Effect of dietary carbohydrate sources on apparent nutrient digestibility of olive flounder (Paralichthys olivaceus) feed

  • Apparent digestibility coefficients (ADCs) of dry matter, crude protein, crude lipid, nitrogen-free extract, and energy in selected carbohydrate sources including wheat flour (WF), ${\alpha}-potato$ starch (PS), ${\alpha}-corn$ starch (CS), Na alginate (AL), dextrin (DEX), and carboxymethyl cellulose (CMC) were determined for olive flounder. The olive flounder averaging $150{\pm}8.0g$ were held in 300-L tanks at a density of 30 fish per tank. Chromic oxide was used as the inert marker. Feces were collected from the flounder by a fecal collector attached to a fish rearing tank. Apparent dry matter and energy digestibilities of flounder fed WF, PS, CS, and DEX diets were significantly higher than those of fish fed AL and CMC diets. Apparent crude protein digestibility coefficients of flounder fed PS and CS diets were significantly higher than those of fish fed AL, DEX, and CMC diets. Apparent crude lipid and nitrogen-free extract digestibility coefficients of flounder fed PS and DEX diets were significantly higher than those of fish fed WF, CS, AL, and CMC diets. The present findings indicate that PS and DEX could be effectively used as dietary carbohydrate energy compared to WF, CS, AL, and CMC for olive flounder.

  • Cysteine-cysteine chemokine receptor 9 (CCR9) homologue cDNA was isolated from olive flounder leukocyte cDNA library. Olive flounder CCR9 homologue consisted of 1709 bp encoding 367amino acid residues. When compared with other known CCR peptide sequences, the most conserved region of the olive flounder CCR9 peptide is the seven transmembranes. A phylogenetic analysis based on the deduced amino acid sequence showed the homologous relationship between the olive flounder CCR9 sequence and that of Mouse CCR9. The olive flounder CCR9 gene was predominantly expressed in the Peripheral blood leukocytes (PBLs), kidney, spleen, and gills.

Molecular Identification of the Fish 4-Aminobutyrate Aminotransferase from Flounder, Paralichthys olivaceus

  • 4-Aminobutyrate aminotransferase plays an essential role in the 4-aminobutyric acid shunt, converting 4-aminobutyrate to succinic semi aldehyde. We isolated and sequenced' a fish cDNA fragment that encodes 4-aminobutyrate aminotransferase. A brain cDNA library from flounder (Paralichthys olivaceus) was constructed using the ZAP- III XR vector and screened for the fish 4-aminobutyrate aminotransferase gene using a probe derived from the conserved sequences of known mammalian 4-aminobutyrate aminotransferases. A partial cDNA for 4-aminobutyrate aminotransferase was cloned and found to be 700 bp in length corresponding to 66 amino acids. Nucleotide sequence of the clone was aligned with NCBI (National Center for Biotechnology Information) DNA sequence data base. The result showed high sequence identity with previously reported mammalian 4-aminobutyrate aminotransferases. The trans­criptional level of flounder 4-aminobutyrate aminotransferase was detected with the presence of mRNA at different flounder tissues by reverse transcription-polymerase chain reaction (RT-PCR). The expression of flounder 4-aminobutyrate aminotransferase was also tested and detected from the flounder tissues of the brain, liver, kidney and pancreas.

제주넙치 양식산업의 경제파급 효과분석 (Economic Impact Effect Analysis of Flounder Aquaculture Industry in Jeju)

  • We have done the input-output analysis to see the over all impact of flounder industry of Jeju region on the domestic economy of Korea. To do the input-output analysis, we have constructed the data set for the input-output table by using the existing data set in the "2003 input- output table of Jeju regional area" published by the joint work of Jeju branch of Korea bank and the Jeju Development Institute, together with some raw data provided by Jejudo Marine Fish-Culture Cooperative. We have also produced input coefficient of flounder industry by making flounder industrial sector exogenous, separated from intermediate demand. To summarize our empirical results, the inducement effect of production, value added, and employment of Jeju flounder aquaculture industry are 300 billion won, 116 billion won and 1,800 people respectively. In conclusion, the results of this study suggest flounder industry of Jeju region contributes powerfully to not only Jeju economy but also all over the Korea economy.

Generation of polyclonal antiserum to olive flounder (Paralichthys olivaceus) immunoglobulin by immunization of rabbit with plasmids containing heavy chain gene of olive flounder immunoglobulin

  • In fish vaccinology, the secondary antibodies against fish immunoglobulins (Igs) are necessary to measure specific humoral immune responses in immunized fish. In the present study, polyclonal antiserum against olive flounder (Paralichthys olivaceus) IgM heavy chain was generated by intramuscular immunization of rabbit with Escherichia coli/eukaryotic shuttle vector containing open reading frame (ORF) of olive flounder IgM heavy chain. Western blot analysis demonstrated the specific activity of the rabbit antiserum with reduced olive flounder serum H chain at dilutions up to 1:1000. Titer of immunized rabbit serum against olive flounder serum was significantly higher than that of pre-immunized rabbit serum when determined by ELISA.

  • The cDNA sequence of the Japanese flounder (Paralychthys olivaceus) IgD has been previously reported (GenBank accession no. AB052658) and this was followed by the detection of IgD mRNA expression in some flounder organ tissues. However, it has not been determined whether the flounder IgD gene is virtually expressed into IgD protein. To characterize the flounder immunoglobulins utilized in elucidating the mechanism, evolution and diversity of the flounder immune system, antibodies specific to IgD and IgM were necessary. In the present study, partial flounder recombinant IgD (rIgD), IgM (rIgM) and the conserved regions of IgD and IgM (rCIg) were produced by cloning the cDNA sequence using isotype specific primers which were designed to produce unique fragments of IgD and IgM specific amino acid sequences. The production of recombinant Igs was ascertained by SDS-gel electrophoresis and immunoblot analysis using anti-T7$\cdot}$Taq antibody. The produced recombinant Igs were purified using affinity columns, and used as immunogens. Antibodies specific to the isotype of flounder Igs were generated by immunizing rabbits with rfIgs and the antibodies produced were identified by enzyme-linked immunosorbent assay (ELISA) and immunoblotting. Specificities of the generated antibodies were evaluated by testing cross-reactivity between recombinant IgM and IgD. By ELISA, rabbit antibodies against the rfIgD fragment (anti-rfIgD) failed to recognize any kind of flounder serum Igs, whereas respective antibodies against rfCIg (anti-rfCIg) and rfIgM fragments (anti-rfIgM) reacted with serum Igs. Likewise, in immunoblot assays, though anti-rfIgD did not, both anti-rfCIg and anti-rfIgM bound with the ~85 kd flounder IgM heavy chain. By flow cytometry analysis, anti-rfCIg, anti-rfIgD and anti-rfIgM reacted with 6%, 3% and 6.5% of cells, respectively, suggesting that flounder IgD is not secreted in serum but expressed on flounder B-like cell surfaces as in mammals. Antibodies produced against recombinant flounder Igs could be used to develop sandwich assay systems for detecting flounder Igs and for further investigating the flounder immune system.

  • The properties of surimi gel from Olive flounder Paralichthys olivaceus were evaluated with addition of various food additives and by heating under different conditions. The optimum heating conditions for get good textured surimi gel from Olive flounder was 40 minutes at $85^{\circ}C$. Optimum texture characteristics such as hardness, cohesiveness, chewiness, gumminess and gel strength of olive flounder fish paste can be acquired by washing the paste 2 times and by adding sodium chloride (2.5%), sodium polyphosphate (0.3%) and starch (4%), respectively. Furthermore, we compared gel texture characteristics and strength of manufactured Olive flounder fish surimi with commercial Alaska pollack to determine their commercial applicability. Texture characteristics and gel strength of Olive flounder surimi were higher than those of the commercial Alaska pollack surimi. In addition, both lightness and whiteness were higher in surimi gel from Olive flounder than from the commercial Alaska pollack.

  • Interspecific hybridization is a beneficial strategy in aquaculture. In this study, diploid hybrids were produced through interspecies hybridization of flounder Platichthys bicoloratus ♀ × starry flounder P. stellatus ♂, which became commercial within a year. These hybrids exhibited an advantage in growth ability, with female hybrids growing twice as large as males at 11 months. Furthermore, hybrid flounders exhibited faster growth rates than starry flounders and reciprocal hybrids. The average growth rate of hybrids was 1.37 and 1.18 times greater than that of starry flounder and reciprocal hybrids at 5.5 months, respectively; Additionally, feed efficiency was 1.21 and 1.10 times higher than that of the experimental groups. The experiments were conducted at five temperatures to determine the optimal fry production method. Six weeks of culture resulted in a significant temperature effect, with maximum growth at 27℃. However, over 90% of the males occurred in the groups at temperatures above 24℃. Therefore, the optimum temperatures for survival, growth, condition factor and frequency of the best-sized fingerling at 13 weeks of age were estimated at 21℃. Thus, hybrid flounders are of great potential for use in the flounder aquaculture industry.

Korean Mistletoe, Viscum album coloratum Induces Non-Specific Immune Responses in Japanese Flounder, Paralichthys olivaceus

  • Effects of Korean mistletoe, Viscum album coloratum on the non-specific immune responses of Japanese flounder, Paralichthys olivaceus were examined. Flounder were inoculated with mistletoe, Freunds complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control into their peritoneal cavities. Reactive oxygen intermediate (ROI) products were more enhanced in mistletoe-injected fish kidney phagocytes than in FCA-injected ones. The level of lysozyme activity detected in the serum of fish 4 d after injection with mistletoe was also significantly higher than that found in the serum of the control fish. The appropriate concentration of mistletoe in eliciting the highest level of serum lysozyme activity was 500 $\mu$m/300 g of fish. In phagocytic activity assays, mistletoe-sen-sitized flounder kidney phagocytes captured more yeasts than those of the control fish. Korean mistletoe appeared to be a good activator of the non-specific immune responses of Japanese flounder.

Compensatory Growth in Juvenile Olive Flounder (Paralichthys olivaceus) in the Spring

  • A feeding trial was conducted to determine compensatory growth of juvenile olive flounder in the spring. Five treatments of fish with 3 replicates were prepared: the control group fish (C) fed twice daily for 8 weeks, the Sl, S2, S3 and S4 fish fed for 7, 6, 5 and 4 weeks after 1, 2, 3 and 4 weeks of starvation, respectively. Survival of olive flounder was not significantly different among treatments. Weight gain of flounder in S2 was significantly (P<0.05) higher than that of fish in S3 or S4, but not significantly different from that of fish in C or Sl. The poorest weight gain was observed in fish of S4 treatment. Specific growth rate (SGR) for flounder in S2 was significantly (P<0.05) higher than that for fish in C or S4, but not significantly different from that of fish in Sl or S3. Feed intake (g/fish) was proportional to duration of days of feeding except for flounder in S2, but not significantly different among C, Sl or S2. Feed efficiency ratio (FER) and protein efficiency ratio (PER) for flounder in S2 were significantly (P<0.05) higher than for fish in C, but not significantly different from those for fish in Sl, S3 or S4. Hepatosomatic index (HSI) and condition factor (CF) for flounder in Sl, S3 and S4 were not significantly different from those for fish in C, but significantly (P<0.05) lower than S2 except for CF in Sl at the end of the experiment. Proximate composition of the whole body of flounder was not significantly different among treatments. In considering results of the experiment, juvenile olive flounder achieved compensatory growth when properly fed after starved up to 2 weeks in the spring. Compensatory growth of fish was supported by improvement in SGR, FER and PER in fish starved.
