Lactobacillus reuteri의 주요 식품 위해 미생물에 대한 항균 효과 (Antimicrobial Activity of Lactobacillus reuteri Against Major Food-Borne Pathogens)

  • 본 연구에서는 5종의 유산균들 (Lactobacillus reuteri, L. acidophillus, L. bulgaricus, L. casei and Bifidobacterium longum)의 식중독 균들에 대한 항균력을 조사하였다. 각 유산균들을 3가지 서로 다른 조건 (MRS+glucose, MRS+0.5 M glycerol, 0.25 M glycerol solution)에서 배양한 후 그 상층액이 포함된 Moulter Hinton Agar에 8종의 유해균들을 접종하였다. MRS+glucose, MRS+0.5 M glycero에서의 상층액을 이용한 실험에서는 L. reuteri의 항균력이 다른 유산균들에 비해 높지 않았으나 0.25 M glycerol solution에서는 탁월하게 높은 결과를 나타내었다. (p<0.05). 0.25 M glycerol solution에서 나타난 높은 항균력은 L. reuteri가 glycerol을 이용하여 생산하는 reuterin이란 물질 때문이라 생각되며, 0.25 M glycerol solution의 상층액을 500 MHz Nuclear Magnetic Resonance (NMR)를 통해 분석한 결과 reuterin의 존재를 확인할 수 있었다. 3가지 조건 하에서의 항균력 시험 결과 가장 높은 항균력을 나타낸 3가지 유산균들 (Lactobacillus resuteri, L. bulgaricus, L. casei)을 선발, 각각 최고의 항균력을 나타낸 조건으로 배양하여 최소사멸농도 (minimum bactericidal concentration)를 측정한 결과, L. reuteri가 생산한 reuterin이 광범위 항균물질임을 확인하였다. 또한 pH 적정, pepsin 혹은 trypsin 처리를 한 후 3가지 유산균들의 항균력 변화를 조사한 결과 L. bulgaricus와 L. casei의 항균력은 이러한 조건에서 상당히 감소한 반면 reuterin의 항균력은 아무런 영향을 받지않았다 (p<0.05). 이상의 결과들로 보아 주요 유산균들 중에서 L. reuteri의 항균력이 가장 우수하면서 광범위하였으며 이는 L. reuteri가 분비하는 항균물질인 reuterin에 기인한 것으로 사료된다. 또한 reuterin의 항균력은 타 유산균과는 달리 장관내 pH나 단백질 분해효소에 의해 영향을 받지 않으므로 인체나 동물 장관에서 가장 높은 효력을 가진다고 판단할 수 있다.

통계적 방법을 이용한 Lactobacillus plantarum과 Lactobacillus reuteri 의 유효 배지 성분의 탐색 (Screening of Effective Medium Composition for the Cultivation of Lactobacillus plantarum and Lactobacillus reuteri Using Statistical Methods)

  • 본 연구는 가축용 생균제로서 이용성이 우수한 $Lactobacillus$ $plantarum$$Lactobacillus$ $reuteri$의 대량생산을 위한 경제적 최적 배지조성을 도출하기 위하여 수행되었다. 두 균주의 배양을 위한 배지성분 요인들은 유산균 배지로 알려진 MRS broth를 기준으로 하여, sucrose, glucose, molasses, yeast extract, corn steep liquor, soy peptone, dipotassium phosphate, manganese chloride, magnesium chloride, tween 80, sodium chloride, sodium acetate, ammonium citrate, sodium sulphate 및 ferrous sulphate 등 총 15가지를 선택하였다. 각 배지성분들이 각 균주들의 세포성장에 미치는 요인분석은 20개의 실험으로 구성된 Plackett-Burman design 방법을 이용하여 분석하였다. 그 결과 $L.$ $plantarum$의 성장에 있어서는 sucrose, glucose, molasses, yeast extract, corn steep liquor, soy peptone, sodium acetate, ammonium citrate 등이 긍정적인 효과(positive effect)를 나타내었다. $L.$ $reuteri$의 경우, yeast extract, soy peptone, $K_2PHO_4$, tween 80에서 수준간의 격차가 큰 긍정적인 효과가 나타났다. 두 가지 균주들에서 얻어진 표준화된 통합적 효과의 주 효과를 분석한 결과, sucrose, yeast extract, soy peptone에서 긍정적인 효과가 나타났다. 최종적으로 $L.$ $plantarum$$L.$ $reuteri$ 두 균주에 공통적으로 적용할 수 있는 유효 배지 성분들로 sucrose (20.0 g/l), glucose (5.0 g/l), soy peptone (11.0 g/l), yeast extract (5.0 g/l), $K_2PHO_4$ (0.2 g/l), $CH_3COONa$ (2 g/l) 및 $MgCl_2$ (0.02 g/l)를 선택하였다.

Lactobacillus reuteri 함유 Probiotics가 구강미생물의 생존 및 biofilm 형성에 미치는 영향 (The effects of Lactobacillus reuteri-containing probiotics on the viability and biofilm formation of oral microorganisms)

  • Objectives: This study aimed to evaluate the inhibitory effects of probiotics containing Lactobacillus reuteri on Streptococcus mutans and Aggregatibacter actinomycetemcomitans. In addition, the degree of biofilm formation, initial acidity, buffering ability, and acid production performance were measured to confirm the dental caries-inducing ability. Methods: S. mutans (KCTC3065) and A. actinomycetemcomitans (KCTC2581) were used as experimental strains. The number of viable cells, degree of biofilm formation, initial pH, buffering capacity, and production performance were measured for comparing L. reuteri-containing probiotics and Bulgaris. Results: The viability of S. mutans in the groups was reduced in the following order: Bulgaris, probiotics, control. The degree of biofilm formation was significantly higher at 0% and gradually reduced at different concentrations (p<0.01). At 2.5%, the absorbance of the probiotics and Bulgaris groups differed significantly (p<0.01). The acid formation ability differed significantly based on the performance of S. mutans in each product (p<0.05). The absorbance of the probiotics group was significantly lower than that of the Bulgaris group (p<0.01). Conclusions: This study suggests that the use of L. reuteri-containing probiotics as an adjuvant for the prevention and decreasing of oral diseases may reduce their incidence, which can be considered one of the benefits of using probiotics.

Heterologous Production of Pediocin PA-1 in Lactobacillus reuteri

  • The recombinant DNA pLR5cat_PSAB, in which pediocin PA-1 structural and immunity genes (pedAB) fused with the promoter and deduced signal sequence of an ${\alpha}$-amylase gene from a bifidobacterial strain were inserted in Escherichia coli-lactobacilli shuttle vector pLR5cat, was transferred to Lactobacillus reuteri KCTC 3679 and the transformant presented bacteriocin activity. The recombinant L. reuteri KCTC 3679 transformed with the shortened pLR5cat(S)_PSAB, where a nonessential region for the lactobacilli replicon was removed, also showed bacteriocin activity. The molecular mass of the secreted pediocin PA-1 from the recombinant bacteria was the same as that of native pediocin PA-1 (~4.6 kDa) from Pediococcus acidilactici K10 on a sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE) gel. In cocultures with Listeria monocytogenes, the recombinant L. reuteri KCTC 3679 effectively reduced the viable cell count of the pathogenic bacterium by a 3 log scale compared with a control where L. monocytogenes was incubated alone.

육계 맹장 유산균의 성장특성에 관한 연구 (Study on Growth Characteristics of Lactobacillus Isolated from Broiler Cecum)

  • This study was investigated to observe Lactobacillus spp. population of poultry digestive organ, duodenum, ilium, cecum and colon by growing steps and to select of valuable lactobacilli as probiotics. Two strains of male broiler commercial chickens, Hybrid and Ross, were used to evaluate population of Lactobacillus spp. in intestinal tracts. Three strains of Lactobacillus were identified, and bile salts environment. The number of lactovacilli was the lowest in duodenum compared to other intestinal tracts which had similar population. Population of Lactobacillus was maintained constantly regardless growing steps after one week of age. Identification of Lactobacillus from cecum resulted in L.reuteri BC5, L. crispatus BC7, L.reuteri BC9. All strains was depressed in pH 1 and 2, although two strains could survive for one hour at pH 2. And they could survive at pH 4 for 4hours. In bile salts tolerance, L.reuteri BC5, L.crispatus BC7 were maintained for 2 hours, but the growth reduced from 2hours. Growth of L.reuteri BC9 was increased continuously. In conclusion, Lactobacillus of intestinal tracts were established at first week, and maintained constant population. They were influenced on severe acidic condition and bile salts. Cecal Lactobacillus has different growth charcteristics by strains.

Assessment of probiotic potential of Lactobacillus reuteri MD5-2 isolated from ceca of Muscovy ducks

  • Lactic acid bacteria (LAB) are commonly used as probiotics in poultry. The present study employed in vitro and in vivo methods to select and test LAB isolated from Muscovy duck ceca as potential probiotics. In the in vitro study, 50 LAB were isolated from Muscovy duck ceca and tested for growth inhibition against Salmonella (S.) Enteritidis. Eleven isolates strongly inhibited S. Enteritidis and only 1 isolate (MD5-2) showing the strongest inhibition was selected for identification. This isolate was called as Lactobacillus (L.) reuteri MD5-2. For the in vivo investigation, 90 1-day-old Muscovy ducks were randomly assigned into three groups of 30 animals each (group 1, control; group 2, treated with $10^8$ colony-forming unit (CFU) of L. reuteri MD5-2 orally once on day 1; and group 3, treated with $10^8CFU$ of L. reuteri MD5-2 orally once daily from days 1 to 5). The ducks were housed in three large cages and raised for 50 days, after which body weight, duodenal villus height and crypt depth were measured. Both villus height and villus height to crypt depth ratio were significantly greater in group 3 than in groups 1 and 2. In conclusion, further investigation of L. reuteri MD5-2 as a potential probiotic strain is warranted.

The role of Lactobacillus reuteri DSM 17938 for the absorption of iron preparations in children with iron deficiency anemia

  • Purpose: To determine whether Lactobacillus reuteri DSM 17938 plays a role in absorption of iron preparations given to children with iron deficiency anemia (IDA). Methods: We performed a quasi-experimental study involving pre- and postintervention tests using a control group in North Sulawesi province, Indonesia, between July and September 2017. We conducted a single-blind controlled trial that included primary school children who were diagnosed with IDA based on reticulocyte hemoglobin equivalent (Ret-He) levels <27.8 pg/L. Results: A total of 66 children were randomized into 2 groups. Thirty-four children received iron preparations with the addition of L. reuteri DSM 17938 (group 1), whereas the other 32 received iron preparations alone (group 2). The baseline Ret-He levels before intervention were similar in both groups. After 14 days of intervention, mean Ret-He level in group 1 changed from $24.43{\pm}1.64$ to $28.21{\pm}1.72pg/L$ (P=0.000). Mean Ret-He level in group 2 changed from $24.31{\pm}1.42$ to $27.03{\pm}2.14pg/L$ (P=0.000). Statistical analysis showed a significant increase in Ret-He levels in both groups; Ret-He levels were significantly higher in the experimental group than in the control group (P<0.05). Conclusion: Children with IDA receiving iron preparations with L. reuteri DSM 17938 for 14 days show higher Ret-He levels than those receiving iron preparations alone.

유산균(L. reuteri)의 첨가가 돼지의 생산성, 분과 결장내 미생물균총 및 육등급에 미치는 영향 (Effects of Dietary Supplementation of Lactobacillus reuteri on Performance of Swine, Fecal and Rectumal Microflora and Carcass Grade)

  • Two hundred [(Duroc${\times}$Yorkshire)${\times}$Landrace] pigs were used in a 117-d growth assay (including four growth stages) to determine the effects of dietary supplementation of Lactobacillus reuteri on performance of swine, fecal and rectumal microflora and carcass grade. Pig diet was divided tow types, commercial diet (Control group) and supplemention of 0.1% Lactobacillus reuteri (Treatmental group). There was tend to increased in average daily gain (ADG) and feed efficiency (Feed/gain) in treatmental group than control group during the whole experimental period. The number of Lactobacilius spp. into rectum and feaces and carcass rate tended to increase in treatmental group than control group. Ammonia emission from excreta were decrease by supplemention of 0.1% Lactobacillus reuteri in feed (P<0.05). These results indicated that the dietary Lactobacillus reuteri were effective in performance, increasing of Lactobacilius spp. into rectum and feaces, decreasing ammonia emission from excreta, and it had also effective the carcass rate in swine.

The Effects of Probiotic Lactobacillus reuteri Pg4 Strain on Intestinal Characteristics and Performance in Broilers

  • This study was conducted to evaluate the feasibility of using L. reuteri Pg4, a strain isolated from the gastrointestinal (GI) tract of healthy broilers, as a probiotic. In preliminary in vitro studies the Pg4 strain was proven capable of tolerating acid and bile salts, inhibiting pathogenic bacteria and can adhere to intestinal epithelial cells. The probiotic properties were then evaluated on the basis of the broiler's growth performance, intestinal microbial population and cecal volatile fatty acid and lactic acid concentrations under conventional feeding. Dietary supplementation of dried L. reuteri Pg4 decreased significantly feed intake in grower chickens and improved significantly the feed conversion by 5% in a 0-6 weeks feeding period compared with the control group. The Lactobacillus counts in the crop, ileum, and cecum of the probiotic group were higher than in the control group. The L. reuteri Pg4 strain was traceable in the GI tract of probiotic supplemented chicks and showed capability of survival in the intestine for a protracted period. The probiotic group had a higher lactic acid concentration and lower pH value in the cecum than the control chicks. Probiotic supplement also affected the histology of the intestinal mucosa of chicks. The present findings demonstrated that L. reuteri Pg4 possesses probiotic characteristics and it is suggested, therefore, that the organism could be a candidate for a new probiotic strain.

개 아토피 피부염에서 3종 프로바이오틱스 복합제의 임상 효능 평가 (Clinical efficacy of L. plantarum, L. reuteri, and Ped. acidilactici probiotic combination in canine atopic dermatitis)

  • Canine atopic dermatitis (CAD) is an inflammatory and pruritic skin disease with a genetic predisposition, characterized by allergic sensitivity. It is known for its distinctive clinical features, including a high recurrence rate and chronic progression. To manage CAD, medications such as steroids and immunosuppressants are commonly used, but consideration should be given to the potential resistance and side effects associated with long-term use. In order to reduce these risks, various adjunctive factors are currently under consideration. One of these adjunctive agents, probiotics have shown effectiveness in regulating atopic dermatitis by modulating immune responses, as demonstrated in several recent studies. In this study, a substance combining three probiotics-L. plantarum, L. reuteri, and Ped. Acidilactici-was used in patients diagnosed with CAD, and its clinical effects and safety were evaluated. The trial involved four groups: a group receiving conventional treatment for atopic dermatitis (A), a group prescribed low-dose probiotics (B), a group prescribed high-dose probiotics (C), and a group prescribed topical probiotics (D). For assessment, the Canine Atopic Dermatitis Extent and Severity Index (CADESI), Trans-Epidermal Water Loss (TEWL) test, gut microbiome, and serum IgE test were conducted. As a result, the CAD severity index (CADESI-4) significantly decreased in the probiotics groups (B & C). In the serum total IgE test, the groups consuming probiotics showed a significant difference, while the group using topical probiotics (D) did not exhibit a significant change. Also, the TEWL test showed improved scores in the probiotics groups (B & C). Therefore, L. plantarum, L. reuteri, and Ped. Acidilactici probiotic combination could be considered as an effective adjunctive treatment, especially for atopic patients with moderate to severe skin lesions.
