inducible testosterone (T) production might be influenced by the ER, ICI 182.780 was used, and it was found that these inhibitory effects of DDT were antagonized by ICI 182.780, implying that the estrogen receptor (ER) mediates the suppressive effects of DDT. Furthermore, the inducible effects of DDT on aromatase gene expression might be influenced by the ER, ICI 182.780 was used, and it was found that these enhancing effects of DDT were antagonized by ICI 182.780, implying that the ER mediates the inducible effects of DDT. Our results indicated that DDT inhibition of luteinizing hormone (LH) -inducible T production in R2C cell is mediated through aromatase. However, the precise mechanisms by which DDT enhance in R2C cell remains unknown. The current study suggests the possibility that DDT might act as a modulator aromatase gene transcription.

Biological Functions of the COOH-Terminal Amino Acids of the $\alpha$-Subunit of Tethered Equine Chorionic Gonadotropin

  • Glycoprotein hormones have a common $\alpha$-subunit that is involved in the signaling pathway together with G protein, adenylcyclase and cAMP induction; however, it is an unclear how this common structure is related to hormonal action. To determine the biological functions of the COOH-terminal amino acids in the $\alpha$-subunit of these glycoprotein hormones, a tethered-molecule was constructed by fusing the $NH_2$-terminus of the $\alpha$-subunit to the COOH-terminus of the $\beta$-subunit of equine chorionic gonadotropin (eCG). The following deletion mutants were created by PCR; Ile was inserted at position 96 to form ${\Delta}96$, Lys was substituted at position 95 to form ${\Delta}95$, His was inserted at position 93 to form ${\Delta}93$ and Tyr was substituted at position 87 to form ${\Delta}87$. Each mutant was transfected into CHO-K1 cells. Tethered-wt eCG, and ${\Delta}96$, ${\Delta}95$, and ${\Delta}93$ mutants were efficiently secreted into the medium but the ${\Delta}87$ mutant was not secreted. Interestingly, the RT-PCR, real-time PCR, and northern blot analyses confirmed that the RNA was transcribed in the ${\Delta}87$ mutant. However, the ${\Delta}87$ mutant protein was not detected in the medium or the intracellular fraction of the cell lysates. The LH- and FSH-like activities of the recombinant proteins were assayed in terms of cAMP production using rat LH/CG and rat FSH receptors. The metabolic clearance rate (MCR) was determined by injecting rec-eCG (2 IU) into the tail vein. The ${\Delta}95$ and ${\Delta}93$ mutants were completely inactive in both the LH- and FSH-like activity assays. The ${\Delta}96$ mutant showed slight activity in the LH-like activity assay. In comparison to the wild type, the activity of the ${\Delta}96$ mutant in the FSH-like activity assay was the highest among all the mutants. The MCR assay in which rec-eCG was injected showed a peak at 10 min in all the treatment groups, which disappeared 4 h after injection. These results imply a direct interaction between the receptor and the COOH-terminal region of the a-subunit. The data also reveal a significant difference in the mechanism by which the eCG hormone interacts with the rLH and rFSH receptors. The COOH-terminal region of the $\alpha$-subunit is very important for the secretion and functioning of this hormone.

Expression and Regulation of Gonadotropin-Releasing Hormone(GnRH) and Its Receptor mRNA Transcripts During the Mouse Ovarian Development

  • The present study examines the expression and regulation of gonadotropin-releasing hormone (GnRH) and its receptor (GnRH-R) mRNA levels during mouse ovarian development. A fully processed, mature GnRH mRNA together with intron-containing primary transcripts was expressed in the immature mouse ovary as determined by Northern blot analysis and reverse transcription-polymerase chain reaction (RT-PCR). The size of ovarian GnRH mRNA was similar to that of hypothalamus, but its amount was much lower than that in the hypothalamus. Quantitative RT-PCR procedure also revealed the expression of GnRH-R mRNA in the ovary, but the estimated amount was a thousand-fold lower than that in the pituitary gland. We also examined the regulation of ovarian GnRH and GnRH-R mRNA levels during the follicular development induced by pregnant mare's serum gonadotropin (PMSG) and/or human chorionic gonadotropin (hCG). Ovarian luteinizing hormone receptor (LH-R) mRNA was abruptly increased st 48 h after the PMSG administration and rapidly decreased to the basal level thereafter. Ovarian GnRH mRNA level was slightly decreased at 48 h after the PMSG administration, and then returned to the basal value. GnRH-R mRNA level began to increase at 24 h after the PMSG treatment, decreased below the uninduced basal level at 48 h, and gradually increased thereafter. HCG administration did not alter ovarian GnRH mRNA level, while it blocked the PMSG-induced increase in GnRH mRNA level. Taken together, the present study demonstrates that the expression of GnRH and GnRH-R mRNA are regulated by gonadotropin during follicular development, suggesting possible intragonadal paracrine roles of GnRH and GnRH-R in the mouse ovarian development.

암컷 흰쥐의 사춘기 개시에 미치는 di(2-ethyl hexyl)phthalate(DEHP)의 효과 (Effect of Di(2-ethyl hexyl)phthalate(DEHP) on the Onset of Puberty in Female Rat)

  • 전세계적으로 매년 대량생산되는 프탈레이트(phthalate)류 물질은 플라스틱처럼 다양한 생활용품의 원료로 사용되기 때문에 인간에 대한 노출 정도가 매우 높은 산업 물질이다. 대표적으로 di(2-ethyl hexyl)phthalate(DEHP)와 그 활성 대사물인 monoethyl hexyl phthalate(MEHP)가 내분비계 장애물질(endocrine disruptor)로 작용하여 인간과 실험 동물의 생식계를 교란한다는 증거들이 급속히 축적되고 있다. 본 연구는 DEHP가 암컷 흰쥐의 사춘기 개시와 이와 관련된 생식 지표들의 변화에 미치는 효과를 조사한 것이다. 생후 25 일부터 corn oil 또는 DEHP(100mg/kg/day i.p.)를 사춘기 개시의 지표인 질구 개방(vaginal opening, VO)이 일어나는 날까지 투여하였다. 다음 날 희생시킨 후 난소와 자궁의 외양과 조직 무게를 측정하여 DEHP가 이들 조직에서의 세포증식에 미치는 효과를 조사하였고, 현미경 관찰을 통해 해부학적인 변화 양상을 조사하였다. 또한 혈액내의 LH 수준을 방사면역측정법(RIA)로 측정하였다. 자궁 성숙의 지표인 프로게스테론 수용체(PR) 발현 양상은 정량적인 RT-PCR로 확인하였다. 결과로, DEHP 투여에 의해 질구 개방이 지연됨을 확인하였다($35.3{\pm}0.7\;day$ in control group vs $37.3{\pm}0.7\;day$ in DEHP group, p<0.05). 흥미롭게도, 첫번째 주사 다음 날(p<0.05)부터 이후 마지막 주사 날(p<0.01)까지 DEHP 군의 체중이 대조군에 비해 유의하게 낮음을 관찰하였다. 난소와 자궁의 크기와 무게, 그리고 혈중 LH 수준도 대조군에 비해 DEHP 군에서 유의하게 감소하였다. 질구 개방이 일어난 대조군의 난소에서는 성숙의 지표인 그라프 난포(Graafian follicle)와 황체가 관찰되었으나 DEHP 군의 난소에서는 미성숙한 1차, 2차 난포 그리고 작은 퇴화 난포(atretic follicle)들만이 다수 관찰되었다. 자궁의 경우도 대조군에서는 내막층과 근막층은 물론 상피층까지 잘 발달된 과다 성장 상태와 함께 분비선 수의 증가가 나타났으나 DEHP 군에서는 모든 세포층의 발달이 미약한 상태였다. 이러한 결과는 DEHP가 과립세포에서의 에스트로겐 합성 분비에 억제적 효과를 나타내는 것에 기인하는 것으로 보인다. RT-PCR 결과 난소(p<0.05)와 자궁(p<0.01)에서의 PR mRNA 수준은 DEHP 투여에 의해 유의한 발현 감소가 나타났다. 결론적으로, 본 연구는 사춘기 전 특정 시기에 단기적인 DEHP 투여에 의해 암컷 흰쥐의 생식계 활성화가 억제되고, 그 결과 사춘기가 지연될 수 있음을 시사한다.

  • Biogenic amine 류에는 catecholamine, indoleamine 그리고 histamine이 있으며, 동물의 다양한 생리 현상과 행동양식 조절에 중요한 역할을 담당한다. 이중 indoleamine 류인 세로토닌(serotonin혹은 5-hydroxytryptamine, 5-HT)의 경우 최근 들어 그 수용체 아형의 유전자 규명과 발현 조절, 중추신경계 및 표적 기관에서의 역할, 특히 항우울 효과와 같이 행동 및 심리적인 영향 등에 대해 광범위하게 연구되고 있다. 본 논문은 5-HT의 합성 경로, 수용체 아형, 생식과 관련된 기능에 대해 서술하였다. 특히 생식과 관련된 5-HT의 신경내분비학적인 역할로 GnRH-LH-sex steroids 축 조절 기능과 이에 관여하는 수용체 아형들, 생식 조절에 있어서 5-HT 효과의 성적 이형 양상, 중추신경계 이외 기관에서의 역할, 성행동의 조절에 대한 연구 결과들을 요약하였다. 5-HT의 기능에 대한 연구는 특히 현재 주목 받고 있는 '삶의 질' 고양과 밀접하게 연관되어있는 유망한 주제 가운데 하나로 판단된다.

Testosterone secretion is affected by receptor tyrosine kinase c-Kit and anoctamin 1 activation in mouse Leydig cells

  • Receptor tyrosine kinase c-Kit, a marker found on interstitial cells of Cajal (ICCs), is expressed in Leydig cells, which are testicular interstitial cells. The expression of other ICC markers has not yet been reported. In this study, we investigated the expression of c-Kit and anoctamin 1 (ANO1), another ICC marker, in mouse testes. In addition, the relationship between c-Kit and ANO1 expression and Leydig cell function was investigated. We observed that c-Kit and ANO1 were predominantly expressed in mouse Leydig cells. The mRNA and protein of c-Kit and ANO1 were expressed in TM3, a mouse Leydig cell line. LH induced an increase in intracellular Ca2+ concentration, membrane depolarization, and testosterone secretion, whereas these signals were inhibited in the presence of c-Kit and ANO1 inhibitors. These results show that c-Kit and ANO1 are expressed in Leydig cells and are involved in testosterone secretion. Our findings suggest that Leydig cells may act as ICCs in testosterone secretion.

수컷 흰쥐 뇌하수체의 생식소자극호르몬 발현에 미치는 Ethane 1,2-Dimethane Sulfonate(EDS)의 효과 (Effect of Ethane 1,2-Dimethane Sulfonate(EDS) on the Expression of Pituitary Gonadotropin in Male Rats)

  • Ethane 1,2-dimethane sulfonate(EDS)은 Leydig cells(LC)만을 선별적 사멸을 유도하는 약물로서 가역적인 테스토스테론(testosterone, T) 결핍 흰쥐 모델을 만드는데 널리 사용된다. 본 연구에서는 수컷 흰쥐 뇌하수체의 생식소자극호르몬인 LH와 FSH의 발현에 미치는 EDS 투여 효과를 조사하였다. 성숙한 수컷 흰쥐(SD strain, $300{\sim}350\;g$ B.W.)에 EDS(75 mg/kg, i.p.)를 1회 복강주사하고 주사 후 0, 1, 2, 3, 4, 5, 6 그리고 7주가 경과한 날 희생시켰다. 뇌하수체로부터 total RNA를 추출한 후 뇌하수체 glycoprotein hormone common alpha subunit($C{\alpha}$), LH beta subunit($LH{\beta}$), FSH beta subunit($FSH{\beta}$) 그리고 GnRH 수용체(GnRH-R)의 발현 변화를 semi-quantitative RT-PCR로 측정하였다. 그 결과, $C{\alpha}$ 전사수준은 주사 후 1주부터 급격히 상승하여 주사 후 4주까지 유의하게 높게 유지되다가 5주 후부터 control 수준으로 회귀하였다. $LH{\beta}$ 전사 수준은 주사 후 2주부터 유의하게 상승하여 주사 후 4주에 최고 수준에 도달하였으며, 5주 후부터 control 수준으로 감소하였다. $FSH{\beta}$ 전사수준은 주사 후 2주부터 유의하게 상승하여 주사 후 3주에 최고 수준에 도달하였으며, 4주 후부터 감소하여 5주 후에 최소치를 보였다. 유사하게, GnRH-R 전사 수준도 주사 후 2주부터 유의하게 상승하여 주사 후 3주에 최고 수준에 도달하였으며, 5주 후부터 control 수준으로 감소하였다. 본 연구는 EDS 주사에 의해 수컷 흰쥐 뇌하수체 전엽에서의 생식소 자극호르몬 subunit들과 GnRH-R의 발현 변화가 가역적으로 유도될 수 있음을 보여준 것이다. EDS 주사 모델은 수컷 흰쥐에서의 시상하부-뇌하수체 신경내분비 축의 호르몬 조절에 대한 기작을 이해하는데 도움이 될 것이다.

$\beta$-Subunit 94~96 Residues of Tethered Recombinant Equine Chorionic Gonadotropin are Important Sites for Luteinizing Hormone and Follicle Stimulating Hormone like Activities

  • Equine chorionic gonadotropin (eCG) is a heavily glycosylated glycoprotein composed of non-covalently linked $\alpha$- and $\beta$-subunits. To study the function and signal transduction of tethered recombinant-eCG (rec-eCG), a single chain eCG molecule was constructed, and the rec-eCG protein was prepared. In this study, we constructed 5 mutants (${\Delta}1$, ${\Delta}2$, ${\Delta}3$, ${\Delta}4$, and ${\Delta}5$) of rec-eCG using data about known glycoprotein hormones to analyze the role of specific follicle stimulating homone (FSH)-like activity. Three amino acids of certain specific sites were replaced with alanine. The expression vectors were transfected into CHO cells and subjected to G418 selection for 2~3 weeks. The media were collected and the quantity of secreted tethered rec-eCGs was quantified by ELISA. The LH- and FSH-like activities were assayed in terms of cAMP production by rat LH/CG and rat FSH receptors. Then, the metabolic clearance rate analyzed by the injection of rec-eCG (5 IU) into the tail vein was analyzed. The mutant eCGs (${\Delta}l$, ${\Delta}4$, and ${\Delta}5$) were transcripted, but not translated into proteins. Rec-eCG A2 was secreted in much lower amounts than the wild type. Only the rec-eCG ${\Delta}3$ ($\beta$-subunit: $Gln^{94}-Ile^{95}-Lys^{96}{\rightarrow}Ala^{94}-Ala^{95}-Ala^{96}$) was efficiently secreted. Although activity is low, its LH-like activity was similar to that of tethered $eCG{\beta\alpha}$. However, the FSH-like activity of rec-$eCG{\beta\alpha\Delta}3$ was completely flat. The result of the analysis of the metabolic clearance rate shoed the persistence of the mutant in the blood until 4 hours after the injection. After then, it almost disappeared at 8 hours. Taken together, these data suggest that 94~96 amino acid sequences in eCG $\beta$-subunit appear to be of utmost importance for signal transduction of the FSH receptor.
