An Experimental Study on the Effect of Immunopotential and the Anticancer Effect of Red Ginseng Extract (한국 홍삼의 면역활성 및 항암효과에 관한 실험적 연구)

  • To evaluate the anticarcinogenic effect and its mechanism of red ginseng, the mice were treated with red ginseng and received subcutaneous Bl6 melanoma cell line injection on the back. Tumor incidence was same (100%) both in water and red ginseng-treated groups, but tumor production was delayed in red ginseng-treated group. Survival time was somewhat longer in red ginseng-treated group. The histopathological findings were similar in both groups, but lymphocytic infiltration around the tumor and melanin production in the tumor cells were prominent in the red ginseng-treated group. Flow cytometric analysis on T lymphocytes and natural killer cells revealed increased $T_H$/$T_S$ ratio and increased NK cells in red ginseng-treated group. These results suggest that the anticarcinogenic effect of red ginseng may be exerted by the increased cell-mediated immunity and natural killer cell activity.

Studies on Ginseng Vinegar (인삼식초에 관한연구)

  • Ginseng-vinegars were produced by the fermentation of 5% ethanol solution contained ginseng, red ginseng, ginseng marc and red ginseng marc using Acetobacter aceti 3281 for 26 days at 35$^{\circ}C$. The ginseng and red ginseng vinegar contained 0.236mg/ml of total sugar 0.236mg/ml of reducing sugar and 0.05% of ethanol and 1.005 of specific gravity 8,58CFU of viable cell count 3,24 of pH and 5.11% of acidity. Whereas the vinegars produced using the water-extracted red ginseng marc and the ethanol-extracted red ginseng marc were consisted of total sugar was 1.27mg/ml and 1.60mg/ml reducing sugar was 0.077mg/ml and 0.725mg/ml specific gravity was 1.001 and 1.004 the number of viable cells was 8.51CFU/ml and 8.1CFU/ml pH was 2.81 and 2.89 acidity was 5.18% and 5.32% respectvely ethanol concentration was 0.05% in both cases. In five-grade scoring test of sensory evaluation, it was estimated favorable that each vinegar made by were-extracted red ginseng marc, ethanol-extracted red ginseng marc ginseng and red ginseng ginseng from 0.5 to 32% of water-and ethanol-extract red ginseng was extracted with 10% white vinegar for 30 days. The best sensory vinegars were obtained that ginseng of 0.4~1.6% above red glnsend of 0.8% water-extracted red ginseng marc of 0.8~1.6% and ethanol-extracted red ginseng marc of 0.4~1.6% added in 10% white vinegar respectively.

  • Browning intensity is a major factor to estimate the quality of red ginseng or red ginseng products. The Maillard type of browning reaction proceeds nonenzymatically during extraction and concentration of red ginseng. The present studies were carried out to investigate the effects of amino acids and sugars on the browning reaction during extraction and concentration of red ginseng. Red ginseng was pulverized to 115 mesh and then tenfold (v/w) of water was added to the powder to make the substrate of red ginseng. Solution (0.1 M) of fourteen amino acids and of folly silgars were added to the substrates of red ginseng powder and these were then extracted and concentrated to examine their browning intensities. Amino acids were more effective than sligars in acrelerating the browning reaction. Acceleration of the browning reaction in the concentrate was in the order of arginine> histidine>glycine>alanine>lysine phenyl alanine>aspartic acid>lelicine>threonine>gllitamic acid>tyrosine>valine>istleucine>methionine for amino acids, and was glucose>frlictose >silcrose, maltose for sugars.

Fermented red ginseng and red ginseng extract diet supplementation on the effect of growth performance, nutrient digestibility, and meat quality of growing-fattening pigs

  • The influence of fermented red ginseng (FRG) and red ginseng extract (RGE) on the growth, nutrient digestibility, and meat quality of growing-fattening pigs was evaluated in the present study. In total, 120 growing-fattening crossbred pigs with a bodyweight of 21.77 kg (SE1.88) were allocated to 3 treatments with 8 repeats of 5 pigs (2 boars and 3 gilts) for a 22-week experimental period. The pigs were supplemented with a basal diet (CON group) or a basal diet containing 0.1% FRG or 0.1% RGE. Pigs fed the FRG diet had a higher (p < 0.05) average daily gain (ADG) from days 1 to 42 and during the whole period. The apparent nutrient digestibility of dry matter (DM) and nitrogen (N) was improved (p < 0.05) in the FRG treatment in week 6 and that of DM in week 10. Moreover, the drip loss of the meat from the FRG pigs was reduced (p < 0.05) on days 1, 5, 7, and 9. Taken together, this study revealed that the supplementation of 0.1% FRG in the diet improved the ADG and the apparent nutrient digestibility of DM and N and decreased the drip loss of the meat. However, the addition of RGE did not affect the growth of the growing-fattening pigs.

방사선에 조사된 생쥐의 간 및 신장에서 홍삼 추출물의 방어효과

  • This study was prepared to observe the charge of tissues weight, glutamate oxaloacetate transminase(GOT) and glutamate pyruvate transminase(GPT) activities in the liver and kidney treated with red ginseng extract injection in the gamma ray(5.0 Gy) irradiated mice groups were divied in to control, red ginseng extract injection, irradiation after saline injection and irradiation after red ginseng extract injection. The GOT activity showed earlier recovery at irradiation after red ginseng extract injection group than irradiation after saling injection group in the liver and kidney. The GPT activity showed earlier recovery at irradiation after red ginseng extract injection group than irradiation after saling injection group in the liver and kidney. The above result suggest that red ginseng extract have the protection effect on the change of GOT and GPT activity after radiation injury in the liver and kidney.

Morphine dependence is attenuated by red ginseng extract and ginsenosides Rh2, Rg3, and compound K

  • Background: Red ginseng and ginsenosides have shown plethoric effects against various ailments. However, little is known regarding the effect of red ginseng on morphine-induced dependence and tolerance. We therefore investigated the effect of red ginseng extract (RGE) and biotransformed ginsenosides Rh2, Rg3, and compound K on morphine-induced dependence in mice and rats. Methods: While mice were pretreated with RGE and then morphine was injected intraperitoneally, rats were infused with ginsenosides and morphine intracranially for 7 days. Naloxone-induced morphine withdrawal syndrome was estimated and conditioned place preference test was performed for physical and psychological dependence, respectively. Western blotting was used to measure protein expressions. Results: Whereas RGE inhibited the number of naloxone-precipitated jumps and reduced conditioned place preference score, it restored the level of glutathione in mice. Likewise, ginsenosides Rh2, Rg3, and compound K attenuated morphine-dependent behavioral patterns such as teeth chattering, grooming, wet-dog shake, and escape behavior in rats. Moreover, activated N-methyl-D-aspartate acid receptor subunit 1 and extracellular signal-regulated kinase in the frontal cortex of rats, and cultured cortical neurons from mice were downregulated by ginsenosides Rh2, Rg3, and compound K despite their differential effects. Conclusion: RGE and biotransformed ginsenosides could be considered as potential therapeutic agents against morphine-induced dependence.

Anti-diabetic Mechannism Study of Korean Red Ginseng by Transcriptomics (전사체 프로파일을 이용한 고려 홍삼의 항당뇨 기전 연구)

  • This study was designed to investigate the anti-diabetic effect and mechanism of Korean red ginseng extract through transcriptomics in C57BL/KsJ db/db mice. The db/db mice were randomly divided into six groups: diabetic control group (DC), red ginseng extract low dose group (RGL, 100 mg/kg), red ginseng extract high dose group (RGH, 200 mg/kg), metformin group (MET, 300 mg/kg), glipizide group (GPZ, 15 mg/kg) and pioglitazone group (PIO, 30 mg/kg), and treated with drugs once per day for 10 weeks. At the end of treatment, we measured blood glucose, insulin, hemoglobin A1c (HbA1c), triglyceride (TG), adiponectin, leptin, non-esterified fatty acid (NEFA). RGL-treated group lowered the blood glucose and HbA1c levels by 19.6% and 11.4% compared to those in diabetic control group. In addition, plasma adiponectin and leptin levels in RGL-treated groups were increased by 20% and 12%, respectively, compared to those in diabetic control. Morphological analyses of liver, pancreas and epidydimal adipose tissue were done by hematoxylin-eosin staining, and pancreatic islet insulin and glucagon levels were detected by double-immunofluorescence staining. RGL-treated group revealed higher insulin contents and lower glucagon contents compared to diabetic control. To elucidate an action mechanism of Korean red ginseng, DNA microarray analyses were performed in liver and fat tissues, and western blot and RT-PCR were conducted in liver for validation. According to hierarchical clustering and principal component analysis of gene expression Korean red ginseng treated groups were close to metformin treated group. In summary, Korean red ginseng lowered the blood glucose level through protecting destruction of islet cells and shifting glucose metabolism from hepatic glucose production to glucose utilization and improving insulin sensitivity through enhancing plasma adiponectin and leptin levels.

  • This experiment was carried out to investigate the effect of red ginseng extract on the growth of Lactobacillus sp. (L acidophilus, L casei, L salivarius), Escherichia coli and Listeria monocytogenes in pH controled medium by $\beta-Glycerol\;PO_4$ buffer. The growth of Lactobacillus sp. was show a similar pattern in control and MRS broth with red ginseng extract $1.0\%$ but was remarkably show inhibiting in MRS broth with over $2.0\%$ red ginseng extracts. The growth of E coli was inhibited in Trypticase soy broth with $1.0\%$ red ginseng extracts. Also the growth of L monocytogenes was inhibited in Trypticase soy broth with $5.0\%$ red ginseng extract The growth of L acidophilus KCTC3150, L casei KCTC3189, L salivarius ssp. salivarius CNU27, and E coli KCTC1039, L monocytogenes KCTC3443 were remarkably inhibited in pH non-control medium and pH control medium with $10\%$ red ginseng extract These results was suggested to effect of inhibition of microorganisms growth not pH decrease by organic acid but another components in red ginseng extract.

Effect of Controlled Atmosphere Storage on the Antioxidative and Cytotoxic Activities of Fresh and Red Ginseng (CA저장이 수삼 및 대삼의 항산화 작용과 항암작용에 미치는 영향)

  • 4-year-old fresh ginseng was stored for 12 weeks at 4$^{\circ}C$ under the CA storage and samples were withdrawn after every 1, 2, 3, 4, 6, 8 and 12 weeks for processing of red ginseng. Antloxidative and cytotoxic activities of steamed red ginseng for the quality evaluation are summarized as follows: Reducible activity of water extract to 1, 1-dipenyl-2-picryl hydrazyl increased with increasing storage period. Antioxldative activity of red ginseng did not change in petroleum ether fraction, while it had a tendency to increase In the fractions of ethyl ether and ethyl acetate with increasing storage period. in contrast to fresh ginseng, malonaldehyde formation In heparin mlcrosome of red ginseng processed from fresh ginseng stored In CO2:O2:Nl (6:4:90) with a 0.5% DF-100 immersion condition decreased but was not changed significantly by the period of storage. Cytotoxic activity against cancer cell at a 40 mg/ml concentration of red ginseng slightly increased with an increase in storage period but no effect was observed at the concentration less than 10 mg/ml.

Extraction of Red Ginseng Extract by Impulse Vacuun System (Impulse-Vacuum System을 이용한 홍삼엑스의 추출)

  • This study was carried out to establish the extraction method of red ginseng extract without saponin decomposition. Red ginseng was extracted with impulse vacuum system and multi-stage extraction method. Crude saponin content of red ginseng extract (RGE) from impulse vacuum system was 5.4-5.9%, while that of RGE from multi-stage extraction method was 8.2-8.3%. However, HPLC Patterns indicated that saponins of RGE from impulse vacuum system were hardly decomposed, while those of RGE from multi-stage extraction method were decomposed, especially in ginsenoside -Rgl and -Re saponin. Also, the yields of red ginseng by impulse vacuum system were 15 to 20 times higher than that of multi-stage extraction method.
