Molecular Mechanism of L-Pyroglutamic Acid Interaction with the Human Sour Receptor

  • Taste is classified into five types, each of which has evolved to play its respective role in mammalian survival. Sour taste is one of the important ways to judge whether food has gone bad, and the sour taste receptor (PKD2L1) is the gene behind it. Here, we investigated whether ʟ-pyroglutamic acid interacts with sour taste receptors through electrophysiology and mutation experiments using Xenopus oocytes. R299 of hPKD2L1 was revealed to be involved in ʟ-pyroglutamic acid binding in a concentration-dependent manner. As a result, it is possible to objectify the change in signal intensity according to the concentration of ʟ-pyroglutamic acid, an active ingredient involved in the taste of kimchi, at the molecular level. Since the taste of other ingredients can also be measured with the method used in this experiment, it is expected that an objective database of taste can be created.

The Effect of Docosahexaenoic Acid on Brain Function and Acetylcholine Level in Cerebral Cortex of Electroconvulsive Shock Induced Mice (Docosahexaenoic acid가 전기충격성 뇌장애 마우스의 기억력 및 Acetylcholine량 변화에 미치는 영향)

  • Electroconvulsive shock (ECS) increases the activity of acetylchohnesterase and decreases in brain acetylcholine levels. A large amount of free fatty acids accumulated in the brain tissue affects cerebral blood flow, brain edema and inflammation and results in brain injury. The present study examined the effect of docosahexaenoic acid (DHA) and D,L-pyroglutamic acid (D,L-PCA) on the learning and memory deficit using the passive avoidance failure technique and on the change of acetylcholine and choline level in the cerebral cortex of ECS-induced mice. The application of ECS (25mA, 0.5sec) induced a significant decrease in memory function for 30 min. ECS-induced a significant decrease in cortical acetylcholine and choline levels 1 min following the ECS application, which were almost recovered to ECS control level after 30 min. DHA (20 mg/kg, i.p.). administered 24 hr before shock. prevented the ECS-induced passive avoidance failure and the decrease of acetylcholine level 1 min following the ECS application. DHA failed to elicit a change in cortical choline level. DHA did not affect memory function and the cortical Ach and choline level of normal mice. The administration of D,L-PCA (500 mg/kg, i.p.) increased the effect of DHA on memory function and the change of cortical acetylcholine level of ECS induced mice. These results suggest that DHA treatment may be contributed to the prevention against memory deficit, and to the activation of cholinergic system in the ECS induced mice.

Physicochemical and Sensory Characterization of a Korean Traditional Rice Wine Prepared from Different Ingredients (첨가원료 종류에 따른 전통발효주의 이화학 및 관능특성)

  • In order to investigate the effect of different ingredients on the quality of Korean rice wines, 30% of rice were substituted by malt, corn, potato, soybean, glutinous millet, unpolished rice, glutinous rice, or non-glutinous rice, when rice wines were prepared. Physicochemical and sensory characteristics of the rice wines from the varying ingredients were evaluated. Sample rice wines were analyzed for ethanol, pH, total acid, amino acid, soluble solid, coloring degree, UV absorbance, reducing sugar, organic acids, and free sugars. After fermentation for 16 days, the ethanol contents ranged from 13.28 to 16.23%, while the total acid levels were within the range of 0.27 to 0.32%. The amino acid contents in eight samples ranged from 0.18 to 0.36%, while the soluble solid contents were within the range of 8.35 to $11.1^{\circ}$Bx. Among the eight samples tested, rice wine prepared with malt showed the highest level of coloring degree, UV absorbance, and reducing sugar levels, while rice wine prepared with potato showed the lowest value. Organic acid contents of rice wines prepared with soybean, glutinous rice, and non-glutinous rice showed the highest levels of succinic acid followed by acetic acid, citric acid, malic acid, and pyroglutamic acid. Free sugar contents of all rice wines showed the higher levels of glucose followed by fructose and maltose. Rice wines prepared with unpolished rice and corn showed the same highest overall sensory preference. By descriptive analysis, the overall mean sensory intensities of samples prepared with glutinous rice and non-glutinous rice, showed similar levels of 'sweetness', 'fruitiness', and 'freshness of aroma', while those samples prepared with soybean, glutinous millet, and potato showed stronger intensities of 'nuruk', 'grain aroma', and 'yellowness'. Rice wine prepared with corn showed middle ranges in all sensory attributes tested.

  • New anthracycline analogues 2-9 as potential anticancer agents have been synthesized from daunomycin (1a) and doxorubicin (1b). Compounds 2 and 6 were prepared by the nucleophilic displacement type esterification of 14-bromodaunomycin (1c) with N-benzoyl-(2R,3S)-phenylisoserine and L-pyroglutamic acid in triethyl-amine, respectively. Compounds 3, 7 and 4, 8 were prepared by the reaction of either daunomycin (1a) or doxorubicin (1b) with one equivalent of the corresponding acids in the presence of EDCI/PP. Compounds 5, 9 were obtained from 1b by reaction with 2.2 equivalents of the corresponding acids in the same manner. The cytotoxic activities of the analogues in comparison with adrimycin on cultured SNU-16 and MCF7 cell were described.

  • We have been isolating various physiologically active substances from non-saponin fraction of Korean Red Ginseng These are adenosine, gyro-glutamic acid, dencichine and acidic polysaccharide. Adenosine and gyro-glutamic acid are loom to inhibit epinephrine-induced lipolysis in fat cells and stimulate the insulin-mediated lipogenesis. In addition to these actions, adenosine was found to inhibit both norepinephrine- and histamine-induced aorta constriction, and pyre·glutamic acid inhibits angiotensin-converting enzyme. Dencichine stimulated histamine-induced aorta constriction. Finally, acidic polysaccharide was found to inhibit both lipolytic and anorexigenic actions of Toxohormone-L. Based on these experimental results, I presented a brief review on these compounds isolated from non- saponin fraction of Korea Red Ginseng. Antibacterial Activity by Lactobacillus bulgaricus SP5 against Pathogenic Bacteria (병원성 미생물에 대한 Lactobacillus bulgaricus SP5의 항균활성)

  • This study was carried out to get basic resources for the industrial use of Lactobacillus bulgaricus SP5. The antibacterial activity of the supernatant obtained from Lactobacillus bulgaricus SP5 was tested against the pathogenic bacteria such as Escherichia coli KCCM 11234, Salmonella enteritidis KCCM 3313, Salmonella enteritidis KCCM 12021, and Salmonella typhimurium KCCM 40253. The supernatant of L. bulgaricus SP5 showed antibacterial activity against tested pathogenic bacteria. The antibacterial activity was examined after adjusting pH and heat treatment of supernatant. Heat treatment of supernatant had antibacterial activity against pathogenic bacteria at all temperature. However, pH changes showed no antibacterial activity. Antibacterial activity of the supernatant was confirmed to be due to organic acids (lactic, acetic, phosphoric, succinic, pyroglutamic, citric, malic, and formic acid).

Alcoholic Fermentation of Bokbunja (Rubus coreanus Miq.) Wine (복분자 발효주의 양조특성)

  • In order to improve wine quality, the selection of yeast strain and of additives in the manufacture of Bokbunja (Rubus coreanus Miq.) wine was investigated. The chemical composition of the edible portions of Bokbunja fruits was 86.5% moisture, 0.2% crude protein, 0.9% crude fat, 6.6% crude fiber, 0.5% ash and $10^{\circ}Brix$ sugar, and was 2.99% fructose, 2.53% glucose and 0.07% sucrose in fruit extract. The predominant organic acids in the fruit were citric acid (14.57 mg/mL) and malic acid (2.24 mg/mL) with smaller amounts of shikimic, pyroglutamic and oxalic acid. During fermentation, citric and malic acid levels decreased, while formic and acetic acid were released. Saccharomyces cerevisiae KCCM 12224 (Sc-24) was more favorable for alcoholic fermentation of Bokbunja and the addition of 200 ppm of potassium metabisulphite to must was more efficient than other $SO_2$ sources with a higher overall acceptability score. Sc-24 increased alcohol production from 9.8 to 14.8% in a sugar concentration dependent manner $(18-28^{\circ}Brix)$. The color value of early stage Bokbunja must was improved by supplementing with Japanese apricot extract, but this did not influence the color value of Bokbunja wine after primary fermentation. The astringent taste of Bokbunja wine was reduced by removing the seed from the fruit. Sugar solution (50%, w/v) was used instead of sugar power to prevent the possibility of undissolved sugar due to insufficient mixing. This substitution did not influence sensory evaluation.

Algicidal Activity of a Dibenzofuran-Degrader Rhodococcus sp.

  • Rhodococcus sp. strain p52, a previously isolated dibenzofuran degrader, could effectively inhibit the growth of cyanobacteria, including species of Microcystis, Anabaena, and Nodularia. When strain p52 was inoculated at the concentration of $7.7{\times}10^7$ CFU/ml, 93.5% of exponentially growing Microcystis aeruginosa ($7.3{\times}10^6$ cells/ml initially) was inhibited after 4 day. The threshold concentration for its algicidal activity against M. aeruginosa was $7.7{\times}10^6$ CFU/ml. Strain p52 exerted algicidal effect by synthesizing extracellular substances, which were identified as trans-3-indoleacrylic acid, DL-pipecolic acid, and L-pyroglutamic acid. The effective concentrations of trans-3-indoleacrylic acid and DL-pipecolic acid against M. aeruginosa were tested to be 0.5 mg/l and 5 mg/l, respectively.

A Comparstive Study of Nutrients and Teste Components in Korean and Imported Red Peppers (시중에서 유통되고 있는 일부 국산 고추가루와 수입산 고추가루의 영양성분 및 맛성분에 관한 비교연구)

  • This study was performed to compare the contents of nutrients and taste components in Korean(King and Dabok) and imported(from Myanma) red pepper powders. Red pepper imported from Myanma was narrower and lighter and showed significantly higher contents of moisture and lipid. Lower amount of fructose and glucose shown in imported red pepper powder seemed related to lower sensory score in sweet taste. Capsaicin was contained much higher in imported red pepper powder and looked related to higher score in pungent tastes. Organic acids like oxalic acid, malonic acid, succinic acid, pyroglutamic acid were significantly lower in imported red pepper powder. Imported red pepper powder showed lower amount of $\beta$-carotene, capsanthin and color value(L, a, b) than Dabok and obtained lower sensory score in color.
