Studies on Milk Protein of Korean Cattle I. Fractionation of Milk Protein on DEAE-Cellulose (한우유(韓牛乳)의 단백질(蛋白質)에 관(關)한 연구(硏究) 제(第)1보(報) DEAE-Cellulose에 의(依)한 한우유단백질(韓牛乳蛋白質)의 분별(分別))

  • An experiment has been carried out in order to analyze the main components of Korean Cattles' milk, and fractionate the milk protein by DEAE-cellulose column. The results obtained were summarized as follow. 1) The average values of specific gravity, pH and acidity of Korean Cattles milk which were negative in alcohol test were 1,036, 6.4 and 0.21, respectively. 2) The average values of total solids, solids-not-fat, protein, lactose and ash contents of Korean Cattles milk were 11.61%, 9.53%, 2.08%, 3.99%, 4.76% and 0.86%, respectively. 3) Distribution of casein, whey protein, N.P.N., protein precipitated in 12% TCA, lactoglobulin and lactalbumin contents of the milk were 3.07%, 1.13%, 0.10%, 4.06%, 0.34% and 0.66%, respectively. 4) Acid casein obtained from Korean Cattles milk was fractionated into four fractions on DEAE-cellulose column with 0.005M tris-citrate buffer containing 6M urea, pH 8.6, and the ratio of the fraction I, II, III and IV was 3.24%, 52.67%, 26.22% and 17.87%, respectively. 5) Whey protein obtained from Korean Cattles milk was also fractionated into four fractions on DEAE-cellulose column with 0.04M phosphate buffer, pH 5.8, and the ratio of the fraction I, II, III and IV was 41.74%, 10.17%, 1.50% and 46.59%, respectively.

Distribution of Renal Artery in the Kidney of Korean Native Cattle (한우신동맥(韓牛腎動脈) 분포(分布)에 관하여)

  • The distribution of renal artery of fifty Korean native cattles (100 kidneys) was observed. Vinylite solution was injected into renal artery of ninety specimens for cast preparation. The angiography was prepared in 10 specimens by injecting thirty percent of barium sulfate solution into renal arteries, and then radiographed on a soft X-ray apparatus (Shimadzu Waltes 60). 1. A. renalis arose from the each side of abdominal aorta in the Korean native cattles. 2. The renal arteries were bifurcated into Ramus cranialis and caudalis (91%), and Ramus cranialis, medius and caudalis(9%) which were ramified 1-4 segmental arteries, respectively. 3. The segmental arteries were originated from R. cranialis and R. caudalis (87%), R. medius (9%) and A. renalis (4%). 4. The kidney were divided separately into 5-7 arterial segments by running of the segmental artery into the parenchyma. Among them six segments were mostly frequent(53%). 5. The Arcus arteriosus renalis was observed at 44% of the left kidney and 14% of the right kidney.

  • The present study was carried out to investigate the agent of foot disease of cattles, to develop the therapeutics. The results obtained were as follows ; 1. Sixty eight heads of cattle affected foot rot during the observation period and the incidence rate shown 3.25%. 2. The high incidence was observed on September and October. 3. The disease was more frequently seen affecting in hindlimbs than forelimbs. 4. The disease was more frequently seen affecting in older cattles and higher milk production cow. 5. Isolated strains wert shown E. coli (20.6%), Staphylococcus SPP (17.6%), stridium SPP (22.1%), Fusobacterium SPP (20.6%), Bacteroid SPP (19.1%). 6. The most susceptible therapeutics are A, B preparation.

Report on The Incidence of rabies in Kangwon-do (강원도내 광견병 발생실태)

  • During the period of 2 years from 1993 to 1994, epidemiological investigation were carried out each time the outbreak of rabies suspected cases were informed and all specimens were transported to the Kangwon-do Veterinary Service Lab. At the Lab., autopsy and laboratory examinations were conducted as well. The results obtained as follows 1. The dogs and cattles were confirmed to be infected with rabies virus by routine histopathological findings, indirect flulrescent antibody technique, and mouth inoculation test. 2. The total numbers of rabies occured in dogs and cattles were 9, 5 heads, respectively. The regional distribution of total of 14 heads were 9 heads in Chulwon, 2 heads in Hwachun, 1 head Yangku, 2 heads in Inje and the highest number of outbreaks were orrured in Chulwon. 3. Most of dogs showed furious type and dumb type found in only one dog. 4. Cattles showed the symtoms of uneasiness feeling, excitement, disturbance howling, the change of voice, sexual accelaration, foreign body swatting and attacked to human and ani-mal and found to be died after two or four days. 5. In the seasonal outbreaks of rabies, 8 heads of the highest incidence were occured in win-ter followed by 4 heads in autumn, 1 head in spring and 1 head in summer. 6. The first outbreaks of rabies was reported in Chulwon and then transmitted into Hwachun, Yangku and Inje in turn. 7 The infectious source was supposed to be wild animals such as laccon dog, badger etc. living around the D. M. Z.

Identification of Hanwoo Meat by DNA Analysis (DNA 분석법에 의한 한우고기 판별)

  • This study was deal with the development of breed-specific DNA marker which is able to identify Hanwoo and European cattle breeds(Non-Hanwoo) meat. Genetic differentiation between Korean cattle(Hanwoo) and European cattle breeds was examined by Random Amplified Polymorphic DNA(RAPD) analysis. The RAPD patterns were identical among Non-Hanwoo, such as Holstein, Hereford, Aberdeen Angus, Brown Swiss, Limousin or Simmental, but the above pattern was different from that of Hanwoo. All bands detected in the Hanwoo samples were observed in Non-Hanwoo cattle samples, but one of the common bands found in samples was not detected in the Hanwoo samples. The band(1.4kb) may be useful as a marker for identifying a meat of Hanwoo from imported cattle meat. Actually, the detection of the DNA marker was tested by DNA analysis with 929 samples which were prepared from bloods of 673 Hanwoo cattles and 141 Holstein cattles, from 115 imported cattle meats. The DNA marker was absent in 644 of 673 Hanwoo cattles (96%) but present in 245 of 256 Non-Hanwoo cattles (95%). These results show that the DNA marker is effective to characterize Hanwoo and Non-Hanwoo meat by its detection. This DNA marker, however, was not useful in detecting unwanted crossbreeding between two cattle breeds, because the band pattern in hybrid cattle shows one of two band patterns in Hanwoo and Non-Hanwoo.

  • Now in Korea Brucellosis was diagnosed by individual periodic blood tests. Author hope that individual periodic blood tests was performed after Milk Ring Test, because if Cattles show negative reaction to Milk Ring Test, Periodic blood tests are needless.
