Comparison of Four-view Abdominal Radiographic Studies on Duodenal Gas Pattern in Dog

  • In this retrospective study the effect of positioning on the radiographic appearance of duodenum and the radiographic differences of duodenal gas pattern between dogs without pancreatitis and dogs with pancreatitis were investigated. Ventrodorsal, dorsoventral as well as both lateral abdominal radiographs of 255 dogs were evaluated. Gas distribution in the duodenum, gastroduodenal angle, accuracy of the positioning was examined. The duodenum was more visible in a left lateral view and a dorsoventral view. The positioning of dorsoventral view was the most inaccurate. Based on this study, there were not significant differences in duodenal gas pattern between dogs without pancreatitis and dogs with pancreatitis.

  • This study was carried out to evaluate the three months subacute intravenous toxicity of water soluble dimethyl dimethoxy biphenylate derivative (DDB-S), a newly formulated therapeutic agent for hepatitis, in Beagle dogs. Groups of 12 male and 12 female dogs were given different dosage of DDB-S, 10 mg/kg/day (high dose group), 5 mg/kg/day (middle dose group), 2.5 mg/kg/day (low dose group) and 0 mg/kg/day (control group) for three months by intravenous route. 1n the three months intravenous toxicity study, there were neither dead animals nor significant changes of body weights during the experimental period. 1n addition to, no significant DDB-S related changes were found in clinical signs, urinalysis and other findings. Statistical changes were observed in hematological. biochemical, partial thromboplastin time (PIT) and organ weight parameters of treated groups. However, these alteration had no relationship with dosage. No histopathological lesions were observed in both control and treated animals. Above data suggest that no observed adverse effect level of test materials in Beagle dogs might be over 10 mg/kg/day in this study.

  • We described a rapid, sensitive and reproducible real-time PCR assay for detection and quantitation of canine parvovirus type 2 in the feces of dogs with parvovirus infection. The method was demonstrated to be highly specific and sensitive, allowing a precise canine parvovirus type-2 quantitation over range of eight orders of magnitude from $10^2\;to\;10^9$ copies of standard DNA. Then, fecal samples from parvovirus infected dogs were analyzed by conventional PCR and real-time PCR. Real-time PCR is more sensitive than conventional PCR, allowing to detect low viral titers of CPV-2 in infected dogs. By real-time PCR, a wide range of parvovirus particles was found in the samples from $1.45\times10^6\;to\;9.45\times10^8$ copies/0.01g of feces. However, when dogs are in infection of parvovirus, it is difficult to prove that the numbers of peripheral blood leukocytes are correlated with those of fecal shedding virus.

  • It is well-known that onion and garlic of Allium family have been cultivated for major food since ancient times. Especially garlic is known for containing protective materials against cardiovascular disease, neoplasm and cancer, and decreasing cholesterol value. Hemolytic anemia was first reported by Goldsmith in onion-fed cattle in 1909. Subsequently, similar effects have been reported in cattle, horses, sheep, dogs and cats. This toxic effect has been attributed to n-propyl disulfide which causes oxidant damage to erythrocyte cell membrane. There are few reports on overconsumption of garlic. Kim et al. reported in 1999 that 45% of Jindo dogs in korea have erythrocytes characterized by high Na-K-ATPase activity with high K and low Na concentrations (HK) which easily results in hemolytic anemia. The purpose of present report is to determine whether garlic-induced hemolytic anemia occurs in normal erythrocytes characterized by complete lack of Na-K-ATPase activity with low K and high Na concentrations (LK) dogs are fed large amounts of garlics. We found in this experiment that Erythrocyte count, PCV and Hemoglobin concentration decreased significantly, and GSH and Met-hemoglobin concentrations increased. Consequently we determined that overconsumption of garlic induces hemolytic anemia in LK dogs.

Toxicity of Novel Solubilizer of Paclitaxel, Aceporol 330, in Beagle Dogs

  • In order to develop an improved paclitaxel formulation vehicle, a micelle forming solubilizer, Aceporol 330 was synthesized. It was previously reported that Aceporol 330 provided the linearity of paclitaxel plasma pharmacokinetics. In this study, the single dose toxicity test and 2-week repeated dose toxicity test of Aceporol 330 was performed in beagle dogs after intravenous administration. Single dose and 2-week repeated dose toxicity test of Aceporol 330 showed fever/generalized erythema, severe vomiting, and diarrhea in beagle dogs. However, those toxicities were less severe than those of Cremophor EL. Blood chemistry analysis of 2-week repeatedly treated beagle dogs with Aceporol 330 showed significant elevation of total cholesterol (TCHO) and triglyceride (TG) compared to that of control group. Cremophor EL also significantly increased total cholesterol (TCHO) and triglyceride (TG) as much as Aceporol 330. Results from this study indicated that Aceporol 330 was less toxic than Cremophor EL. Based on the pharmacokinetic advantages and the low toxicity of Aceporol 330 in single dose and 2-week repeated dose toxicity test, Aceporol 330 has a potential for use as a safer solubilizer for paclitaxel than Cremophor EL.

Pediococcus spp.-fermented chicken meat for dogs

  • An experiment was conducted to evaluate Pediococcus spp.-fermented chicken meat as a snack for dogs. The fermented or non-fermented snacks used in this study were prepared through the following process; meat mixtures containing 52.8% MDCM, 35.2% chicken breast meat (CBM) and 9.7% corn starch were inoculated with or without Pediococcus spp., incubated at 37℃ for 24 h and then sterilized at 121℃ for 20 min. During the 24-h fermentation, the pH of fermented chicken snack dropped rapidly with concomitant increase in number of lactic acid bacteria. The nutritional composition was not altered by fermentation. In vitro pepsin nitrogen digestibility was higher (p < 0.05) in the fermented snack compared with the non-fermented snack. Upon storage at room temperature for 14 days, bacteria grew slowly in fermented vs. non-fermented snack samples. In a palatability trial, dogs preferred non-fermented over fermented snack food. In 12-d-long feeding trial, fecal ammonia content was lowered, but fecal lactic acid content was increased in dogs fed the fermented vs. non-fermented snack food. Our study shows that the fermented MDCM-based snack exhibited good preservability upon storage, and improved in vitro nitrogen digestibility and fecal characteristics in dogs.

Treatment of Calcinosis Cutis with Minocycline in Five Dogs

  • Calcinosis cutis is a chronic condition characterized by insoluble calcified deposits in the skin and subcutaneous tissue. Although there is no uniformly effective treatment for calcinosis cutis, minocycline therapy has demonstrated varying degrees of benefit in humans. Five client-owned dogs with calcinosis cutis were included. Minocycline was administered orally in a dose of 10 mg/kg bodyweight twice a day. Treatment was repeated every day until complete remission. The efficacy of minocycline was evaluated within this period. The side effects of minocycline were monitored and reported by the owners and veterinarians. Of the 5 dogs with calcinosis cutis, which was classified as the dystrophic form, four dogs had a complete remission of calcinosis cutis and one dog had a partial response. The major improvement was a reduction in the size of the calcified deposits and reduction in inflammation associated with them. The duration of remission was $9.1{\pm}2.2$ weeks. The adverse effects, observed in one dog, were anorexia and vomiting. Minocycline may be effective in the control of calcinosis cutis in dogs.

Cross-reactivity between sera from dogs experimentally infected with Dirofilaria immitis and crude extract of Toxocara canis

  • This study was performed to investigate whethere there is cross-reactivity between Dirofilaria immitis and three intestinal nematodes of dogs. In ELISA, D. immitis- infected dog sera obtained at the 4th molting stage (9-11 weeks) and microfilaremic stage (25-30 weeks) were shown to be highly reactive with crude extract of T. canis. In immunoblotting, some antigenic fractions, 44, 57 88, 100 kDa of crude extract of T. canis, were found to be positive reaction with sera of dogs infected with D. immitis. However, little or no cross-reaction were observed between sera of D. immitis-infected dogs and crude extract antigen of T. vulpis or A. caninum. These result suggest that there are partial cross reaction between sera of D. immitis-infected dogs and the antigen of T. canis.

Safety and Effects on General Health, Skin Condition and Lipid Profile of a Soy Lecithin-Based Dietary Supplement in Healthy Dogs

  • The aim of this study was to evaluate the safety and effects of a soy lecithin-based supplement on the general health, skin condition, and blood lipid levels of dogs. Twenty-five, healthy, privately-owned dogs were enrolled in the study. Oral supplementation was administered for 8 weeks in the treatment group. Evaluation items included a thorough physical examination, owner responses to a questionnaire, blood analysis (CBC, CRP, serum chemistry, lipid profile). The results showed a significant improvement in amount of exercise, water intake, skin exfoliation in the treatment group (P < 0.05) but no significant changes in the other evaluation items, including no significant changes in blood component results following administration of the supplement. Most dogs well tolerated the treatment, and there were no serious adverse events. We suggest that soy lecithin has potential as a nutraceutical for the positive effect of general health condition such as activity and skin condition. Further studies are needed to establish the appropriate dose level and administration frequency of soy lecithin in dogs.

Combination of Stades Forced Granulation Method and Hotz-Celsus Procedure for Treatment of Upper and Lower Eyelid Entropion-Trichiasis in Three Dogs

  • Three dogs (a 30-month-old spayed female English bulldog; a 12-month-old castrated male English bulldog; a 19-month-old castrated male Shar-pei dog) were presented to the Dana Animal Hospital Eye Center with clinical signs of blepharospasm, epiphora, and ocular discharge (serous to mucopurulent) in one or both eyes. Through ophthalmic examination, the dogs (5 eyes) were diagnosed with trichiasis caused by upper and lower eyelid entropion, including the lateral canthus. To correct upper and lower eyelid entropion, modification using a combination of the Stades forced granulation method for upper eyelid entropion and Hotz-Celsus procedure for lower eyelid entropion was performed in all cases. At the last follow-up, mean 274 days after surgery, clinical signs disappeared and no recurrences of entropion were observed in any of the 3 dogs (5 eyes). Therefore, the combination of the Stades forced granulation method and Hotz-Celsus procedure may be an effective treatment for trichiasis caused by upper and lower eyelid entropion in dogs.
