한국산 마의 당질 분석 (Carbohydrates Analyses of Korean Yam(Dioscorea) Tubers)

  • 한국산 마의 주요 품종인 D. batatas, D. aimadoimo 및 D. japonica 등 3가지 품종의 일반성분을 분석하여 비교하였고 각 시료들을 동결건조하여 분말로 만든 다음 분획하여 HPLC에 의한 탄수화물의 조성과 함량, DNS법에 의한 환원당을 정량하였다. 탄수화물 중 유리당을 구성하는 주된 조성은 fructose, glucose, sucrose 및 maltose 등이었고 이 중 D. batatas에서는 glucose와 sucrose, D. aimadoimo와 D. japonica에서는 fructose와 sucrose가 가장 많은 부분을 차지하고 있었다. 총당은 주로 mannose와 glucose로 구성 되었고 3가지 품종 모두 mannose가 80% 이상으로, 이는 마가 감자나 고구마보다 끈끈한 점질물로 알려진 mannan이 많기 때문인 것으로 생각된다. Soluble fraction과 전체 마에서의 환원당 정량결과 soluble fraction에서 당함량은 D. aimadoimo가 가장 많았고, insoluble fraction에서 당함량은 D. japonica가 가장 많았다.

Effects of Gibberellic Acid and Gibberellin Biosynthesis Retardants on Ethylene Production, Batatasins, and Free Sugars in Dormant Tubers of Chinese Yam

  • Gibberellic acid did not affect ethylene production, whereas gibberellin biosynthesis inhibitors triggered ethylene production in dormant tubers. Gibberellic acid did not induce sprouting of dormant tubers, however, treatment of gibberellin biosynthesis retardants enhanced sprouting rates. Sprouting rate in ancymidol-treated tubers was highest among gibberellin biosynthesis retardants. Sprouting rate of tubers treated with ancymidol increased to $91.4\%$. Batatasin-III content in $GA_3$ treated tuber was increased in the highest concentration $(30{\mu}g\;I^{-1})$. Tubers treated with mepiquat chloride, Batatasin-I was increased steadily, but contents of Batatasin-III and V showed dramatic decrease at the $ 1,000{\mu}g\;I^{-1})$ concentration. This infers that gibberellin biosynthesis retardants play key roles in promoting breaking dormancy on dormant tubers of Chinese yam.

Effects of the applications of excessive irrigation water and acetaldehyde on Chinese yam tubers at byobusan area of Aomori prefecture in Japan

  • Byobusan area of Aomori prefecture in Japan was a marshy sand dune and had developed for agricultural land use with a large-scale sprinkler system. Recently, it becomes an agricultural problem at this area that distinctive damage with browning maculation and fissures frequently occurs in Chinese yam tubers. Acetaldehyde is one of the factor candidates of underground part damage in plants. In this study, incidence rate of the tuber damage, and the morphological character and elemental composition of the damage parts in tubers were investigated with applications of excessive irrigation water or acetaldehyde water solution into the yam field. The incidence rate of the distinctive tuber damage increased as the input amount of irrigation water was increased. At the browning maculation parts of the tubers, many fissures and damages of cork layer were observed under scanning electron microscopy. In addition, the periderm of tubers was significantly thicker in damaged parts than in non-damaged parts. Funguses, bacterium and nematodes were not observed in the damaged part under scanning electron microscopy. The weight ratio of each constituent element in an analyzed area relative to the total weight of major essential elements was measured with energy dispersive X-ray spectrometry. The results showed that the weight ratios of boron, carbon, phosphorus, sulfur and calcium were higher in damaged parts than in non-damaged parts whereas the weight ratios of oxygen and chlorine were lower in damaged parts than in non-damaged parts. It was also shown by this spectrometry that iron, cadmium, lead and zinc were not directly involved in occurrence of the tuber damage. In this study, there was no remarkable difference of tuber appearance between non-acetaldehyde and acetaldehyde application treatments. From the above results, it is shown that the damage would be a physiological disorder induced by the input of a large quantity of water in the sandy field.

Foliar Application of Extract from an Azalomycin-Producing Streptomyces malaysiensis Strain MJM1968 Suppresses Yam Anthracnose Caused by Colletotrichum gloeosporioides

  • Yam anthracnose caused by Colletotrichum gloeosporioides (C.g) is the most devastating disease of yam (Dioscorea sp.). In the present study, we evaluated the culture filtrate extract (CFE) of azalomycin-producing Streptomyces malaysiensis strain MJM1968 for the control of yam anthracnose. MJM1968 showed strong antagonistic activity against C.g in vitro. Furthermore, the MJM1968 CFE was tested for inhibition of spore germination in C.g, where it completely inhibited spore germination at a concentration of 50 μg/ml. To assess the in planta efficacy of the CFE and spores of MJM1968 against C.g, a detached leaf bioassay was conducted, which showed both the treatments suppressed anthracnose development on detached yam leaves. Furthermore, a greenhouse study was conducted to evaluate the CFE from MJM1968 as a fungicide for the control of yam anthracnose. The CFE non-treated plants showed a disease severity of >92% after 90 days of artificial inoculation with C.g, whereas the disease severity of CFE-treated and benomyl-treated yam plants was reduced to 26% and 15%, respectively, after 90 days. Analysis of the yam tubers from the CFE-treated and non-treated groups showed that tubers from the CFE-treated plants were larger than that of non-treated plants, which produced abnormal smaller tubers typical of anthracnose. This study demonstrated the utility of the CFE from S. malaysiensis strain MJM1968 as a biofungicide for the control of yam anthracnose.

Dormancy-related Change in Endogenous ABA, Batatasin, and Sugar in Stored Tuber and Bulbil of Chinese Yam

  • Endogenous ABA and batatasins were first quantified in the stored tubers and bulbils of the Chinese yam, Dioscorea opposita Thunb. cv. 'Tsukune' by GC-MS with comparison of internal standard, and sugar content was also analyzed by HPLC with comparison of external standard. Endogenous ABA content in stored tubers and bulbils was rapidly decreased as storage period prolonged. ABA content of the bulbils was always higher than that of tuber during storage periods. All batatasins of stored tubers and bulbils at 30 days after storage decreased steadily compared to 120 days after storage. On the contrary, batatasin-Ⅲ of the stored tuber and bulbils was only decreased at 60 days after storage. In Dioscorea opposita Thunb. cv. 'Tsukune' like D. japonica, D. alata, and D. bulbifera, may be controlled by endogenous batatasins and ABA, In these compounds, batatasins rather than ABA might be closely related to dormancy-inducing compound during dormancy of the Chinese yam, Dioscorea opposita Thunb. cv. Tsukune.

둥근마(Dioscorea opposita Thunb.)의 괴경비대 및 성분특성 (Tuber Enlargement and Chemical Components of Yams (Dioscorea opposita Thunb.))

  • 둥근마(Dioscorea opposita L.) 계통 중 품질이 우수하고 이용 가치가 기대되는 단파마를 우리나라에서 재배시 그 가능성과 제 기관의 생육특성을 밝히기 위하여 실시하였다. 두 D. opposita의 괴경의 수량성 및 성분특성을 비교하여 재배 가능성을 시험하였다. 둥근마의 괴경형성은 정식 60일 정도에서 일어나, 그 후 지속적으로 비대 생장이 일어나 160일까지 비대한 후 등숙하였다. 200일째 단마의 괴경중이 512g으로 둥근마 498g보다 약간 높았으나 건물중은 108/127g으로 둥근마가 높아 모양과 건물중이 우수하였다. 둥근마의 일반성분을 측정한 결과 단백질 함량에서 단마$(2.10\%)$보다 $3.62\%$로 높았으며 섬유, 지방의 함량이 낮았다. 수분함량은 $ 64.53\%로 단마$(79.38\%)$보다 건물량이 높았다. 둥근마의 경도를 보면 2787.6로 단마의 4946.9 보다 약 2배정도 낮고, 색도 중 명도(L)는 둥근마가 77.37으로 단마보다 높았다. 둥근마의 괴경으로부터 추출한 물질은 diosgenin의 표준물질과 일치하는 피크를 얻었으며, retention time은 약 5.9에서 나타났다. 둥근마의 괴경에서 diosgenin은 $3.32\%$의 함량을 나타낸 반면 단마에서는 $2.61\%$로 낮은 함량치를 나타냈다.

Effect of Proton Beam Radiation on Bulbil Yield and Gibberellins of Chinese Yam (Dioscorea opposita Thunb.)

  • The study was carried out to evaluate the affect of proton beam radiation on production of bulbil and tuber including change of endogenous gibberellins, of Dioscorea opposita Thunb. The yield of bulbils and tubers from non- and irradiated D. opposita Thunb at doses of 5, 10, 15 and 20 Gy were determined. Endogenous gibberellins were also quantified by GC/MS analysis. D. opposita tubers irradiated at 15 Gy produced higher bulbil production than non-irradiated plants. Enlarged bulbil (above size diameter 4 mm) was significantly increased at 15 Gy. Bioactive endogenous $GA_4$ was dominant in bulbils and tubers irradiated with proton beam rather than $GA_1$. Major gibberellins biosynthetic pathways in bulbils and tubers of D. opposita plants were non C-13 hydroxylation route. From the results of this study, 15 Gy proton beam radiation was suggested as an optimal dose that can produce high amounts of bulbil for mass production of D. opposita plant.

마 주아를 이용한 종묘생산 재배기술 (Cultural Practices of Bulbils for Seed-Tuber Production in Dioscorea batatas Decne)

  • 본 연구는 마의 지상부에서 형성되는 주아를 이용하여 이듬해 종서로 사용 가능한 씨마 생산을 위한 주아 재배법을 개발하기 위하여 수행하였으며, 파종기, 파종량, 파종주아 크기에 따른 씨마 수량을 검토하였다. 1. 주아 파종기가 늦어질수록 생장 일수가 짧아져 괴경장이 짧아지고 평균 괴경중이 가벼워지는 등 괴경의 크기가 작아졌다. 5월 11일 파종의 경우 씨마용으로 사용할 수 없는 무게가 20g 이하 되는 괴경수가 많아짐으로 파종은 늦어도 5월 1일까지 이루어져야 할 것으로 판단된다. 2. 파종량이 많을수록 괴경장이 짧아지고 평균 괴경중이 가벼워지는 등 괴경의 크기가 작아지므로 마로 사용 가능한 $21{\sim}4kg$ 크기의 괴경수를 고려할 때 200kg/10a가 파종 적량으로 판단되었다. 3. 파종주아의 크기가 클수록 출아율과 평균 괴경중이 높아져 단위 면적당 괴경 수량이 높게 나타났다.

영여자 절편체 배양 및 액체배지 첨가에 의한 둥근마의 기내 대량번식 (In Vitro Micropropagation of Chinese Yam (Dioscorea opposita Thunb.) through the Culture of Micro-tuber Sections and by Addition of Liquid Medium)

  • Shoot tips of chinese yam (Dioscorea opposita Thunb.) were cultured on MS medium containing 0.5 mg/L BA to produce micro-tubers in vitro. To stimulate the formation of shoots and micro-tubers, and produce large micro-tubers, the sections of micro-tubers were cultured on MS media with BA and IAA. The shoot multiplication, and the micro-tuber formation and growth were very effective on the media containing 2.0 mg/L BA and 0.5~1.0 mg/L IAA. Sucrose added to MS medium with 2.0 mg/L BA and 0.5 mg/L IAA to stimulate more micro-tuber growth. The medium added 50 g/L sucrose was very effective in the increase of plant fresh weight and micro-tuber growth. After 4 weeks' culture of micro-tuber sections on the medium with 2.0 mg/L BA, 0.5 mg/L IAA and 50 g/L sucrose, the liquid media were added into the same vessels. The micro-tuber growth was stimulated remarkably by the addition of liquid medium. The addition of 25 $m{\ell}$ liquid medium containing 10 g/L activated charcoal, 3x MS salts and 250 g/L sucrose was the most effective in micro-tuber growth.

Changes of Endogenous Gibberellins in Tubers of Chinese Yam(Dioscorea opposita) during Storage Period

  • For storage periods of tubers in Chinese yam, the levels of $\textrm{GA}_{44}$ and $\textrm{GA}_{20}$ was constant, meanwhile both $\textrm{GA}_{53}$ and $\textrm{GA}_{19}$ level were always higher than that of $\textrm{GA}_{44}$ and $\textrm{GA}_{20}$$\textrm{GA}_9$ content as precursor of $\textrm{GA}_4$ was not changed during storage. $\textrm{GA}_{24}$ content was low to below 0.2 ng for 90 days after storage, $\textrm{GA}_{36}$ content as precursor of $\textrm{GA}_4$ like $\textrm{GA}_9$ was about 6-8 fold higher than that of $\textrm{GA}_9$ during storage. GA contents of the two gibberellin biosynthetic pathways were gradually increased when storage periods were progressed. Bioactive GA$_1$ content as the GA members of an early C-13 hydroxylation was always constant, and its content was very low as below 0.1ng per dry weight, meanwhile, bioactive $\textrm{GA}_4$ content as the GA members of non C-13 hydroxylation was drastically increased, also, its content was highest at 90 days after storage, and then decreased at 120 days after storage. Consequently, we suggest that $\textrm{GA}_4$ may be involved in controlling tuber sprouting in Chinese yam.
