Effects of several factors on pollen germination in Platycodon grandiflorum

  • Pollen germination and its' viability in bellflower hybrid system are of great importance. The present study was conducted to investigate the several factors underlying the pollen germination of Platycodon grandiflorum and obtain the basic data for effective artificial pollination for the production of sound specifies. The pollens of Platycodon grandiflorum started germination from one hour after planting, and the germination was actively progressed as time dependent manner. For lighting conditions, the germination of pollens under the light was faster by more than twice than that without the light. Furthermore, the germination was better in the high temperature rather than in the low temperature. The germination rate was higher in the $30^{\circ}C$. For the carbon source, the germination rate was better at the concentration of 15% regardless of the kinds. In particular, the highest value was observed with glucose. The germination rate was decreased substantially as the increasing with the higher pH. The dynamic germination of pollens was observed at the pH 5. With respect to the growth regulator, the higher concentration of NAA induced the higher the germination rate. $GA_3$ showed a good germination rate in $0.05mg{\cdot}L^{-1}$. Meanwhile, for kinetin, lower concentration increased the germination rate, unlike NAA. The higher concentrations of boric acid degraded the germination rate, and the addition of boric acid of $10mg{\cdot}L^{-1}$ demonstrated higher germination rate than the addition of other growth regulators. Notably, the addition of asparaginic acid exhibited the similar results in all test sectors regardless of concentration, whereas a little higher result was observed in the high concentration sector. Taken together, the results concluded that the boric acid was considered as one of the essential minerals that played an important role on the germination of pollens.

Characteristics and Temporal Distribution of Airborne Pollen in an Urban Area of Japan

  • Using a sampling device of our own making, airborne pollen has been monitored in Kyoto, Japan from the middle of February to the end of May 2004. From the morphological analysis of pollen grains by Scanning Electron Microscope (SEM), it was possible to identify some pollen types like Cryptomeria, Pine, Alder, Cyclobalanopsis, Chamaecyparis, and Equisetum. Daily average airborne pollen counts show strong variations from the day to day which makes the appropriate daily forecasts that could be of practical use for patients difficult. Diurnal variation of airborne pollen grains at our local sampling site is very irregular and shows no similarity between pollen types. The highest concentrations of Cryptomeria and Alder pollens in the south -west wind directions might be attributed to the airborne pollen transport, while the increase in Pine pollen grain in the southern wind direction was probably due to the local spread. Prevailing wind direction (SW) during the pollinating periods of Cryptomeria and Alder pollens could suggest a long-distance transport from a distant mountain.

Immature Pollen-Derived Plant Regeneration in Anther Cultures of Ranunculus japonicus Thunb (미나리아재비(Ranuculus japonicus Thunb.)의 약배양에 의한 미숙 화분 유래의 식물체 재분화)

  • In order to induce immature pollen derived plants, anthers of Ranunculus japonicus Thunb. were cultured on Murashige and Skoog's medium supplemented with various combinations of auxins and cytokinins. The combinations of NAA and BA were more effective than those of 2,4-D and kinetin in the formation of calli and embryos. Up to 5t5% of the anthers cultured on medium containing 0.5 mg/L NAA and 1.0 mg/L BA gave rise to plantlets. The most suitable stage for anther culture in the induction of calli and/or embryos from immature pollens was at the uninucleate and early binucleate stage (3 days before anthesis). Immature pollens developed into embryos by repeated division of the vegetative nucleate after 60days of culture.

Effects of Flavonoids on Pollen Tube Growth in Arabidopsis thaliana

  • Arabidopsis mutants deficient in flavonoid and sinapate ester (tt4 and fah1-7, respectively) were evaluated in vivo and in vitro to study the possible role of flavonoid compounds in pollen tube growth. In vivo, we investigated pollen tube growth in the pistils of the mutants and wild type(Ler). The growth of pollen tubes was significantly different among the three genotypes. In the fal1-7 pistils, the tubes grew to a greater length relative to those of the wild type or tt4. To examine in vitro pollen tube growth, a solid medium was devised for pollen germination and subsequent growth. In vitro, the identical result was obtained; fahl-7 pollens developed the longest tubes and elongated most rapidly. Therefore, the growth response of pollen tubes to phenolic compounds was examined by adding quercetin or sinapate ester in various concentrations to the media. Quercetin enhanced both germination rate and tube growth in the pollens of the mutants and the wild type, especially in tt4. In contrast, sinapate ester inhibits pollen germination and pollen tube growth in three genotypes. These results suggest that flavonoids and related phenolic compounds have physiological role in the plant reproductive system.

  • Plants pollination are conducted through various pollinators such as wind, animals, and insects. Recently, the necessity for artificial pollination is drawing attention as the proportion of natural pollinators involved is decreasing over the years. Likewise, the trade in pollen for artificial pollination is also increasing worldwide. Through these imported pollens, many unknown microorganisms can flow from foreign countries. Among them, spores of various fungi present in the particles of pollen can be dispersed throughout the orchard. Therefore, in this study, the composition of fungal communities in imported pollen was revealed, and potential ecological characteristics of the fungi were investigated in four types of imported pollen. Top 10 operational taxonomic unit (OTU) of fungi were ranked among the following groups: Alternaria sp., Cladosporium sp., and Didymella glomerata which belong to many pathogenic species. Through FUNGuild analysis, the proportion of OTUs, which is assumed to be potentially plant pathogens, was higher than 50%, except for apple pollen in 2018. Based on this study of fungal structure, this information can suggest the direction of the pollen quarantine process and contribute to fungal biology in pollen

Pollen Morphology of Genus Sedum in Korea

  • Pollens of 20 species of Sedum were investigated with a scanning electron microscope. The pollen morphology of Sedum was rather variable, within particular species or even within a single inflorescence. Differences occurred in the number and shape of apertures and surface sculpture. Besides 3-colporate, various aperture types including 2-syncolporate, 3-syncolporate, 40stephanocolporate, 5-stephanocolporate, zonate, and irregular types were found in a single specimen. Also, striate-rugulose and psilate sculpture were found in S. viviparum. No correlation was found between the pollen morphology and the floral formula. Pollen characters appeared to be not useful for infrageneric classification of Korean Sedum.

  • Cone and seed development after self-pollination in 14-year-old Pinus rigida Mill. was investigated in an $F_1$-hybrid seed orchard that consists of Pinus rigida Mill. and Pinus taeda L. in alternative rows. On an average, 83.8% of the self-pollinated receptive female strobili developed into cones. This value appears to be normal compared to cone development after open-pollination in other pine species such as in Pinus sylvestris L. However, there was a great variation in filled seed development after self-pollination among the investigated individual trees (Table 1-1). The 34.5%, a ratio of filled seeds after self-pollination was significantly lower than 91.7%, that by the open-pollination. Some of the investigated trees developed 70.0%-83.5% of filled seeds after self-pollination. Thus, those are not desirable as trees for seed parent in establishing $F_1$-hybrid seed orchards in future. A further study on the rate of filled seed development by various pollination systems consisting of self-pollination, self-and cross-pollination with intraspecific, mixed (tree's own and foreign pollens) pollens and cross-pollination with interspecific, mixed pollens of the two parental tree species is necessary to determine the quality of seed parent trees in establishing $F_1$-hybrid seed orchards.

Extraction Conditions for Rhododendron mucronulatum Pollen (추출조건에 따른 진달래 화분의 이화학적 특성)

  • The physiochemical properties of Rhododendron mucronulatum pollens were examined after the use of various extraction conditions. The levels of phenolic compounds and electron donating abilities (DPPH) were better after 80% (v/v) ethanol extraction than after water extraction. The content of phenolic compounds and the DPPH were high when the solvent ratio was 20X. The content of phenolic compounds was highest at $45^{\circ}C$ (347.60 mg/100 g). The DPPH was highest, at 67.93%, when extraction was performed at $25^{\circ}C$. An extraction time of 6 hr yielded the highest content of phenolic compounds (312.63 mg/100 g). The DPPH did not vary with extraction time. Both the levels of phenolic compounds and DPPH values rose when extractions were performed twice. In summary, a solvent ratio of 20X, an extraction temperature of $25-45^{\circ}C$, double extraction and an extraction time of 6 hare optimal for extraction, with maximal DPPH and phenolic content, of Jindalrae pollens.

  • Interspecific hybrid seedlings by artificial crossing of Genus Quercus (Q. serrata, Q. dentata, Q. mongolica var. crispula, Q. aliena) were planted at nursery in Tottori University. Seedlings of hybrids by natural crossing(Q. fabri and $Q.{\times}mccormickii$) were selected and planted at Hiruzen district in Tottori University. Artificial interspecific hybrid $F_1$ and natural hybrid $F_1$ bloomed when they were 4 years old and 3 years old, respectively. The pollen fertility and seed viability were investigated from the bloomed individuals in 2001. The germination percentages of the pollens of artificial interspecific hybrid were more than 84% except one individual, and the extension of pollen tubes was normal. The normal seed percentages of artificial interspecific hybrid were more than 90% similar to parents. Germination percentages of normal seed of natural crossing family were more than 64%, respectively, and selfed offsprings of Q. fabri, and $Q.{\times}mccormickii$ hade high reproductive ability.

  • In order to improve the efficiency of F$_1$ hybrid seeds production(KF 114), some experiments were carried out in a greenhouse this year. Mother plant(MSNC567) and pollen plant(NC) were grown in some pots (30 x 30cm, WxH). The gathered pollens were mixed with celite, pollen deluent dusts and stored in refrigerator at l$0^{\circ}C$ before pollination. To establish the critical range of the mixed rate and the storage period of pollens, the change in a percent of capsule set, number of seed and seed weight per capsule, weight of 1000 seeds and germination percent of seed resulted of pollination with pollen deluent dusts were investigated. The results are as follows; Percent of capsule set showed the mixed rate 1:5 resulted in no difference and those of 1:10 and 1:20 decreased about 5 ~15% when compared with trials using pollen alone. There was no difference among duration of storage in a same mixed rate. Numbers of seed per capsule showed a significant difference among the mixed rates and among durations of storage. Numbers of seed per capsule were decreased about 55 ~ 90 % as the mixed rate increase, about 2 ~ 28 % as the duration of storage increase. Weight of seed per capsule decreased about 16 ~ 23 % as the duration increase and decreased about 3 ~ 89 % as the mixed rate increase when compared with trials using pollen alone. Weight of 1000 seeds showed non significant difference among duration of storage but were higher than trials used pollen alone as the mixed rate increase.
