Serological Study on Q Fever by Detection of Complement Fixation Antibodies to Coxiella burnetii in Dairy Cattle in Kyuuggi Province, Korea (Coxiella burnetii 보체결합항체 검사에 의한 경기지역 젖소에 있어서의 Q열에 대한 혈청학적 조사연구)

  • Regarding to Q fever which is one of the most important zoonoses in Food (Milk) Hygiene and in Environmental Public Health, a sero-epidemiological study was carried out to detect the complement fixation antibodies to Coxiella burnetii Nine Mile strain phase II antigen among the milking cows in Kyunggi Province. The results obtained were summarized as follows: 1. The overall prevalence of the CF antibodies to Q fever among 989 milking cows of 75 dairy farms in Kyunggi Province was revealed as high as 58.7% by the farms and 27.8% by the individual cows with higher prevalence in Kyunggi Central and Kyunggi East regions than any other regions in the Province. 2. Anticomplementary reactions were appeared as 7.5% (74/989) and it ranged from 1.0% to 16.0% according to the regions investigated. 3. In the titration of the positively reacted sera, the figures of 16.7%, 37.5%, 29.8%, 9.5%, 2.9% and 3.6% at the serum dilutions of 1: 10, 1: 20, 1: 40, 1: 80, 1: 160 and higher than 1: 160, respectively. 4. It was recognized that the relatively higher cumulated frequency distribution of the CF antibodies was shown in the sera collected from the regions with higher prevalence of Q fever. 5. There was a high correlation between the prevalence of Q fever CF antibodies and the age which is usually equivalent to one year older than the calving history of the milking cows.

Non-Pathogenic Factors Affecting Somatic Cell Counts of Goat Milk (산양유의 체세포수에 영향을 미치는 비병원성 요인)

  • Somatic cell counts (SCCs) of goat milk can vary widely depending on the counting methods used and non-pathogenic factors; the goat milk industry can be threatened by establishment of a legal standard based on the findings in cow milk. In Korea, SCCs have been excluded from the items that are analyzed under the "Livestock Products Processing and Composition Standards" in accordance with a recent NVRQS Notice amendment. From April to October, SCCs of 150 goat milk samples from 2 farms were analyzed using a Somascope calibrated with standard goat milk samples. Average SCCs of the samples was 598,000/mL, and significant differences were not found between farms and between breeds. SCCs increased from 3 to 8 months after delivery.

Development of an Automatic Body Temperature Measuring System for Dairy Cattle (젖소의 자동 체온 측정 시스템 개발)

  • In this paper the development of an automatic body temperature measuring system which can be attached to the milking machine has been studied. Since the disease is highly related to the body temperature of the cattle, early detection of the abnormal temperature would prevent the severe problems which may occur in dairy farms. The electronic component AD590 is used as a temperature sensor for the system. The device is highly robust against the noise since the output signal is the current. So it can be applied to the long distance sensing. The resolution of the signal is $0.1{\circ}C$ and the current is 10 mV. Also the A/D converter is designed for interfacing the sensor with a computer. A temperature measuring experiment using the developed system has been done for measuring the temperature of human beings and the system was proven to be useful for measuring the body temperature of the dairy cattle properly.

Fecal and Molecular Survey of Neospora caninum in Farm and Household Dogs in Mashhad Area, Khorasan Province, Iran

  • Neospora caninum is an important cause of abortion in dairy cattle worldwide. Dog is the definitive host for N. caninum and can infect dairy cattle. The aim of this study is to determine the prevalence of Neospora oocysts in feces of dogs from dairy farms. A total of 174 fecal samples was collected from 89 farm dogs and 85 household dogs during 2006 and 2008. Fecal samples of dogs were microscopically examined for detecting Hammondia Neospora-like oocysts (HNLO) by Mini $Parasep^{(R)}SF$ fecal parasite concentrator. HNLO were microscopically detected in 4 fecal samples (2.2%). The fecal samples with HNLO were examined by N. caninum-specific PCR. Two of the samples were positive for N. caninum. The 2 positive fecal samples were selected for inoculation to calves. Two inoculated calves were seronegative by ELISA for 4 months post-infection. This is the first report of finding N. caninum DNA in feces of farm dogs in Mashhad area, Iran.

Investigation of Ophthalmic Diseases in Dairy Cattle (젖소의 안과질환 조사)

  • This study was performed to investigate the ophthalmic diseases in 269 Holstein dairy cattle of 10 farms in Kyunggido, Korea. The acerage prevalence rate of ophthalmec diseases was 25.7%, ranging of from 4.8% to 47.6%. The highest incidence age was between 3 and 4 years old, 41.9% and then in order of between 5 and 6 years, between 2 and 3 years, over 6 years, between 4 and 5 years, between 1 and 2 years, and up to 1 year were shown the incidence of 33.3%, 29.4%, 27.7%, 23.8%, 19.8% and 12.5%, respectively. The prevalence rate of cataracts was higher than any other ophthalmic diseases, 10.4% and then in order of chorioretinitis, iris posterior synechiae, corneal scar, conjunctivitis, blindness, third eyelid carcinoma and globe anomaly were diagnosed as the incidence of 4.1%, 3.0%, 1.7%, 1.3%, 0.4%, 0.2% and 0.2%, respectively.

Detection and molecular characteristics of bovine rotavirus A in dairy calves in China

  • Background: Bovine group A rotavirus (BoRVA) is a major cause of severe gastroenteritis in newborn dairy calves. Only one study has investigated the G and P genotypes among dairy calves in a few regions of China, which were G6 and P[5]. Therefore, data on the prevalence and molecular characteristics of BoRVA in dairy calves in China remains limited. Objectives: The purpose of this study was to investigate the prevalence and molecular characteristics of BoRVA in dairy calves in China. Methods: 269 dairy calves diarrheic samples from 23 farms in six provinces in China were collected to detect BoRVA using reverse transcription polymerase chain reaction. Results: 71% of samples were determined to be BoRVA-positive. Two G genotypes (G6, G10) and two P genotypes (P[1], P[5]) were identified, and G6P[1] BoRVA was the predominant strain. Moreover, the VP7 and VP4 gene sequences of these dairy calf BoRVA strains revealed abundant genetic diversity. Interestingly, eight out of 17 complete G6 VP7 sequences were clustered into G6 lineage VI and analysis showed the strains were closely related to Chinese yak BoRVA strains. Conclusions: The results of this study show that BoRVA circulates widely among dairy calves in China, and the dominant genotype in circulation is G6P[1], first report on molecular characteristics of complete P[5] VP4 genes in chinese dairy calves. These results will help us to further understand the prevalence and genetic evolution of BoRVA among dairy calves in China and, thus, prevent the disease more effectively.

  • The objective of this study was to compare the accuracy analysis and the effect of field application of a newly developed automatic heat detector in dairy cows. From 2009 to 2010, we used 48 Holstein cows (mounting cows : 38 heads, standing cows : 10 heads) raised in experimental barn of National Livestock Research Institute (RDA) for the accuracy analysis of automatic heat detector, and 14 Holstein cows raised in three commercial dairy farms of Cheonan and Pochun area for comparison of the effect of field application. The accuracy of response in cows attached with automatic heat detector was 86.8% (33/38) displayed on board when mounting activity observed, and 100% (10/10) when standing activity observed, and on average, 90.0% (43/48) displayed on board. The accuracy of automatic heat detector in on-farm test was 85.7% (12/14), and conception rate was 75.0% (9/12).

Work-related Injuries in Dairy Farm in Gyeonggi Province (경기 지역 낙농작업자의 재해 발생 현황과 요인)

  • Objectives : Although dairy farming ranks as the industry with the highest injury incidence rate, the information on the injuries is limited in Gyeonggi Province. The purpose of this study was to investigate the occurrence of farm accidents and injuries. Methods : The occurrence of accidents among dairy farmers due to work-related injuries were investigated from 2,799 dairy farms of Seoul Dairy Cooperative. Among 171 total accidents, the number of accidents for human was 108. In this study, the 108 human accidents were investigated. Results : The results of this study were as follows; First, the injury rate in dairy farming was 1.60%. Second, the highest injury in dairy farm was occurred in the spring and at 6~8 p.m. Third, a variety of injuries occurred when doing the milking. Fourth, cow, machinery and falls were among the most common causes. Fifth, the most common injuries was fracture (52.8%) and the most common body part of injuries were torso (25.9%), legs (22.2%) and hand (18.5%). Sixth, 45% of the accidents indicated the lost work time from 4 weeks to 3 months. Recommendations: With the results, it is recommended that practical control methods to prevent accidents in dairy farming, for instance, wearing adequate personal protective equipment (PPE) or designing a moderate floor of works etc. be studied and developed.

Seroprevalence of Coxiella burnetii in bulk-tank milk and dairy cattle in Gyeongbuk province, Korea (경북지역 집합유와 젖소에서 큐열 항체 보유율 조사)

  • Q fever is a rickettsial infection caused by Coxiella burnetii that is transmitted from animals to humans. Modes of transmission to humans include inhalation, tick bites and ingestion of unpasteurized milk or dairy products. This survey was aimed at monitoring the seroprevalence of C. burnetii in bulk-tank milk (BTM) in Gyeongbuk province. In addition, the seroprevalence of C. burnetii was investigated at the herd level of dairy cattle in eastern Gyeongbuk province in which many dairy cattle are reared. Among 324 BTM samples collected from 20 country areas, 175 (54%) BTM samples from 15 (75%) country areas were positive for C. burnetii by ELISA. By regions, the seroprevalence of BTM samples in eastern, central, western and northern areas of Gyeongbuk province were 62.7%, 48.4%, 45.1%, and 41.4%, respectively. When analyzed in the dairy cattle reared in the eastern area where high seroprevalence occurred in BTM samples, 119 (24.2%) out of 492 dairy cattle were positive for C. burnetii. Seroprevalence of C. burnetii in dairy cattle was increased with daily milk yield of farm (P<0.05) and age (P<0.001). Since seroprevalence of C. burnetii is relatively high in both BTM samples and dairy cattle reared in Gyeongbuk province, further studies on the high risk farms and herds are needed to evaluate infection status and appropriate control programs in this region.

Studies on the brucellosis in Kyongbuk area (경북지역의 부루셀라병에 관한 연구)

  • The present study was carried out to investigate the prevalence of brucellosis in Kyungbuk area for the 3 years from 1966 to 1998. Collective milk samples were routinely screened to detect positive farms by using the milk ring test(MRT), and serum agglutination test was performed to detect sero-positive individuals in the MRT positive farms. Attempt were made to isolate the causative organismas from slaughtered sero-positive reactors and some biochemical and polymerase chain reation characters of the isolates were also made to identify the organisms. Seroprevalence to brucellosis in peoples who are close contact with infected dairy herds was also investigated. Brucellosis of dairy cattle was rare before 1997, but has been broken more frequently since early 1998. By the MRT for dairy herds, positive rate was gradually increased every year : 0.6% in 1996, 1.5% in 1997, 3.9% in 1998. Among 262 MRT-positive herds, only 21 herds(8.0%) showed positive brucellosis in serological test. The isolation rates of Brucella sp from tested materials were 51.2% in supramammary glands, 39.5% in milks, and 50.0% in pulmonary Iymphnode, respectively. Isolated strain and biotype were Brucella(B) arbortus biotype 1 in 26 heads, and were B suis biotype 1 in 2 heads. Isolated strain and vaccine strain were very similar in their colony morphology and staining. In drug susceptibility, isolated stains(B abortus) and vaccine strain(B abortus RB-51) were sensitive to ampicillin, gentamycin, kanamycin, neomycin, penicillin, streptomycin, and to tetracycline, but resistant to erythromycin. In the PCR, field strains reacted to BA and IS711 primers, and vaccine strain reacted to BA, IS711, and RB5l primers. In the plate agglutination test of 96 sera of human contacted with animals, serum antibody titer detected 1 : 100 in one person, 1 : 200 in one, and below 1 : 25 in the others.
