The efficiency of gamma irradiation (0, 1, 5, 10, 15, 20, and 30 kGy) as a sterilization method of corn samples (30 g) artificially contaminated with Fusarium moniliforme stored at normal condition ($25^{\circ}C$ with approximate relative humidity (RH) of 55%) and optimal condition ($25^{\circ}C$ with a controlled RH of 97%) was studied. The results showed that the fungal growth and the amount of fumonisin were decreased as the dose of gamma irradiation increased. Gamma irradiation at 1-5 kGy treatment significantly inhibited the growth of F. moniliforme by 1-2 log reduction on corn samples (P < 0.05). Sublethal effect of gamma irradiation was observed at 10-20 kGy doses after storage, and a complete inactivation required 30 kGy. Fungal growth and fumonisin production increased with higher humidity and longer storage time in all corn samples. This study also demonstrated that there was no strict correlation between fungal growth and fumonisin production. Storage at normal condition significantly resulted in lower growth and fumonisin production of F. moniliforme as compared with those stored at optimal condition (P < 0.05). Gamma irradiation with the dose of ${\geq}5$ kGy followed by storage at normal condition successfully prolonged the shelf life of irradiated corns, intended for human and animal consumptions, up to 7 weeks.
The rational utilization of crop straw as a raw material for natural gas production is of economic significance. In order to increase the efficiency of biogas production from agricultural straw, seasonal restrictions must be overcome. Therefore, the potential for biogas production via anaerobic straw digestion was assessed by exposing fresh, silage, and dry yellow corn straw to cow dung liquid extract as a nitrogen source. The characteristics of anaerobic corn straw digestion were comprehensively evaluated by measuring the pH, gas production, chemical oxygen demand, methane production, and volatile fatty acid content, as well as applying a modified Gompertz model and high-throughput sequencing technology to the resident microbial community. The efficiency of biogas production from fresh straw (433.8 ml/g) was higher than that of production from straw silage and dry yellow straw (46.55 ml/g and 68.75 ml/g, respectively). The cumulative biogas production from fresh straw, silage straw, and dry yellow straw was 365 l-1 g-1 VS, 322 l-1 g-1 VS, and 304 l-1 g-1 VS, respectively, whereas cumulative methane production was 1,426.33%, 1,351.35%, and 1,286.14%, respectively, and potential biogas production was 470.06 ml-1 g-1 VS, 461.73 ml-1 g-1 VS, and 451.76 ml-1 g-1 VS, respectively. Microbial community analysis showed that the corn straw was mainly metabolized by acetate-utilizing methanogens, with Methanosaeta as the dominant archaeal community. These findings provide important guidance to the biogas industry and farmers with respect to rational and efficient utilization of crop straw resources as material for biogas production.
The objective of this study was to examine the effects of seven dietary oils on the serum lipid patterns of rats. Seventy weanling Wistar Kyoto rats were divided into seven groups of ten rats each. Walnut oil (rich in PUFA), wheat germ oil (rich in PUFA), corn oil (rich in PUFA), canola oil (rich in monounsaturated fatty acids), fish oil (rich in PUFA), primrose oil (rich in PUFA), and palm oil (rich in saturated fatty acids) were employed for 21 days. Serum total cholesterol concentrations for rats fed palm oil, walnut oil, and wheat germ oil were significantly higher than were concentrations for rats receiving corn oil. fish oil, and primrose oil. The mean serum LDL cholesterol values for rats fed fish oil, primrose oil, and corn oil were significantly lower than those for rats fed walnut oil, wheat germ oil, canola oil, and palm oil. HDL cholesterol concentrations were the highest when wheat germ oil was fed and the lowest when fish oil was fed. The feeding of wheat germ oil and palm oil to rats resulted in considerably higher serum triglyceride levels than did all other treatments. The feeding of wheat germ oil to rats resulted in considerably higher serum phospholipid levels. Serum phospholipid concentrations were significantly lower in rats fed the canola oil, fish oil, ,and primrose oil diets, when compared to concentrations achieved with the feeding of walnut oil, wheat germ oil, corn oil, and palm oil. Palm oil, which has a high ratio of saturated to polyunsaturated fatty acids, resulted in the highest serum total cholesterol and highest LDL cholesterol levels, while fish oil, primrose oil, and corn oil produced the lowest total cholesterol and LDL cholesterol. Wheat germ oil produced the highest values for HDL cholesterol, triglycerides, and phospholipids. In general, feeding oils rich in polyunsaturated fatty acids produced more favorable responses than feeding oils containing large amounts of monounsaturated or saturated fatty acids.
Corn starch was saccharified without cooking in an agitated bead reaction system. Uncooked corn starch was effectively hydrolyzed even at the concentration as high as 39%(w/v). After 24 hours. the extent of saccharification reached at 92%, which corresponds glucose concentration of 425g/L. Fed-batch feeding of starch was more effective than batch feeding for saccharification of uncooked corn starch. The composition of hydrolysated of uncooked starch was analyzed. which was composed of 95% glucose, 0.7% of maltose, and 4.5% of high saccharide, similar with that of cooked starch. The hydrolysate can be successfully utilized for HFCS manufacture. The starch liquefying and saccharifying enzyme was relatively stable even be the physical impact of the attrition-milling media. The enzyme stabilizer, $Ca^{++}$, played an essential role in preventing the enzyme deactivation caused by the physical impact.
The present work was designed to determine whether change in fluidity of the mitochondrial membrane affects mitochondrial {TEX}$F_{1}${/TEX}{TEX}$F_{0}${/TEX}ATPase characteristics in NIDDM-prone BHE/Cdb rat. Isolated mitochondria fom BHE/Cdb rat fed a 6% coconut oil or corn oil were functionally tested by an analysis of its respiration and the coupling of this process to ATP synthesis in presence of oligomycin, a specific inhibitor of oxidative phosphorylation (OXPHOS), that binds to the {TEX}$F_{1}${/TEX}{TEX}$F_{0}${/TEX}ATPase. Mitochondria from rats fed coconut oil were more responsive to the inhibitory action of oligomycin with respect to state 3 respiration, respiratory control (RC) ratio and ADP:P (P/O) ratio than were mitochondria from rats fed corn oil. In state 3 respiration, mitochondria from rats fed coconut oil consumed less oxygen than did mitochondria from rats fed corn oil. RC ratio was lower in the mitochondria from rats fed coconut oil than was mitochondria from rats fed corn oil. In P/O ratio, the mitochondria from rats fed coconut oil had a lower P/O ratio than did mitochondria from rats fed corn oil. The data showed that the chang influidity of the mitochondrial membrane by dietary fat affected mitochondrial {TEX}$F_{1}${/TEX}{TEX}$F_{0}${/TEX}ATPase characteristics. The present study on diet differences in {TEX}$F_{1}${/TEX}{TEX}$F_{0}${/TEX}ATPase characteristics provides considerable insight into the role diets play in the control of mitochondrial function, expecially OXPHOS in NIDDM with mitochondrial defects.
This study was conducted to compare the effects of perilla oil or corn oil on lipid peroxidation and eicosanoid productions which are associated with the promotion of carcinogenesis. in liver or blood in rats. Male Sprague-Dawley8 weaning rats were fed on semisynthetic diets containing 15%(w/w) beef fat(BF). corn oil(CO) or perilla oil(PO) Three weeks after the half of rats in each diet group were injected with a single dose of 50mg 2-acetylaminofluorene (AAF)/Kg BW hepatocarcinogen intraperitoneally 3 times at 2-day interval and all of the rats were sacrificed after 8 weeks from the first injection. The rats fed on different dietary fats without 2-AAF treatment had not different MDA produc-tion and conjugated diene content in liver microsome. CO+AAf group had significantly higher conjugated diene content than BF+AAF and PO+AAF groups. and lower glucose-6-phospha-tase activity than BF+AAF group But PO+AAF had similar conjugated diene content to BF+AAF group and significantly lower MDA production than BF+AAF and CO+AAF groups. The hepatic mocrosomal lipid peroxidation was slightly greater in CO group than in PO group though perilla oil(P/S=9.67) has much more polyunsaturated fatty acids than corn oil(P/S=2.92) PG E2 level in liver and TX B2 level in plasma were significantly higher in CO group than in BF and PO groups. TX B2 level was lowered in CO and BF groups by 2-AAF treatment. These results reach to the contclousion than the type of dietary fatty acid as well as the P/S ratio has effect on hepatic microsomal lipid peroxidation and eicosanoid production and perilla oil or linolenic acid(n3) might be less effective on lipid peroxidation or PG E2 and TX B2 mediated tumor promotion than corn oil or linoleic acid(n6).

Individual immunochromatographic assays (ICG) for ochratoxin A (OTA) and zearalenone (ZEA) were optimized and used in the development of a one-step simultaneous immunochromatographic assay (OS-ICG) for the rapid multianalysis of two mycotoxins in corn samples. The nitrocellulose membrane of the OS-ICG was treated with OTA-bovine serum albumin (BSA), ZEA-ovalbumin (OVA), and anti-mouse IgG in the OTA test, ZEA test, and control zones, respectively. Monoclonal antibody-gold conjugates (OTA3 MAb-gold and ZEA2C5 MAb-gold) were sprayed onto the conjugate pad. The visual detection limits were 2.5 and 5 ng/ml for OTA and ZEA, respectively, and the results were obtained within 15 min after starting the analysis. An efficient, simple, and rapid extraction method using 30% MeOH/PBS was established and validated by analyzing the corn samples spiked with OTA/ZEA mixtures (0/0, 5/10, 10/20, and $20/30\;{\mu}g/kg$). The cut-off values of the OS-ICG for the spiked corn were 5 and $10\;{\mu}g/kg$ for OTA and ZEA, respectively. Natural corn samples were analyzed by OS-ICG, direct competitive enzyme-linked immunosorbent assay (DC-ELISA), and HPLC. Results of the OS-ICG were in good agreement with those obtained by DC-ELISA and HPLC. The developed OS-ICG offers a rapid, easy-to-use, and portable analytical system and can be used as a convenient qualitative tool for the on-site simultaneous determination of OTA and ZEA in cereals, food, and agricultural products in one analytical cycle.

Several species belonging to the Gibberella fujikuroi species (Gf ) complex are commonly associated with rice and corn, not only causing serious diseases, but also producing fumonisins, a group of mycotoxins harmful to animals and humans. To characterize the population structure of the putative fumonisin-producing Gf complex in Korea, we obtained 276 candidate isolates from rice and corn harvested in 2009 and 2010 by diagnostic polymerase chain reaction with several specific primer sets. Phylogenetic trees were constructed using multilocus sequences (combined RPB2 and EF1A, totaling 1.6 kb) from these isolates. Among the 135 isolates from rice, F. fujikuroi (teleomorph: G. fujikuroi; 59.3%) and F. proliferatum (G. intermedia; 13.3%) were predominant, followed by F. concentricum (5.9%). Additionally, twenty-five (18.5%) rice isolates belonged in a distinct subclade of F. commune, a non-member of the Gf complex. In contrast, F. verticillioides was the most predominant species (38.3%) among the 141 corn isolates, and followed by F. fujikuroi (27.7%), F. proliferatum (14.9%), F. subglutinans (7.1%), and F. concentricum (2.8%). A single mating type (MAT1-1) was found predominantly among the Gf complex isolates examined. Possible distinct subclades were detected within the populations of F. fujikuroi and F. proliferatum; however, this needs further confirmation. This is the first reported population-level characterization of putative fumonisin-producing Gf complex associated with rice and corn in Korea.
As corn starches with different amylose content were heated at different temperature $(100^{\circ}C,\;121^{\circ}C)$ with starch / water ratio (1:3.5, 1:9) and heating-cooling treatment was repeated up to 4 times, the yield of RS(resistant starch) from heated corn starches was investigated by the enzymatic-gravimetric method and the ${\alpha}-amylase$ treatment. Compared to ${\alpha}-amylase$ method, enzymatic-gravimetric method was more effective to hydrolyze the amorphous region of heated corn starch. With increasing the amylose content and the number of heating-cooling treatment, the yield of RS increased, regardless of isolation method. Heated corn starches formed at $121^{\circ}C$ provided higher yield of RS than those formed at $100^{\circ}C$. Higher RS yield was also observed in the case of starch/water ratio (1:3.5) compared to the case of ratio (1:9).
The objectives were to assess the use of near-infrared reflectance spectroscopy (NIRS) as a tool for estimating nutrient compositions of corn kernel, and to apply an NIRS-based indium gallium arsenide array detector to the system for collecting spectra and analyzing calibration equations using equipments designed for field application. Partial Least Squares Regression (PLSR) was employed to develop calibration equations based on representative data sets. The kennard-stone algorithm was applied to induce a calibration set and a validation set. As a result, the method for structuring a calibration set significantly affected prediction accuracy. The prediction of chemical composition of corn kernel resulted in the following (kennard-stone algorithm: relative) moisture ($R^2=0.82$, RMSEP=0.183), crude protein ($R^2=0.80$, RMSEP=0.142), crude fat ($R^2=0.84$, RMSEP=0.098), crude fiber ($R^2=0.74$, RMSEP=0.098), and crude ash ($R^2=0.81$, RMSEP=0.048). Result of this experiment showed the potential of NIRS to predict the chemical composition of corn kernel.
