납 및 구리화합물이 Carassius auratus(goldfish)에 미치는 독성에 관한 연구 (A Study on the Toxicity of Pb and Cu Compound in Carassius auratus(goldfish))

  • In order to investigate acute toxicity and bioconcentration of heavy metals for a freshwater fish, the fish used in this experiment was goldfish, Carassius auratus. Each ten goldfish was accommonidated in a water and was treated with different concentration of Pb and Cu compound. The 24 $hr-LC_{50}$ was obtained by plotting on the log-normal distribution graph. Furthermore, the combined effect of Pb and Cu was also investigated the fish was treated with Pb or Cu compound only, and Pb and Cu compound together, respectively. These results were summarized as follows: 1. The 24 $hr-LC_{50}'s$ of Pb and Cu were 7.48 mg/l and 0.666 mg/l, respectively. 2. When single or/and combined treatment with Pb(7.0 mg/l) or/and Cu(0.6 mg/l) to Carassius auratus for 24 hours were performed, there was significant difference between the single or/and the combined treatment in their bioaccumulated Cu concentrations. Cu concentrations in goldfish were higher in the combined treatment than in the single treatment. 3. When Carassius auratus was exposed to 0.748 mg/l (1/10 of 24 $hr-LC_{50}$) and 1.496 mg/l of Pb (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 79.14 and 100.11 for Pb, respectively. The BCF of Pb was obtained as a linearity according to the concentration and exposure time as follows log BCF=1.014 log $P\cdot T$+1.011 ($r^2$=0.9041) where, P: pollutant concentration(mg/l) T: exposure time(day) 4. When Carassius auratus was pxposed to 0.0666 mg/l (1/10 of 24 $hr-LC_{50}$) and 0.1332 mg/l of Cu (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 55.42 and 63.24 for Cu respectively. The BCF of Cu was obtained as a linearity according to the concentration and exposure time as follows log BCF=0.571 log $P\cdot T$+1.823 ($r^2$=0.8974) where, P: polutant concentration(mg/l) T: exposure time(day)

  • 최근 개발된 유한차분법의 하나인 선형특성 (LC) 법을 절단오차의 분석을 통하여 계단형 차분 (SD)법, 다이아몬드형차분 (DD)법, 그리고 계단형특성 (SC) 법과 비교했다. 제시된 4가지 방법들이 선속 계산문제, 유효증배계수 계산문제, 그리고 임계평판 두께 계산문제에 적용되었으며, 그때 계산된 결과들은 몇몇의 절단오차 측정 기준들에 의해 평가되어졌다. 본 연구의 주요 결론으로, 모든 경우에 있어서 어느 정도 기대했던 바와 같이 선형특성법이 가장 우수하다는 것이 밝혀졌으며, 특히, 선형특성법은 유효증배계수와 임계평판두께 같은 적분 인자들을 정확하게 계산하는데, 이는 선형특성법이 다른 방법들에 비해 오차소거(Error Cancellation)작용이 효과적으로 일어나기 때문으로 밝혀졌다.

Fodder Productivity and Growth Persistency of Three Local Cassava Varieties

  • Three cassava varieties, namely MM 92 (MM), Black Twig (BT) and Local (LC), were arranged in a randomized complete block design to evaluate their dry fodder and crude protein (CP) productivity as well as growth persistency. Cassava plants grown in small plots of $5m{\times}10m$ at a planting distance of $25cm{\times}25cm$ were harvested every 6 weeks starting from 3 months after planting. Dry fodder yields of MM, BT and LC over the 8 harvests were 8.55, 8.01 and 6.15 t/ha, respectively. All varieties produced more leaves than stems with average leaf:stem ratios of 5, 5.9 and 4.8 for MM, BT and LC, respectively. In terms of CP production, MM was the highest yielder (272 kg/ha/harvest), followed by BT and LC (238 and 184 kg/ha/harvest, respectively). The total accumulative CP amounts over the 8 harvests were 2179, 1903 and 1474 kg/ha for MM, BT and LC, respectively. The mortality rates were 9.91, 14.01 and 13.98% for MM, BT and LC, respectively. Phosphorus content was more stable than potassium content during defoliation. MM, BT and LC had whole plant phosphorus contents of 0.41, 0.41 and 0.39%, respectively; whole plant potassium contents were 1.25, 1.38 and 1.20%.

이중배양법에 따른 Lactococcus lactis의 아토피 유발인자 억제 효과 증대 (Double-culture Method Enhances the in Vitro Inhibition of Atopy-inducing Factors by Lactococcus lactis)

  • 유산균이 활성화된 비만세포에서 발현하는 IL-4와 IL-13을 조절할 수 있는지를 분석하였고, GATA-1, GATA-2, NF-AT1, NF-AT2, NF-κB p65 전사인자의 활성을 억제하는지를 실험적으로 규명하였다. 이전 연구에서 T cell에서 CD4+/CD25+/foxp3+ 증가를 실험하여 항아토피 기능성이 있는 유산균을 탐색하였고, Staphylococcus aureus에 대한 항균력을 증가시키는 유산균의 이중배양법을 확인하였다. 여기서는 RBL-2H3 비만세포를 이용하여 이 배양법으로 배양한 유산균이 아토피 피부염의 원인이 되는 allergy 염증반응에서 얼마나 억제능을 갖는지 알아보았다. 그 결과 Lc. lactis culture with medium containing Lb. plantarum supernatants > Lc. lactis > Lc. lactis culture broth with medium containing Lb. plantarum culture broth > Lb. plantarum의 순으로 나타났다. 이 cell 수준(level of mast cells)에서의 순서는 이전 연구의 level of microorganisms (anti-S. aureus)에서의 아토피 유발인자 억제능 순서와 같다. 따라서 세포수준에서도 Lb. plantarum 배양상층액을 첨가한 배지에 Lc. lactis 배양한 경우가 활성화된 비만세포의 allergy 반응으로의 분화 및 활성을 가장 잘 억제하고 관련 유전자 발현을 선택적으로 조절하는 anti-allergy 효과를 나타낸다고 사료된다.

LC-ESI/MS/MS를 이용한 소고기와 우유에서의 클렌부테롤 분석 (Analysis of clenbuterol in bovine muscle and milk by LC-ESI/MS/MS)

  • 본 연구에서는 소고기와 우유에서의 클렌부테롤의 확인과 정량을 위하여 LC-ESI/MS/MS을 이용한 방법을 개발하였다. 클렌부테롤과 내부표준물질로 클렌부테롤-D9을 사용한 시료를 가수분해 한 후 에틸아세테이트로 추출하고 농축하였다. 그리고 추출물을 20% 메탄올에 용해하여 HLB 카트리지로 정제한 후 $C_{18}$ 칼럼을 이용한 LC-ESI/MS/MS로 분석하였다. 높은 감도를 얻기 위하여 positive ion mode에서 SRM(selected reaction monitoring)방법을 이용하였다. MS/MS의 SRM방법으로 클렌부테롤과 클렌부테롤-D9의 선구이온(precursor ion), 생성이온(product ion)을 각각 m/z 227${\rightarrow}$203, m/z 286${\rightarrow}$204로 분석한 결과 소고기에서의 클렌부테롤의 정량한계와 회수율은 각각 $0.2{\mu}g/kg$과 84.3~91.1%이었고, 우유에서의 정량한계와 회수율은 각각 $0.05{\mu}g/kg$과 87.7~98.3%이었다.

Analysis of Myosin Heavy Chain Isoforms from Longissimus Thoracis Muscle of Hanwoo Steer by Electrophoresis and LC-MS/MS

  • The purpose of this study was to analyze myosin heavy chain (MHC) isoforms in bovine longissimus thoracis (LT) muscle by liquid chromatography (LC) and mass spectrometry (MS). LT muscles taken from Hanwoo (Korean native cattle) steer (n=3) used to separate myosin bands by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The peptide queries were obtained from the myosin bands by LC-MS/MS analysis following in-gel digestion with trypsin. A total of 33 and 43 queries were identified as common and unique peptides, respectively, of MHC isoforms (individual ions scores >43 indicate identity or extensive homology, p<0.05). MHC-1 (IIx), -2 (IIa), -4 (IIb), and -7 (slow/I) were identified based on the Mowse score (5118, 3951, 2526, and 2541 for MHC-1, -2, -4, and -7, respectively). However, more analysis is needed to confirm the expression of MHC-4 in bovine LT muscle because any query identified as a unique peptide of MHC-4 was not found. The queries that were identified as unique peptides could be used as peptide markers to confirm MHC-1 (14 queries), -2 (8 queries), and -7 (21 queries) in bovine LT muscle; no query identified as a unique peptide of MHC-4 was found. LC-MS/MS analysis is a useful approach to study MHC isoforms at the protein level.

  • A photoalignment material based N-(phenyl)maileimide were synthesized and the liquid crystal (LC) aligning capabilities on the photopolymer layer were studied. A goof LC alignment with UV exposure on the PM15CA(N-(phenyl)maileimide with 5-carbon chain cinnamoyl group) can be obtained However, the LC alignment defects were observed on the PM13CA (N-(phenyl)maileimide with 3-carbon cam cinnamoyl group) and PMIF (N-(Phenyl)maileimide including fluoro cinnamoyl group). Also, the good LC alignment with UV exposure on the PM15CA surface was observed at until $150^{\circ}C$ of annealing temperature. The LC aliging ability on the photopolymer layer based N-(phenyl)maleimide depends on the side chin length of photopolymer.

유전율 이방성이 양인 액정을 이용한 fringe-Field 구동형 반사형 Hybrid Aligned Nematic 액정 디스플레이의 전기-광학 특성 (Electro-optic Characteristics of the fringe-field Driven Reflective Hybrid Aligned Nematic Liquid Crystal Cell using a Liquid Crystal with Positive Dielectric Anisotropy)

  • Electro-optic characteristics of reflective hybrid aligned liquid crystal (LC) cell driven by fringe field using a nematic LC with positive dielectric anisotropy have been studied. Optimized optical configurations are achieved by using a single polarizer, half-wave film and a cell with quarter-wave retardation. The simulation results shows an optimum cell retardation of $0.30{\mu}m$. This value may allow a practical cell gap larger than $3{\mu}m$, which makes it easy to control in the manufacturing process. Furthermore, this LC cell with optimized cell parameters shows low wavelength dispersion and the contrast ratio greater than 5 over exists about $100{\circ}$ in vertical direction and $160{\circ}$ in horizontal direction. Also, when using the LC with positive dielectric anisotropy rather than negative dielectric anisotropy, the display shows low power consumption and fast response time.

Effects of L-Carnitine and Nicotinic Acid on Sperm Characteristics in Miniature Pigs

  • This study investigated the effects of L-carnitine (LC) and nicotinic acid (NA) on sperm viability during liquid storage at $18^{\circ}C$ in miniature pigs. $10{\mu}M$ LC and 30 mM NA, combined LC and NA (LN) were treated in fresh semen for 3, 7, and 10 days. In results, sperm survival increased in NA- and LN-treated semen on 7 and 10 days (p<0.05), mitochondrial integrity of live sperm increased in LN-treated semen on 7 days (p<0.05), but not NA-treated semen. In addition, we examined the acrosome reaction of sperm in miniature pigs. LC and NA did not influence on acrosome reaction of boar sperm. In conclusion, LC and NA effectively maintained the viability and quality of sperm during long-term storage in miniature pigs, suggesting that the combined LN may be useful for improving the semen extender for long-term liquid storage in pigs.

주택용 단상 ESS-PCS의 LC 필터위치에 따른 상용변압기의 특성 (Characteristics of Utility Transformer on Household Single-Phase ESS-PCS According to LC Filter Location)

  • Shortage of electric power occurs frequently along with increased electric power demand. ESS is a precaution to solve this issue. Household ESS has a capacity of approximately 3 kW/7 kWh. Household ESS batteries are typically designed with nominal voltages between 40 and 50 V. To connect household ESS with a 220 V AC system, low battery voltages in power conditioning system (PCS) should be boosted. To boost low battery voltage and match it with AC grid voltage, the use of a transformer for a commercial frequency can be considered. To attenuate switching harmonics of the household single-phase ESS-PCS, LC filter can be installed in two positions: on the primary side or on the secondary side of a transformer. A method has been used generally in single-phase inverters for the ESS-PCS. In another method, however, the output efficiency of the ESS-PCS may be decreased. Parasitic components of the transformer can affect voltage losses, when the square wave with the switching frequency in the ESS-PCS is passed through the transformer windings. In this work, the characteristics of the transformer according to the position of an LC filter are investigated for household single-phase ESS-PCS.
