COMPARATIVE STUDIES ON INDIGENOUS STARTER CULTURE STRAINS FOR THEIR RATE OF ACID PRODUCTION

  • Three strains each of L. bulgaricus and S. thermophilus isolated from indigenous dahi were examined for their rate of acid production and corresponding pH values in skim milk medium incubated at $40^{\circ}C$. No significant increase in titrable acidity or decrease in pH were recorded from initial period up to 2nd hour in both the strains. Following this period, however, there was a continuous increase in titrable acidity and a decrease in pH value in the milk for all the tested strains. Further it was observed that there was a variation among these strains for their acid production rate. High rate of acid production was recorded for the L. bulgaricus as compared to S. thermophilus. The results further, suggested that efforts should be made to select a proper pair of L. bulgaricus and S. thermophilus according to their rate of acid production, at a particular temperature in order to produce a good quality product.

The Effects of Freeze-Dried Potato Flour Addition on the Fermentation Characteristics of Yogurt (감마선 조사된 감자분말 첨가가 요구르트의 발효특성에 미치는 영향)

  • This study was performed to characterize the mechanism of the acceleration of lactic acid fermentation in milk incubated with potato flour irradiated by ${\gamma}$-ray and two types of lactobacillus, Lactobacillus bulgaricus and Streptococcus thermophillus, and to examine its effect on the quality of yogurt by assessing the acidity of lactic acid, viscosity, free amino acid, and sensory. In lactic acid fermented in the presence of 0.5% (w/w) potato flour, its acidity was higher, its pH was lower, and its viscosity was increased than lactic acid fermented in the absence of potato flour. In the sample fermented with the mixture of S. thermophillus and L. bulgaricus, the acid production rate was highest. In the samples fermented with a single type of bacteria, the acid production rate of S. thermophillus was higher than that the L. bulgaricus. Fermented a single type of bacteria with potato flour, the lysine utilization rate of L. bulgaricus and S. thermophillus was high. L. bulgaricus produced a large quantity of glutamic acid whereas S. thermophillus consumed glutamic acid. In sensory evaluation of yogurt fermented with potato flour, the preference of texture was improved while its color, taste, flavor, and overall preference were decreased. The data suggest that L. bulgaricus stimulates the growth of S. thermophillus by providing free glutamic acid that is required by S. thermophillus and thus the addition of potato flour shortens the fermentation period of yogurt.

Changes in the Microbiological Characteristics of Oat Extract by Lactic-bacterial Fermentation (귀리추출물의 젖산발효에 의한 미생물학적 특성 변화)

  • The effect of fermentation with mixed cultures of Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus salivarius subsp. thermophilus on the microbiological characteristics of oat extract was investigated. Changes in pH, titratable acidity and viable cell populations indicated that growth was better in mixed cultures of L. delbrueckii subsp. bulgaricus and S. salivarius subsp. thermophilus. Growth of S. salivarius subsp. thermophilus in oat extract was more rapid than growth of L. delbrueckii subsp. bulgaricus. Cooperative interaction between two cultures during fermentation of oat extract as in yogurt from cow's milk was observed, but the intensity was relatively weak.

Growth of Lactic Acid Bacteria in Soy Milk and Flavor of Soy Yogurt (두유(豆乳)에서 젖산균의 생육(生育)과 대두(大豆)요구르트의 향미(香味))

  • Soy milk prepared from soy protein concentrate was fermented with Lactobacillus acidophilus, L. bulgaricus, L. casci, Streptococcus lactis or S. cremoris. Growth and acid production of each organism in soy milk and flavor of soy yogurt beverages were investigated. Volatile compounds in soy milk and soy yogurts were also determined. Among the five organisms tested, L. bulgaricus produced most amount of acid in soy milk while S. cremoris produced least amount of acid in soy mile. Sensory evaluation showed that the flavor of soy yogurt beverages was inferior to that of milk yogurt beverage and the flavor of soy yogurt beverage prepared by L. bulgaricus was better than that of other soy yogurt beverages. Soy milk fermented with L. bulgaricus was more acceptable than unfermented soy milk. Lactic fermentation reduced n-hexanal in soy milk while it produced diacetyl that was not detected in unfermented soy milk.

Studies on Antioxidative Effect and Lactic Acid Bacteria Growth of Persimmon Leaf Extracts (감잎 추출물의 유산균 생육 및 항산화 효과에 관한 연구)

  • This study was investigated the growth effect of persimmon leaf extracts on various Lactic acid bacteria, the effect as freeze-dry cryoprotectan, by the changes of vitamin C content, electron donating ability, total cell count and SOD-like activity. The total cell count of L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 in the absence of persimmon leaf extracts(10%) at 37℃ after 48hr were 3.2×10/sup 9/cfu/mL, 2.9×10/sup 9/cfu/mL, 1.1×10/sup 9/cfu/mL, 1.6×10/sup 9/cfu/mL and 3×10/sup 9/cfu/mL, respectively, and pH and total acidity of those were 3.82∼3.88, and 1.697∼1.842. On the other hand, the total cell count of L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 in the presence of persimmon leaf extracts at 37℃ after 48hr were 4.4×10/sup 9/cfu/mL, 4.3×10/sup 9/cfu/mL, 2×10/sup 9/cfu/mL, 3.3×10/sup 9/cfu/mL, and 3.4×10/sup 9/cfu/mL, respectively, and pH and total acidity of those were 3.74∼3.82 and 1.528∼1.805. The total cell counter of lactic acid bacteria in the presence of persimmon leaf extracts(10%) at 37℃ after 48hr higher than those in the absence of persimmon leaf extracts. In freeze-dry chryoprotectan, the survival rate of L. acidphilus MG501 decreased about 30%. In case of L. brevis MG19 and L. bulgaricus MG515, the survival rate decreased about 10%. However, the survival rate of L. lactis MG530 increased about 10% and L. casei MG311 showed the almost same effect. The changes of vitamin C content were the highest in L. lactis MG530(190.26mg/mL) and the lowest in L. acidophilus MG501(56.05 mg/mL). The electron donating ability indicated to L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 added by 10% persimmon leaf extracts, respectively. when 10% persimmon leaf extracts were added L. brevis MG19, the electron donating ability is the highest of all lactic acid bacteria. The SOD-like activity of L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 showed lower than that of control.

Fermentation and Quality Characteristics of Soy Yogurt incorporating Insoluble Components of Domestic Soybeans (국내산 대두의 불용성 물질을 포함한 두유 요구르트의 발효 및 품질 특성)

  • The objective of this research was to determine the fermentation and quality properties of soy yogurt incorporating insoluble components fermented with Streptococcus thermophilus (S. thermophilus) and Lactobacillus bulgaricus (L. bulgaricus) for 24h. Boiled soybeans were ground and reconstituted into equivalent total solids (18%). After soymilk was homogenized with sugar, it was pasteurized in a water bath at $90^{\circ}C$ for 30 min. Two kinds of lactic acid bacteria were inoculated into two types of soymilk after cooling down $40{\sim}45^{\circ}C$ and fermentation at $37^{\circ}C$ for 24h. Titrable acidity, pH, viscosity, sugar content, and number of viable cells were determined in triplicate. Soy yogurt fermented with L. bulgaricus did not reach pH 4.5, where as S. thermophilus was considered good for achieving pH 4.08 and titratable acidity of 2.27% for 24 h. Soy yogurt fermented with S. thermophilus had a higher viscosity and lower sugar contents than that fermented with L. bulgaricusas incubation time increased. Total viable count was $1.80{\times}10^{10}CFU/mL$ on S. thermophilus and $2.16{\times}10^8CFU/mL$ on L. bulgaricus after 24 h at $37^{\circ}C$. However, there was no significant difference in sensory intensities and preference between the two samples. As a result, S. thermophilus was identified as a better culture than L. bulgaricus for the manufacture of soy yogurt incorporating insoluble components.

Quality Characteristics of Frozen Soy Yogurt Prepared with Different Proteolytic Enzymes and Starter Culture (단백분해효소 전처리 및 starter culture의 종류에 따른 frozen soy yogurt의 품질특성)

  • This study was carried out to evaluate the quality characteristics of frozen soy yogurts prepared with different proteolytic enzymes and starter culture. The viable cell counts of lactic acid bacteria in frozen soy yogurts were measured $10^8$ CFU/g by the single culture method, while $10^9$ CFU/g by the mixed culture method except ${\alpha}-chymotrypsin$ treatment. The viable cell counts of lactic acid bacteria did not decrease after freezing for 30 min in ice cream maker. The lactic acid bacteria from the mixed culture showed better bile acid tolerance than those from the single culture. The lactic acid bacteria from the frozen soy yogurt prepared with ${\alpha}-chymotrypsin$ and mixed culture of Bifidobacterium bifidum and Lactobacillus bulgaricus showed better acid tolerance and bile acid tolerance. The highest(73.45%) overrun was observed in the frozen soy yogurt treated with ${\alpha}-chymotrypsin$ and mixed culture of B. bifidum and L. bulgaricus. The melt-down percent was higher in the single culture than the mixed culture. In sensory test, the frozen soy yogurt prepared with ${\alpha}-chymotrypsin$ and mixed culture of B. bifidum and L. bulgaricus was the most desirable, the highest scores in sourness, bitterness and mouthfeel.

A Study on the Improvement of Stretchability of Mozzarella Cheese (모짜렐라 치즈의 신전성 향상에 관한 연구)

  • This study was carried out to investigate the improvement of stretchability in Mozzarella cheese based on the influence of various factors such as starter type, fat content, salt content and stretching temperature. To do this study, stretchability, meltability, fat leakage and the change of soluble nitrogen compound were measured during storage of the Mozzarella cheese at 4$^{\circ}C$ for 10 weeks. When L bulgaricus starter was added, stretchability of the cheese was excellent and cheese manufactured with starter of L bulgaricus and L. bulgaricus : Str. thermophilus = 1:2 improved meltability because of high soluble nitrogen compound. When salt content of cheese was 0.5%, this resulted in extreme deterioration of quality in regard to stretchability, meltability and fat leakage at five weeks and when salt content of cheese was 0.5, 1.0 and 1.5%, soluble nitrogen compound was 4.49, 4.45 and 2.61 % at one week and 19.71, 19.44% and 17.07% at eight weeks, respectively. Stretchability was good at high stretching temperature, and cheese with stretching at 60'C showed poor meltability at the first stage, but this cheese had good meltability in process of time because increase in soluble nitrogen compound was high at low stretching temperature.

Preparation of Soymilk Yogurt and the Related Studies (고형 두유 요구르트의 제조에 관한 연구)

  • This study was carried out to investigate the feasibility of production of soy yogurt, which is inexpensive and high protein product with an acceptable flavor, from soy milk using lactic acid bacteria. Utilization of various carbohydrates by Lactobacillus acidophilus, Streptococcus thermophilus and Lactobacillus bulgaricus was stuied. Sucrose, the major carbohydrate in soymilk, was fermented by L. acidophilus and S. thermophilus. None of the testing microorganisms was able to ferment melibiose and raffinose. Growth of lactic acid bacteria in soymilk was examined every 4 hour. L. acidophilus exhibited the highest growth rate during the early stage. After 16 hours of incubation, however, all the cultures with the exception of L. bulgaricus grew at nearly equal rates. Microscopic examination of mixed cultures showed that the presence of S. thermophilus was much more pronounced than that of L. bulgaricus. All the cultures with the exception of L. bulgaricus formed acid rapidly during 16 hours of incubation, bringing the titratable acidity to 0.6% and pH to 4.3, which was sufficient to cause coagulation of soymilk. L. bulgaricus produced acid to a much lesser extent and caused coagulation of soymitk after 30 hours at earliest. Three kinds of yogurts were prepared from 100% soymilk, 100% milk and 50% soy-50% milk combination by S. thermophilus and were evaluated by taste panel. Soy yogurt received a significantly lower mean odor score than milk yogurt. Soy yogurt had custardlike texture, while milk yogurt was syrupy. However the scores for texture as well as those for color and flavor did not differ significantly among the treatment. The mean total scores for yogurts were not significantly different and were equivalent to ratings between good and high fair.

Studies on the Electrofusion between Lactobacillus acidiophilus 88 and Lactobacillus bulgaricus IFO 13953 (Lactobacillus acidophilus 88과 Lactobacillus bulgaricus IFO 13953간의 Electrofusion에 관한 연구)

  • 균주개량 방법의 일환으로 electrofusion 법을 이용하여 protease 화성, lipase 활성, 내열성, 내산성 등이 우수한 L. bulgaricus와 bacteriocin을 생산하는 L. acidophilus 간의 융합조건을 검토하였다. Electrofusion에 의해 생성된 융합주는 식별하기 위해 streptomycin(2mg/ml)과 lincomycin(5$\mu\textrm{g}$/ml)에 내성을 나타내는 L. bulgaricus 변이주이와 kanamucin (600$\mu\textrm{g}$/ml)에 내성을 나타내는 L. acidophilus 88 변이주를 분리하였다. Electrofusion을 500V/cm, 100mses(49phms, 2300 capacotance)에서 수행했을 때 가장 융합이 잘 일어났으며 전기장의 세시와 시간이 증가할수록 융합요율이 현저히 감소하였다. 2가 양이온은 대체적으로 융합효율을 감속시키는 경향을 나타내었으나 1mM MgCl$_2$에서 융합 효율이 약간 증가하였다. PEG 400을 elec-trogusion buffer에 30%되도록 첨가했을 때, 가장 양호한 융합효율을 나타내었으며 그 이상에서는 오히려 융합 효율이 감소하였다. Electrofusion법과 PEG 매개에 의한 융합법의 융합 효율을 비교한 결과, 거의 비슷한 융합 효율을 나타내었으나 electrofusion 법과 PEG매개에 의한 융합법을 혼합하였을 때 가장 양호한 융합 효율을 나타내었다.
