New distribution records of two rare species of Cynanchum (Apocynaceae) in South Korea: Cynanchum thesioides (Freyn) K. Schum. and Cynanchum chinense R. Br.

  • Cynanchum L. in the subfamily Asclepiadoideae (Apocynaceae) includes four recognized species on the Korean peninsula, two of which are native to South Korea. However, the species ranges in South Korea are poorly defined. During a field survey, we discovered C. thesioides, previously unrecorded in South Korea, in Gimpo-si, Gyeonggi-do, and found an additional population of C. chinense, for which only one population has been reported in South Korea. The two taxa are considered rare species with extremely restricted distributions in South Korea, especially C. thesioides. We provide fundamental information, including descriptions, images of the habitats and morphological characters, and a taxonomic key for identification and assessments of the conservation status of Cynanchum species in Korea.

Plant Regeneration from Leaf and Internode Segment Cultures of Boxthorn (Lycium chinense Mill.) (구기자나무의 잎과 마디절편체 배양에 의한 식물체 재생)

  • Callus and shoot formation from medicinal crop, Lycium chinense Mill. cv. 'Cheongyang', as influenced by various media, explant sources and plant growth regulators were investigated. The rate of shoots formation, number of shoots, and fresh weight of shoots were the best on MS medium followed by B$_{5}$, WPH, and SH. Callus induction was more effective in leaf than internode segments, and was the best on MS medium containing 0.5 mg/L NAA with 0.2 mg/L BA. Effects of plant growth regulators in shoot formation were more effective in BA than TDA combined with NAA. Shoot formation from callus induced in leaf and internode segments was the best on MS medium containing 0.01 mg/L NAA with 0.2 mg/L BA.

  • This experimental study was done to research effects of a number of extract on the anti-allergic effect. The results were obtained as follows: 1. In effect of herb-extract on compound $\frac{48}{80}$-mediated histamine release from Evans blue skin assay, Isatis Japonica Miquel, Dictamnus dasycarpus Turcz, Spirodela polyrhiza, Cimicifuga heracleifolia, Bupleurum chinense, Magnolia liliflora, Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(MtOH), Trichosanthes kirilowii, Phellodendron amurense Rupr, Schizonepeta tenuifolia Var, Betula platyphylla show considerable visible anti-allergic effect. In the result of quantification of histamine induced compound $\frac{48}{80}$, Spirodela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively. 2. In effect of Herb-Extract on compound $\frac{48}{80}$-mediated histamine release from RPMC, Spirodcla polyrhiza, Cimicifuga heracleifolia inhibit histamine release effectively. 3. In effect of Herb-Extract on anti-DNP IgE-mediated histamine release from Evansblue skin assay. Spirodela polyrhiza, Cimicifuga heracleifolia(0.1mg/ml). Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(0.1mg/ml), Trichosanthes kirilowii(0.1mg/ml) show considerable visible anti-allergic effect. In the result of quantification of histamine induced anti-DNP IgE, Spiradela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively 4. In the result of genetic manifestative inhibition about the human mast cell-1(HMC-1), Cimicifuga heracleifolia has considerable effect in IL-4 in IL-5, and Tussilage farfara L. has in IL-4. According to the above results, it is suggested that several Herb-Extract have anti-allergic effect.

Isolation of the Constituents from Clinopodium chinense var. shibetchense and Inhibition Activity on Cancer Cell Growth and Nitric Oxide Production (산층층이꽃 추출물로부터 성분 분리 및 암세포성장 및 NO 생성 억제활성)

  • This study was performed to find anti-inflammatory or antitumor compounds from the polar fraction obtained from the extract of Clinopodium chinense var. shibetchense (H. Lev) Koidz (Labiatae). Chromatography of the BuOH fraction yielded two flavonoid glycosides (compounds 1 and 2) and two saponins (compounds 3 and 4). On the basis of spectroscopic data, compounds 1 and 2 were identified to be ponciretin 7-O-α-L-rhamnopyranosyl-(1→6)-α-D-glucopyranoside (neoponcirin) and naringenin 7-O-α-L-rhamnopyranosyl-(1→6)-α-D-glucopyranoside (isonaringin). Compounds 3 and 4 were identified to be 3-O-{β-D-glucopyranosyl-(1→2)-[β-D-glucopyranosyl-(1→3)]-β-D-fucopyranosyl}-saikogenin F (buddlejasaponin IV) and 3-O-{β-D-glucopyranosyl-(1→2)-[β-D-glucopyranosyl-(1→3)]-β-D-fucopyranosyl}-21β-hydroxysaikogenin F (clinoposaponin XV). In addition, ursolic acid (5) was isolated and identified from the CHCl3 fraction. Inducible nitric oxide synthase (iNOS) assay and sulforhodamine B (SRB) assay were performed to lead a potential anti-inflammatory or anti-tumor compounds from C. chinense var. shibetchense. Of the four compounds (1 - 4), compound 3 considerably inhibited cancer cell growth and NO production (IC50s, 5.59 μM in iNOS assay and 6.62 - 14.88 μM in SRB assay).

  • Volatile components were extracted from leaves of two Boxthorn (Lycium chinense M.) cultivars by using simultaneous distillation and extraction, analyzed by gas chromatography-mass spectrometry. Seventy components were identified : 13 acids, 15 alcohols, 18 hydrocarbons, 13 carbonyls, three esters, three ionones, and five others. The principal volatile components (and their peak area percentage) were n-pentanol (11.2~30.2%), phytol (14.5~28.3%), hexadecanoic acid (13.5~17.1%) 2,3-dihydrobenzofuran (1.5~4.2%), benzyl alcohol (1.9-4.8%), phenylacetaldehyde (1.8~3.2%), and octadecadienoic acid (1.7~10.7%). Fresh leaves showed much higher peak area than that of dried leaf in n-pentanol, n-hexanol, cis-2-penten-l-ol, cis-3-hexen-l-ol, benzyl alcohol, and $\beta$-phenylethyl alcohol, while dried leaves showed much higher content than that of fresh leaves in 9-hydroxytheaspran A, octadecanoic acid and octadecadienic acid.

Factors Affecting Introduction of rolC Gene in Lycium chinense Mill. (구기자나무(Lycium chinense Mill.)로의 rolC유전자 도입에 미치는 요인)

  • Transformation system of rolC gene, dwarf gene in Lycium chinenese Mill. established by using system. Pin-punctured leaves induced numerous adventious buds in abaxial side when cultured on 3/2 MS medium containing 2.0 mg/L zeatin. Survival rate and shoot regeneration frequency of leaf explants decreased as kanamycin sulfate level increased. Shoot buds were not regenerated on 3/2 MS medium containing 10 mg/L kanamycin sulfate and 2.0 mg/L zeaein. Of the level tested, 10 mg/L of kanamycin sulfate was optimum in selection of kanamycin sulfate resistant plant. Co-culture time of bacteria and leaf explants was affected at the frequency of shoot regeneration and survival of leaf explants. Leaf explants co-cultivated during above 48hr severely decreased survival rate and shooting rate. Best result on survival rate and shooting rate were obtained when exposed for 24 h. 80 explants of 105 leaf explants survived on 3/2 MS medium containing 2.0 mg/L zeatin and 10 mg/L kanamycin sulfate, and 15 shoots was regenerated on the same medium. To select kanamycin sulfate resistant plant, regenerate as cultured on 3/2 MS medium containing 10 mg/L kanamycin sulfate, and obtained 5 kanamycin resistant plants. Southern blot analysis conformed that the rolC gene was incorporated into the genomic DNA of kanamycin resistant plants.

Occurrence patterns of insect pests in the field of Lycium chinense under environment-friendly management (친환경 구기자재배지에서 다년간 해충발생양상)

  • The occurrence of insect pests of Lycium chinense was monitored at 4 organic farms that was cultivated with environment-friendly management at Cheongyang, Chungnam from May, 2012 to September, 2014. Total 4 orders and 18 species occurred in the L. chinense. Among them, 4 major pests and Lepidopteran and Hemipteran insect pests were investigated the occurrence pattern. During the budding, the population density of Myzus persicae was very high in late-May and early-June, and the injury by Lema decempunctata was increased from mid-June. The injuries by Henosepilachna vigintioctomaculata and Hedma lycia sp. were continued throughout the growing season. From the fruit held in late-July, hemipteran insects including Plautia stali and Halyomorpba balys were sucking the fruit, and their injury to L. chinense were gradually increased. In the case of Eriophys macrodonis densities to increase gradually from early July. The population density of E. macrodonis in 2014 was rapidly increased more than 5 times compared with 2012 and 2013. It was supposed that the initial control of mites or the removing of over-wintering density in the past year might be neglected. However, there is a need to more review the factors for the mass occurs.

Developmental characteristics of Henosepilachna vigintioctomaculata under different temperatures and control effect of eco-friendly agricultural materials on Lycium chinense (구기자에서 큰이십팔점박이무당벌레(Henosepilachna vigintioctomaculata) 의 온도조건에 따른 발육특성과 친환경제재에 의한 방제효과)

  • Host plants of the Henosepilachna vigintioctomaculata are known as Solanaceae plants such as potato, tomato, eggplant and Lycium chinense. Henosepilachna vigintioctomaculata (Motschulsky) cause continuous damage from early June to mid-August in leaves of L. chinense under environmental-friendly management in Cheongyang, Chungnam. The developmental period of egg and larva of H. vigintioctomaculata were investigated. H. vigintioctomaculata were reared on L. chinense at several temperatures, for example, 15, 20, 25 and $30^{\circ}C$ in the laboratory. In addition, the control effect of 5 eco-friendly agricultural materials were examined. As results, hatching rate is the highest to 91% at $25^{\circ}C$. Egg periods were 10.8, 7.7, 5.1 and 3.7 days at each tested temperature, respectively. And the total developmental periods from egg to adult were 42.7, 26.3, 18.4 and 19.4 days at each tested temperature, respectively. Otherwise, their eclosion rates from pupa to adult were showed 58%, 77%, 85%, 65% at each tested temperature, respectively. As a result of the control effect, both adults and larvae of H. vigintioctomaculata were shown a high mortality to mixture with extract of Sophora flavescens, Chenopodium ambrosioides and Melia azedarach. Additional, larvae is shown high mortality to extract of Azadirachta indica and Cymbopogon nardus. Thus H. vigintioctomaculata might be controled by using plant extract of eco-friendly agricultural materials in due season.

Resistance to Anthracnose Caused by Colletotrichum acutatum in Chili Pepper(Capsicum annuum L.)

  • Pepper fruit anthracnose, caused by Colletotrichum acutatum, results in serious yield loss and affects crop quality in many Asian countries, making it a disease of economic consequence. A source resistant to C. acutatum was identified by the AVRDC within the line Capsicum chinense Jacq. PBC932. The resistant breeding line C. annuum AR is the $BC_3F_6$ generation derived from C. chinense Jacq. PBC932. The inheritance of resistance to C. acutatum was analyzed in segregating populations derived from the two crosses HN 11$\times$AR and Daepoong-cho$\times$AR. Detached mature green fruits were inoculated using microinjection method. The disease response was evaluated as the disease incidence at 7 DAI. The segregation ratios of resistance and susceptibility to C. acutatum in the $F_2$ and $BC_R$ populations derived from the two crosses fit significantly to a 1:3 Mendelian model. This indicates that the resistance of AR to C. acutatum is controlled by a single recessive gene.

Plant Regeneration through Leaf Explant Culture of Boxthorn (Lycium chinense Mill.)

  • This study was conducted to investigate the optimal plant growth regulator level for the shoot formation of Lycium chinense Mill. In vitro plant propagation was developed for leaf explants of boxthorn. Leaf explants were cultured on MS medium supplemented with cytokinins (BA and 2-iP) alone. Plants were successfully regenerated through in vitro culture by using leaf explants of boxthorn grown in the field. After 4 weeks of culture, 58% of shoot formation had developed from the leaf explants. The shoot formation rate of 'Jangmyeong' was highest followed by 'Myeongan', ;Cheondae', and 'Bullo'. The use of 0.2mg/L BA was critical for enhanced production of shoot formation and resulted in 58% of the culture producing shoot formations. Regenerated plantlets transplanted to pots were developed and successfully acclimatized to greenhouse.
