Parallux, a solid-phase fluorescence immunoassay (SPFIA) developed for detection antibiotics residue in milk, was applied for analysis of antibiotics in muscle tissue of olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major). Fishes were dipped in neomycin 140 mg/ton water, the recommended therapeutic dose, for 24 h. Muscle samples were obtained on 1st, 2nd, 3rd, 4th and 5th day after drug treatment. The concentration of neomycin in muscle was determined using an internal standard (100 ppb as neomycin). The absorbance ratio of sample to internal standard (S/C) was employed as an index to determine the muscle residues in fishes. To investigate the recovery rate, the standard solutions were added to muscle samples to give final concentrations in muscle of 0.2 and 0.5 mg/ml. The recovery rates of all spiked samples were > 85% of the spiked value. Neomycin was detected in muscles of fishes treated after the 1st day of withdrawal period. On the 2nd day after drug treatment, all muscle samples showed negative reaction (S/C ration ${\leq}$ 1.0). The present study showed that the SPFIA can be applied for predicting residues of neomycin in muscle tissues of farmed fishes.
A feeding experiment was conducted to investigate the effects of replacement of dietary fish meal by frozen whole krill (FWK) and/or krill meal (KM) on growth performance and body composition of juvenile black rockfish, Sebastes schlegeli. The basal experimental diet (ED) contained 58.4% fish meal (FM) as a control, the other five EDs were formulated with the protein replacement ratio of fish meal by as 0, 10, 20, 30, and 40%, respectively (FWKM0, FWK10, FWK20, FWK20KM10, FWK20KM20 and FWK20KM20HP). Each diet was fed to juvenile rockfish initially weighing 3.09±0.02 g/fish in a flow-through system. The control ED containing 58.4% FM and 10% FWK diet showed significantly higher weight gain and feed efficiency than all the other EDs containing 20, 30, and 40% FWK and/or KM. But the survivals, whole-body moistures, crude proteins and lipids of black rockfish fed all EDs were similar in all EDs. The only whole-body ashes of juvenile black rockfish were significantly differentiated by feeding the various EDs containing different levels of FWK and/or KM with a protein and lipid levels adjustment. These results suggest that dietary frozen whole Antarctic krill and/or krill meal inclusions could not improve the growth but change the whole-body ashes in juvenile black rockfish. Therefore, this study further indicates that dietary more than 20% of FWK and/or KM supplementation could not be a potential source of animal protein to replace fishmeal in juvenile black rockfish.

Changes in plasma levels of thyroid and sex steroid Hormones were examined during maturation and parturition periods in rockfish (Sebastes schlegeli) cultured in net pens. Plasma L-thyroxine levels were $35.2{\pm}5.7\;ng/m{\ell}\;(n=\;5;\;mean\;{\pm}\;sem)$ at vitellogenesis stage and then significantly decreased to $20.5\;{\pm}\;4.2\;ng/m{\ell}$ at parturition stage (P<0.05), and rapidly returned to high level, $44.9{\pm}\;7.2\;ng/m{\ell}$ at resting stage. Plasma 3,5,3'-triiodo-L-thyronine levels were 12.9 $\pm$ 1.6 ng/ml at vitellogenesis stage, but significantly decreased to $3.7{\pm}0.7\;ng/m{\ell}$ at ovulation stage (P<0.05) and increased to $52.9{\pm}7.0\;ng/m{\ell}$ at jesting stage. Plasma estradiol-17 $\beta$ level showed the highest value ($4.3{\pm}0.9\;ng/m{\ell}$) at vitellogenesis stage, but the level significantly decreased to $0.3{\pm}0.1\;ng/m{\ell}$, during parturition stage (P<0.05). In vitellogenesis and ovulation stages, plasma testosterone levels were $1.8{\pm}0.3\;ng/m{\ell}$ and $2.1{\pm}0.7\;ng/m{\ell}$, respectively, thereafter the level significantly decreased to $0.1{\pm}0.1\;ng/m{\ell}$ at parturition stage (P<0.05). These findings suggest that thyroid hormones may have relation to maturation and parturition of mother rockfish.

The objective of this study was to evaluate the effects of acute hypoxia on the physiological stress responses (plasma cortisol as the primary response, and hematocrit, hemoglobin, plasma glucose, sodium, chloride, osmolality, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) as the secondary responses) of Korean rockfish. The mortality in Korean rockfish started at $0.7mg\;L^{-1}$ and $0.8mg\;L^{-1}$ levels at $12^{\circ}C$ and $20^{\circ}C$, respectively. And the time required for the mortality in experimental fish was 274 and 148 minutes at $12^{\circ}C$ and $20^{\circ}C$, respectively. Levels of hematocrit, hemoglobin, AST, ALT, ion concentration, osmolality, glucose and cortisol had significantly increased with decreasing dissolved oxygen at $12^{\circ}C$ and $20^{\circ}C$.

Effect of artificial illumination on feeding by the rockfish Sebastes schlegeli receiving no synthetic feed in the cage of the Tongyong Marine Ranch was investigated by analysing the stomach contents of 20 individuals every month from October 1998 to July 1999. The fish was found to have fed on naturally available zooplanktons, i.e. Calanus sinicu, zoeae, amphipods and copepodites in October, fish larvae in November-December, nematodes in January, amphipods in February and again fish larvae from May to July. Despite increase in the weight of stomach contents from 260 mg/fish during the initial period of study to 2,173 mg/fish, as many as 70% of the fish were found to have empty stomach during March-April. Weight of stomach contents did not increase with increasing age of the fish. Occurrence frequency was 83-90% for C. sinicus during October-November and >50% for fish larvae during March-April and June-July.

Withdrawal time of erythromycin in the cultured black rockfish (Sebastes schlegeli) was investigated to provide regulatory authority and fishery industry with full information needed to secure food safety of the fish treated with erythromycin. Medication was carried out using the experimental diet containing erythromycin of which concentration was 200 mg/kg diet and its daily dose was about 40 mg/kg body weight. The withdrawal time needed to reduce antibiotic contents around 10.0 mg/kg accumulated during 9 days medication below 0.2 mg/kg was identified about 10 days. The antibiotic with 13.7 mg/kg of fish on the $9^{th}$ days of medication was completely depleted after 30 days from stop of medication.
Myostatin (MSTN; also known as GDF8) is a member of the transforming growth factor ${\beta}-superfamily$ of proteins. MSTN negatively regulates mammalian skeletal muscle growth and development by inhibiting myoblast proliferation. Mice and cattle possessing mutant MSTN alleles display a 'double muscling' phenotype characterized by extreme skeletal muscle hypertrophy and/or hyperplasia. We isolated the full-length cDNA of a novel MSTN gene from S. schlegeli muscle tissue and examined its expression pattern in various tissues. The full-length gene (GenBank DQ423474) consists of 1941bp with an open reading frame of 1134 bp, encoding 377 amino acids that show 62-92% amino acid similarity to other vertebrate MSTNs. The predicted protein contains a conserved proteolytic cleavage site (RXRR) and nine conserved cysteine residues at the C terminus. RT-PCR revealed that the unprocessed and prodomain myostatin mRNAs were predominantly present in muscle, with limited expression in other tissues. However, the mature myostatin mRNA was highly expressed in brain and muscle, intermediately expressed in the gills, intestine, heart, and kidney, and weakly expressed in the liver and spleen.

This study was conducted to identify the general conditions for the isolation and in vitro culture of ovary-derived cells in rockfish (Sebastes schlegeli). The effects of three different enzymes on cell retrieval from ovarian tissues were evaluated first, and then the ovary-dissociated cells were cultured under various culture conditions, with varying basal media and culture temperatures, addition of growth factors, and/or culture types. We found that collagenase type I treatment was effective for cell isolation from ovarian tissues. From a total of 42 trials to evaluate the effects of basal media and culture temperatures on cell culture of ovary-dissociated cells, we observed that Leibovitz's L15 medium was more supportive than Dulbecco's modified Eagle's medium for culture, and the cells could grow at all three temperatures tested, 15, 20, and $25^{\circ}C$, at least up to passage 2. However, growth factor addition did not improve cell growth. Introduction of suspension culture after monolayer culture expanded the culture period significantly more than did monolayer culture alone. Our results may provide a basis for developing an in vitro system for S. schlegeli germline cell culture, which will ultimately lead to improvement of the species.
In order to estimate energy budget of the rockfish, Sebastes schlegeli juvenile, during the period from parturition to juvenile, of rockfish were reared at constant condition of water temperature($16^{\circ}C$ and $20^{\circ}C$). Energy used by the reared juveniles were calculated from estimates of data on growth, oxygen consumption, nitrogen excretion, feeding and energy content. After parturition, total length of juvenile was 4.56~5.49mm(average 4.97mm, n=20), and the average dally growth rates were 0.50mm at $20^{\circ}C$. The weight-specific oxygen consumption and nitrogen excretion rates decreased with increasing body weight. Temperature significantly affected oxygen consumption and nitrogen execretion rates, with the higher rates at $20^{\circ}C$ than $16^{\circ}C$. During the 25 days from parturition to jllveniles, feeding rates were 65.8cal at $16^{\circ}C$, and 89.2cal at $20^{\circ}C$. The assimilation effeciency estimated by nitrogen content of food and egested feces were 85.21% at $16^{\circ}C$ and $20^{\circ}C$. During the period from parturition to juvenile, energy was used the higher in body growth than in oxygen consumption. The gross growth efficiencies($K_1$)and net growth efficiencies($K_2$) of the rockfish, Sebastes schlegeli juvenile ranged from 43% to 47% and from 50% to 55%, respectively.
We report on the variation of infection and histopathological change of Microcotyle sebastisci parasitic on cultured rockfish, Sebastes schlegeli, in Namhae Islands and Kamak Bay from April to October in 1995. Microcotylosis due to Microcotyle sebastisci occurred among cultured rockfish, Sebastes schlegeli. This parasite was not found on the host fish from June to July. Parasite sites were mainly consist of 2nd and 3rd gill arch`s filaments of rockfish. Also, the sites were secreted in large quantity of mucus with a very small bleeding. In Namhae Islands, maximum values of prevalence, relative density and mean intensity were found on September 1995, as 40.0%, 30.7 and 76.8, respectively. In Kamak Bay, maximum values of prevalence, relative density and mean intensity were obtained on October 1995, as 46.0%, 40.5 and 88.0, respectively. Histopathological changes of the heavily infested gills were showed necrosis, epithelium of the gill filaments underwent hyperplasia with fusion of the lamella and filamental clubbing. And a bacterial colony is invaded on the surface of lamella epithelium.
