흰쥐황체에서 MCP-1과 큰포식세포아형의 역할에 관한 면역조직화학적 연구 (Immunohistochemical Study on Role of the Monocyte Chemoattractant Protein-1 and Macrophage Subpopulations in the Rat Corpora Luteum)

  • 큰포식세포나 혈관의 내피세포 등에서 분비되는 monocyte chemoattractant protein-1(MCP-1)은 큰포식세포의 활성을 조절하고 황체의 용해시기에는 용해를 개시, 촉진시키는 작용을 하는 것으로 알려져 있다. 그러나 아직 임신 황체나 출산 후의 황체의 발달과 유지에 대한 MCP-1의 작용기전은 확실히 알려져 있지 않다. 난포발달과정에서 큰포식세포의 역할을 알아보기 위해서 흰쥐를 실험동물로 임신시기별, 출산 후 황체에서 TUNEL 염색, ED1, ED2 및 MCP-1에 대한 면역조직화학을 실시하였다. 출산 후 황체에서 큰포식세포의 수가 의미 있게 증가하였으며, 큰포식세포에 대한 ED1, ED2의 면역반응성이 증가하였고, MCP-1의 면역반응성도 크게 증가하였다. 본 연구의 결과 출산 후 황체에서는 큰포식세포가 주로 탐식작용을 하게 되지만, 임신 황체에서는 황체의 구조와 기능을 유지하는데 주로 관여할 것으로 생각된다.

Biochemical Characterization of 20α-Hydroxysteroid Dehydrogenase

  • In this review, we have tried to summarize the evidence and molecular characterization indicating that $20{\alpha}$-hydroxysteroid dehydrogenase ($20{\alpha}$-HSD) is a group of the aldo-keto reductase (AKR) family, and it plays roles in the modulation and regulation of steroid hormones. This enzyme plays a critical role in the regulation of luteal function in female mammals. We have studied the molecular expression and regulation of $20{\alpha}$-HSD in cows, pigs, deer, and monkeys. The specific antibody against bovine $20{\alpha}$-HSD was generated in a rabbit immunized with the purified recombinant protein. The mRNA expression levels increased gradually throughout the estrous cycle, the highest being in the corpus luteum (CL) 1 stage. The mRNA was also specifically detected in the placental and ovarian tissues during pregnancy. The $20{\alpha}$-HSD protein was intensively localized in the large luteal cells and placental cytotrophoblast villus, glandular epithelial cells of the endometrium, syncytiotrophoblast of the placenta, the isthmus cells of the oviduct, and the basal part of the primary chorionic villi and chorionic stem villus of the placenta and large luteal cells of the CL in many mammalian species. Further studies are needed to determine the functional significance of the $20{\alpha}$-HSD molecule during ovulation, pregnancy, and parturition. This article will review how fundamental information of these enzymes can be exploited for a better understanding of the reproductive organs during ovulation and pregnancy.

말에서 조기 임신진단을 위한 동음파영상 이용 방법에 관한 연구 (Studies on the Ultrasonographic Images for the Early Pregnancy Diagnosis in Horses)

  • To investigate the estrual changes of ovaries, appropriate time of mating, diagnosis of early pregnancy after mating, and diagnosis of twin fetuses, ultrasonography was performed in 61 thorough-bred mares (5-12 years old) which had been raised in Chrju island. The followings are the results obtained: 1. The average size of a follicle of mares was $3.2{\times} 3.8cm$ at the beginning of estrus and $4.2{\times} 3.5 cm$ from the middle of estrus to ovulations respectively, and the average size of a follicle and a corpus luteum was $1.8{\times} 1.4 cm and 3.1{\times}2.3 cm$ in the middle of diestrus, respectively. 2. In the result of pregnancy according to mating time after ultrasonographic examination of follicle size, the mares having a follicle of 4.6-5.0 cm (diameter) were mated on the day of ultrasonographic examination or the next day rind all were conceived (3 mares) and another mares having a follicle of 4.1- 4.5cm (diameter) on the 1st or 2nd day after examination and all were conceived (4 mares). 3. By ultrasonographyi early pregnancy in the mares was diagnosed from 12 days of pregnancy and the equine fetus was directly detectable from 23 days of pregnancy, whereas the heartbeat of a fetus was detectable from 28 days of pregnancy. 4. Diagnosis of earl pregnancy of twin was possible from 16 days of pregnancy. These result indicated that ultrasonography is applicable in mares to diagnose the estrual changes of ovaries and the time of ovulation or mating, and to diagnose early pregnancy from 12 days of pregnancy and early pregnancy of twin fetuses.

자궁수종이 있는 개에서 체내 성숙 난자의 회수 (Recovery of In Vivo Matured Oocytes from a Bitch with Hydrometra)

  • 한살령의 잡종 암캐에서 체내 성숙된 난자를 회수하기 위한 실험이 진행되었다. 혈청 프로게스테론 농도를 이용하여 배란 후 약 72시간 뒤에 개복술을 시행하였고, 비정상적으로 확장된 왼쪽 자궁각이 발견되었다. 양쪽 난소는 각각 여덟 개의 황체를 가지고 있었고, 배지를 난관에 관류시켜서 총 16개의 난자를 회수하였다. 모든 난자는 극체가 돌출되어 있었고, 25 um 미만의 난황주위 공간과 제 2중기의 핵을 가지고 있었다. 본 증례는 자궁수종을 가진 암캐에서 체내 성숙된 난자를 확인한 최초의 연구이며, 생식기 관련 질병을 가진 개과 동물에서 회수된 난자가 보조 생식 기술에 사용될 수 있음을 의미한다.

LHMM기반 영어 형태소 품사 태거의 도메인 적응 방법 (Domain Adaptation Method for LHMM-based English Part-of-Speech Tagger)

  • 형태소 품사 태거는 언어처리 시스템의 전처리기로 많이 활용되고 있다. 형태소 품사 태거의 성능 향상은 언어처리 시스템의 전체 성능 향상에 크게 기여할 수 있다. 자동번역과 같이 복잡도가 놓은 언어처리 시스템은 최근 특정 도메인에서 좋은 성능을 나타내는 시스템을 개발하고자 한다. 본 논문에서는 기존 일반도메인에서 학습된 LHMM이나 HMM 기반의 영어 형태소 품사 태거를 특정 도메인에 적응하여 높은 성능을 나타내는 방법을 제안한다. 제안하는 방법은 특정도메인에 대한 원시코퍼스를 이용하여 HMM이나 LHMM의 기학습된 전이확률과 출력확률을 도메인에 적합하게 반자동으로 변경하는 도메인 적응 방법이다. 특허도메인에 적응하는 실험을 통하여 단어단위 태깅 정확률 98.87%와 문장단위 태깅 정확률 78.5%의 성능을 보였으며, 도메인 적응하지 않은 형태소 태거보다 단어단위 태깅 정확률 2.24% 향상(ERR: 6.4%)고 문장단위 태깅 정확률 41.0% 향상(ERR: 65.6%)을 보였다.

토픽 모델링과 네트워크 분석을 활용한 사물주소 도입에 대한 언론보도 분석 (An Analysis of the Media's Report on the Adoption of the Address of Things using Topic Modeling and Network Analysis)

  • 본 연구는 주소를 둘러싼 국내외 환경변화 속에서 관계 법령 개정 및 시범사업 등에 의해 본격적으로 도입이 이루어지고 있는 사물주소에 대한 언론보도를 분석하였다. 네이버 뉴스 플랫폼에서 2018년 4월부터 2020년 9월까지 기간동안 '사물주소'라고 검색하여 수집된 언론보도기사의 제목과 원문을 수집하여 토픽 모델링 및 네트워크 분석을 실시하였다. 분석 결과, 보도주제는 4가지 유형으로 '사물주소체계 추진', '사물주소 부여대상 실증', '도로명주소 사용 개선', '주소 활성화를 위한 교육·홍보'로 나타났으며, 해당 기간동안 '사물주소 부여 실증' 주제가 주요 의제였음을 확인하였다. 분석 결과를 행정안전부의 「제3차 주소정책 기본계획(2018-2022)」과 비교하여 정책적 시사점을 제시하였다.

Effects of Repeated Ovarian Stimulation on Ovarian Function and Aging in Mice

  • Controlled ovarian hyperstimulation (COH) is routinely used in the in vitro fertilization and embryo transfer (IVF-ET) cycles to increase the number of retrieved mature oocytes. However, the relationship between repeated COH and ovarian function is still controversial. Therefore, we investigated whether repeated ovarian stimulation affects ovarian aging and function, including follicular development, autophagy, and apoptosis in follicles. Ovarian hyperstimulation in mice was induced by intraperitoneal injection with pregnant mare serum gonadotropin (PMSG) and human chorionic gonadotropin (hCG). Mice subjected to ovarian stimulation once were used as a control group and 10 times as an experimental group. Repeated injections with PMSG and hCG significantly reduced the number of primary follicles compared to a single injection. The number of secondary and antral follicles increased slightly, while the number of corpus luteum increased significantly with repeated injections. On the other hand, repeated injections did not affect apoptosis in follicles associated with follicular atresia. The expression of autophagy-related genes Atg5, Atg12, LC3B, and Beclin1, cell proliferation-related genes mTOR, apoptosis-related genes Fas, and FasL was not significantly different between the two groups. In addition, the expression of the aging-related genes Dnmt1, Dnmt3a, and AMH were also not significantly different. In this study, we demonstrated that repeated ovarian stimulation in mice affects follicular development, but not autophagy, apoptosis, aging in ovary. These results suggest that repetition of COH in the IVF-ET cycle may not result in ovarian aging, such as a decrease in ovarian reserve in adult women.

양측 천외음부동맥 천공지 피판을 이용한 음경둘레 연조직 결손의 재건 (Reconstruction of a Circumferential Penile Shaft Defect Using Bilateral Superficial External Pudendal Artery Perforator Flaps)

  • 음경 둘레 연조직 결손은 다양한 공여부를 이용하여 재건할 수 있으며, 유리피판술 및 국소피판술을 응용할 수 있다. 유리피판술의 경우 성공적인 결과를 위해 수술 과정 및 술 후 관리에 기술적인 어려움이 있으며, 기존 국소피판술은 결손 부위의 크기나 모양에 제약이 따르게 된다. 이에 양측 천외음부동맥 천공지피판을 이용한 음경둘레 연조직 결손 재건의 성공적인 사례를 발표하고자 한다. 음경 둘레 연조직 결손은 음낭근막까지 관찰되었으며, 변연절제술 시행 이후 심부근막과 해면체는 보존하였다. 이후 양측 천외음부동맥 천공지 피판을 이용하여 음경 전체 둘레연조직 결손 부위를 재건하였다. 27개월의 추적 관찰 기간 동안 음경의 형태와 기능은 보존되었으며, 창상치유와 연관된 문제는 관찰되지 않았다.

An MRI-Based Quantification for Correlation of Imaging Biomarker and Clinical Performance in Chronic Phase of Carbon Monoxide Poisoning

  • Purpose: The purpose of this study was to determine the relation between quantitative magnetic resonance imaging biomarkers, and clinical performances in chronic phase of carbon monoxide intoxication. Materials and Methods: Eighteen magnetic resonance scans and cognitive evaluations were performed, on patients with carbon monoxide intoxication in chronic phase. Apparent diffusion coefficient (ADC) ratios of affected versus unaffected centrum semiovale, and corpus callosum were obtained. Signal intensity (SI) ratios between affected centrum semiovale, and normal pons in T2-FLAIR (fluid-attenuated inversion recovery) images were obtained. The Mini-Mental State Exam, and clinical outcome scores were assessed. Correlation coefficients were calculated, between MRI and clinical markers. Patients were further classified into poor-outcome and good-outcome groups based on clinical performance, and imaging parameters were compared. T2-SI ratio of centrum semiovale was compared, with that of 18 sex-matched and age-matched controls. Results: T2-SI ratio of centrum semiovale was significantly higher in the poor-outcome group, than that in the good-outcome group and was strongly inversely correlated, with results from the Mini-Mental State Exam. ADC ratios of centrum semiovale were significantly lower in the poor outcome group than in the good outcome group, and were moderately correlated with the Mini-Mental State Exam score. Conclusion: A higher T2-SI and a lower ratio of ADC values in the centrum semiovale, may indicate presence of more severe white matter injury and clinical impairment. T2-SI ratio and ADC values in the centrum semiovale, are useful quantitative imaging biomarkers for correlation with clinical performance in individuals with carbon monoxide intoxication.

Induction of superovulation in mature mice and rats using serum of spayed female dogs

  • The following experiments were designed to examine the effect of serum of spayed dogs on superovulation response in mice and rats. In Experiment 1, female mice at diestrus (n=30) were divided into three equal groups and superovulated with either administration of 5 IU pregnant mare serum gonadotropin (PMSG) or recombinant follicle stimulating hormone (rFSH) (reducing dose from 2.5 to 0.5 IU) and 5 IU human chorionic gonadotropin (hCG) administered 48h later. Serum of spayed dogs was administered intraperitoneally at a reduced dose from 0.1 to 0.025 mL in a 48 h period. In Experiment 2, female rats (n=30) at diestrus stage were divided into three equal groups. Superovulation was induced using either 30 IU PMSG, or a dose reduced from 5 to 1 IU rFSH and 25 IU hCG administered 48h later. Serum of spayed dogs was administered in a reduced dose from 0.6 to 0.1 mL in a 48 hour period. Female mice and rats were mated 24 h following hCG administration. On day 14 after mating, animals were euthanized and ovarian sections were fixed for histopathological evaluation and corpus luteum (CL) counting. No significant difference observed in mean (${\pm}SEM$) number of CLs between the PMSG group and the mice that received serum of spayed dog ($10.4{\pm}1.3$ vs $9.2{\pm}1.0$). Mean (${\pm}SEM$) number of CLs tended to be lower in rats that received serum of spayed dog than those of rats which received either PMSG or rFSH ($15.1{\pm}1.9$ vs $23.6{\pm}3.1$ and $23.1{\pm}2.9$, P=0.06, respectively). In conclusion, serum of spayed dogs is able to induce a superovulatory response in mice and rats.
