The Production of Xylanase and $\beta$-Xylosidase by Aspergillus niger NRC 107 (Asperillus niger NRC 107에서의 Xylanase와 $\beta$-Xylosidase의 생산)

  • The production of xylanase and $\beta$-xylosidase was investigated in submerged culture of Aspergillus niger NRC 107. The maximum production occurred when the pH was controlled at 6.0 during the fermentation. Among the various carbon sources investigated, corn-cob xylan (1.5%, w/v) yielded maximal production of the enzymes. The $NaNO_{3}$ was the most favorable nitrogen source for enzyme production and $KH_2P0_4$ concentration at 0.3%(w/v) was found to be optimum. Incorporation of wheat bran to the culture medium improved xylanase production. Addition of L( -) sorbose to the culture medium promoted the secretion of $\beta$-xylosidase. It was possible to increase the production of xylanase (39.43 units/ml) and that of $\beta$-xylosidase (4.2 unitslml) by submerged culturing the A. niger NRC 107 in the modified medium.

Yeasts in the Flowers of Wild Fleabane [Erigeron annus (L.) Pers.]

  • BACKGROUND: Yeasts associated with fleabane flowers were identified using isolation methods previously applied in yeast biotechnology. A culture-based approach was required for isolation of many yeast strains associated with fleabane. METHODS AND RESULTS: We spread homogenized fleabane flowers onto GPY medium containing chloramphenicol, streptomycin, Triton X-100, and L-sorbose. We isolated 79 yeast strains from the flowers of wild fleabane, and identified the yeasts via phylogenetic analysis of isolates from agar plates. The yeast species included 39 isolates of Aureobasidium pullulans, 17 of the genus Candida, 14 of the genus Rhodosporidium, 6 of the genus Cryptococcus, and 3 of the genus Rhodotorula. CONCLUSION: Yeast isolates associated with fleabane flowers included A. pullulans (39 isolates) and other yeast species (40 isolates). Such yeast isolates may have biotechnological potential.

  • 유산균의 균주개량방법의 일환으로 photoplast fu-sion 기법과 electrofusion법을 이용하여 protease 활성, lipase 활성, 내열성, 내산성 등이 우수한 L. bulgaricus와 bacteriocin을 생산하는 L. acidophilus 간의 융합시켜 얻은 융합주들의 생리학적 성질을 검토하였다. 산 생성능, 내열성, 내산성 protease, lipase 활성 등 L. bulgaricus가 L. acidophilus보다 우수하였다. L. bulgaricus는 lactose와 sorbos를 이용하였으나. mal-tose 와 dorbitol을 이용하지 못하는 반면 , L. acidophilus는 matose를 이용하고 lactose 와 sorbose를 이용하지 못하였다. 융합주 가운데서는 367810번이 모균주의 발효능 특성을 함께 지님으로서 재조합체임을 확인 할 수 있었으며, sorbitol의 경우 모균주에서는 발효능이 전혀 나타나지 않았음에도 불구하고 융합주 4, 7번이 발효능을 나타내어 융합과정에서 새로운 형질을 획득하기도 한다는 사실을 알 수 있었다. Lactase 활성은 모균주 모두 높은 $\beta-galactosidase$ 활성을 보였으나, $phospho-\beta-galactosidase$ 활성은 거의 없었으며 융합주들도 다소 차이는 있었지만 모균주와 유사한 효소활성을 나타내었다. 발효에 있어서 key enzyme으로 작용하는 protease. lipase 등의 효소활성도 모균주의 활성보다 우수한 융합주도 존재하였다.

Yeasts in Internal Roots of the Rare Plant Dendropanax morbifera

  • To isolate and identify the yeast strains associated with D. morbifera, homogenized D. morbifera root samples were spread onto GPY, DG18, SCG and DOB agar media containing antibiotics, Triton X-100, and l-sorbose. Total 81 yeast isolates were analyzed by sequencing of internal transcribed spacer (ITS) region of the ribosomal DNA. The results showed that the root-associated yeast species were composed of the genera Vanderwaltozyma (40 isolates), Cryptococcus (40 isolates), and Kluyveromyces (one isolate). Moreover, the Kluyveromyces isolate exhibited high bioethanol productivity. In addition, the Vanderwaltozyma and Cryptococcus were dominant in D. morbifera roots. The specific yeast community associated with D. morbifera roots was identified by phylogenetic sequence analyses. These yeast isolates may have industrial applications as biosurfactant and bioethanol.

Yeasts Associated with Roots of the Endemic Plant Mankyua chejuense

  • BACKGROUND: Identification of endophytic yeasts inhabiting the internal roots of the Mankyua chejuense tree requires techniques involving biotechnology. There is a need for a culture-based method to isolate and identify yeast strains associated with M. chejuense.METHODS AND RESULTS: We spread homogenized M. chejuense root samples onto glucose-peptone- yeast agar containing antibiotics, Triton X-100, and L-sorbose. A total of 152 yeast isolates were obtained and identified via phylogenetic analysis based on ITS gene sequencing. The results revealed that the root-associated yeast species included the genera Cyberlindnera (140 isolates), Candida (11 isolates), and Kluyveromyces (one isolate). Additionally, three yeast isolates showed high bioethanol production.CONCLUSION: We identified the specific yeast community associated with M. chejuense roots. These yeast isolates may have industrial applications as bioethanol producers. Our findings revealed that Cyberlindnera isolates included C. suaverolens and C. satumus, while Kluyveromyces isolates showed high bioethanol production.

Nonvolatile Flavor Components in Chinese Quince Fruits, Chaenomeles sinensis koehne (모과의 비휘발성 Flavor 성분에 관한 연구)

  • This study was performed to identify detailed informations on the nonvolatile flavor of Chinese quince fruits, Chaenomeles sinensis koehne. About 72% of the free amino acids were shown to be valine, asparagine, ${\gamma}-aminobutyric\;acid$, aspartic acid and serine. Arginine, tyrosine, methionine and tryptophan were not present. Glutamic acid and glutamine as a amino acid for peptides were the major components, whereas cysteic acid, methionine sulfone and tryptophan were not detected. The nucleotides attained were composed of cytosine, uridine-5'-monophosphate and cytidine-5'-monophosphate, and these were proved to be a very small quantity. Guanosine-5'-monophosphate, inosine-5'-monophosphate and adenosine-5'-monophosphate were not present. The major sugars were shown to be glucose, sorbose, sucrose and fructose. Fructose was the most abundant one among them. A total of 11 organic acids were identified by capillary gas chromatography and capillary gas chromatography-mass spectrometry. The major components identified were tartaric acid and α-ketoglutaric acid. The total content of vitamin C determined was 386.6mg%, and those of ascorbic, dehydroascorbic, and 2, 3-diketo-L-gulonic acid were 28.8mg%, 154.5mg% and 197.3mg%, respectively. Calcium and phosphorus were the major components, while heavy metals such as cadmium, copper and lead were determined to be a small amount. In the result of organoleptic test on the natural and synthetic extract of Chinese quince fruits, the principal taste components consisted of free amino acids, sugars, organic acids, vitamin C and minerals. Five groups mentioned would have a favorable influence upon the taste of fresh Chinese quince fruits.

Endophytic Yeasts Colonize Roots of Ulmus parvifolia Jacq. and Quercus salicina Blume

  • BACKGROUND: Identification and characterization of endophytic yeasts inhabiting the roots of Ulmus parvifolia Jacq. and Quercus salicina Blume require biotechnological and culture-based techniques. METHODS AND RESULTS: Homogenized U. parvifolia and Q. salicina root samples were spread onto four types of agar medium containing ancgtibiotics, L-sorbose, and Triton X-100. In total, 25 yeast strains were isolated and subjected to phylogenetic analysis based on their internal transcribed spacer region sequences. The results revealed that the yeast genera Cyberlindnera (12 isolates) and Cryptococcus (1 isolate) were associated with roots of U. parvifolia; and the genera Rhodotorula (8 isolates), Trichosporon (3 isolates), and Kluyveromyces (1 isolate) were associated with roots of Q. salicina. Additionally, a Kluyveromyces isolate produced a detectable level of bioethanol. The yeast strains reported herein may be used in industrial production of biosurfactants and bioethanol. CONCLUSION: Our findings revealed that the endophytic yeast genera Cyberlindnera and Cryptococcus predominated in roots of U. parvifolia; and the genera Rhodotorula (8 isolates), Trichosporon (3 isolates), and Kluyveromyces (1 isolate) predominated in roots of Q. salicina. Additionally, Kluyveromyces isolates produced a detectable level of bioethanol.

Interaction Between the Quorum Sensing and Stringent Response Regulation Systems in the Enterohemorrhagic Escherichia coli O157:H7 EDL933 Strain

  • Quorum sensing and the stringent response are well-known regulation systems for the expression of virulence genes in enterohemorrhagic Escherichia coli (EHEC). However, how these two systems interact is not well known. E. coli strains with mutations in two regulation systems, ${\Delta}luxS$ (ECM101) and ${\Delta}luxS{\Delta}relA{\Delta}spoT$ (ECM201), and the ${\Delta}luxS$ complement strain to ECM201 (ECM202) were created from EHEC O157:H7 EDL933 to investigate how the regulatory systems interact. The phenotypic changes of the mutant strains were characterized and compared with the wild type. The mutant strains exhibited no obvious growth defects, although acid resistance and cellular cytotoxicity were decreased significantly in all the mutant strains. Phenotypic characterization revealed that mutations in the stringent response system (ECM201 and ECM202) influenced the metabolic (defective utilization of arabinose and L-sorbose) and enzymatic activities (decreased trypsin activity, and increased ${\alpha}$-glucosidase activity). In contrast, the quorum sensing system mutant (ECM101) did not display these phenotypes. The motility of the quorum sensing system mutant (ECM101) was unchanged, but mutation in the stringent response system influenced the motility. Our results suggest that quorum sensing interacts with the stringent response regulation system.

Synthesis lit Alcohol-oxidase in Hansenula polymoypha on Various Carbon Sources (여러가지 탄소원에 의한 Hansenula polymoypha의 Alcohol-oxidase합성)

  • The regulation of the synthesis of alcohol-oxidase (E.C.1.1.3.13.) was investigated in the methanol-utilizing yeasts during growth on different carbon sources. For this experiment, Hansenula polymoypha CBS 4132, Hansenula polymoypha CBM 11 and Hansenula polymoypha Cooney were cultured in mineral salt medium by changing its carbon sources. The production of alcohol-oxidase was varied by the carbon sources. For exmaple, alcohol-oxidase was undetectable: in all strains submitted to the test in the medium with glucose, but its production was rapidly increased when the carbon source was changed from glucose to methanol after 30 hrs of incubation. Moreover, this enzyme was not synthesized during growth on the primary aliphatic alcohols alone (ethanol, propanol, butanol or pentanol) or on the mixed substrates (0.5% methanol + 0.5% primary aliphatic alcohols). When cells were grown on the various carbon sources (glucose, xylose, lactose, glycerol, galactose, saccharose, sorbose, lactic acid or acetic acid), the alcohol-oxidase was about one-tenth of the activity found in cells grown on methanol alone. These carbon sources together with methanol yielded far better synthesis of alcohol-oxidase than in case of carbon sources alone. Especially, the alcohol-oxidase activity of the cells grown on lactose or lactic acid together with methanol was far better or similar than that of cells grown on methanol alone. The synthetic activity of alcohol-oxidase of Hansenula polymoypha CBS 4132 was the strongest among the three strains tested in every respect.

Detection of Auxotrophic Mutants form Valsa ceratosperma, the Causal Fungus of Apple Canker (사과나무 부란병균(腐爛病菌) Valsa ceratosperma에서의 Auxotrophic Mutants의 검출(檢出))

  • This study was conducted to elucidate the most appropriate method to obtain auxotrophic mutants from Valsa ceratosperma, the causal fungus of apple canker, which may be used as a gene marker in detecting the transfer of the factors of avirulent strains to virulent strains. Among the 3 kinds of synthetic media tested, each have two formula for minimal and complete, the medium which has been used in study of Endothia parasitica (E. P medium) was turned out to be most appropriate for the growth of V. ceratosperma. A medium for single colony formation from pycnidiospore of this fungus was developed by adding 0.5% L - sorbose to the E. P minimal medium. The period of incubation in dark for preventing the photoreactivation after U. V irradiation was estimated as about 60hrs at which most of the spores become binucleate. Largest number of putative auxotrophs were obtained at about 50second of irradiation to the spores smeared on the medium for single colony formation, at which the survival rate of spores was 5 to 6 percent. With these method developed in this experiment, 161 isolates of putative auxotrophs were detected among which the nutrient requirement for 10 isolates were determined. Five out of 10 mutants were still virulent to apple tree and all but one could not sporulate.
