Studies on Production of Heteropolysaccharide by Mutant of Xanthomonas malvacearum (Xanthomonas malvacearum 돌연변이주(突然變異株)의 Heteropolysaccharide 생산성(生産性)에 관(關)하여)

  • The mutant with high productivity, X. malvacearum SNUF 560-6, was acquired from the X. malvacearum SNUF 560 with low productivity by UV-light irradiation. It was preserved is lyophilized stock culture and it was transferred to PDA slant to maintain viability fortnightly. Fermentations were started by retransfering to MY agar slant from PDA stok culture. The experiments for optimal xanthan gum production were studied in a chemically defined medium. Of the carbon and nitrogen sources tested, 0.4% sucrose medium and 10mM glutamic acid medium yielded the highest xanthan gun production respectively. The addition of 10g/l succinic acid stimulated xanthan gum production. Also 65mM $PO_4\;^{-3}\;(12.6g/l\;KH_2PO_4)$ was effective on xanthan gum production. Finally, medium 1 and medium 2 which have high xanthan gum production potencies were achieved in this stud. The components of medium 1 and medium 2 were as follows: Medium 1 : sucrose 40g/l glutamate 10mM $PO_4\;^{-3}\;54mM\;(KH_2PO_4\;12.65g/l)$ Citrate 2g/l $MgSO_4{\cdot}7H_2O\;0.2g/l$ $H_3BO_3\;0.005/l$ ZnO 0.006/l $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$ Medium 2 : $Sucrose\;40g/l\;(NH_4)_2SO_4\;2g/l$ $PO_4\;^{-3}\;65mM\;(KH_2PO_4\;12.65g/l)$ Succinate 10g/l $MgSO_4{\cdot}7H_2O\;0.02g/l$ $H_3BO_3\;0.06g/l$ ZnO 0.006g/l $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$.

Plant Regeneration Through Adventitious Bud Formation and Callus Induction from Scales of Lilium lancifolium Thunb. (참나리 (Lilium lancifolium Thunb.) 인편으로부터 부정아 발생과 캘러스 유도를 통한 식물체 재생)

  • This study was performed to investigate the effects of various media compositions in regeneration of Lilium lancifolium. The adventitious bud initiation from microscale was the best on MS medium supplemented with BAP 1.0 mg/L and NAA 0.1 mg/L after 4 weeks of culture. However, from bulbscales, adventitious bud initiation was the best in dark condition on MS medium supplemented with BAP 0.5 mg/L and NAA 0.1 mg/L. On the other hand, callus induction was found to be the best from the microscales incubated in complete dark condition for 8 weeks on MS medium supplemented with 2,4-D 1.0 mg/L and BAP 0.1 mg/L. The highest plantlet regeneration from callus was obtained after incubation in the light condition for 8 weeks on MS medium supplemented with NAA 0.5 mg/L and BAP 0.1 mg/L. Rooting of shoots was obtained easily on MS medium and the plantlets were transferred to soil pots after 8 weeks. The chromosome analysis of the root tip cells was revealed that the callus-derived plantlets had normal chromosome number, 2n=24. No variation was observed in the morphology of the plantlets.

  • From the roots of Puisatiila koreana, three monodesmosides(pulsatilla saponins A, B and D) and two bisdesmosides(pulsatilla saponins F and H) were isolated. The structure of these saponins have been determined as hederagenin 3-O-${\beta}$-L-rhamnopyranosyl($1{\to}2$)- ${\alpha}$-L-arabinopyranoside(A), hederagenin 3-O-${\beta}$-D-glucopyrano syl($1{\to}4$) - ${\alpha}$-L-arabinopyranoside(B), hederagenin 3-O- ${\alpha}$-L-rhamnopyranosyl ($1{\to}2$)-[${\beta}$-D-glucopyranosyl($1{\to}4$]-${\alpha}$-L-arabinopyranoside(D), 3-O-${\alpha}$-L-rhamnopyranosyl($1{\to}2$)-{${\alpha}$-L-arabinopyranosyl hederagenin 28-O-${\alpha}$-L-rhamnopyrano syl($1{\to}4$)-${\beta}$-D-glucopyrano syl($1{\to}6$)-${\beta}$-D-glucopyranosyI ester (F) and 3-O-${\alpha}$-L-rhamnopyranosyl($1{\to}2$)-[${\beta}$-D-glucopyranosyl($1{\to}4$)]- ${\alpha}$-L-arabinopyranosyl hederagenin 28-O-${\alpha}$-L-lharnnopyranosyl($1{\to}4$)-${\beta}$-D-glucopyranosyl($1{\to}6$)-${\beta}$-D-glucop yranosyl ester(H) on the basis of chemical and spectral studies. Pulsatilla saponin B is the first report of its presence in plants but saponins A, D, F, and H have recently been isolated from the same genus p. cernua.

  • This paper measured a proper ratio of the size of demand fetch cache $L_1$ to that of prefetch cache $L_P$ by imulation when the size of $L_1$ and $L_P$ are constant which organize space-limited level 1 cache of a single microprocessor chip. The analysis of our experiment showed that in the condition of the sum of the size of $L_1$ and $L_P$ are 16 KB, the level 1 cache organization by constituting $L_P$ with 4 KB and employing OBL and FIFO as a prefetch technique and a cache replacement policy respectively resulted in the best performance. Also, this analysis showed that in the condition of the sum of the size of $L_1$ and $L_P$ are over 32 KB, employing dynamic filtering as prefetch technique of $L_P$ are more advantageous and splitting level 1 cache by constituting $L_1$ with 28 KB and $L_P$ with 4 KB in the case of 32 KB of space are available, by constituting $L_1$ with 48 KB and $L_P$ with 16 KB in the case of 64 KB elicited the best performance.

Appropriate Block Level in Neurolytic Lumbar Sympathetic Ganglion Block (신경파괴적 요부교감신경절 차단의 적정 부위)

  • Background: The lumbar sympathetic ganglia are variable in both position and numbers. The aim of this study was to detect the appropriate lumbar vertebral level where the lumbar sympathetic ganglia primarily aggregate. Methods: Forty patients comprising of hyperhidrosis (26 cases), complex regional pain syndrome (10 cases), peripheral artery occlusion disease (3 cases) and postherpetic neuralgia (1 case) underwent lumbar sympathetic block. We randomly selected one of two (L2 or L3) levels and the L4 level. The position of the needle tip and distribution of dye was verified by injection of a mixture of radio-opaque dye (1.5 ml) and 4% lidocaine (1.5 ml) and subsequently confirmed by L-spine anteroposterior and a lateral view X-ray. We considered the response positive when the skin temperature increased more than $1^{\circ}C$ in 5 min. Results: In general, the positive response ratio was greater when the needle tip located at the L2 or L4 level vice L3 and when the drug was distributed on the lower half of the L4 body and in the L4/5 intervertebral disc space. In a right side block, the positive response ratio was greater when the drug was distributed on the lower half of the L4 body and in the L4/5 intervertebral disc space, although in a left side block there was no significant difference seen between the levels. The complications of the neurolytic block were alcohol neuritis (7.5%) and hypoesthesia (5%) on the L1 or L2 dermatome. Conclusions: The best effect with least chance of complication may be induced by spreading the drug on the lower half of the L4 body and/or into the L4/5 intervertebral disc space by placing the needle tip on the L4 body.

Effect of plant growth regulators and carbon source on the shoot regeneration and rooting of 'Wonhwang' pear (Pyrus pyrifolia L.) (배 '원황'의 재분화와 발근에 미치는 식물생장조절물질과 탄소원의 영향)

  • The aim of this research is to establish shoot regeneration system for 'Wonhwang' pear (Pyrus pyrifolia L.) using various concentrations of 1-Naphthaleneacetic acid (NAA) 0.01, 0.05, 0.1, 0.5 mg/L in combination with benzylaminopurine (BA) 3, 5, 10 mg/L. Medium containing 4.4 g/L of Murashige and Skoog (MS) medium with vitamins containing 8 g/L of plant agar and 30 g/L of sucrose with NAA 0.05 mg/L and BA 3 mg/L showed 13.3% of shoot regeneration rate. 'Wonhwang' showed no root growth on existing rooting media of P. pyrifolia cv. Niitaka, 'Whang-keumbae' and 'Bae Yun No. 3'. We evaluated the effect of concentration and kinds of plant growth regulators and carbon source to establish efficient rooting condition for 'Wonhwang' pear. In the result of using various concentrations of NAA 0.5 mg/L and 1.0 mg/L in combination with indolebutyric acid (IBA) 3, 5, 10 mg/L, rooting rate of 24% was observed using 1/4 Linsmaier and Skoog (LS) medium supplemented with 7.5 g/L glucose as carbon source and IBA 1.0 mg/L with NAA 1.0 mg/L.

Effect of Plant Growth Regulator on Antioxidant Activity of Rosmarinus officinalis L. and Lavandula spica L. (로즈마리(Rosmarinus officinalis L.)와 라벤더(Lavandula spica L.)의 항산화 활성에 미치는 식물생장물질의 영향)

  • This experiment was conducted to investigate the induction rate of callus and antioxidative activity of rosemary and lavender. Callus induction rate of rosemary was 92% and 94% at 0.5mg/L and 1.0mg/L of 2,4-D in the media, respectively. The antioxidative activities of rosemary appeared differently according to the callus cultured under various concentrations of plant growth regulators. The strongest effect was observed at 0.1mg/L of 2,4-D extract, with $43{\pm}1.5{\mu}g/mL$ of an RC50 value. Callus induction rate of lavender was 98% and 99% at 1.0mg/L of 2,4-D and 1.0mg/L of NAA in the media, respectively. The regeneration rate of roots lavender was 85% at 1.0mg/L of NAA in the media. Antioxidative activities of lavender appeared diffently according to the callus cultured under plant growth regulators. The strongest effect was observed in 0.1mg/L of 2,4-D extract, with $37.6{\pm}0.9{\mu}g/mL$ of an RC50 value, which could be compared with value of leaf.

Prevalence of Leptospiral Antibodies in Sows (모돈에서의 Leptospira속균에 대한 혈중항체조사)

  • A serological survey was conducted to detect the type of Leptospirosis in sows. Antigen(live antigen) tested were L.icterohemorrhagiae, L.pomonal, L.canicola, L.Hardjo, L.australis, L.autumnalis, L.grippotyphosa, L.pyrogens, L.bataviae, L.hebdomadis. The survey was performed from J one 1992 to December 1992 by microscopic-agglutination test. The results were as follows 1, Among the serum samples of 92 heads of the sows, 6 heads of the sow(6.5%) were positive. 2. Among the positive samples of 6 heads, L.icterihemorrhagiae were 4 heads(4.3%) and L. pomona 2 heads (2.2%), respectively. 3. Antibody titers of positive sera were ranging from 1:200 to 1:1600. 4. The seroprevalence of leptospira in Chechon city, Chechon county, Danyang county that Chechon county(3.3%) was higher than that of other districts(1% -2.2%).

  • Seasonal variations of the contents of some chemical constituents of the estuary water at two definite stations of the laver bed in Nack Dong River have been determined over one tidal cycle in spring tide from Nov. 1962 to Oct. 1963. The ranges of annual variations of the contents at station 1 and station 2 are as follows: water temp. $2.2-30.8^{\circ}C$, $3.3-28.0^{\circ}C$; pH 7.8-8.5, 7.9-8.4; chlorosity 0.025-19.66 g/l, 4.31-19.56 g/l; magnesium 0.00355-1.565 g/l, -1.524 g/l; calcium 0.00557-0.482 g/l, - -0.590 g/l; saturation % of dissolved oxygen 71.8-123.2%, 88.2-113.8%; silicate-Si 8.00-125.5 ${\mu}$g-at./l, 6.70-100.5 ${\mu}$g-at./l; phosphate-P 0.12-1.47 ${\mu}$g-at./l, 0.11-1.09 ${\mu}$g-at./l; ammonia-N 4.88-25.45 ${\mu}$g-at./l, 4.12-17.58 ${\mu}$g-at./l; nitrite-N 0.07-0.75 ${\mu}$g-at./l, 0.08-0.58 ${\mu}$g-at./l; nitrate-N 2.11-6.89 ${\mu}$g-at./l, 1.85-7.43 ${\mu}$g-at./l each. The annual tidal variations of the constituents at station 1 are more remarkable than of station 2. The chlorosity, magnesium and calcium contents are decreased nearing the slack after ebb, and increased abruptly then one hour after the slack. The contents of the other constituents are varied according to the chlorosity variety. The values of pH, chlorosity, magnesium and calcium contents are lower in summer than winter, while the difference of seasonal variations of the % saturation of dissolved oxygen is not remarkable. The phosphate-P and total nitrogen contents have a tendency of increasing within a definite range, while the silicate-Si increase proportionally, to the increasing of mixing percentage of fresh water. The average values of Si/P and N/P are several times greater than of the normal in sea water. The chemical composition considered from the value of Mg/Cl or Ca/Cl of estuarine water varies according to the variety of chlorosity, even at the high chlorosity of 19 g/l.

  • The neutral compounds [$MCl_3L_2$(MeCN)] (M = Mo, V: L = $PPh_3$, 1/2 phda) have been prepared from the reaction of starting material $MCl_z$ (M = Mo; z = 5, M = V; z = 3) with N,P-donating ligands in acetonitrile solution. Addition of $AgClO_4$ to these neutral monomeric compounds in acetone solution were produced [$MCl_3-_nL_2(MeCN)(S)_n$]$(ClO_4)_n$ (n = 1, 2 : s = solvent). Finally treatment of bivalent cationic compound and neutral compound was formed chloride bridged dinuclear complex $[(MeCN)(L)_2ClM({\mu}-Cl)_2M'Cl(L)_2(MeCN)](ClO_4)_2$ and treatment of univalent cationic compound with half equivalent pyrazine to pyrazine bridged complex $[(MeCN)(L)_2Cl_2M({\mu}-pyz)M'Cl_2(L)_2(MeCN)](ClO_4)_2$. These complexes are characterized by elemental analysis, $^1H$, $^{13}C$ NMR, IR, Far-IR and UV-Vis spectroscopy.
