Differential Effects of Nitric Oxide Synthase Inhibitors in Rats

  • We investigated the action of NOS inhibitors on NOS in rats. Both of nitric oxide synthase inhibitors, $N^G$-monomethyl-L-arginine $(L-NMMA,\;3\;{\mu}M)$ or $N^G$-nitro-L-arginine methylester $(L-NAME,\;30\;{\mu}M),$ augmented phenylephrine $(PE,\;10^{-7}\;M)-induced$ contraction which was inhibited by acetylcholine (ACh) in rat thoracic aorta. This augmentation by L-NAME or L-NMMA was attenuated with the treatment of NO precursor, arginine. ACh, however, decreased the augmentation induced by L-NMMA, but not by L-NAME. Superoxide dismutase (SOD, 50 u/ml) potentiated an inhibitory effect of ACh on the PE $(10^{-7}\;M)-induced$ contraction. It has been known that platelet activating factor itself induces iNOS. Platelet activating factor $(PAF,\;10^{-7}\;M)$ inhibited PE $(10^{-7}\;M)-induced$ contraction. Pretreatment with L-NMMA (30 mM) or L-NAME (30 mM) significantly blocked the inhibitory action of PAF on PE-induced contraction. L-NMMA (100 mM) or L-NAME (100 mM) reduced nerve conduction velocity (NCV) relevant to nNOS in rat sciatic nerve. ACh attenuated the reduction of NCV by L-NMMA-, but not by L-NAME-induced reduction of NCV. These results suggest that L-NMMA and/or L-NAME have different action on three types of NOS in rats.

Effects on the thermal change of the Taeyon(L1) and the Chungbu(L1) area following acupuncture stimulation on Taeyon(L9) in man (태연(太淵)(L9)자침(刺鍼)이 태연(太淵)(L9)과 중부(中府)(L1)영역(領域)의 온도변화(溫度變化)에 미치는 영향(影響))

  • Backgrounds and purpose : The acupuncture of oriental medicine is very important in treatments. Until now it was been researched according to the meridian and qi xue(氣血) phenomenon of oriental medicine's theory. Acupuncture will show more objective index to observe the meridian. And then, I studied the effects on the thermal change of the Taeyon($L_9$) and Chungbu($L_1$) following acupuncture stimulation. Objective and Methods : This study was performed from December 1999 to February 2000 on 60 healthy students. The objective was divided into two groups, those are the control group A(N=30) that no acupuncture stimulation, the group B(N=30) of acupuncture stimulation on Taeyon($L_9$). First, in the control group A, I took a picture Taeyon($L_9$) Chungbu($L_1$) Taenung($P_7$) Chondol($CV_{22}$) area for 30 men without any stimulation with the Digital Infrared Thermograph Imaging(D.I.T.l.) and did same area, 10min after. Second, in the acupuncture stimulation group B, we took a picture Taeyon($L_9$) Chungbu($L_1$) Taenung($P_7$) Chondol($CV_{22}$) area for 30 men without any stimulation with the Digital Infrared Th - ermograph Imaging(D.I.T.I.), and then stimulate acupuncture on Taeyon($L_9$) and took a picture same area, 10min after. Results : 1. In healthy men, average skin temperture on Taeyon($L_9$) area was lower than Chungbu($L_1$) area about $3.0^{\circ}C$, in the Lt. Taeyon($L_9$) and Chungbu($L_1$) area was lower than Rt. Taeyon($L_9$) and Chungbu($L_1$) area. 2. In the acupuncture stimulation group B, the skin temperature of both side Taeyon($L_9$) area showed the increase or decrease significantly. But both Taenung($P_7$) area did not showed significantly. 3. In the acupuncture stimulation group B, the skin temperature of both side Chungbu($L_1$) area showed the increase or decrease significantly. But both Chondol($CV_{22}$) area did not showed significantly. 4. The thermal changes of the area which is a meridian point in the Lung Meridian of the acupuncture stimulation group on Taeyon($L_9$) different from other Meridian with significantly change. Conclusion : The acupuncture stimulation on Taeyon($L_9$) affected the thermal change of the area which is a meridian point, in the Lung Meridian. And then I could relate these results with the existence of the meridian and meridian point. Thus, continuous thermographic study will be needed for the existence of the meridian and meridian point.

Anti-oxidative Activity and Moisturizing Effect of Fermented Puer Tea Extract (발효보이차 추출물의 항산화활성 및 보습효과)

  • The fermented puer tea leaves were high concentrated extracted with 50% ethanolic solution in this study. Also, evaluating the anti-oxidative activity and anti-aging effect of this extract, we expected to apply the cosmetic industry. The yield of fermented puer tea extract was 17.9%. The total polyphenol content was 37.5%, tannin content was 7.5%, polysaccharide was 17.9%, unknown compound was 22%, and water content was 8%. Anti-oxidative activity (in-vitro) of fermented puer tea extract by DPPH method was $33.7{\pm}1.8%$ in 30mg/mL, $39.4{\pm}2.2%$ in 50mg/mL respectively. Therefore, we could know that anti-oxidative activity of fermented puer tea extract was effect higher than tocopheryl acetate and greentea extract. The collagen synthesis activity (in-vitro) of fermented puer tea extract was increased with $102.9{\pm}9.9%$ in 1 mg/mL, $111.5{\pm}9.9%$ in 5 mg/mL, $122.7{\pm}12.2%$ in 10 mg/mL, $131.5{\pm}13.7%$ in 30 mg/mL (*p-value£0.05, n=3). Skin moisturizing activity of fermented puer tea extract after application 8 hours was increased 38.5% higher than control samples both tocopheryl acetate and greentea extract. Total moisturizing effect was increased about 32.7% compared to before treatment. Fermented puer tea extract of this study can be applied to the skin care cosmetics industry.

Study of optimization of natural nitrite source production from spinach (시금치 유래 천연 아질산염 생산의 최적화 연구)

  • This study investigated the screening and optimization of nitrite production from fermented spinach extract using different lactic acid bacteria, fermentation temperature, and time. Spinach extract was fermented using various lactic acid bacteria at 24, 30, and $36^{\circ}C$ for 6, 12, 18, 24, 36, 48, 72, and 96 h in the presence of different carbohydrates (glucose, sucrose, fructose, and lactose). Lactobacillus farciminis (KCTC 3618) produced the highest amount of nitrite using fermented spinach extract at $30^{\circ}C$ for 28 h compared to Staphylococcus carnosus, L. coryniformis (KCTC 3167), L. fructosus (KCTC 3544), L. reuteri (KCTC 3677), L. amylophilus (KCTC 3160), L. hilgardii (KCTC 3500), L. delbrueckii (KCTC 1058), L. fermentum (KCTC 3112), L. plantarum (KCTC 3104), and L. brevis (KCTC 3498). Comparison of the yield at different fermentation temperatures showed that the highest amount of nitrite was produced using fermented spinach extract at $30^{\circ}C$. Similarly, maximum nitrite yield was observed after 36 h fermentationin in the presence of sucrose. Therefore, maximum nitrite production was observed upon L. farciminis-mediated fermentation of spinach extractat $30^{\circ}C$ for 36 h in the presence of sucrose.

Effects of Photoperiod and Water Temperature on Male Sex Steroid Levels in Cultured Small Yellow Croaker (Larimichthys polyactis) (광주기와 수온이 양식산 수컷 참조기(Larimichthys polyactis) 성성숙에 미치는 영향)

  • In the present study, we investigated the effect of photoperiod and water temperature on the gonadosomatic index (GSI), histological stage of the testes, and plasma levels of sex steroid (testosterone, T; 11-ketotestosterone, 11-KT) hormones in cultured male small yellow croakers (Larimichthys polyactis). In a photoperiod experiment, small yellow croakers were reared under a natural photoperiod (NP, 10L:14D-11L:13D), long photoperiod (LP, 14L:10D), and short photoperiod (SP, 10L:14D) at 17℃ for 90 days. The GSI was significantly higher in the LP group than in the other groups at 30 and 60 days. The plasma 11-KT levels were significantly higher in the LP group than in the other groups at 30 days. In a water temperature experiment, small yellow croakers were reared under natural water temperature (NT, 19.1-15.0℃), or at 17℃, 21℃, or 25℃ under a LP (14L:10D) for 60 days. The GSI was significantly lower for the 25℃ group than for the other groups at 30 and 60 days. The plasma 11-KT levels were significantly lower for the 25℃ group than for the other groups at 60 days. Therefore, the sexual maturation of cultured male yellow croakers was promoted by LP and inhibited at water temperatures above 25℃. These findings suggest that the sexual maturation of cultured male small yellow croakers is controlled by both the photoperiod and the water temperature.

  • The effect of levulinic acid (LA) and biosynthetic precursors of ${\delta}$-aminolevulinic acid (ALA) on the production of extracellular ALA was examined for the cells of soil derived Rhodospirillum rubrum N-1 belonged to the genus Rhodospirillaceae. The extracellular yield of ALA was increased to 23 fold (45 mg/l) from the basal condition (Lascelles' medium without L-glutamate) by successive addition of LA at initial (10 mM) and mid-log stage (30 mM) of cell cultivation. In addition to initial/mid-log mutual supplementations of LA (10 mM/30 mM) and glutamate (30/30 mM), respectively, by means of alternative feeding 10 mM $C_4$-precursors at mid-log phase of culture the extracellular ALA content was reached to 75 mg/l (40 fold).

Properties of jeonbyeong prepared with Artemisia annua L. powder (개똥쑥 분말을 첨가한 전병의 특성)

  • This study was conducted to investigate the effects of addition of Artemisia annua L. powder in jeonbyeong. Contents of crude ash increased as the amount of Artemisia annua L. powder increased, whereas crude fat contents decreased. According to increasing contents of Artemisia annua L. powder, total amino acid contents significantly increased. Hunter's L value increased as contents of Artemisia annua L. powder increased, whereas total free sugar contents decreased. Addition of Artemisia annua L. powder to jeonbyeong increased dietary fiber contents. The sensory score of jeonbyeong containing 100 g of Artemisia annua L. powder was the highest of all jeonbyeong tested. According to the results, addition of Artemisia annua L. powder could improve the quality and sensory characteristics of jeonbyeong.

Neuroprotective Effects of Herbal Ethanol Extracts from Gynostemma pentaphyllum on L-DOPA Therapy in 6-hydroxydopamine-lesioned Rat Model of Parkinson's Disease (돌외 에탄올 추출물 엑스가 6-hydroxydopamine-유도 파킨슨병 백서 모델에서의 L-DOPA 요법에 미치는 영향)

  • The neuroprotective effects of herbal ethanol extracts from Gynostemma pentaphyllum (GP-EX) in 6-hydroxydopamine (6-OHDA)-lesioned rat model of Parkinson's disease treated with L-DOPA were investigated. Rats were prepared for the Parkinson's disease model by 6-OHDA-lesioning for 14 days. The rats were then treated with L-DOPA (10 and 20 mg/kg) with or without the oral administration of GP-EX (30 mg/kg, daily) for 28 days. L-DOPA (20 mg/kg) treatment for 28 days enhanced dopaminergic neuronal cell death in 6-OHDA-lesioned rat groups, but L-DOPA (10 mg/kg) did not. However, the oral administration of GP-EX (30 mg/kg) for 28 days ameliorated the enhanced neurotoxic effects induced by chronic L-DOPA treatment in 6-OHDA-lesioned rat groups by increasing tyrosine hydroxylase (TH)-immunohistochemical staining and the number of TH-immunopositive cells surviving in the substantia nigra. In addition, GP-EX administration (30 mg/kg) for 28 days recovered the levels of dopamine and norepinephrine of the striatum in 6-OHDA-lesioned rat groups, which were markedly reduced by L-DOPA treatment (20 mg/kg). GP-EX (30 mg/kg) did not produce any signs of toxicity, such as weight loss, diarrhea, or vomiting in rats during the 28-day treatment period. These results suggest that GP-EX has protective functions against chronic L-DOPA-induced neurotoxic reactions in dopaminergic neurons in the 6-OHDA-lesioned rat model of Parkinson's disease. Therefore, GP-EX may be beneficial in the prevention of adverse symptoms in parkisonian patients.

Development of L-Lysine Producing Strains from Cellulosic Substrate by the Intergeneric Protoplast Fusion - Conditions for Fusion and Properties of Fusants- (속간 원형질체 융합에 의한 섬유질 기질로부터 L-Lysine 생산균주 개발 -융합조건 및 융합체의 성질 -)

  • To produce L-lysine from cellulosic substrate, the intergeneric protoplast fusion between Cellulomonas flavigena and Corynebacterium glutamicum, Cellulomonas flavigena and Brevibacterium flavum was performed. The fusion frequencies were 1.9$\times$10$^{-6}$ to 2.1$\times$10$^{-6}$ for the regenerated protoplasts when two parental strains were treated with 30% of polyethyleneglycol (M.W.6000) containing 5 mM EDTA at 3$0^{\circ}C$ for 30 min. Two fusants, FCB3 and FCC 19 were finally selected by comparision of their genetic stability and L-lysine productivity. The properties of fusants-DNA con-tent, G+C content and L-lysine productivity-were investigated. The DNA content of fusants was greater than those of the parental strain and their G+C contents are equal to half of total G+C con-tent of two parental strains. The fusants showed high productivity of L-lysine from carboxy methyl cellulose as substrate.

Enclosure Experiments on the Effects of Various Plants on Algae (경안천 현장실험조에서 식물체를 이용한 조류 증식억제)

  • Enclosure experiments to reduce the growth of Cyanophyceae were carried out using plants in Kyongan stream. Wet plants put into the enclosure at a rate of 2.5 g wet wt/l and at that time, the average concentration of chlorophyll a was ranged from 30 to $50\;{\mu}g/l$. The dominant species was Microcystis aeruginosa. Ginkgo, big cone pine and pine needles significantly inhibited the growth of Microcystis from the early days to the stages of log-Phase. Waterchestnut was the most inhibitory to the growth of Microcystis. Pine needles inhibited in 85% of the algal growth: ginkgo in 80%; big cone pine in 75%; waterchestnut in 78%; wildrice in 59%; and iris in 30%. At the treatment with 0.25 g dried plants/l, algae was declined at a rate of 90% by waterchestnut: 53% by pine needles. Phenolic compounds were purifled from decomposing big cone pine and waterchestnut.
