pH values in all samples were in the range of 5.04 to 5.32. pH of kanjang prepared with normal pressure(NPK) was higher than that of kanjang prepared with high pressure(HPK) and kanjang prepared with steam(SPK). Titratable acidity(TA) in all samples were found to be 4.26~7.72 mL. TA in HPK(5.99~6.96 mL) was lower than that in SPK(4.26~5.61 mL) and NPK(7.32 mL). Reducing sugar(RS) contents in all samples were in the range of 25.18 to 194.81 mg%. RS contents of HPK and SPK were a similar pattern and were higher than that of NPK. Major free sugars(FS) were glucose, fructose and xylose. In total FS, SPK was higher than that of NPK and HPK. Major organic acids(OA) were succinic acid, oxalic acid, tartaric acid, malic acid and citric acid. In total OA contents, SPK(40.38~76.96 mg%) was higher than that of NPK(55.85 mg%)and HPK(27.78~34.21mg%). OA contents in HPK and SPK were decreased slightly according to the heating time of whole black bean. The highest percentage of fatty acid (FA) in all samples was palmitic acid(33.28~43.51 % as area % of GC chromatogram), followed in a decreasing order by stearic acid(14.15~19.16%) and myristic acid(5.93~19.14%). The composition ratios of three FA to total FA ranged from 55.97 to 72.85&. In all samples, there was myristic acid that was not contained in raw soybean.
A bacterium strain (K-173-10) which was isolated from waste soil of Korea brewing factory, could be grown on acetate as the sole carbon source and accumulated a considerable amount of L-glutamic acid in the medium. This strain was identified as the new species Brevibacterium ammoniagenes. This study was concerned not only with the culture condition for the production of L-glutamic acid and the cell growth, but also with the effects on concentration of various kind of organic substances, growth factors and penicillin. The results obtained were summarized as follow; 1. It was found that the concentrations of acetate and ammonium ions affected the growth of the bacterium as well as its L-glutamate accumulation. The optimum conditions of the composition of grown media for the growth of the bacterium and its glutamic acid production was found to be 40 g/l of total acetate, $100\;{\mu}g/l$ thiamine, $0.5\;{\mu}g/l$ biotin and $1{\sim}2g/l$ corn steep liquor as the growth factors. 2. Organic acid such as succinic acid, malic acid and ${\alpha}-ketoglutaric$ acid inhibited the cell growth as well as its L-glutamic acid production. 3. The penicillin (20 units/ml) stimulated the production of glutamic acid at appropriate incubation period. 4. It was found that this strain could grow in the presence of urea and ammonium acetate but not in other nitrogen sources.
This study was conducted to enhance the shelf-life of Kimchi and to make the unique taste of Kimchi by fermentation control. Kimchis, Prepared by win baechu soaked in 10% salt solutions containing 0.2-0.3% organic acids (OS-Kimchi), acetic acid, citric acid (CA-Kimchi), lactic acid, its mixtures and formic acid+acetic acid+fumalic acid+malic acid+citric acid(FAFMC), were examined for pH, titratable acidify, sensory evaluation, the number of total microbe and lactic acid bacteria, content of organic acids and texture during fermentation at 1$0^{\circ}C$. The decrease of pH and the increase in acidity, CA-Kimchi showed lower than those of control and various OS-Kimchi. Total microbe, lactic acid bacteria, content of lactic acid of CA-Kimchi were lower than those of control. The hardness of CA-Kimchi measured instrumentally was higher than that of control. Sensory scores of CA-Kimchi were also lower than those of control, so the Kimchi maintained good crispness and overall taste.
The present work is aimed to evaluate the conditions of the hydride generation (HG) for germanium analysis by inductively coupled plasma (ICP)-atomic emission spectrometry (AES). Twelve different kinds of acids were used such as phosphoric, hydrochloric, nitric, sulfuric, perchloric, boric, tartaric, malic, oxalic, tannic, citric, and acetic acid. It was found that phosphoric acid yielded the maximum efficiency of hydride generation. Also, efficient hydride generation was obtined with the buffer solutions containing phosphate ions over a wide range of pH. In addition, in the presence of phosphoric acid the interference caused by metals was suppressed in the hydride generation of germanium. As the concentrations of a reducing agent and a stabilizing increased the hydride generation efficiency and the acid concentration proviaing the maximum intensity were increased. By using an analytical method developed in this study, the contents of germanium in water and rock samples were determined. The detection limit of germanium in the presence of phosphoric acid was $0.03{\mu}g/L$.
The experiment was conducted to find changes of growth response, free proline content and organic acids on manganese chloride toxicity($4,000mg\;l^{-1}$) for germination and early growth in rice. Root growth was increased in kinetin $10^{-3}M$ compared with control and germination rate was also increased in kinetin $10^{-3}M$ as 89%. Chlorophyll contents was slightly increased in kinetin $10^{-3}M$. Free proline content at 3days in Mn $4,000mg\;l^{-1}$ was higher than at 7 and 10 days, and all kinetin concentrations promoted free proline content. In organic acids, particularly, malic acid was remarkably increased in kinetin $10^{-3}M$ compared with control and Mn $4,000mg\;l^{-1}$.
The chemical components and quality of Takjoo mash were examined on the product brewed by the case of commercial amylase, Lactobacillus bulgaricus and the yeast. 1. Lactic acid was produced at the range of 917~1,350 mg per 100 ml of the mash brewed using wheat flour, corn flour, rice and pressed barley, while succinic acid was formed at the range of 6~32 mg per 100m1 of the mash. 2. Citric acid was formed at the concentration of 6.5mg per 100m1 of the mash on the use of pressed barley, and malic acid formed at the concentration of 1.7 mg and 1.4 mg per 100m1 of the mash respectively, on the use of wheat flour and corn flour. 3. No remarkable differences between the kind of raw materials and commercial amylases and compared with the mash brewed with Nurook were observed in the components of fusel oil formed in the mash. 4. Seventeen kinds of the amino acids of the mash brewed with commercial amylase, Lactobacillus and the yeast were detected and its content was 31mg per 100ml, which is less than that of the conventional takjoo, but the contents of glycine and alanine which pray as the component of seasoning in takjoo were sufficient.

This study evaluated the physicochemical characteristics and nutritional components of Goroshoe (Ace rmono Max.) sap related with collection periods. The changes of browning index, turbidity, pH, total acidity, organic acids, free sugar, crude protein, crude ash, and minerals were investigated. Browning index and turbidity were slightly increased, while pH and total acidity were not changed as collection periods increased. Citric acid decreased slightly from 0.081 to 0.045 mg/mL at middle collection periods. Malic acid also decreased from 0.494 to 0.416 mg/mL at middle collection periods but then afterward increased to 0.674 mg/mL as collection periods increased. Sucrose decreased from 1.759 to 1.000%, while fructose increased from 0.023 to 0.044% as collection periods increased. Crude protein increased remarkably from 0.80 to 17.49 mg% as collection periods increased. Crude ash decreased slightly from 0.04 to 0.03% at middle collection periods, and then increased afterward to 0.06% as collection periods increased. Potassium increased from 13.43 to 49.75 mg/L as collection periods increased. These results indicate that the useful components of Goroshoe sap increased with increasing collection periods, but turbidity could be reduced.
This research investigated the physicochemical properties and antioxidant activities of hot air-dried red mustard (Brassica juncea L.) leaf (HR) and freeze-dried red mustard leaf (FR). Crude protein content was highest in FR, and crude fat and carbohydrate contents were highest in HR. However, moisture and crude ash contents were not significantly different between the two drying methods. Total free sugars were higher in HR compared with FR. Sucrose, fructose, and glucose were the major free sugars in both HR and FR. Contents of essential and non-essential amino acids were higher in HR compared with FR. The major organic acid of FR was malic acid, and the major organic acid of HR was malic acid. The contents of saturated and unsaturated fatty acids were higher in HR than in FR. Total mineral contents were higher in FR (10,187.22 mg%) compared with HR (9,815.80 mg%). Major minerals were K, Ca, and Na in the two drying methods. The contents of vitamins C and E in HR were higher than those in FR. Total polyphenol contents showed no significant difference between the two methods. However, total flavonoid contents in HR were higher than in FR. The $IC_{50}$ values of FR and HR in ABTS assay were 0.89 mg/mL and 0.65 mg/mL, respectively. The results of all experiments suggest that HR and FR can be natural candidates as a rich source of antioxidants for further chemical investigation.
This study was carried out to improve the analytical methods for determination of organic acids in tobacco leaf by HPLC. The samples for HPLC analysis were pre-treated by means of SPE. The calibration curve for each acid was linear and $R^2$ values ranged from 0.9990 to 1.0000. The limit of detection were obtained from the calibration curves and their values were between 1.39 to $4.87{\mu}g/mL$. Recovery rates of organic acids were between 88.6 % to 98.5 %. The concentrations of organic acids among the various tobacco leaves were compared to the concentration of organic acids, were in the order oriental, burley, flue-cured tobacco. In the case of flue-cured and oriental tobacco leaves, the order of concentration of organic acids was malic acid, oxalic acid, citric acid. But in the case of burley tobacco leaves, the order of concentration of organic acids was citric acid, oxalic acid, malic acid.
Immature and mature plums(Prunus salicina L.) were coated with calcium-added methylcellulose(CaMC) and hydroxypropylmethyl-cellulose-15(CaHPMC-15). Physicochemical, nutritional and sensory characteristics were compared during the 8-day storage period at $30^{\circ}C$. Irrespective of maturity, pH of plums increased and the titratable acidity decreased during storage. Free sugars such as glucose, fructose, sucrose and sorbitol were eventually decreased during storage, but Ca-added cellulose coatings influenced on the prevention of free sugars from decreasing especially in mature plums. Vitamin C contents in immature and mature plums became decreased during storage, but the coatings took effective on its retention. Changes of malic acid, the major organic acid in plums were appeared the same tendency as vitamin C during storage. Results of sensory evaluation showed the development of red color in peel and the softening of plum tissue, and also showed that the overall eating quality and appearance generally decreased during storage. However, the coated plums either CaMC or CaHPMC-15 exhibited the better sensory characteristics. As though the significant difference in postharvest storage quality were not found between the two coatings, CaMC and CaHPMC-15 coatings significantly contributed to the improvement of overall storage quality, especially the retention of vitamin C and organic acids only on the mature plums.
