Isozyme Variation in Two Species of Freshwater Pleurocerid Snails in Korea: Koreanomelania nodifila and Koreoleptoxis globus ovalis (한국산 다슬기과 (Family Pleuroceridae) 2 종의 등위효소 변이)

  • A genetic analysis using starch gel electrophoresis was peformed to clarify the degrees of genetic differentiation among two species of the genus Koreanomelania and Koreoleptoxis (Gastropoda, Pleuroceridae). The average genetic similarity coefficients (S) among the populations of K. nodifila was S=0.848 (0.805-0.905) and K. globus ovalis was S : 0.755 (0.666-0.860). The genetic similarity coefficients between Semisulcospira gottschei and K. nodifila, S. gottschei and K. globus ovalis, and K. nodifila and K. giobus ovalis were 0.194, 0.301, and 0.301, respectively. So between K. nodifila and K. globus ovalis seems to be genetically two distinct taxa.

Study on Radulae of Seven Species of the Family Pleuroceridae in Korea (한국산 다슬기 과 7 종의 치설 연구)

  • These studies were conducted to compare the external radulae of seven species (Semisulcospira gottschei, S. coreana, S. forticosta, S. tegulata, S. ibertina, Koreanomelania nodifila and Koreoleptoxis globus ovalis) of freshwater snails in three genera of the family Pleuroceridae in Korea. Under a microscope of 100 magnifications, observation of seven species can be distinguished sharply between three genera of Semisuicospira, Koreanomelania and Koreoleptoxis, and the morphological characters of radulae can be used as taxonomic tool in genus level of Pleuroceridae. Besides SEM photograph of the radulae based on the morphological characters in seven species can be verified a new criteria for classification of genus' unit grade in accord with firsthand drawing sketch under a microscope of 100 magnifications. But it is difficult that all of the species can be detected sharply the difference of interspecies by comparisons of radulae in the family Pleuroceridae in Korea. Therefore, the morphological characters of radulae can not be set a criteria for classification of species level.

  • Koreoleptoxis nodifila (Martens, 1886) is an endangered species only living in the central and north streams of South Korea. However, there is a lack of information on physical habitat characteristics of K. nodifila. We aimed to determine preference ranges for water depth, current velocity, streambed substrate of K. nodifila. The weibull model was used to estimate the habitat suitability based on distribution of individual abundance by physical factors. Optimal depth preferences ranged from 0.53~17.17 cm, current preferences ranged from 48.40~81.03 cm s-1 and substrate (𝜱m) preferences ranged from -4.36~ -2.26. Median values of central tendency were determined as follows: water depth 16.73 cm, current velocity 65.23 cm s-1, substrate -3.51. Mean values of central tendency were determined as follows: water depth 21.32 cm, current velocity 65.65 cm s-1, substrate -3.63. Mode values of central tendency were determined as follows: water depth 5.17 cm, current velocity 64.77 cm s-1, substrate -3.24. Based on the habitat suitability analysis, the microhabitat types of K. nodifila were determined as riffle and coarse-grained streambed.

Biological Activities of Seven Melania Snails in Korea (국내산 7종 다슬기 추출물의 생리활성 특성 비교)

  • This study was conducted to compare the biological activities of 7 melania snails from the family Pleuroceridae (Semisulcospira coreana, Koreanomelania nodifila, Semisulcospira forticosta, Koreoleptoxis globus ovalis, Semisulcospira libertina, Semisulcospira tegulata and Semisulcospira gottschei) in Korea. Among the 7 species, S. coreana, Korean. nodifila, S. forticosta and S. gottschei showed over 80% cytotoxicities on three cancer cell lines (SNU-1, A549 and Hep 3B) compared to the non-treatment, whereas S. libertina and S. tegulata showed almost no growth inhibition activities on the same cancer cell lines. In relation to ACE inhibition activity, only S. coreana, Korean. nodifila, and S. forticosta showed over 60% ACE inhibition activities, whereas other melania snails exhibited inhibition activities of lower than 25%. DPPH radical scavenging activities were also determined, and used to categories melania snails into three groups based on Duncan's multiple range test at P<0.05. The amount of TNF-${\alpha}$ produced by in vitro mouse peritoneal macrophage was determined according to bioactivity on L-929 cells. Three melania snails, S. coreana, Korean. nodifila and S. gottschei, exhibited 95.2%, 89.7% and 93.7% cell death(%) on L-929 cells, respectively. Glucose-6-phosphate dehydrogenase inhibitory activity was also obtained in the extract of S. coreana (31.9%) and Korean. nodifila (28.1%), showing that these extracts can be used as supplemental dietary health foods. In conclusion, we believe that the extracts of melania snails should be given due consideration in functional health food development.
