  • Neocaridina denticulata is a small freshwater shrimp indigenous to Korea. As an indigenous species has long-adapted to particular water environments, the species can be a suitable indicator to assess environmental risks caused by hazard chemicals in the particular site. Thus Neocaridina denticulata, a small freshwater shrimp indigenous to Korea, is worth considering for a test species for such purpose. N. denticulata were exposed to pentachlorophenol sodium salt, sodium azide and potassium dichromate using automatic flow-through system for 96 hours. The 96 hr lethal concentrations ($LC_{50}$) of these chemicals were calculated as $0.53{\pm}0.09\;mg/L$, $2.40{\pm}0.61\;mg/L$ and $1.21{\pm}0.09\;mg/L$ respectively and showed relatively small deviation from repetitive test results. When compared with the toxicity values of other species for each chemical, N. denticulata had moderate or high sensitivity to the toxicity of these chemicals. It can be concluded that N. denticulata is a good test species to evaluate acute toxicity of various hazardous chemicals.

  • Although some degree of mechanical properties of suede fabrics mainly related to non-woven suede fabric has some researched, the thin suede fabric has rarely been researched. In this study, polyester(DTY 50/72) was used for warp, and after producing latent yarn and sea-island yarn for weft, two yarns were compounded to produce sea-island DTY yarn. By using the two produced yarns for warp and weft, we produced thin suede fabric with stretch function. For weft 2ply, weft density 85, 90, 100(picks/in) were applied to weave fabric, and for weft 1ply, weft density 125, 135, 140(picks/in) were applied to produce weft face 5-end satin weave. The mechanical properties of the produced fabric were researched. The result are as followed. The weight loss ratio of the suede fabric produced for this experiment reached 15% on the conditions of temperature $90^{\circ}C$ and 20 minutes, so that island parts were completely separated. The strength of weft 1ply applied suede fabric was about 7.5kg and that of 2ply suede fabric and about 3.5kg. But the strain of two samples ranged from about 40 to 43%. Although Hari was high when weft was denser. The values of Koshi and Kisimi were low. And shear stiffness was high when sea-island DTY yarn was used. The WC value was higher in the case of 2ply than in that of 1ply sea-island DTY yarn for weft, so that we may conclude that Fukurami was more affected in the 2ply case.

  • Primary culture of cerebellar neuronal cells derived from 8-day old Long-Evans rats was used. Pure granule cells, astrocytes or mixed cells culture systems were prepared. These cells were differentiated and developed synaptic connections. And the astrocytes were identified by immunostaining with glial fibrillary acidic protein (GFAP). Methamphetamine (MAP), which acts on dopaminergic system and cadmium (Cd), a toxic heavy metal, were applied and biochemical assays and electrophysiological studies were performed. $LC_50$ values estimated by MTT assay of MAP and Cd were 3 mM and 2$\mu M$ respectively. Cells were treated with 1 mM or 2 mM MAP and 1$\mu M$ $CdCl_2$ for 48 hour, and the incubation media were analyzed for the content of released LDH. MAP (2 mM) and Cd significantly increased the LDH release. Cell viability was decreased in both groups and some cytopathological changes like cell swelling or vacuolization were seen. The cerebellar granule cells were used for measuring membrane currents using whole-cell clamp technique. Sodium and potassium currents were not affected by MAP neither Cd, but calcium current was significantly reduced by Cd but not affected by MAP. Therefore, in vitro neurotoxicity test system using neuronaI cells and astrocytes cultures were established and can be used in screening of potential neurotoxic chemicals.

  • DC23, a triazolothione resorcinol analogue, is known to inhibit heat shock protein 90 and pyruvate dehydrogenase kinase which are up-regulated in cancer and diabetes, respectively. This study was performed to elucidate the metabolism of DC23 in human liver microsomes (HLMs). HLMs incubated with DC23 in the presence of uridine 5'-diphosphoglucuronic acid (UDPGA) and/or ${\beta}$-nicotinamide adenine dinucleotide phosphate (NADPH) resulted in the formation of four metabolites, M1-M4. M1 was identified as DC23-N-Oxide, on the basis of LC-MS/MS analysis. DC23 was further metabolized to its glucuronide conjugates (M2, M3, and M4). In vitro metabolic stability studies conducted with DC23 in HLMs revealed significant glucuronide conjugation with a $t_{1/2}$ value of 1.3 min. The inhibitory potency of DC23 on five human cytochrome P450s was also investigated in HLMs. In these experiments, DC23 inhibited CYP2C9-mediated tolbutamide hydroxylase activity with an $IC_{50}$ value of $8.7{\mu}M$, which could have implications for drug interactions.

  • The current study was undertaken to determine the effects of the ratios of dietary fat to carbohydrate and energy restriction on insulin sensitivity in the growing rats. Male rats weighting 80-90g were fed experimental diets for two weeks. Rats were killed and epiddymal adipose tissue were removed and sliced. Explants of adipose tissues were incubated for 2 hours in KRB(Krebs-Ringer bicarbonate) buffer containing various concentrations of human insulin and [U-14C]glucose. Insulin sensitivity was determined as glucose conversion to total lipids (lipogenesis) during 2 hr incubation. Exp't I : Effects of Ratios of Fat to Carbohydrate on Insulin Sensitivity. Eighteen male rats were fed 3 diets for 2 weeks. Diet 1 was low fat-high carbohydrate (4% soybean oil and 66.5% cornstarch) ; diet 2, medium fat-medium sarbohydrate(12% soybean oil and 58.5% cornstarch) ; diet 3, high fat-low carbohydrate (20% soybean oil and 50.5% cornstarch). Insulin sensitivity was higher in the order of LF-HC, MF-HC and HF-LC diet groups (p<0.05), i.e, lipogenesis was higher at all insuline concentration in the explants from rats fed LF-HC diet. However, thers was no significant difference in body weight gain and epididymal adipose tissue weight among treatments. Exp't II ; Effects of Energy Restriction on Insulin Sensitivity. Twelve rats were grouped into ad libitum feeding and restricted feeding(70% of ad libitum). The experimental diet was medium fat-medium carbohydrat diet as used in the Exp't I. Restricted feeding group tended to show higher insulin sensitivity compared to ad libitum group. However, there was no statistical difference between two groups. As expected, body weight gain and epididymal adipose tissue were higher in the ad libitum group. In summary, the resutls of the current study showed that the epididymal adipose tissue taken from the rats fed low fat-high carbohydrate diet showed higher insulin sensitivity compared to those fed high fat-low carbohydrate, and that resticted feeding tended to elevate insulin sensitivity in these tissues.

  • Human cytochrome P450 2C9 is a highly polymorphic enzyme that is required for drug and xenobiotic metabolism. Here, we studied eleven P450 2C9 genetic variants-including three novel variants F69S, L310V, and Q324X-that were clinically identified in Korean patients. P450 2C9 variant enzymes were expressed in Escherichia coli and their bicistronic membrane fractions were prepared The CO-binding spectra were obtained for nine enzyme variants, indicating P450 holoenzymes, but not for the M02 (L90P) variant. The M11 (Q324X) variant could not be expressed due to an early nonsense mutation. LC-MS/MS analysis was performed to measure the catalytic activities of the P450 2C9 variants, using diclofenac as a substrate. Steady-state kinetic analysis revealed that the catalytic efficiency of all nine P450 2C9 variants was lower than that of the wild type P450 2C9 enzyme. The M05 (R150L) and M06 (P279T) variants showed high $k_{cat}$ values; however, their $K_m$ values were also high. As the M01 (F69S), M03 (R124Q), M04 (R125H), M08 (I359L), M09 (I359T), and M10 (A477T) variants exhibited higher $K_m$ and lower $k_{cat}$ values than that of the wild type enzyme, their catalytic efficiency decreased by approximately 50-fold compared to the wild type enzyme. Furthermore, the novel variant M07 (L310V) showed lower $k_{cat}$ and $K_m$ values than the wild type enzyme, which resulted in its decreased (80%) catalytic efficiency. The X-ray crystal structure of P450 2C9 revealed the presence of mutations in the residues surrounding the substrate-binding cavity. Functional characterization of these genetic variants can help understand the pharmacogenetic outcomes.

  • BACKGROUND: Agricultural use and pest control purposes of pesticides may lead to livestock products contamination. Thiodicarb and its degraded product, methomyl, are carbamate insecticides that protect soya bean, maize, fruit, and vegetables and control flies in animal and poultry farms. For maximum residue limit enforcement and monitoring, the JMPR residue definition of thiodicarb in animal products is the sum of thiodicarb and methomyl, expressed as methomyl. This residue definition was set to consider the fact that thiodicarb was readily degraded to methomyl in animal commodities. And therefore the simultaneous analytical method of thiodicarb and methomyl is required for monitoring in livestock products. METHODS AND RESULTS: The study was conducted using a quick, easy, cheap, effective, rugged, and safe (QuEChERS) method and HPLC-MS/MS to determine the thiodicarb and methomyl in livestock products. The limit of quantitation (LOQ) was 0.01 mg/kg for livestock products, including beef, pork, chicken, milk, and egg. The coefficient of determinations (r2) for the calibration curve were > 0.99, which was acceptable values for linearity. Average recoveries at spiked levels (LOQ, 10LOQ, and 50LOQ, n=5) in triplicate ranged from 73.2% to 102.1% and relative standard deviations (RSDs) were less than 10% in all matrices. CONCLUSION: The analytical method was validated for the performance parameters (specificity, linearity, accuracy, and precision) in livestock products to be acceptable by the CODEX guidelines.

  • Triclosan (TCS) is a phenolic antimicrobial chemical used in consumer products and medical devices. Evidence from in vitro and in vivo animal studies has linked TCS to numerous health problems, including allergic, cardiovascular, and neurodegenerative disease. Using Caenorhabditis elegans as a model system, we here show that short-term TCS treatment ($LC_{50}$: ~0.2 mM) significantly induced mortality in a dose-dependent manner. Notably, TCS-induced mortality was dramatically suppressed by co-treatment with non-ionic surfactants (NISs: e.g., Tween 20, Tween 80, NP-40, and Triton X-100), but not with anionic surfactants (e.g., sodium dodecyl sulfate). To identify the range of compounds susceptible to NIS inhibition, other structurally related chemical compounds were also examined. Of the compounds tested, only the toxicity of phenolic compounds (bisphenol A and benzyl 4-hydroxybenzoic acid) was significantly abrogated by NISs. Mechanistic analyses using TCS revealed that NISs appear to interfere with TCS-mediated mortality by micellar solubilization. Once internalized, the TCS-micelle complex is inefficiently exported in worms lacking PMP-3 (encoding an ATP-binding cassette (ABC) transporter) transmembrane protein, resulting in overt toxicity. Since many EDCs and surfactants are extensively used in commercial products, findings from this study provide valuable insights to devise safer pharmaceutical and nutritional preparations.

  • The study of metabolomics in natural products using the diverse analytical instruments including GC-MS, LC-MS, and NMR is useful for the exploration of physiological and biological effects and the investigation of drug discovery and health functional foods. Cordyceps militaris has been very attractive to natural medicine as a traditional Chinese medicine, due to its various bioactive properties including anti-cancer and anti-oxidant effects. In this study, we analyzed the metabolite profile in 50% ethanol extracts of C. militaris fruit bodies from three development periods (growth period, matured period, and aging period) using $^1H-NMR$, and identified 44 metabolites, which are classified as 16 amino acids, 10 organic acids, 5 carbohydrates, 3 nucleotide derivatives, and 10 other compounds. Among the three development periods of the C. militaris fruit body, the aging period showed significantly higher levels of metabolites including cordycepin, mannitol (cordycepic acid), and ${\beta}-glucan$. Interestingly, these bioactive metabolites are positively correlated with antitumor growth effect; the extract of the aging period showed significant inhibition of HepG2 hepatic cancer cell proliferation. These results showed that the aging period during the development of C. militaris fruit bodies was more highly enriched with bioactive metabolites that are associated with cancer cell growth inhibition.
