곰보배추 추출물이 항산화 및 피부미백효과에 미치는 영향 (Effects of Salvia plebeia Herb Extracts on Anti-oxidant Activity and Whitening action)

  • 본 논문은 재배가 용이하고 민간요법에서 다양하게 이용되고 있는 곰보배추의 에탄올 추출물이 화장품 소재로서 가능성이 있는지를 알아보기 위해 항산화, 미백효능에 대하여 관찰하였다. B16F10 세포에서 곰보배추 에탄올 추출물 25, 50, $100{\mu}g/mL$ 농도에서 독성이 나타나지 않았다. DPPH radical 소거능을 관찰한 결과 모든 농도에서 소거능을 보여주었고, $50{\mu}g/mL$의 농도에서 65.2%, $100{\mu}g/mL$에서 77.6%의 강한 항산화 효능을 나타냈다. Raw 264.7 세포 내에서 ROS 생성 저해능을 관찰한 결과 농도 의존적으로 유의하게 나타냈고, $100{\mu}g/mL$ 농도에서는 39.1% 억제하는 것으로 나타났다. NO생성 억제를 관찰하기 위해 Raw 264.7 세포에 곰보배추 에탄올 추출물을 25, 50, $100{\mu}g/mL$ 농도별로 첨가하여 관찰한 결과 농도 의존적으로 NO생성을 억제하였다. 시험관 내에서 L-DOPA와 L-tyrosine을 이용하여 곰보배추 에탄올 추출물이 tyrosinase activity를 농도 의존적으로 억제하는 것을 나타냈다. 세포 내에서 MSH를 가한 B16F10 세포에 곰보배추 에탄올 추출물을 25, 50, $100{\mu}g/mL$의 melanin 함량을 관찰한 결과 농도 의존적으로 억제하는 것을 보여주며 $100{\mu}g/mL$에서 30.7%로 억제하였다. 따라서 곰보배추 에탄올 추출물이 항산화 기능이 있는 미백 기능성 화장품의 소재로서 개발 가능성이 충분히 있는 것으로 사료된다.

위암에서 정량적 역전사 중합효소연쇄반응을 이용한 다중 표지자 분석 (Multiple Genetic Marker Analysis with Using Quantitative RT-PCR in Gastric Cancer)

  • 목적: 위암 세포주 및 조직에서 다중 표지자 mRNA 발현을 정량적 RT-PCR 검사를 통해 확인함으로써 이들 표지자를 이용하여 위암의 복강내 미세전이 진단이 가능한가를 평가하고자 본 연구를 시행하였다. 대상 및 방법: 12개의 인체 위암 세포주와 10개의 위암 조직을 대상으로 Carcinoembryonic antigen (CEA), Cytokeratin 20 (CK20), Dopa decarboxylase (DDC), L-3-phosphoserine phosphatase (L3PP)의 네 가지 mRNA를 이용한 정량적 RT-PCR 다중 표지자 분석을 시행하였다. 결과: 12개의 인체 위암 세포주 중 CEA는 4개(33%), CK-20는 1개(8%), DDC는 6개(50%), L3PP는 12개 세포주 모두(100%)에서 과발현되었다. 10개의 위암 조직 중 CEA는 9개, CK20은 3개, DDC는 9개, L3PP는 10개 조직 모두에서 과발현되었다. L3PP는 모든 위암 세포주와 조직에서 과발현을 나타내었으나 과발현 정도는 비교적 낮게 측정된 반면, CEA와 DDC는 일부 위암 세포주 및 조직에서만 과발현을 나타내었지만 파발현 시 충분한 발현도를 나타내었다. 결론: 위암 환자에서 하나 이상의 암 특이적 유전자를 이용한 다중 표지자 분석은 단일 표지자 분석이 가지는 단점을 보완할 수 있을 것으로 예상되며, CEA, DDC, 및 L3PP의 세 가지 mRNA가 후보 유전자로 사용될 수 있을 것으로 생각한다.

Antioxidant and Tyrosinase Inhibitory Activities from Seed Coat of Brown Soybean

  • Soybeans with brown, black, and yellow seed coats were compared to total phenolic contents and antioxidant activities including 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radicals. Also, 3 seed coats were examined for inhibitory activities on tyrosinase and lipoxygenase-1 on the basis of spectrophotometric and polarographic methods. Among seed coat extracts, 80% methanol extract of brown soybean seed coat showed the highest total phenolic contents ($68.9{\pm}3.29\;mg$ GAE/g) as well as exhibited potent scavenging effects on the DPPH ($IC_{50}=4.3\;{\mu}g/mL$) and ABTS ($IC_{50}=3.7\;{\mu}g/mL$) radicals. In a polarographic experiment, this extract was potentially inhibited the oxidation of L-tyrosine and L-3,4-dihydroxy-phenylalanin (L-DOPA) catalyzed by mushroom tyrosinase with $IC_{50}$ values of 12.4 and $63.7\;{\mu}g/mL$, respectively. It was also detected inhibition of the tyrosinase catalyzed oxidation of L-DOPA with an $IC_{50}$ value of 120.3 mg/mL in UV spectrophotometric experiment. In addition, this extract inhibited the linoleic acid peroxidation catalyzed by lipoxygenase-1 with an $IC_{50}$ value of $4.0\;{\mu}g/mL$. These results suggest that brown soybean may possess more beneficial effect on human health than black and yellow soybeans.

황기 약침액의 사람 섬유아 세포에서 Collagen 생성과 Tyrosinage 활성에 미치는 영향 (Effects of Astragali Radix Herbal Acupuncture Extracts on the Collagenase Activity and Procollagen Synthesis in HS68 Human Fibroblasts and Tyrosinase Activity)

  • 목적 : 본 연구는 황기약침액의 collagen 대사과정과 tyrosinage 활성에 대한 효과를 관찰하여 항노화 약침소재로의 개발 가능성을 알아보고자 계획되었다. 방법 : 황기 약침액이 사람 정상 섬유아세포 HS68에 UVB 조사 후 type I procollagen의 생산량 회복과 collagenage 활성에 미치는 효과를 ELISA법을 이용하여 측정하였다. 황기약침액의 tyrosinage 활성도에 미치는 영향을 측정하였다. 결과 : 황기 약침액은 사람 섬유아세포 HS68 실험에서 UVB 조사로 감소된 type I procollagen생성을 회복시키고, 통계적으로 유의하게 collagenage 활성을 억제하는 것을 관찰하였고 tyrosinage 활성을 통계적으로 유의하게 억제하지만, L-DOPA 산화는 억제하는 경향이 있는 것을 관찰하였다. 결론 : 본 연구 결과에 의하면, 황기약침액은 collagenage 활성을 억제하고, tyrosinage 활성을 억제하므로 주름개선과 미백효과가 있어 미용약침 소재로 개발가능성이 있을 것으로 사료된다.

Berberine이 백서의 6-hydroxydopamine-유도 파킨슨병 모델에 미치는 영향 (Effects of Berberine on 6-hydroxydopamine-induced Parkinsonism in Rats)

  • Many isoquinoline alkaloids including berberine lower dopamine content by reducing tyrosine hydroxylase (TH) activity and aggravate L-DOPA-induced cytotoxicity in PC12 cells. In this study, the effects of berberine on 6-hydroxydopamine (6-OHDA)-induced cytotoxicity in PC12 cells and on unilateral 6-OHDA-lesioned rat models were investigated. Berberine at 10-30 ${\mu}M$ did not affect cell viability in PC12 cells. However, berberine at concentrations higher than $50{\mu}M$ caused cytotoxicity at 24 h. Berberine (10-50 ${\mu}M$) also enhanced 6-OHDA (10-50 ${\mu}M$)-induced cytotoxicity at 24 h compared to 6-OHDA alone with an apoptotic process. In addition, treatment with berberine (5 and 30 mg/kg, i.p.) for three weeks showed a dopaminergic cell loss in substantia nigra of 6-OHDA-lesioned rats: 30 mg/kg berberine was more intensive cytotoxic. The levels of dopamine were also decreased by berberine (5 and 30 mg/kg) in the ipsilateral substantia nigra of 6-OHDA-lesioned rats. These results suggest that berberine aggravated 6-OHDA-induced cytotoxicity in PC12 cells and treatment with berberine (5 and 30 mg/kg) in 6-OHDA-lesioned rats also enhanced the degeneration of dopaminergic cell death and the decrease in dopamine levels in substantia nigra. Therefore, the long-term L-DOPA therapeutic patients with isoquinoline compounds including berberine may need to be checked for the adverse symptoms.

THE STUDY ON TISSUE CULTURED WILD MOUNTAIN GINSENG(Panax Ginseng C.A. Meyer) ADVENTITIOUS ROOTS EXTRACT AS A COSMETIC INGREDIENT

  • Korean ginseng(Panax Ginseng C.A. Meyer) known as a oriental miracle drug is an important medicinal plant. Ginseng has been used for geriatric, tonic, stomachic, and aphrodisiac treatments for thousands years. Also, it is an antibiotic and has therapeutic properties against stress and cancer. Ginseng is widely distributed all over the world. Among them, Korean mountain ginseng has the most valuable effect on pharmaceuticals. The roots of mountain ginseng contained several kinds of ginsenosides that have many active functions for the human body. However, the study of mountain ginseng has a limit because the mountain ginseng is very expensive and rare. So, we artificially cultured mountain ginseng adventitious roots using the bioreactor culture system. We induced callus from original mountain ginseng, directly dug up in mountain and aged about one hundred ten years. Separated adventitious roots were precultured in 500ml conical flasks and then, transferred in 20L bioreactors. The adventitious roots of mountain ginseng were harvested after culturing for 40days, dried and then, extracted with several solvents. In this study, we investigated the whitening effect, anti-wrinkle effect and the safety of tissue cultured adventitious roots extract of mountain ginseng in order to identify the merit as a cosmetic ingredient. Particularly, extract of mountain ginseng adventitious roots showed whitening and anti-wrinkle effects. The inhibitory effect of this extract on the melanogenesis was examined using B-16 melanoma cell. When B-16 melanoma cells were cultured with adventitious root extract, there was a dramatically decrease in melanin contents of 8-16 melanoma cell. And we identified this extract inhibited Dopa auto-oxidation significantly. Also, when transformed mouse fibroblast L929 cells were treated with this extract, there was a significant increase in collagen synthesis. The results show significant inhibited melanization and wrinkle without inhibiting cell viability.

  • 저자들은 생후 3개월에 전형적인 페닐케톤뇨증으로 진단받고 식사요법을 유지하였으나 지연된 발달 소견 및 지능 저하를 보이고 경련 증상이 있었던 9세 남자 환아에서 효소 검사와 유전자분석으로 dihydropterine reductase (DHPR) 결핍증을 진단하였다. 그리고 $BH_4$, 신경전달물질 전구체 투여 및 엽산 보충으로 DHPR 결핍증을 치료한 1례를 경험하였기에 문헌고찰과 함께 보고한다.

한국 BH4 결핍증 환자의 장기 추적관찰 (Long-term Follow-up of Patients with BH4 Deficiency in Korea)

  • Background and objectives: A deficiency of BH4 (tetrahydrobiopterin) not only causes the classical phenylketonuric phenotype, but also is the source of neurological signs and symptoms due to impaired syntheses of L-Dopa and serotonin. The treatment of BH4 deficiency usually consists of replacement with BH4 and the neurotransmitters. We performed this study to finding out long-term follow-up clinical symptoms and prognosis of BH4 deficiency. Methods: Clinical and biochemical, genetic analysis were done retrospectively from January 1999 to July 2015 in Soonchunhyang University Hospital. Results: In our study, total 207 patients were confirmed to hyperphenylalaninemia. Among them, 10 patients were BH4 deficiency. 9 patients were 6-pyruvoyl-tetrahydropterin (PTPS) deficiency and one patient was dihydropteridine reductase (DHPR) deficiency. The patients who received delayed treatment, most of our patients suffered from severe psychomotor retardation, hypotonia and seizure. c.259C>T mutation was identified most commonly in PTPS gene analysis. A patient with DHPR deficiency had a mental retardation, dystonia, seizure. His seizure semiology was dialeptic feature. His EEG showed generalized spike wave patterns. All patients had treated with tolerate L-Dopa, BH4 and 5-hydroxytryptophan. Most of the early treated patients have a good tolerance for drugs well. But some patients had neurologic symptoms, despite early detection and treatment. Conclusion: BH4 deficiency patients who had delayed treatment tend to have severe psychomotor problem and neurologic deficits.

참나물로부터 추출한 polyphenol oxidase의 부분정제 및 성질 (Purification and some properties of polyphenol oxidase from Spuriopimpinella bracycarpa)

  • 참나물로부터 추출한 polyphenol oxidase의 부분정제를 $(NH_4)_2SO_4$, 처리 및 Sephadex G-150 column chromatography에 의해 분리하였다. Polyphenoloxidase의 최적 pH와 최적온도는 7.5와$30^{\circ}C$였으며, pH 6.5에서 $70^{\circ}C$에서 30분간 처리시와 $80^{\circ}C$에서 5분간 처리시 완전 실활하였다. Polyphenol oxidase는 ascorbic acid, glutathione, potassium cyanide(0.1mM)에 의해 불활성화되었으며 L-cysteine, potassium cyanide, ascorbic acid, glutathione(0.5, 1mM)에 의해서는 완전 실활되었다. Catechol과 3,4-dihydroxytoluene의 기질은 높은 특이성을 나타낸 반면 pyrogallol, dopamine, DL-dopa의 기질은 강하게 활성을 억제하였다. Polyphenol oxidase의 $K_m$ 값은 86.5mM이었다.
