올리고당 첨가식이가 흰쥐의 지질대사에 미치는 영향 (Effect of Fructooligosaccharide on Lipid Metabolism in Hypercholoesterolemic Rat)

  • This study was conducted to investigate the effect of fructooligosaccharide on intestinal flora, lipid metabolism and immune response. Thirty two male rats of Sprague-Dawley strain were divided into two groups according to body weight. Each group was fed the diet containing 2% cholesterol or the normal diet, respectively for 4 weeks. Each group was again divided into two sub-groups and they were fed with the diet containing 5% of sucrose and fructooligosaccharide, respectively for 8 weeks. The number of bifidobacteria slight increased, but not significantly, in oligosaccharide groups. Plasma total lipid concentration in cholesterol group was signficantly increased compared to the value in normal group. Dietary oligosaccharide decreased plasma total lipid concentration and triglyceride concentrations in normal group, but not in cholesterol group. Fecal lipid excretion was higher in cholesterol group than in normal group. Fecal cholesterol concentration in cholesterol-oligosaccharide group was significantly increased compared to other groups. Cholesterol had slight effects on TBARS and the immune status, but dietary oligosaccharide had no effects on these variables. The results in this study suggest that dietary oligosaccharide had no effect on serum and liver profiles of rats fed high cholesterol diet but increases the fecal cholesterol excretion.

한국인 성인 여자의 칼슘대사에 관한 연구 (I) - 서울 지방 - (Dietary Intake of Calcium and Absorption Rate of College Women in Korea ( I ) - Seoul Area -)

  • This study was designed to investigate calcium intake, calcium excretion, and calcium absorption rate of the college women in Korea. The subjects of this experiment were five healthy college women aged from 18 to 23 years who lived in Seoul. The period of this experiment was two weeks, the first week was spent in having an ordinary diet and the next week for the experimental diet. Two weeks later, the feces of subjects were collected. Results obtained by the present experiment were as follows: 1) The total food intake from the experimental diet daily was about $1600{\sim}2000g$. 2) The total calcium intake was about $270{\sim}585mg$ and $710{\sim}880mg$ for the ordinary diet and the experimental diet respectively. 3) The daily calcium excretion of feces was about $219{\sim}295mg$. 4) The calcium absorption rate of subjects was about $60{\sim}70%$.

Effect of Capsaicin on the Body fat and Adipocyte in the Diet induced-obese Mice

  • As the obesity has been known to be related with the hyperlipidemia, cardiovascular disease, cerebral apoplexy, fatty liver, and other chronic diseases, recent researches have focused on the functional food materials and their anti-obesity activities. This study was performed to study the effects of vanilloid family capsaicin, major pungent ingredient of hot chillies and peppers, on anti-obesity activities. ICR male mice were fed one of the pellet diet, basal diet, and high fat diet with capsaicin (45 $\mu\textrm{g}$/day) solution for 5 days. Mice in the corresponding control groups were given water for 5 days. In results, capsaicin reduced body weights in any diet groups. Percent weight and cell size of the abdominal white adipose tissue in mice on the high fat diet with capcaicin were significantly lower compared with those in mice on the high fat diet with water. However, percent brown adipose tissue weight per body weight in mice on the high fat diet was not affected by capsaicin. Capsaicin reduced the levels of s-triglyceride and s-total cholesterol in the pellet diet or high fat diet groups. There was no difference in the s-protein levels between the capsaicin group and the control water group. These data indicate that 1) orally administered capsaicin has a reducing effect on the blood triglyceride and total cholesterol levels, and 2) capsaicin has lowering effects on the body weight, percent weight and cell size of the abdominal white adipose tissue.

고지혈증 흰쥐에서 운동훈련이 혈액, 대사 변인 및 유전자 발현에 미치는 영향 (The Effect of Exercise Training on Blood and Metabolic Variances, and Genes Expressions in Hyperlipidemic Rats)

  • This study was conducted to investigate the effect of exercise training on blood and metabolic variances and genes expressions in hyperlipidemic rats. Three weeks-old male rats were randomly assigned into chow (n=7), high-fat diet (HF, n=7) and HF+exercise (HF+EX, n=7) groups. Exercise training consisted of the treadmill running 5 times per week during 8 weeks (0% grade, 30 min/time for first 4 weeks and 0% grade, 60 min/time the other 4weeks). The levels of triglyceride and total -cholesterol were increased in HF diet compared with chow group, and recovered to level of chow group by exercise training. Plasma glucose and insulin concentrations increased by 40 and 50%, respectively in HF diet compared with chow diet group, and these increases returned to the level of chow group by exercise training (p<.05). Body weight and abdominal fat mass were increased by high-fat diet compared with chow diet, and recovered to level of chow group by exercise training. Long-chain fatty acid oxidation rate and AMPK protein expression was not changed by HF diet, but increased by exercise training compared with high-fat diet (p<.05). UCP3 protein expression was not changed by either high-fat diet or exercise training compared with chow group. There was high correlation between plasma triglyceride and total cholesterol concentrations(p<.01). Plasma triglyceride or total cholesterol level showed correlation with following factors; plasma insulin and glucose levels, body weight, abdominal fat weight, UCP3 protein expression and long-chain fatty acid oxidation rate. These results showed that exercise training on the treadmill recovered hyperlipidemia, hyperglycemia and hyperinsulinemia induced by high-fat diet for 8 weeks. These exercise effects may be related with decreased body weight and abdominal fat mass, and increased long-chain fatty acid oxidation rate.

Hypocholesterolemic effects of curcumin via up-regulation of cholesterol 7a-hydroxylase in rats fed a high fat diet

  • There is an increasing interest in curcumin (Curcuma longa L.) as a cardiovascular disease (CVD) protective agent via decreased blood total cholesterol and low-density lipoprotein-cholesterol (LDL-cholesterol) level. The aim of this study was to investigate further the potential mechanism in the hypocholesterolemic effect of curcumin by measuring cholesterol 7a-hydroxylase (CYP7A1), a rate limiting enzyme in the biosynthesis of bile acid from cholesterol, at the mRNA level. Male Sprague-Dawley rats were fed a 45% high fat diet or same diet supplemented with curcumin (0.1% wt/wt) for 8 weeks. The curcumin diet significantly decreased serum triglyceride (TG) by 27%, total cholesterol (TC) by 33.8%, and LDL-cholesterol by 56%, respectively as compared to control group. The curcumin-supplemented diet also significantly lowered the atherogenic index (AI) by 48% as compared to control group. Hepatic TG level was significantly reduced by 41% in rats fed with curcumin-supplemented diet in comparison with control group (P < 0.05). Conversely, the curcumin diet significantly increased fecal TG and TC. The curcumin diet up-regulated hepatic CYP7A1 mRNA level by 2.16-fold, compared to control group p (P < 0.05). These findings suggested that the increases in the CYP7A1 gene expression may partially account for the hypocholesterolemic effect of curcumin.

고지방 식이로 유도된 비만 쥐에서 실리빈 (Silybin)이 체중 및 내당 능에 미치는 영향 (Effect of Silybin on Body Weight and Glucose Tolerance in High-fat-diet Induced Obese Mice)

  • In this study, we investigated the effects of silybin on body weight and glucose tolerance in mice fed high fat diet mice. We found that body weight, plasma TG contents, fat size, glycerol, 3-hydroxybutyrate and total cholesterol were significantly decreased in silybin (500 mg/kg) supplemented groups compared to high fat diet group. Whereas, total food intake was not changed between high fat diet group and high fat diet plus silybin group. Futhermore, supplement of high fat elevated the glucose intolerance and was improved in silybin supplement group. Finally, we examined the effect of silybin on circulating adipocytokine level to explore the possible mechanism by which silybin improves high fat diet-induced obesity and diabetes. The silybin supplement significantly reduced the level of adipocytokine, such as leptin, resistin, IL-6, and MCP-1 induced by high fat diet. These results suggest that silybin can be used to improve obesity and diabetes.

Antioxidant Effects of Cranberry Powder in Lipopolysaccharide Treated Hypercholesterolemic Rats

  • This study was conducted to investigate the effects of cranberry power on antioxidant defense system in rats fed an atherogenic diet and injected with lipopolysaccharide (LPS). Sprague-Dawley rats were divided into the following 5 groups: normal diet+saline (NS), atherogenic diet+saline (AS), atherogenic diet+LPS (AL), atherogenic diet with 5% cranberry powder+LPS (AL-C5), and atherogenic diet with 10% cranberry powder+LPS (AL-C10). Total antioxidant status measured by ferric reducing ability of plasma (FRAP) was significantly reduced by LPS injection (24%) and was restored by the cranberry powder treatment (P<0.05). In addition, the mean level of plasma total phenolics was significantly decreased by LPS injection (P<0.05) and tended to be increased when cranberry powder was incorporated in to the diet. Activity of serum superoxide dismutase (SOD) tended to be lowered by LPS injection and declined further in cranberry powder fortified groups. Overall results indicate that dietary cranberry powder may provide appropriate antioxidants to counter the diminished antioxidant status induced by exposing hypercholesterolemic rats to LPS.

가시발새우 키토산 식이가 식이성 고지혈증 흰쥐의 지질대사에 미치는 영향 (Effects of Dietary Lobster Shrimp Chitosan on Lipid Metabolism in Diet-Induced Hyperlipidemic Rats)

  • This study was designed to investigate effects of lobster shrimp chitosan on serum and liver lipid levels in rats fed high fat and cholesterol diets. Male Sprague-Dawley rats of 4-weeks old, weighing 70$\pm$5g, were fed experimental diets for 3 weeks: basal diet+5% cellulose(group 1), high lipid diet+5% cellulose(group 2, control group), high lipid diet+5% balloon flower root(group 3), high lipid diet+5% cellulose(group 4, high lipid diet+5% lobster shrimp chitosan(group 5). Concentrations of total cholesterol, HDL-cholesterol, LDL-cholesterol, free cholesterol, cholesteryl ester, triglyceride, phospholipid in serum and liver were determined. The results obtained were as followes: Serum total cholesterol concentration were significantly reduced in all supplemented group compared with control. HDL-cholesterol concentrations were higher in groups 1, 4, and 5 and the ratios of HDL-cholesterol to total cholesterol concentration were higher in groups 1, 4, while the atherogenic-indices were lower in groups 1, and 4 than control. Concentrations of LDL, LDL-cholesterol and free cholesterol, cholesteryl ester were significantly lower in all supplemented groups than the control group. Concentrations of phospholipid and triglyceride were lower in groups 3, 4, 5, while activities of GPT and GOT were significantly lower in groups 4, 5 than the other group. Concentrations of calcium, magnesium and phosphorous were high in all treatment groups compared to the control group. Liver contents of total cholesterol, free cholesterol and cholesteryl ester were lower in the groups 1, 3, 4, and 5 than in the control group, especially lower in groups 1, and 5. Liver triglyceride and phospholipid concentrations were significantly lower in the groups 1, and 5 than in the other groups. The results from the data suggested that the experimental diet containing 5% lobster shrimp chitosan was effective to reduce the serum and liver lipids.

고지방식이를 급여한 흰쥐의 패류식이 급여 효과 (The Effects of Dietary Shellfish on Lipid Metabolism in Rats Fed High-Fat Supplement)

  • This study was conducted to investigate the effects of shellfish on lipid metabolism in rats fed high fat supplements. Male sprague-dawley rats weighting approximately 165g were fed a basal diet, a high fat diet, or a high fat diet plus shellfish for 4 weeks. The shellfishes on the were oyster, hard-shelled mussel, little neck clam, and march clam. Alfter 4 weeks high fat diet, supplementation of 20% lard significantly increased plasma. GOT, GPT, ${\gamma}$-GTP , and liver triglyceride (TG). Plasma GOT, GPT, ${\gamma}$-GTP , triglyceride, and total cholestrerol levels were significantly lower in shellfish groups than in basal and high-fat groups regardless of high-fat supplementation (p<0.05). The total lipid and cholesterol content in liver showed similar results(p<0.05). There were no differences in glucose, HDL-cholesterol in plasma and total cholesterol and total lipid in liver between basal and high-fat supplemented diets. Liong chain fatty acids, specific components of shellfishes group, were exclusively higher than in basal and high-fat diets, and were most well-reflected in liver and plasma. From the above results, the hypolipidemic effects of shellfish were detected in the process of inducing hyperlipidemia by high-fat supplement.

쑥 첨가급식이 흰쥐의 혈청 성분에 미치는 영향 (Effect of Feeding Basal Diet Supplemented with Mugwort Powder on the Serum Components in Rat)

  • This study was designed to observe the body growth and components of edible mugwort(Artemisia sp.) and medicinal mugwort(Kanghwa medicinal mugwort) . Twenty-four young rats of Sprague Dawley strain, body weight of about 89g were used in this study. They were fed on the basal diet(control diet) supplemented with 5% edible mugwort powder ( EM diet) and 5% medicinal mugwort powder( MM diet) for 4 weeks respectively. In proximate composition of nutrients of mugwort in dry basis(100g). crude protein (16.4g) and crude ash(11.8g) contents of EM were higher to about 2% than that of MM, but crude lipid content(4.3g) of EM was lower to about 2% than that of MM. However, the contents in calcium(6.9g) of MM was higher to 5.3 times than that of EM. but in Mn(17mg), Zn(0.5mg), Fe(131mg), Mg(337mg) of EM were higher to 2.8∼2.3 times and vitamin A(39,776 IU) of EM was higher to 2.9 times than that of MM respectively. Body wight gain rate and diet efficiency ratio of EM and MM diet group were similar to that of the control group. The contents of total protein, albumin, urea nitrogen. creatinine, uric acid, total cholesterol, HDL-C, LDL-C, glucose, amylase, transaminase (GOT, GPT) in serum exhibited no remarkable difference among of the EM and MM diet group but the level of LDH activity of MM diet group were significantly lower than that of the control group and EM diet group.
