Structural Investigation of Lignins in Three Different Ferns (Pteridopbytes)

  • This paper examines the structural characteristics of fern lignins (deer fern (Blechnum spicant), sword fern (Polystichum munitum) and maidenhair fern (Adiantum pedatum)) by chemical degradation methods of thioacidolysis and nitrobenzene oxidation as well as 13C NMR. Phloroglucinol-HCI staining indicates that the lignins are specifically accumulated at the sclerenchyma cells beneath the epidermis and vascular bundles. The fern lignins consist of only guaiacyl units. Remarkably, the frequency of the -O-4 linkages is extremely low in fern lignins (only 9 to 11 %). Furthermore, the presence of lignin is ambiguous in maidenhair fern, due to very rare amount of -O-4 linkage. Biphenyl (5-5) and 1,2 bis arylpropane (-1) are main condensed dimeric substructures in fern lignins over 70%. In addition, 13C NMR analysis strongly evidenced the integration of phenolics or their derivatives into the fern lignins.

Report of fern spore-feeding Calicotis latebrifica Terada, 2016 (Lepidoptera, Stathmopodidae) new to Korea

  • A fern spore-feeding stathmopodid species, Calicotis latebrifica Terada, 2016 is reported for the first time from Korea on the basis of one male and three females from Island Chujado, Jeju Province. This record represents the third example of the Korean Lepidoptera associated with fern spores for larval food items. The species is similar to Cuprina fuscella Sinev, 1988 in feeding on fern spores but differs from the latter in the body color and the presence of cilia on the ventral side of antennal flagellum. The genus Calicotis is introduced for the first time to the Korean fauna. Calicotis latebrifica has been known exclusively from Japan before this study. Larval habits for feeding and shelter construction were demonstrated from field observation and rearing in captivity. A new host plant of C. latebrifica, Dryopteris nipponensis Koidz. is recorded. The habitus and genitalia of both sexes are briefly described for C. latebrifica with photographs.

  • Fern gametophytes are a good model system to study plant morphogenesis, because of their simple organization and various photocontrolled responses. We studied fern photomorphogenesis including chloroplast photoorientation using Adiantum gametophytes to analyze signal transduction pathways of plant photomorphogenesis. Chloroplast photoorientation will be shown in detail and molecular structure of fern phytochromes and blue light absorbing pigments will also be discussed.

  • The global environment is deteriorating at an alarming rate despite of enhanced international environmental regulation. Many studies have been performed to reduce pollutants. Recently, phytoremediation, plant-based technology for the removal of toxic contaminants from soil, water, and air, has been receiving large attention. Arsenic-contaminated soil is one of the major pollutant sources fur drinking water. The fern brake (Pteris erotica) has been reported as a hyper-accumulate arsenic from soils. In this study, we investigated the arsenic absorption effect on sap flow inside xylem vessels of a fern brake. The synchrotron X-ray micro-imaging technique was employed to monitor flow inside the plant non-invasively. The captured phase-contrast X-ray images show both anatomy and transport of water inside the fern brake. The refilling process of water containing arsenic inside the xylem vessels of fern brake's leaves and stems was clearly observed. These results would provide important information needed fur understanding the mechanisms of accumulation, translocation, and transformation of toxic materials in plants.

고사리의 독성물질 Ptaquiloside 제거방법 연구 (Study on Processing Methods to Remove Toxic Ptaquiloside from Bracken Fern)

  • Ptaquiloside는 고사리에 들어 있는 발암성 물질이다. 본 연구는 다양한 조리법에 따라 고사리에 함유된 ptaquiloside을 측정해 보고자 하였다. 조리법은 가열, 물에 침지, 물교환 횟수 등이었다. 알칼리 가수분해와 용매분획은 ptaquiloside를 화학적으로 안정한 물질인 pterosin B로 전환시켜 주었다. Pterosin B 함량은 이동상용매로 3 mM ammonium과 메탄올 용매를 사용하여 UPLC-MS/MS로 분석하였다. 조리과정이 없는 생고사리의 $H_2O$추출물에 함유된 pterosin B함량은 81 mg/kg으로 검출되어 독성물질 ptaquiloside는 46.4 mg/kg이었다. 5분의 가열 시간 후 고사리의 pterosin B 함량은 약 60%정도 제거시켜주며, 이 중 2/3는 ptaquiloside에서 전환된 pterosin B가 아닌 고사리 고유의 ptersoin B 함량이었다. 12시간 침지를 추가한 고사리는 처리하지 않은 고사리에 비해 약 88%, 또한 매시간 물교환 작업을 추가시 99.5%까지 제거됨을 알 수 있었다. 따라서 독성물질 ptaquiloside의 제거는 가열시간과 더불어 침지와 물 교환 횟수도 동시에 이루어져야 함을 알 수 있었다.

마우스의 실험적(實驗的) 고사리 중독(中毒)에 관한 병리학적(病理學的) 연구(硏究) (Pathologic Studies of Experimental Bracken Poisoning in Mice)

  • In order to investigate the effects of bracken fern (Pteridium aquilinum) on animals, a total of 105 mice given a ration containing different percentage of bracken fern were studied pathologically during the experimental period of 90 days. The results obtained were as follow: 1. Mice in the group, given a ration containing 15% of bracken fern, showed 85% of death rate in 90 days, and mice in the group, given a ration containing 25% of it, showed 100% in 30 days 2. The experimental mice showed decreased myeloid cells and significant reduction in erythrocytes, leukocytes, and thrombocytes. 3. Severe congestion and hemorrhages were observed in lungs, liver, kidney, spleen, stomach, intestine, brain, adrenal and testis from the ones that died during the experimental period, but the necropsies of the latter period indicated mild chanties comparatively. 4. Mice in the group, given a ration containing 15% blacken fern showed a decreasing tendency in body weight.

Neopestalotiopsis Leaf Blight, an Emerging Concern on Leatherleaf Fern in Indonesia

  • Leatherleaf fern (Rumohra adiantiformis) is an important ornamental plant in Indonesia and global. Green fern leaves with bold dark green color with long shelf-life, attract florists as decoration. Indonesia is one important leatherleaf fern exporters, however currently an outbreak of leaf blight decreased production significantly. Initial symptom was reddish brown spots from edge of leaf, which was gradually followed by dark-brown necrotic lesions causing leaf blight and dried. This is a study to do Koch-Postulate approach and molecular identification, to identify the pathogen of the "new emerging disease" reported. Based on multigene analysis using primers from ITS, β-tub and tef1-α gene markers, the pathogen was identified as Neopestalotiopsis sp. All sequences have been deposited in GenBank with accession number of OR905551 (ITS), OR899817 (ßtubulin) and OR899816 (TEF). This Neopestalotiopsis leaf blight causes an emerging concern in leatherleaf fern in Indonesia and global biosecurity because it infected an export commodity.

제주도의 상록양치식물 자생지 환경특성 및 식생구조에 관한 연구 (Habitat Characteristics and Vegetation Structure of the Evergreen Fern in Jejudo, Korea)

  • The research was carried out to define the environmental characteristics and vegetation structure of the evergreen fern habitat in Jejudo. The growth conditions of evergreen fern in various habitats was surveyed, including topographical features, vegetation structure, air temperature, humidity, intensity of light, water content and organic matter content in the soil. In the direction of the native habitat, 70% of research area was located in the southeast-facing slopes, 30% was located in southern slopes. The gradient ranged from $0^{\circ}$ to $30^{\circ}$. Temperature ranged from $16^{\circ}C$ to $28^{\circ}C$, and $22.3^{\circ}C$ was the average. Humidity ranged from 20 to 68%, and 36% was the average. In the native habitat, the highest light intensities reached 60,000 to 80,0001ux, but in general ranged from 300 to 40001ux. Water content in the soil ranged from 32% to 59%, organic matter content ranged from 8 to 13%. Within a unit of 25$m^2$, there were tall-tree layer such as Quercus galuca and Castanopsis cuspidata with a covering of rate 40~80%, a sub-tall-tree layer such as Camellia japonica, Staphylea bumalda and Sambucus williamsii with the covering rate of 3~5%, a shrub layer with the covering rate of 5~20%, and a grass layer with the covering rate of 40~95%. This research provides the basic data about the native habitat environment of the evergreen fern plant. Continuous monitoring and accumulation of data is necessary for the use of evergreen fern as vegetation materials.

천연기념물 칠곡 금무봉 나무고사리 화석산지에 대한 고찰 (Reconsideration of the Natural Monument Geummubong Petrified Tree Fern Fossil Site, Ghilgok, Korea)

  • 칠곡 금무봉 나무고사리 화석산지는 1925년 Tateiwa가 중생대 나무고사리 줄기 화석(Cyathocaulis naktongensis)을 처음 발견한 모식산지로서, 그 학술적 가치를 인정받아 일제강점기인 1930년대부터 천연기념물로 지정, 약 90여년 간 보존되어 왔다. 하지만, 발견 당시 나무고사리 화석 산출 층준을 찾지 못해, 전석 상태의 표본을 기반으로 낙동층 내 역암/사암층을 함화석층으로 해석하면서, 금무봉 일대가 광범위하게 문화재구역으로 설정되었다. 2003년 이후, 지정구역 인근 노두에서 나무고사리 화석이 발견되면서 산출지역의 퇴적암상이 특정되었으며, 나무고사리 화석의 외형과 해부학적 특징에 대한 연구가 추가적으로 이루어졌다. 이를 통해 Ogura가 국내에서 보고한 Cyathocaulis속 2종의 분류 기준을 재검토하고, C. naktongensis와 C. tateiwai가 동종일 가능성을 고찰하였다. 또한, 오랫동안 이어져 온 각종 규제에 따른 갈등과 민원 해결을 위해, 현재의 문화재 구역은 시대환경 변화에 맞게 적정성 검토를 통한 큰 폭의 재조정이 필요하다고 판단된다.

일엽초의 페놀성 물질 (Phenolic Compound from Lepisorus thunbergianus)

  • Two phenylpropanoids and one flavan 3-ol were isolated from Lepisorus thunbergianus (Polypodiaceae, fern), which is used as folkmedicine. Phenylpropanoids were identified as caffeic acid and chlorogenic acid, and flavan 3-ol was elucidated as (-)-epicatechin 7-O-${\beta}$-D-glucoside by physico-chemical and spectral evidences (HMQC, NOESY).
