The Antioxidant Activity of Various Cultivars of Grape Skin Extract

  • The aim of this study was to analyze the antioxidant properties of different cultivars of grape skin extract in an in vitro system. The extracts were prepared from eight grape cultivars: 'Campbell Early' (CE), 'Kyoho' (K), 'New Kyoho' (NK), 'Muscat of Alexandria' (MOA), 'Seibel' (S), 'Morgen Schow' (MS), 'Gold Finger' (GF), and 'Meru' (M). The total phenolic acid contents were highest in MS and K. Resveratrol content was high in NK (50.88 mg/l g of coat), and quercetin content was significantly higher in K (0.68 mg/l g of coat) than in the other grape species (0.21-0.44 mg/l g of coat). The K and MS grape species, in which total phenol content was comparatively high (K: $24.15\;{\mu}g/mL$, MS: $25.52\;{\mu}g/mL$), also showed a high level of electron donating activity (K, 53%; MS, 59%). The hydrogen radical scavenging activity of M (50.36%) was significantly higher than the other grape species, including the S (50.21%), MS (49.43%), and K (49.06%) cultivars. Antioxidant activity varied depending on grape species, but overall it was highest in the MS and K cultivars.

  • This experiment was conducted to find the effect of a non-purine cytokinin, thidiazuron(TDZ), on the increase of berry size and fruit quality in 'Kyoho' grapes. For the comparison, the effect of forchlorfenuron(CPPU) was also examined The number of shot berries per cluster was significantly increased by the application of TDZ at 5 days after calyptra fall, resulting in the increase of cluster weight but no increase of each berry weight. The increase of berry weight was found when TDZ was applied at 10 to 15 days after calyptra fall. Gibberellin(GA $25mg{\cdot}L^{-1}$) was also effective on the increase of berry weight and the combined application of GA and TDZ further increased the berry weight. TDZ showed stronger effect on the increase of berry weight than CPPU at the same concentration. Berry ripening was more delayed at higher concentration of TDZ but no chemical injury was observed even at $20mg{\cdot}L^{-1}$. Results indicated that TDZ was effective on the quality improvement through berry weight increase and the recommended concentration TDZ for 'Kyoho' grapes was 5 to $10mg{\cdot}L^{-1}$.

Root zone environments in two cropping system within a year for Kyoho grapes (포도 '거봉'의 2기작 재배에서 근권환경 특성)

  • This study was performed to investigate the behaviour of root zone environments under the control of soil temperature and tension of soil moisture near the root Bone of 'Kyoho' grapes tree grown on restricted root zone system in plastic greenhouse. Maximum diurnal air temperature inside plastic greenhouse ranged between 25.1 and 32.7$^{\circ}C$, and the average of nocturnal air temperature inside plastic greenhouse maintained at 18$^{\circ}C$ in winter season. Also the minimum diurnal relative humidity ranged between 50 and 55%, and the maximum nocturnal relative humidity ranged between 84 to 87%. At a depth of 15cm from soil surface, the average soil temperature maintained at 25.6$^{\circ}C$ for under-ground heating, and appeared to 17.4$^{\circ}C$ for unheated condition. Although the tension of soil moisture just after irrigation sharply decreased to pF 1.5, the tension of soil moisture at the depth of 15cm maintained at pF 2.0~2.2. It is suggested that the tension of soil moisture at the depth of 15cm might be used as the standard for the determination of irrigation set point. Effective drainage system is needed to prevent the spindly and succulent growth of vine trees grown in restricted root zone system.

Physicochemical Properties of Different Grape Varieties Cultivated in Korea (국내에서 재배한 포도 품종간의 이화학적 특성 비교)

  • The aim of this study was to describe the effects of genetic factors on the chemical composition or metabolites of grapes harvested within the same region. Grapes were separated into pulp, skin, and seed, and physicochemical characteristics were compared among seven grape varieties. The sugar concentrations of the grape musts ranged from $15.17-20.93^{\circ}Bx$ with Seibel variety being highest at $20.93^{\circ}Bx$. pH ranges of grape musts were 3.46-4.02 and total acidity was highest with 1.05 in Steuben variety. Tartaric acid content was highest with 146.68, 500.10 mg/L in pulp and skin extract of Merlot variety. Malic acid content was highest in pulp extract of Seibel variety (1127.14 mg/L) and skin extract of Chardonnay variety (1720.06 mg/L). K content was highest with 379.13 and 828.01 mg/L in pulp and skin extract of Chardonnay variety. Ca content was highest in pulp extract of Kyoho variety (6.98 mg/L) and skin extract of Campbell Early variety (12.26 mg/L).

Fruit Quality and Freezing Damage of 'Kyoho' Grapes by Girdling (환상박피처리에 의한 '거봉' 포도의 과실 품질 및 동해 피해)

  • The effects of girdling on fruit quality and cold resistance of 'Kyoho' grapes were investigated. Girdling treatment was conducted on the trunk at 10 cm above ground with 1 cm width and grapes were harvested at 90 and 110 days after full bloom to compare the fruit quality. First harvesting rate in girdling treatment was higher than that in non-girdling treatment and coloration was also higher in girdled vines at the final harvest. In other words, coloring process of grape was promoted and enhanced by girdling, but this effect of coloring improvement was decreased after successive girdling treatment. Fruit quality showed no difference between the treated and non-treated berries, but fruit cracking rate was lower in girdled treated berries. Girdled trees were weakened and suffered from freezing damage. Especially, most grapevines withered up after being girdled for three consecutive years. Although girdling had effect on improving the berry coloring significantly, the effect wore off with continuous girdling. And it was possible that consecutive girdling leaded to wither and growth suppression especially in grapevines. These adverse effects may make the continuous girdling technique unsuitable in practice for 'Kyoho' grape.

Phonology and Minimum Temperature as Dual Determinants of Late Frost Risk at Vineyards (발아시기 정밀추정에 의한 포도 만상해 경보방법 개선)

  • An accurate prediction of budburst in grapevines is indispensable for vineyard frost warning system operations in spring because cold tolerance depends heavily on phonology. However, existing frost warning systems utilize only daily minimum temperature forecasts since there is no way to estimate the site-specific phonology of grapevines. A budburst estimation model based on thermal time was used to project budburst dates of two grapevine cultivars (Kyoho and Campbell Early), and advisories were issued depending on phonology as well as temperature. A 'warning' is issued if two conditions are met: the forecasted daily minimum temperature falls below $-1.5^{\circ}C$ and the estimated phonology is within the budburst period. A 'watch' is issued for a temperature range of -1.5 to $+1.5^{\circ}C$ with the same phonology condition. Validation experiments were done at 8 vineyards in Anseong in spring 2005, and the results showed a good agreement with the observations. This method was applied to the climatological normal year (1971-2000) to determine sites with high frost risk at a 30 m grid cell resolution. Among 608,585 grid cells constituting Anseong, 1,059 cells were identified as high risk for growing Kyoho and 2,788 cells for Campbell Early.

Effects of Bagging Periods on Pericarp Characteristics and Berry Cracking in 'Kyoho' Grape (Vitis sp.) (괘대시기가 '거봉' 포도의 과피 특성과 열과에 미치는 영향)

  • The berry growth and pericarp characteristics were characterized to confirm the effects of bagging periods on berry cracking during berry development in 'Kyoho' grape. The berry weight was the highest at 13.4 g in late period of bagging treated at 7 to 9 weeks after full bloom (WAFB) as compared with the lowest of 12.3 g in total period bagging. The berry cracking rate under critical turgor pressure in the non-bagging control was 53.3%, while those of bagging treatments were decreased in the order of 42.7%, 37.3%, 33.3%, and 18.7% in bagged during 3 to 9, 3 to 5, 5 to 7, and 7 to 9 WAFB, respectively. In the results of observation on histological characteristics of pericarp, berry lenticels of whole bagging treatments had smaller and normal shape compared with non-bagging control treatment. Especially on the pericarp of late period bagged during 7 to 9 WAFB, suberization around stomata and micro-cracking were not observed and structural strength of pericarp was increased with thicker sub-epidermal layer and cell wall. Therefore, the results indicate that bagging treatment for two weeks just before the veraison when the day length and daylight is relatively longer and stronger can effectively reduce berry cracking by strengthening structure of pericarp in 'Kyoho' grape.
