잔디의 약배양에 있어서 살정제 처리에 의한 Larger-sized Pollen의 발생빈도 증가 및 캘러스 유도 (Increase of Larger-sized Pollen Number by Gametocide and Callus Induction in Anther Culture of Zoysia japonica Steud.)

  • 잔디에 있어서 larger-sized pollen의 발생빈도에 대한 살정제 처리효과 및 캘러스 유기 조건을 조사하기 위하여 실험을 수행하였다. 약 안의 화분을 절취하여 화분의 생존력 및 dimorphism현상을 조사한 결과, 2가지 화분의 형태가 관찰되었다. 하나는 크기에 있어서 상대적으로 크며 (직경 30~36 $\mu\textrm{m}$) 연하게 염색되었고 크기가 작은 다른 화분 (직경 15~20$\mu\textrm{m}$)은 진하게 염색되었으며 amyloplast가 많이 산재해 있는 것으로 나타났다. 잔디의 약에서 배발생화분으로 추정되는 크기가 큰 화분 (larger-sized pollen)은 존재하고 있으나 그 수에 있어서는 약 1% 이하로 극히 적은 것으로 나타났다. 크기가 큰 화분의 발생빈도수를 높이기 위하여 약 4$0^{\circ}C$의 주간과 15$^{\circ}C$이상의 야간 온도 조건에서 재배된 잔디의 mid booting stage에서 GA$_3$50mg/L+0.1 mL/L Tween 20을 충분하게 엽면에 살포하였다. GA$_3$의 경우 mid booting stage에서 50mg/L의 농도로 처리하였을 때 크기가 큰 화분의 빈도가 25.4%로 가장 높게 나타났다. 위의 조건에서 처리된 약에서만 AA기본고체배지+8.0 mg/L 2,4-D+0.2 mg/L kinetin 배지에서 캘러스가 형성되었으나 캘러스 형성률은 약 1.0%로 매우 저조한 것으로 나타났다.

일별 꽃가루농도에 대한 기상영향 (Meteorological Impact on Daily Concentration of Pollens in Korea)

  • 세계적으로 기후변화에 의한 온난화 현상과 대기오염증가와 아울러 알레르기 유발성 꽃가루의 증가로 천식, 비염, 결막염 등 알레르기 질환 환자가 급증하고 있다. 본 연구에서는 꽃가루 농도의 변화에 따른 영향을 체계적으로 표준화하기 위하여 한국의 꽃가루 특성을 분석하였다. 관측된 꽃가루는 1998~2010년(단, 2006년은 제외)의 알레르기를 일으키는 소나무, 수목류(소나무 제외), 잡초류를 서울, 부산, 대구, 광주, 강릉, 제주지역을 중심으로 분석하였다. 꽃가루는 지난 12년간 서울을 포함한 대도시 지역에서는 장기적으로 증가하고 있으며, 앞으로도 장기적으로 증가할 것으로 판단된다. 꽃가루 농도와 기상요소는 매우 높은 상관관계가 있으며 특히 기온과 양의 상관관계를 가진다. 지역별로는 꽃가루 농도와 기상요소는 서로 다른 상관관계를 가진다. 꽃가루농도에 영향을 주는 기상요소 중저온누적도와의 관계를 이용하여 꽃가루 개화기간의 예측식을 도출하였다. 분석결과로부터 수목류의 경우, 충분히 낮은 기온이 만족되면, 개화기간이 줄어드는 결론을 얻었다. 이 모형에 의한 결과로부터 알레르기 유발 꽃가루의 유행시기변화를 생물계절모형으로 예측 가능함을 판단할 수 있다.

Varietal Responses of Pollen Development to Salt Stress in Barley

  • Thirty-nine out of eighty-five barley varieties/strains survived until heading stage in the saline experimental field $(0.03-0.05\%\;salt)$ and they were used for pollen study. Light and scanning electron microscopic observations revealed two distinctive types of barley pollens: one transparent and small in size and the other dark and larger. In addition, both types of pollens were stained with Alexander's stain and it was found that the smaller and transparent pollen was cytoplasm-devoid (CD) while the larger pollen was cytoplasm-rich (CR). Sixteen out of 39 barley varieties/lines grown in the saline soil had CR pollens, which were rarely observed in the barley plants grown in the non-saline soil. Moreover, it was observed that salt stress severely reduced seed setting in the varieties having degenerated pollens. These results suggest that salt stress affects the fertility of barley pollen. The sterile pollen was undersized and lack of cytoplasm probably due to abortion. Furthermore, a varietal difference existed in the response of pollen development to salt stress.

Size Determination of Pollens Using Gravitational and Sedimentation Field-Flow Fractionation

  • Pollens are known to be an allergen. They penetrate human respiratory system, triggering a type of seasonal allergic rhinitis called pollen allergy (hey fever). The purpose of this study is to test two field-flow fractionation (FFF) techniques, gravitational FFF (GrFFF) and sedimentation FFF (SdFFF), for their applicability to sizecharacterization of micron-sized pollens. Both GrFFF and SdFFF are elution techniques, providing sequential elution of particles based on size. They allow the size distribution as well as the mean size of the sample to be determined from the elution time. In this study, GrFFF and SdFFF were used to determine the size distribution of Paper Mulberry and Bermuda Grass pollens. For the Paper Mulberry pollen, the mean size obtained by GrFFF is 12.7 μm, and agrees rather well with the OM data with the relative error of 8.0%. For the Bermuda Grass pollen, the mean size obtained by GrFFF is 32.6 μm with the relative error of 12.3%. The mean sizes determined by SdFFF are 12.4 (relative error = 10.1%) and 27.1 μm (relative error = 5.2%) for the Paper Mulberry and the Bermuda Grass pollen, respectively. Although SdFFF tends to yield more accurate size distribution due to lower band broadening under the field strength higher than 1 G, the sizes determined by GrFFF were not significantly different from those by SdFFF.

Effect of different pollen sources on fruit characteristics and quality in 'Niitaka' pears (Pyrus pyrifolia Nakai)

  • This study was conducted to explore which pollen type could be an effective pollen source for self-incompatible 'Niitaka' pear (Pyrus pyrifolia Nakai), when different pollinizers were to be applied. The pollens of five different pears with similar flowering times were applied to bagged 'Niitaka' flowers grown in Cheonan city, Korea. Pollen types used were of different origins including Korean pears ('Wonhwang' and 'Chuhwangbae'), Japanese pears ('Chojuuro' and 'Okusankichi'), and Chinese pear ('Yali'). Results show that the amount of pollen produced by flowers of each cultivar was the largest in Korean pear 'Wonhwang' (234 g/100 flowers) and the smallest in Japanese pear 'Okusankichi' (126 g/100 flowers). A high proportion of large fruits (> 600 g), 71.4% of total production, was found in the fruits treated with 'Yali' pollen. However, these fruits showed an unfavorable external appearance. Based on individual fruit width differences, 'Chojuuro' treatment produced the smallest difference in fruit diameters (3.2 mm) and 'Yali' treatment' gave the largest (4.8 mm). Also, the difference in fruit length was the largest in 'Yali' (7.5 mm), which also produced the most uneven fruits among the pollinizers used. Meanwhile, the use of pollens of 'Wonhwang' and 'Chuhwangbae' improved productivity by increasing the fruit shape index and the ratio of bigger fruits in total production. Moreover, those pollens also decreased the development of lenticels on fruit skins which gave them a smoother appearance.

Morphological Features of Pollen Grains in Portulaca

  • The morphological features of pollen grains collected from the broad-leaved and cylindrical-leaved Portulaca species of the Hawaiian Islands were investigated. The variation in size and surface pattern were examined using scanning electron microscopy and statistical analysis. Pollens of the Portulaca were apolar grains of monads exhibiting apertures, colpi, and relatively thin echini. Of particular interest was the differing size of pollen grains among the species studied. Pollens of the broad-leaved Portulaca were considerable in size with a length of about $73{\sim}86{\mu}m$, while the cylindrical-leaved Portulaca tended to have smaller pollen grains ranging from 50~65 in diameter. The smallest pollens were found in cultivar 2 having an average of $43{\mu}m$. The pollen was intectate, and the exine surface appeared granulous, having sculptured elements of spinules and puncta. Two types of puncta were discerned; one forming an operculum, and the other, a simple perforation. Numerous spinules and small puncta were found throughout the cylindrical-leaved Portulaca. In the present study, morphological features of pollen grains from ten Hawaiian Portulaca species, including endemic and unknown taxa, revealed the aforementioned pattern of variation. This paper aim to provide morphological information that could be of phylogenetic value within the Hawaiian Portulaca.

봉밀(蜂蜜) 및 화분하(花粉荷)의 기원식물(基源植物) (Identification of Floral Type for Honey and Pollen Load)

  • The purpose of this study is concerned with identification of floral type of commercially available honeys and pollen loads. Ikuse's technique was applied to identify the pollen species of honey and pollen loads. Among six kinds of commercially available honeys in Taegu, Korea i.e. Robinia Honey, Brassica Honey, Castanea Honey, Rhododendron Honey, Lespedeza Honey and Multipant Honey, all honeys contained pollens, which were coincident with their trade names given by the apirists, except Lespedeza Honey. The trade name of imported honeys were not given by the plant, from which pollen material and nectar were collected. Among four kinds of these products, pollen was not identified in Clover Honey. A spectrum of honey products added by pollen loads was, of course, wider than simple honey products. The pollen load showed spectrum of pollens including not only those which are specialized for wind pollination, eq. Pinus species and Gramineae, but also contained for insect pollination, eq. rose and leguminous plants.

시판 천연화분 및 가공화분의 품질에 관한 비교 연구 (Comparison of Quality of Natural Pollen and Processed Pollen)

  • Dissolution rate , periduium state, nutrient component and fatty acid composition of 5 species of natural pollen and 3 species of processed pollen were carried out for evaluation of quality. Natural pollens had average 11.5% of moistures, and had native peridium may be responsible for allergy. The average dissolution rate of natural pollens were low as 66.3%. One among 3 species of processed pollen was observed to have not peridium, resulted in higher dissolution rate. Major fatty acid of pollen was palmitic acid but content of oleic acid was lower than that of other plant seeds.

설악산 식물군락 유기물층의 화분분석학적 연구 (Pollen Analytical Study on Humus Accumulated at Plant Communities of Mt. Seolag)

  • A pollen analytical study was carried out on the accumulated humus samples collected from 27 sites under the different plant communities of Mt. Seolag. These pollen spectra were then compared with the actual vegetation. The aboreal pollen(AP) and non-abnoreal pollen(NAP) were 1 Family 25 Genus and 5 Family 5 Genus, respectively. Among the aboreal pollen identified, the pollen of Pinus, Quercus, Betula and Acer were appeared in abundance and the coniferous pollen of Pinus was occupied at least 20%-65% through all the sites studied. The pollens of Abies, Betula, accer, Quercus, Carpinus, Corylus, Fraxinus, Styrax, Prunus and Ericaceae were in accordance with the present vegetation but the other pollens were inconsistent with the actual vegetaton. It suggests that it was caused by the difference between Wehseolag and Namseolag of subalpine zone in Mt. Seolag was 200 m by pollen spectra. Quercus was distributed at the lower part of Namseolag and the subalpine boreal elements were increased with increasing altitudes. The pollen spectra of each site was put together into 5 groups of pollen assemblage by cluster analysis and the community coefficient (CC) was over 60% between all sites studied.

Physio-chemical impacts on efficient germination in Platycodon grandiflorum for. duplex pollen culture

  • The highly valued ornamental plant, Platycodon grandiflorum for. duplex was generated by petaloidy of a calyx of Platycodon grandiflorum. The present investigation was executed to explore the several factors having effects on the germination of pollens with a view to acquire the underlying data for the artificial crossing to cultivate the species of Platycodon grandiflorum for. duplex. Both low and high temperature impaired the germination of Platycodon grandiflorum for. duplex pollens. The good germination rate was observed at the temperature of $25^{\circ}C$. The types and concentrations of carbon sources induced the differences in germination rate. The germination rate increased with the increasing concentration of sucrose and glucose except for fructose. Sucrose and glucose showed the highest results at the concentration of 20%. While fructose demonstrated the similar tendency to sucrose and glucose, it reduced the germination rate at the concentration of 20%. The highest germination rate was observed at the concentration of 15%. The appropriate carbon course for germination of pollens of Platycodon grandiflorum for. duplex was glucose of which germination rate was twice as high as that of sucrose and fructose. The germination rate was reduced substantially when the pH was close to alkali, and the potential germination rate was obtained at pH 6. Boric acid enhanced the germination rate at a lower concentration than the higher concentration.
