Highly-sensitive Detection of Salvianolic Acid B using Alumina Microfibers-modified Electrode

  • Alumina microfibers with porous structures were prepared through hydrothermal reaction, and then used to modify the surface of carbon paste electrode (CPE). After modification with alumina microfibers, the electrochemical activity of CPE was found to be greatly improved. On the surface of alumina microfibers-modified CPE, the oxidation peak current of salvianolic acid B, a main bioactive compound in Danshen with anti-oxidative and anti-inflammatory effects, was remarkably increased compared with that on the bare CPE surface. The influences of pH value, amount of alumina microfibers and accumulation time were studied. Based on the strong signal amplification effects of alumina microfibers, a novel electrochemical method was developed for the detection of salvianolic acid B. The linear range was from 5 ${\mu}gL^{-1}$ to 0.3 mg $L^{-1}$, and the detection limit was 2 ${\mu}gL^{-1}$ (2.78 nM) after 1-min accumulation. The new method was successfully used to detect salvianolic acid B in ShuangDan oral liquid samples, and the recovery was over the range from 97.4% to 102.9%.

Yield and Free Sugar Contents of Burdock (Arctium lappa L.) depending on Nitrogen Levels

  • Excessive nitrogen fertilization influences crop yields and quality as well as environmental pollution. In this study, yields, nitrogen use efficiency and free sugar contents of burdock (Arctium lappa L.) were evaluated at different levels of nitrogen fertilization. Nitrogen fertilizer was applied at 5 levels (0, 50, 100, 150, 200%) based on the conventional fertilization ($N=230kg\;ha^{-1}$), and phosphate and potassium fertilizer were treated by conventional P and K fertilization ($P_2O_5-K_2O=140-210kg\;ha^{-1}$) in all plots. The root yields of burdock were the highest in N 100~150% treatment plots. Nitrogen use efficiency and nitrogen recovery decreased from over N 150% treatment. Nitrogen uptake of root was greater than that of shoot in N 50~200% treatments. Fructose contents in root were inversely proportional to the level of nitrogen fertilization. As considering nitrogen recovery and root quality, economical burdock yield was obtained in N $230kg\;ha^{-1}$.

Measured aerodynamic coefficients of without and with spiked blunt body at Mach 6

  • A spike attached to a blunt nosed body significantly alters its flow field and influences the aerodynamic coefficients at hypersonic speed. The basic body is an axisymmetric, with a hemisphere nose followed by a cylindrical portion. Five different types of spikes, namely, conical aerospike, hemisphere aerospike, flat-face aerospike, hemisphere aerodisk and flat-face aerodisk are attached to the basic body in order to assess the aerodynamic characteristic. The spiked blunt body without the aerospike or aerodisk has been set to be a basic model. The coefficients of drag, lift and pitching moment were measured with and without blunt spike body for the length-to-diameter ratio (L/D) of 0.5, 1.0, 1.5 and 2.0, at Mach 6 and angle of attack up to 8 degrees using a strain gauge balance. The measured forces and moment data are employed to determine the relative performance of the aerodynamic with respect to the basic model. A maximum of 77 percent drag reduction was achieved with hemisphere aerospike of L/D = 2.0. The comparison of aerodynamic coefficients between the basic model and the spiked blunt body reveals that the aerodynamic drag and pitching moment coefficients decrease with increasing the L/D ratio and angle of attack but the lift coefficient has increasing characteristics.

Effects of various medium on mass propagation of in vitro cultured Platycodon grandiflorum with yellow green petals

  • Propagation by crown division in Platycodon grandiflorum is too slow for producing many plants in a short time although the plants are uniform. This study was performed to enhance the mass propagation for Platycodon grandiflorum containing yellow green petals via various medium compositions and the growth regulators. The nodes containing yellow green petals were used as materials to execute the study with a variety of MS medium concentrations. The 1/4MS medium showed the best development of adventitious root, while the 1/2MS medium exhibited the potential growth. The higher the concentration of sucrose showed the better development and growth of both shoots and adventitious roots. Many adventitious roots were developed at the controlled culture medium at pH 4.8 with a tendency of suppression with higher levels of pH. Also, the cultivated node and leaf explants with the treatments of simple and combined applications with auxin and cytokinin at the 1/4MS culture medium with adding 5% of sucrose were used to identify the influences of growth regulators. The regeneration of plantlets at the 234single application showed a good result with the addition of BA $1mg{\cdot}L^{-1}$ and the development and growth of adventitious roots appeared to be good at the addition of NAA $1mg{\cdot}L^{-1}$. For the combined applications, the regeneration of plantlets and the development of adventitious roots were prosperous at the combined applications with BA $0.1mg{\cdot}L^{-1}$ and IAA $0.5mg{\cdot}L^{-1}$. The addition of IAA for the leaf explants induced a number of plantlets that showed the potential regeneration, and the highest results was obtained from the combined applications of both BA $1mg{\cdot}L^{-1}$ and IAA $2.5mg{\cdot}L^{-1}$. In addition, the development of adventitious roots showed the satisfactory results at the combined application of both BA $1mg{\cdot}L^{-1}$ and IAA $0.5mg{\cdot}L^{-1}$.

형산강 수계의 생태 서식지, 화학적 수질 및 어류의 다변수모델 평가 (Evaluations of Ecological Habitat, Chemical Water Quality, and Fish Multi-Metric Model in Hyeongsan River Watershed)

  • 본 연구는 형산강 수계 6개 지점을 선정하고, 2009년 4월과 9월에 조사를 실시하여 이 화학적 수질, 물리적 서식지 분석을 통하여 어류 분포특성 및 생태 건강도를 진단하였다. 생물통합지수(Index of Biological Integrity, IBI) 모델 분석은 국내 하천의 특성에 맞게 수정 보완하여 8개 다변수 메트릭 모델을 이용하였고, 물리적 서식지 평가지수(Qualitative Habitat Evaluation Index, QHEI)분석은 11개의 다변수 메트릭 모델을 적용하였다. 형산강의 1, 2차 생태 건강도 조사 결과 평균 25.4(n=6)로 "보통상태" (C)로 나타났다. 1차 조사 결과 평균 25로 "보통상태" (C), 2차 조사 결과 평균 25.7로 "보통상태" (C)로 나타났다. 물리적 서식지 평가 결과 최상류 H1에서 150.5로 "양호상태" (B)로 나타났으며, 하류인 H6에서 76으로 "보통상태" (C)로 나타나 하류로 갈수록 감소하는 경향을 보였다. 이 화학적 수질 분석은 형산강 수계의 환경부 수질 측정망 자료 중 2000년부터 2009년까지 10년간의 자료를 이용하여 분석하였다. 형산강의 지난 10년간 평균 BOD 값은 2.4 $mgL^{-1}$ (범위: 0.3~13.8 $mgL^{-1}$)로서 우리나라 수질기준(2010년 1월의 환경부 기준고시)에 의거할 때 II (약간 좋음) 등급을 보였고, COD 역시 큰 변이를 보이며(범위: 0.6~12.8 $mgL^{-1}$), BOD와 유사한 경향을 보였다. TN의 평균값은 3.0 $mgL^{-1}$ (범위: 0.3~10.4 $mgL^{-1}$)이고, TN 역시 형산강이 경주시를 지나면서 TN이 증가한 후 감소하는 경향을 보였다. TP의 평균값은 103.5 ${\mu}gL^{-1}$(범위: 0~606 ${\mu}gL^{-1}$)로 TN과 비슷한 양상을 보였다. SS는 7.5 $mgL^{-1}$ (범위: 0.1~25.2 $mgL^{-1}$)로 하류로 갈수록 증가하는 경향을 보였다. 전체적으로 본류 지점은 하류로 갈수록 건강성이 악화되는 것으로 나타났는데 이는 경주시를 관통하면서 수질이 악화되었고, 생활하수, 산업폐수 및 분뇨처리수가 유입되어 이와 같은 결과가 나타난 것으로 사료되었다.

하천 생태계에서 유기탄소 기질 제거에 조류와 세균의 공생작용이 미치는 영향 (Effect of Bacterial and Algal Symbiotic Reaction on the Removal of Organic Carbon in River Ecosystem)

  • It have been investigated how algal and bacterial symbiotic reaction influences on removal of organic carbon in river ecosystem. And artificial experimentation apparatus was made for algae'and bacteia'culture as lab scale. Investigating and researching minutely the change of concentration of organic carbon substrate and the change of population density of algae'and of bacteria'with this artificial experimentation apparatus, the next results could be obtained. 1. Successful decrease of DOC(dissolved organic carbon) could not be expected unless algal and bacterial biomass floe was nut formed effectively and unless biosorption was not proceeded effectively in the very culture system in which artificial synthetic wastewater was supplied continuously at constant rate. 2. In conditions of culture liquid of 1335 glucnse mg/L(type 1) and of 267 glucose mg:L(type 2), the algal dominant species was always Chlorella vulgaris in both types in which artificial synthetic wastewater were supplied continuously at constant rate and algae population density was around maximum 107 cells/mL. 3. It was around 108 ~ 107 cells/mL that the population density of heterotrophic bacterium. In culture medium systems type 1 and type 2 in which artificial wastewater were supplied continuously at constant rate, the same density appeared initially when using the population density of Escherichia coli w 3110 as indirect indicator. And this density decreased rapidly till the culturing date 35 days were passed away, while this density increased with gentle slope after same date and then the trend of change at type 2 was more severe than one at type 1. 4. When seeing such a change of population density of Escherichia coli w 3110, the growth of heterotrophic bacterium appeared as survival instinct pattern of broader requirement of nutrient at condition of low concentration of organic carbon substrate than condition of high concentration of same substrate.

의류점포의 서비스품질이 지각된 전환장벽과 점포충성도에 미치는 영향 (The Effect of Fashion Store Service Quality on Perceived Switching Barriers and Store Loyalty)

  • This study attempts to understand perceived switching barriers of customers and fashion stores service quality as a core of store loyalty. Medium variations of switching barriers are interpersonal relationships, attractiveness of alternatives and switching costs. Date were collected from 485 subjects who finished their shopping with a self-administered questionnaire. The results were as follows: 1. Classified service quality of clothing stores into 4 ways: VMD of stores or atmosphere, store policy which are involved in store service, sales persons respond and sales persons conviction which are involved in sales service. 2. Service quality of fashion stores is related with switching barrier medium variations. 3. Service quality have an influence on switching barrier medium variations. Its mutual influences increase switching barriers and it gets to be related to the store loyalty. 4. Compared influences of variations to store loyalty between the H department store and the L department store which are research objects. The main difference is that the H department store evaluated personal relationships among service quality, customers and sales persons high compared with the L department store. As the results showed, service quality consists of store service of nonpersonal service area and sales service of personal service. Service quality is related with interpersonal relationships, attractiveness of alternatives and switching costs. It also affects switching barriers and store loyalty.

Effect of Suboptimal Nutritional Status on Mineral Uptake and Carbohydrate Metabolism in Tomato Plants

  • A suitable supply of mineral elements into shoot via a root system from growth media makes plants favorable growth and yield. The shortage or surplus of minerals directly affects overall physiological reactions to plants and, especially, strongly influences carbohydrate metabolism as a primary response. We have studied mineral uptake and synthesis and translocation of soluble carbohydrates in N, P or K-deficient tomato plants, and examined the interaction between soluble carbohydrates and mineral elements. Four-weeks-old tomato plants were grown in a hydroponic growth container adjusted with suboptimal N ($0.5mmol\;L^{-1}\;Ca(NO_3)2{\cdot}4H_2O$ and $0.5mmol\;L^{-1}\;KNO_3$), P ($0.05mmol\;L^{-1}\;KH_2PO_4$), and K ($0.5mmol\;L^{-1}\;KNO_3$) for 30 days. The deficiency of specific mineral element led to a significant decrease in its concentration and affected the concentration of other elements with increasing treatment period. The appearance of the reduction, however, differed slightly between elements. The ratios of N uptake of each treatment to that in NPK sufficient tomato shoots were 4 (N deficient), 50 (P deficient), and 50% (K deficient). The P uptake ratios were 21 (N deficient), 19 (P deficient), and 28% (K deficient) and K uptake ratios were 11 (N deficient), 46 (P deficient), and 7% (K deficient). The deficiency of mineral elements also influenced on carbohydrate metabolism; soluble sugar and starch was substantially enhanced, especially in N or K deficiency. In conclusion, mineral deficiency leads to an adverse carbohydrate metabolism such as immoderate accumulation and restricted translocation as well as reduced mineral uptake and thus results in the reduced plant growth.

Dietary L-carnitine Influences Broiler Thigh Yield

  • L-carnitine promotes mitochondrial ${\beta}$-oxidation of long chain fatty acids and their subsequent transport across the inner mitochondrial membrane. Although the role of L-carnitine in fatty acid metabolism has been extensively studied, its role in live performance and carcass responses of commercial broilers is less understood. The objective of this research was to determine if Lcarnitine fed at various levels in diets differing in CP and amino acids impacted on live performance and carcass characteristics of commercial broilers. Two floor pen experiments were conducted to assess the effect of dietary L-carnitine in grower diets. In Exp. 1, Ross${\times}$Hubbard Ultra Yield broilers were placed in 48 floor pens (12 birds/pen) and fed common diets to d 14. A two (0 or 50 ppm Lcarnitine) by three (173, 187, and 202 g/kg CP) factorial arrangement of treatments was employed from 15 to 35 d of age (8 replications/treatment). An interaction (p<0.05) in carcass yield indicated that increasing CP (187 g/kg) resulted in improved yield in the presence of L-carnitine. Increasing CP from 173 to 202 g/kg increased (p<0.05) BW gain and decreased (p<0.05) feed conversion and percentage abdominal fat. Feeding dietary L-carnitine increased back-half carcass yield which was attributable to an increase (p<0.05) in thigh, but not drumstick, yield relative to carcass. In Exp. 2, $Ross{\times}Ross$ 708 broilers were fed common diets until 29 d. From 30 to 42 d of age, birds were fed one of seven diets: i) 200 g/kg CP, 0 ppm L-carnitine; ii) 200 g/kg CP, 40 ppm L-carnitine; iii) 180 g/kg CP, 0 ppm L-carnitine; iv) 180 g/kg CP, 10 ppm L-carnitine; v) 180 g/kg CP, 20 ppm L-carnitine; vi) 180 g/kg CP, 30 ppm L-carnitine; and vii) 180 g/kg CP, 40 ppm L-carnitine (6 replications of 12 birds each). BW gain, feed conversion, mortality (30 to 42 d), and carcass traits (42 d) were measured on all birds by pen. There were no treatment differences (p<0.05). However, the addition of 40 ppm L-carnitine in the 200 g CP/kg diet increased (p = 0.06) thigh yields relative to BW in comparison to birds fed diets without L-carnitine, which was further confirmed via a contrast analysis (0 vs. 40 ppm L-carnitine in the 200 and 180 g CP/kg diets; p<0.05). These results indicated that dietary L-carnitine may heighten metabolism in dark meat of commercial broilers resulting in increased relative thigh tissue accretion without compromising breast accretion.

Assessing the removal efficiency of Synedra sp. through analysis of field data from water treatment plants

  • Prechlorination could increase the removal efficiency of Synedra, but there was no significant effect of increasing the amount of chlorine added. However, a removal efficiency of greater than 80% was noted when ozone was injected at concentrations greater than 2 mg/L. Also, it was found that on addition of polyamine, a removal efficiency of 80% or more could be achieved. As a result of the analysis of field operation data from the water treatment plants G and B, it was found that at water treatment plant G, the filter run time decreased to 10 hours or less when only coagulant was injected, but the filter run time increased to around 40 hours when polyamine (3 mg/L) was also injected. The Synedra population in the raw water subsequently increased to 2,340 cells/mL, and the filter continued running for more than 20 hours. At water treatment plant B, the average Synedra removal efficiency was 56% when only coagulant was injected, and the filter run time decreased drastically with the increasing population of Synedra in the raw water. However, the removal efficiency of Synedra reached 79% when polyamine was injected together with the coagulant, 90% when ozone was also injected, and 95% when polyamine and ozone were injected together and the filter continued running for over 50 hours. The filter run time was maintained at 60 hours when a Synedra population of 6,890 cells/mL flowed into the Paldang water source, but the filter run time with Synedra at 1,960 cells/mL decreased rapidly from 65 hours to 35 hours when the ratio of the size of the individual Synedra reaching 250 ㎛ or more, increased from 38% to 94%. Therefore, the size of the Synedra is considered to be a factor that significantly influences filter clogging, as well as the size of the Synedra population.
