한국산(韓國産) 겨우살이류(類)의 당류(糖類)와 Triterpenoids의 화학적(化學的) 조성(組成) (III) -한국산 참나무겨우살이(Loranthus yadoriki Sieb.)의 수용성 다당류의 구조적 특성 - (Chemical Constituents of Saccharides and Triterpenoids in the Korean Native Mistletoes (III) - Structural Features of Water-soluble Polysaccharides from Korean Oak Mistletoe(Loranthus yadoriki Sieb.) -)

  • This experiment was carried out to elucidate the sugar composition of polysaccharides and the structural features of water-soluble polysaccharides(WSP) isolated from Korean oak mistletoe, Loranthus yadoriki Sieb. The 48-hours ball-milled meals of extractive-free dried mistletoe sawdusts were extracted with distilled water for $24hrs{\times}2$ at room temperature. The extracts poured into 95% ethyl alcohol to precipitate. The separated precipitate of WSP, in form of yellowish white powder by lyophilization, was fractionated into four subfractions of WSP-1, WSP-2, WSP-3 and WSP-4 by anion exchange chromatography on DEAE-cellulose column. The sugar composition of WSPs was analyzed by GLC in form of their glycitol acetates, and the structure of polysaccharides in Fractions WSP-1 and WSP-2 was determined by FT-IR and GC-MS after methylation through and acetylation. The sugars of WSPs from Korean oak mistletoe, Loranthus yadoriki, are majorly arabinose and galactose in stem, galactose in leaves very high in content and showed difference in composition and monomeric units between stems and leaves. D-galactose, D-glucose and L-arabinose are the simple sugars consisting of polysaccharides in WSP-1. ($1{\rightarrow}3$)-Linked galactan is the bakcbone with side chain of ($1{\rightarrow}5$)- -L-arabinofuranosyl residues and ($1{\rightarrow}6$)- -D-galactopyranosyl residues, and ($1{\rightarrow}4$)-linked glucan also presents. ($1{\rightarrow}4$)-Linked rhamnogalacturonan and ($1{\rightarrow}4$)- and ($1{\rightarrow}3$)-linked galactan present in WSP-2.

Effects of Mono- and Polysaccharides on In Vitro Fertility of Boar Spermatozoa

  • This study was conducted to examine the effect of several saccharides on the induction of capacitation and acrosome reaction (AR) and to examine the effects of mono and polysaccharides on the penetration activity of boar spermatozoa. Spermatozoa were inseminated in medium with fucose, galactose and mannose as monosaccharide, and fucoicIan. galactan and marman as polysaccharide. The penetration rates were significantly (p<0.05) lower in medium with galactose (40.6%), mannose (38.1%), fucose (41.6%) and fucoidan (36.6%) compared with control (56.7%). The rates of AR were increased (40.7 to 59.8%) by the preincubation periods prolonged from 0 to 4 hr (p<0.05). Similar tendencies were observed in AR when spermatozoa were treated with monosaccharides, but not significantly differ among the groups treated with different time of preincubation with some exception of galactose. When spermatozoa were treated with polysaccharides, the rates of AR were significantly (p<0.05) increased by preincubation time prolonged from 0 to 4 hr with an exception of fucoidan. In conclusion, the present study suggests that penetration rate of spermatozoa is higher in presence of polysaccharides than monosaccharides. Also, it may resume that the comparing to control, the all saccharides (L-fucose, D-galactose, D-mannose, fucoidan. galactan and mannan)-treated groups slightly increase the AR pattern as preincubation time prolonged.

The Effect of Carbon Sources on Nisin Z Biosynthesis in Lactococcus lactis subsp. lactis A164

  • The effect of carbon sources on nisin Z biosynthesis in Lactococcus lactis subsp. lactis A164 was studied in batch culture using M17 broth containing different carbon sources. Among the eleven carbon sources tested, glucose, sucrose, and lactose were suitable carbon sources for cell growth of L. lactis A164. In particular, cells grown on lactose produced at least 3-fold greater amount of nisin Z than those on other carbon sources. Galactose resulted in less amount of cell mass than did sucrose or glucose, but gave a higher level of nisin Z activity. Northern blot analysis revealed. that lactose increased the transcription of the nisZ pre-peptide gene. Although galactose was less efficient than lactose, it increased the transcription of nisZ along with a higher level of nisin Z than did sucrose and glucose. These results suggest that the increased nisin Z production is correlated with the induction of nisZ by lactose and galactose. Among all the carbon sources tested, no remarkable differences were observed in nisRK and nisFEG transcripts, indicating that the lactose- or galactose-mediated induction is unique to the nisZ promoter.

Production of Chlorphenesin Galactoside by Whole Cells of ${\beta}$-Galactosidase-Containing Escherichia coli

  • We investigated the transgalactosylation reaction of chlorphenesin (CPN) using ${\beta}$-galactosidase (${\beta}$-gal)-containing Escherichia coli (E. coli) cells, in which galactose from lactose was transferred to CPN. The optimal CPN concentration for CPN galactoside (CPN-G) synthesis was observed at 40 mM under the conditions that lactose and ${\beta}$-gal (as E. coli cells) were 400 g/l and 4.8 U/ml, respectively, and the pH and temperature were 7.0 and $40^{\circ}C$, respectively. The time-course profile of CPN-G synthesis under these optimal conditions showed that CPN-G synthesis from 40 mM CPN reached a maximum of about 27 mM at 12 h. This value corresponded to an about 67% conversion of CPN to CPN-G, which was 4.47-5.36-fold higher than values in previous reports. In addition, we demonstrated by thin-layer chromatography to detect the sugar moiety that galactose was mainly transferred from lactose to CPN. Liquid chromatography-mass spectrometry revealed that CPN-G and CPN-GG (CPN galactoside, which accepted two galactose molecules) were definitively identified as the synthesized products using ${\beta}$-gal-containing E. coli cells. In particular, because we did not use purified ${\beta}$-gal, our ${\beta}$-gal-containing E. coli cells might be practical and cost-effective for enzymatically synthesizing CPN-G. It is expected that the use of ${\beta}$-gal-containing E. coli will be extended to galactose derivatization of other drugs to improve their functionality.

남해안 해수로부터 원유 분해 세균의 분리 및 특성 (Isolation and Characterization of Oil Degrading Bacteria from Southern Sea of Korea)

  • 남해안 유류 오염지역으로부터 유류분해능이 우수한 균주를 선별하여 동정한 결과 Pseudomonas aeruginosa로 동정되었으며, Pseudomonas aeruginosa BYK-2로 명명하였다. 균 성장 및 생물 유화제 생산을 위한 최적배양온도, 시간, pH, NaCl 농도는 각각 $25^{\circ}C$, 48시간, 7.0 이었으며, NaCl은 첨가하지 않았을 때 가장 많은 양의 생물유화체가 생산되었다. 본 균주의 경우, 1%(w/v) arabian light 원유와 bunker C oil을 기질로 7일간 배양한 결과 92.1%(w/w)와 76.2%(w/w)가 생분해되었다. n-hexadccane 에 대한 세포부착능이 실험에서는 탄화수소화합물들(arabian light 원유, kuwait 원유, bunker C 유, n-paraffine, n-hexadecane, n-tetradecane)을 기질로 배양했을때는 모두 80% 이상의 높은 세포부착능을 나타냈으며, 지방산들(oleic acid, olive oil, lecithin)중에는 lecithin을 기질로 배양했을 때 91.5%로 가장 높은 세포부착능을 나타냈어다. 또한 당들(arabinose, trehalose, dextrose, galactose, lactose, fructose, maltose, sorbitol, sucrose)을 기질로 했을 경우 arabinose, galactose, sorbitol, sucrose를 제외하고는 대부분 70%이하의 세포부착능을 나타냈다.

가열처리, 당의 첨가 및 발효에 의한 한국산 겨우살이의 세포독성변화 (Effects of Heat Treatment, Sugar Addition and Fermentation on Cytotoxicity of Korean Mistletoe)

  • 한국산 겨우살이를 이용한 암예방 기능성 식품의 개발을 위한 기초연구로써, 한국산 겨우살이가 Balb 3T3 cell line으로부터 얻어진 비종양성 A3l 세포와 종양성 MSV 세포에 미치는 세포독성 효과를 조사하였다. 겨우살이 생즙은 $ID_{50}$ 치가 $3.94\;{\mu}g/mL$로 강한 세포독성을 나타낸 반면, 열처리즙은 $30\;{\mu}g/mL$까지 세포독성을 나타내지 않았다. 한편, 낮은 농도의 열처리즙은 비종양성 A3l 세포의 증식을 촉진시키면서 종양성 MSV 세포의 증식을 억제시키는 효과를 나타내었다. 또한 galactose, lactose, glucose, mannose, fructose, sucrose 및 starch 등과 같은 탄수화물이 겨우살이의 세포독성에 미치는 영향을 조사한 결과, 생즙과 탄수화물의 처리는 큰 변화를 주지 못하였으나, 열처리즙과 lactose, galactose, glucose 및 fructose등과 같은 당을 함께 처리하였을 때 A3l 세포의 증식촉진, MSV 세포의 증식억제효과는 더욱 증가됨을 알 수 있었다. 이러한 결과에서 알 수 있듯이, 한국산 겨우살이가 당의 첨가나 첨가 없이 가열처리공정에 의해서 암 예방 기능성 식품의 재료로써 이용될 수 있음을 암시해준다. 그리고 발효에 의한 한국산 겨우살이의 세포독성 변화를 조사하기 위하여, 겨우살이 생즙과 열처리즙을 각각 Lactobacillus plantarum을 접종하여 발효시켰다. 생즙발효가 1, 3, 5, 7일간 진행되는 동안 종양성 MSV 세포의 세포독성에는 변화가 없었으나 비종양성 A3l 세포에서는 점차적인 증식촉진효과를 나타내었다. 그러나 열처리즙 발효의 경우에는 발효초기에 열처리즙 고유의 A3l 세포 증식촉진효과가 발효가 진행됨에 따라 그 효과가 점차 감소됨을 나타내었다.

부위별 고려엉겅퀴(Cirsium setidens Nakai)의 이화학적 성상 및 항산화 활성 효과 (Composition Analysis and Antioxidative Activity from Different Organs of Cirsium setidens Nakai)

  • 고려엉겅퀴(Cirsium setidens Nakai)의 생리활성물질을 탐색하기위하여 고려엉겅퀴의 일반성분 분석과 항산화 활성을 조사하였다. 각 부위별추출물 중 탄수화물의 함량이 가장 높았으며, 그 밖에 성분은 조단백질, 수분, 조회분, 조지방 순으로 높게 나타났으며, 부위별 무기성분은 모두 일곱 종이 검사되었고, 칼륨의 함량이 가장 높았다. 부위별 K의 함량은 잎 부위에서 5,371.97 mg/100 g, 삶은 및 부위에서 1,770.62 mg/100 g, 꽃 부위는 1,983 mg/100 g, 줄기 부위는 6,096.74 mg/100 g, 뿌리 부위는 1,604.2 mg/100 g로 나타났다. 구성당은 xylose, galactose, glucose 총 3종이 함유되었으며 xylose는 꽃과 줄기부위에서만 분리, 동정되었고, galactose는 줄기 부위에서만, glucose는 전체 부위에서 분리, 동정되었다. 지방산 중 linolenic acid(C18:3)와 linoleic acid(C18:2)가 가장 많이 존재했다. linolenic acid(C18:3)는 잎, 삶은 잎, 꽃 부위에서 각각 41.63%, 34.01%, 18.83%로 가장 많이 나타났으며, linoleic acid(C18:2)는 뿌리, 줄기, 삶은 일 부위에서 각각 49.59, 40.72, 39.24%로 가장 많이 나타났다. 각 용매 별 추출 수율은 잎 부위의 물 추출물에서 27.22%, 삶은 잎 부위의 75% 메탄올과 물 추출물에서 14.2%, 꽃 부위의 75% 메탄올 추출물에서 17.85%, 줄기의 물 추출물에서 14.8%, 뿌리 부위의 75% 메탄올 추출물에서 36.64%로 가장 높은 수율을 보였다. 고려엉겅퀴의 부위별 항산화 효과에서 꽃의 물 추출물에서 $53.47\;{\mu}g/mL$, 삶은 잎의 물 추출물에서 $75.84\;{\mu}g/mL$, 뿌리의 에탄올 추출물에서 $88.22\;{\mu}g/mL$, 잎의 에탄올 추출물에서 $111.19\;{\mu}g/mL$L, 줄기의 물 추출물에서 $257.48\;{\mu}g/mL$의 순으로 높게 나타났다.

Optimum Conditions for the Production of Tetramethylpyrazine Flavor Compound by Aerobic Fed-batch Culture of Lactococcus lactis subsp. lactis biovar. diacetylactis FC1

  • Optimum conditions for the production of acetoin and ammonia as the precursors of tetramethylpyrazine(TMP) were determined using Lactococcus lactis subsp. lactis biovar. diacetylactis FC1 in a modified Lactose-citrate broth containing galactose, citrate, and arginine. The cell growth and the productivity of acetoin and ammonia were remarkably increased in an aerobic culture with 10 $\mu M$ of hematin. For the optimum conditions of acetoin and ammonia production, the concentration of citrate and arginine were adjusted to 156 mM and 50 mM after 18 hr cultivation, and citrate and galactose to 156 mM and 50 mM after 36 hr cultivation, respectively. In these conditions, acetoin and ammonia were produced to the final concentration of 127 mM and 195 mM, which were the highest concentations, respectively. The optimum conditions of the TMP production were also determined as follows; 4 hours at 121, pH 8.3, and the maximal yield of TMP under these conditions was 0.81 g/l.

Aspergillus niger ATCC 16513과 대두(Glycine max. L) $\alpha$-galactosidase의 kinetic 성질 (Kinetic Properties of $\alpha$-Galactosidase from Aspergillus niger ATCC 16513 and Soybean(Glycine max. L))

  • Aspergillus niger ATCC 16513과 대두(Glycine max. L)의 정제 $\alpha$-galactosidase를 사용 하여 몇가지 이들의 kinetic성질을 조사 하였다. Asp. niger $\alpha$-galactosidase의 raffinose와 stachyose에 대한 Km값은 각각 37.0mM과 55.5mM, 대두 $\alpha$-galactosidase는 50.0mM과 55.5mM로서 PNPG보다 이들에 대한 친화성이 적었다. 또한 galactose는 ASP. niger와 대두 $\alpha$-galactosidase 모두의 활성을 저해 하였으나 2-mereaptoethanol과 L-cystene은 대두 $\alpha$-galatosidase의 활성만을 약간 저해 하였다. Asp. niger와 대두 $\alpha$-galactosidase의 활성에 관여하는 아미노산은 diethyl pyrocarbonate에 의한 화학수식에 의하여 histidine임이 확인 되었고 Asp. niger $\alpha$-galactosidase의 1mole당 아미노산 잔기수는 모두 902개, 대두$\alpha$-galactosidase는 286개 이었다.
