Objective: This study was conducted to investigate the lipid-lowering effect and safety of Daeshiho-tang in patients with uncontrolled lipid levels by statins. Methods: We investigated patients who had an abnormal lipid profile even when taking statins and who were administered Daeshiho-tang at Kyung-Hee University Korean Medical Hospital for at least one day between January 2008 and December 2018. Their basal characteristics and examinations were reviewed retrospectively with respect to lipid profile, AST, ALT, GGT, BUN, and creatinine. The lipid profile was composed of total cholesterol, triglyceride, LDL-cholesterol, and HDL-cholesterol. Subgroup analysis was performed on each component of dyslipidemia. Results: Among 20 participants, there were 10 males and 10 females. The mean BMI was 23.52. Eighty five percent of the participants were diagnosed as having cerebral infarction. After the administration of Daeshiho-tang, total cholesterol and LDL cholesterol were significantly reduced, to 41.3 mg/dl and 33.95 mg/dl, respectively. In subgroup analyses, total cholesterol and LDL cholesterol were significantly decreased, to 63 mg/dl and 54.6 mg/dl, respectively. Liver and kidney function showed no significant difference after taking Daeshiho-tang. Conclusions: Daeshiho-tang as a decoction or powder had significant lipid-lowering effects on total cholesterol and LDL cholesterol in patients with dyslipidemia. The lipid-lowering effect on total and LDL cholesterol increased in patients with hypercholesterolemia and hyper-LDL-cholesterolemia, respectively. Based on the minimal changes in the liver and kidney function test, Daeshiho-tang would be safe enough to be used in clinics.
The aim of this study was to investigate the effects of L-carnitine on the components of serum and liver and the effects on the anti-oxidant system. For this purpose, five experimental groups were setup. For fat source, perilla oil enough with unsaturated fatty acid and beeftallow enough with saturated fatty acid were supplemented together with L-carnitine to the rats. Five experimental groups kept eight Sprague-Dawley rats respectively, They were co group supplemented with basic diet or AIN-93, PO group supplemented with perilla oil, POC group supplemented with perilla oil and L-carnitine, BT group supplemented with beeftallow, and BTC group supplemented with beeftallow and L- carnitine. The results are. 1) Weight gain, food intake and FER were not different significantly among the experimental groups. 2) Significant difference was observed in serum total lipid(P<0.05), serum triglyceride(P<0.05), serum total cholesterol (P<0.05)and serum LDL cholesterol(P<0.05). Serum total lipid and serum triglyceride were significantly low in the groups supplemented with L-carnitine. Serum total cholesterol showed difference with the supplementation of L-carnitine in BTC only. LDL cholesterol showed no significant difference with the supplementation of L-carnatine, but total values of LDL-cholesterol were high in groups supplemented with beeftallow. 3) Total cholesterol in liver was low in POC group with the supplementation of L-carnitine however, there was no difference in BTC group with the supplementation of L-carnitine. In summary, dietary L-carnitine did not influence the weight gain, food intake and food efficiency ratio among the experimental groups, but had an effect of lowering the serum total lipid and triglyceride significantly in both groups which were supplemented with L-carnitine. The effect of lowering of sew total cholesterol with the supplementation of L-carnitine in beeftallow group(BTC) only. The effect of lowering of liver total cholesterol with the supplementation of L-carnitine in perilla oil group(POC) only.
This study was conducted to find out the effect of fermented milk on lowering of serum and liver cholesterol in high diet. Sprague-Dawley male rats of 200g initial average weight were assigned to four experimental group : (1) high fat diet + milk (2) high fat diet + bifidobacteria (3) high fat diet + microcapsulated bifidobacteria (4) high fat diet + fermented milk by bifidobacteria. Milk, bifidobacteria, microcapsulated bifidobacteria and fermented milk by bifidobacteria were given 2ml per day. According to the result, there was lowering effect of serum and liver total cholesterol in fermented milk feeding group, Serum LDL-cholesterol was also low tendency in fermented milk by bifidobacteria feeding group. Accordingly, it was confirmed that the material that caused hypocholesteremic effect was not the bifidobacteria itself in fermented milk but the metabolic material produced in the fermentation, and it inhibited cholesterol synthesis in liver.

In India, Tectona grandis is traditionally used in the treatment of diabetes mellitus and lipid disorder. In the present study, the antihyperglycemic and lipid lowering effect of ethanolic extract of bark of Tectona grandis (TG) was evaluated using alloxan induced diabetes in rats. Alloxan was given at the dose of 140 mg/kg, i.p. After induction of diabetes, TG was administered for 42 days p. o. and simultaneously different biochemical parameters like plasma glucose, liver glycogen content, serum triglyceride, cholesterol, LDL-cholesterol and HDL-cholesterol were estimated. Diabetic control showed significant increase (P < 0.01) in plasma glucose, serum triglyceride, cholesterol, LDL-cholesterol and significant decrease (P < 0.01) in serum HDL-cholesterol and liver glycogen content. Treatment with TG showed significant reduction (P < 0.01) in plasma glucose when compared with diabetic control. The elevated levels of serum triglyceride and cholesterol levels were significantly reduced (P < 0.01) by TG. TG treatment for 42 days showed significant decrease in serum LDL-cholesterol (P < 0.01) and significant increase in serum HDL-cholesterol level (P < 0.01). Moreover, diabetic control there was significant decrease in liver glycogen content which was significantly increased (P < 0.05) by treatment with TG. Hence, from the result obtained in the present study it can be concluded that Tectona grandis has the potential to treat diabetes condition and associated lipid disorder.
Statins are a class of lipid-lowering drugs commonly used in the prevention of cardiovascular diseases. However, statin therapy presents many limitations, which have led to an increased interest in non-drug therapies, such as probiotics, to improve blood cholesterol levels. Indeed, probiotic strains such as Lactobacillus acidophilus have been found to improve blood lipid profiles, especially in reducing total cholesterol and LDL-C levels. In this study, we established a high-cholesterol rat model and studied the effect of Lactobacillus acidophilus administration alone or in combination with rosuvastatin. We were able to show that Lactobacillus exerts a cholesterol-lowering effect. Additionally, we observed that when administered together, rosuvastin and Lactobacillus exert a combined cholesterol-lowering effect. Altogether, our data advocate for the possibility of establishing probiotics as non-drug supplements for the treatment of hypercholesterolemia.
The cholesterol-lowering effect of a fermented milk was studied for long-term period (56 weeks) in 206 Korean adults (106 normal cholesterol group and 100 hypercholesterol group) 300 $m\ell$ of the fermented milk was taken daily to all subjects for 10 weeks. 24 subjects drinking the fermented milk for 56 weeks and 42 subjects no-drinking for 40 weeks and then re-drinking from 50 th week for 6 weeks were followed up. Paired t-test, Duncan's multiple range test, and GLM repeated measure were used for statistical analysis. The level of total blood cholesterol and LDL after drinking the fermented milk for 10 weeks were decreased significantly in both group and affected by total cholesterol level at baseline, BMI, smoking and drinking. there is no significant change in triglyceride, HDL, and LDL/HDL ratio by drinking the fermented milk. The fermented milk intake for long-term period (56 weeks) did not show any more effect after 10 week on the level of TC, TG, HDL, LDL.
The aim of this study is to investigate the effects of hot water soluble extract from Angelicae Radix on the components of serum and liver and the effects on the antioxidant system. For this purpose, five experimental groups were set up. And for fat source, perila oil enough with unsaturated fatty acid and beef tallow enough with saturated fatty acid were supplemented to the rats together with hot water soluble extract from Angelicae Radixs. Five experimental groups kept eight Sprague-Dawley rats respectively. They were CO group supplemented with basic diet of AIN-93, PO group supplemented with perila oil, POA group supplemented with perila oil and hot water soluble extract from Angelicae Radix, BT group supplemented with beef tallow, and BTA group supplemented with beef tallow and hot water soluble extract from Angelicae Radix. The results were; 1) Final weight, weight gain, fluid intake and FER were not different significantly among the experimental groups, 2) Significant difference of food intake was observed(p<0.05) in BTA group only, 3) No significant difference was observed in serum total lipid, serum triglyceride and HDL cholesterol among experimental groups. Serum total cholesterol and LDL cholesterol were significantly low(p<0.05) in the group supplemented with beef tallow which was with hot water soluble extract from Angelicae Radix (BTA group). 4)Liver total cholesterol in liver was low in groups supplemented with perila oil and hot water soluble extract from Angelicae Radix. In summary, hot water soluble extract from Angelicae Radix did not affect the weight gain, fluid intake and food efficiency ratio among the experimental groups, but had an effect of lowering food intake, serum total cholesterol and serum LDL cholesterol significantly in the groups which were supplemented with beef tallow and hot water soluble extract from Angelicae Radix. The effect of lowering liver total cholesterol with the supplementation of hot water soluble extract from AnRelicae Radix was observed in perila oil group only. The effect of lowering cholesterol with the supplementation of hot water soluble extract from Angelicae Radix was observed both in serum and in liver.
The antioxidative effects of solvent fractions of kimchi on LDL oxidatiojn in vitro as well as hypolipidemic effects of these fractions in rabbit fed atherogenic diet were studied. Methanol extract of deffated kimchi was fractionated sequentially with dichloromethane, ethylacetate, butanol and water. All solvent fractions of kimchi inhibited Cu2+-induced LDL oxidation. Among these fractions, the dicholoromethane fraction at the concentration of 25$\mu\textrm{g}$/mL showed the highest antioxidant effects against LDL oxidation in the aspect of inhibiting TBARS production by 28.03% or prolonged lag phase duration 2-fold compared to those of control. Based on the results from in vitro study, New Zealand White Rabbits grouped six each were fed for 8 weeks either basal diet containing 1% cholesterol or experimental diet containing dichloromethane, ethylacetate or water fraciton added to the basal diet. The amount of solvent fraction of kimchi added to the experimental diet was equivalent to 5% of freeze-dried kimchi. The hypolipidemic effects was observed from all experimental gropus, especially from dichloromethane fraction added group. The plasma and LDL cholesterol levels of this group were decreased by 49% and 47%, respectively while that of HDL increased by 91% compared to those of control. The calculated atherogenic index for the dichloromethane group was the lowest among groups. However, TG lowering effect of experimental group was not observed since solbent fraction of kimchi was used instead of freeze-dried kimchi. The TBARS concentration of LDL isolated from rabbit fed dichloromethane fraction was decreased 21% than that of control. These results indicate that active principles responsible for inhibiting LDL oxidation and lowering plasma cholesterol may present abundantly in dichloromethane fraction of kimchi.
The present study was designed to investigate the serum cholesterol lowering effect oj the phytosterol derivative (LPSS) on high cholesterol (HC) diet-induced hypercholesterolemia in male weaning Sprague-Dawley (SD) rats. Rats were fed with HC diet containing 1 % cholesterol and 0.5% cholic acid for 1 week. After 1 week, the LPSS oil suspension (0.32 g/kg B. W.) was orally administered to the rats fed with either basal diet or HC diet groups for 7 days. In addition, the LPSS powder (0.14%) mixed with basal diet or HC diet was Jed to the rats for 7 days. Serum total cholesterol and LDL-cholesterol contents were not altered by administration of the LPSS oil suspension with basal diet. However, they were significantly decreased by administration of the LPSS oil suspension with HC diet at day 14. Also, they were significantly decreased by the LPSS powder mixed with basal diet or HC diet at day 9, 11, 14. HDL-cholesterol contents were not altered by the LPSS oil suspension or LPSS powder. These results indicated that the phytosterol derivative(LPSS) might decrease serum total cholesterol and LDL-cholesterol contents in rats.
여러가지 농도의 cholesterol을 함유한 배지에서 배양한 CHO 세포내로 흡입된 cholesterol 양을 조사한 결과 흡입량이 배지의 cholesterol 농도에 비례하였으므로 cholesterol 흡입은 확산에 의한 것으로 생각된다. $^{125}I$으로 표지된 저밀도 지방단백질($^{125}I$-LDL)을 이용하여 여러가지 농도의 cholesterol을 함유한 배지에서 배양한 CHO 세포와 cholesterol이 없는 정상배지에서 배양한 CHO 세포에서의 LDL 수용체의 합성양상을 조사한 결과 배지에 가해준 cholesterol이 LDL 수용체 합성을 억제함을 확인하였다. Cholesterol의 LDL 수용체 생합성 억제작용에 미치는 ginsenoside의 영향을 조사하기 위해 ginsenoside와 cholesterol을 함께 함유한 배지에서 배양한 CHO 세포(시험군)과 cholesterol만을 포함한 배지에서 배양한 CHO 세포(대조군)에서의 LDL 수용체의 활성양상을 분석한 결과 대조군에 비해 시험군에서의 LDL 수용체 활성이 크게 증가하였다. RNA 및 단백질 합성도 시험군이 대조군보다 증가하였음을 관찰하였다. 그러나 이와 같은 실험조건하에서의 대조군과 시험군의 cholesterol농도를 측정한 결과 시험군의 cholesterol 농도가 대조군보다 훨씬 저하되고 있었다. Ginsenoside의 cholesterol농도 저하작용을 관찰하기 위해 CHO세포에서의 cholesterol의 steroid hormone(estradiol, progesterone)으로의 전환에 미치는 ginsenoside의 영향을 조사한 결과 ginsenoside는 cholesterol의 hormone으로의 전환을 촉진하였음이 확인되었다. 위와 같은 실험결과로 볼 때 ginsenoside는 CHO 세포내부에서의 cholesterol의 LDL 수용체 합성억제를 완화시켜 주는 것이라고 생각된다.
